Analysis of Sclerotia and Sporophores of Pleurotus tuber-regium Fr. an edible mushroom in Nigeria

  • Chemical analysis of both sclerotia and the fruits of Pleurotus tuber-regium showed higher values for such elements as calcium(Ca), iron(Fe), zine(Zn) in the fruits than in the sclerotia. On the contrary magnessium(Mg) was found to be higher in sclerotia than in the fruits. protein and carbohydrate were also found to be more in the fruits. There was no significant difference between the chemical values of old(1 year) and fresh sclerotia. Oil palm fruit fibre substrate produced sporophores with higher values for the minerals, protein and carbohydrate than those on riversand substrate.

  • Dietary protein is the most expensive component in flounder feed because of the high protein requirement of this species. The non-protein energy sources may also influence the protein utilization of fish. The inclusion of adequate levels of non-protein energy sources in diets can minimize use of protein as an energy source. Protein sparing effect by fat and carbohydrate has been studied in other fish.   • A series of experiment was conducted to investigate the effects of the level of dietary Protein (soybean meal) and carbohydrate (sucrose) on the growth rate, feed efficiency, nutrients digestibility and amylase activity of larvae at 5th instar. The results obtained from the experimentation are summarized as follow: (1) It was found that the body weight gain, cocoon quality and feed efficiency of the 5th instar larvae were significantly affected by the level of protein and carbohydrate in the artificial diet. Present data revealed that the proper level of dietary protein and carbohydrate would be the most important factor for the optimum growth of larvae. (2) In the amount of diet consumption, the group of larvae gained more body weight consumed more diet than the group gained less amount of body weight. (3) Digestibility of nutrients was improved as the level of protein in the diet increased, although the level of dietary carbohydrate did not exert any positive results. (4) It was observed that the amylase activity in digestive juice was stimulated by the increasement of dietary carbohydrate. However, no correlation between the level of dietary protein and carbohydrate was detected in the amylase activity. (5) Analytical data indicated that the amount of protein or carbohydrate excreted through feces and urine was proportional to the content of protein or carbohydrate in artificial diet. The group o( larvae consumed the diet contained high protein excreted more protein and less nitrogen free extract than did the group received low protein diet. Likewise, the group of larvae fed high dietary carbohydrate excreted more nitrogen free extract and less protein through excreta than the group consumed low carbohydrate diet group. (6) Although the amylase activity of the larvae received the artificial diet was higher than the larvae received natural diet (mulberry leaf), the rate of body weight gam and the quality of cocoon produced from the natural diet group was better than the artificial diet group. It is, thus, concluded that feeding the larvae by the artificial diet may net be recommendable. (7) It was determined that the best level of protein (soybean meal) and carbohydrate (sucrose) in artificial diet was found to be 40g and 0g, respectively when it was fed to the 5th instar larvae. (8) It may be concluded from the results obtained that the artificial diet that could stimulate the activity of amylase nay not be recommendable for the practical larvae feeding Purpose due simply to the Poor economic return from this diet than natural diet.

Quantitation of relationship and development of nutrient prediction with vibrational molecular structure spectral profiles of feedstocks and co-products from canola bio-oil processing

  • Objective: This program aimed to reveal the association of feed intrinsic molecular structure with nutrient supply to animals from canola feedstocks and co-products from bio-oil processing. The special objective of this study was to quantify the relationship between molecular spectral feature and nutrient availability and develop nutrient prediction equation with vibrational molecular structure spectral profiles. Methods: The samples of feedstock (canola oil seeds) and co-products (meals and pellets) from different bio-oil processing plants in Canada (CA) and China (CH) were submitted to this molecular spectroscopic technique and their protein and carbohydrate related molecular spectral features were associated with the nutritional results obtained through the conventional methods of analyses for chemical and nutrient profiles, rumen degradable and intestinal digestible parameters. Results: The results showed that the spectral structural carbohydrates spectral peak area (ca. 1,487.8 to 1,190.8 cm-1) was the carbohydrate structure that was most significant when related to various carbohydrate parameters of canola meals (p<0.05, r>0.50). And spectral total carbohydrate area (ca. 1,198.5 to 934.3 cm-1) was most significant when studying the various carbohydrate parameters of canola seeds (p<0.05, r>0.50). The spectral amide structures (ca. 1,721.2 to 1,480.1 cm-1) were related to a few chemical and nutrient profiles, Cornell Net Carbohydrate and Protein System (CNCPS) fractions, truly absorbable nutrient supply based on the Dutch protein system (DVE/OEB), and NRC systems, and intestinal in vitro protein-related parameters in co-products (canola meals). Besides the spectral amide structures, α-helix height (ca. 1,650.8 to 1,643.1 cm-1) and β-sheet height (ca. 1,633.4 to 1,625.7 cm-1), and the ratio between them have shown to be related to many protein-related parameters in feedstock (canola oil seeds). Multi-regression analysis resulted in moderate to high R2 values for some protein related equations for feedstock (canola seeds). Protein related equations for canola meals and carbohydrate related equations for canola meals and seeds resulted in weak R2 and low p values (p<0.05). Conclusion: In conclusion, the attenuated total reflectance Fourier transform infrared spectroscopy vibrational molecular spectroscopy can be a useful resource to predict carbohydrate and protein-relates nutritional aspects of canola seeds and meals.

  • Growing male rats were allowed to have food from two dishes containing isocaloric, isoprotein 15% or 70% carbohydrate diet for 4, 8, 12 or 24 hours a day at the beginning of the dark period. Four hours of feeding were not enough to grow though the first 4 hours of intake were significantly higher than other periods. 4-hour and 8-hour feeding groups consumed significantly less carbohydrate calorie than 12-hour and ad libitum feeding groups, which hence, showed lower ratio of carbohydrate to protein in the total food consumed. The fasting brain tryptophan and serotonin levels among groups were not significantly different. It appears therefore that the manipulation of the feeding period could be valuable in suppression of carbohydrate appetitie, especially carbohydrate carving.

Physicochemical Properties of Recominant Hepatitis B Surface Antigen Expressed in Mammalian Cell(C127)

  • The physicochmical properties of recombinant hepatitis B surface antigen (r-HBsAg), which was expressed in C127 mammalian cell were studied. Using roller bottle culture in DMEM supplemented with fetal bovine serum, 10-15 mg/L of r-HBsAg was produced with about 31% of purification yield. The purity of r-HBsAg by HPLC was 99.8% and electron microscopic examination showed homogeneous spherical particle with 22 nm in diameter, a morphological characteristic of HBsAg. The density of r-HBsAg by CsCI density gradient method was 1.19g/ml and the isoelectric point by Mono $P^{TM}$ HR 5/20 column was 4.6. The analysis of subunit protein pattern using SDS-PAGE followed by scanning densitometry gave 81.3% of S protein and 18.7% of pre-S protein. fluorophore-assisted-carbohydrate-electrophoresis analysis showed the relative amount of carbohydrate to protein was 1.7% and it smajr component was N-acetyl glucosamine, which was about 39% of total carbohydrate. The relative amount of lipid to protein determined by vanillin phosphoric acid method was 32.5% and its major component was phospholipid, which was about 70% of total lipid. The physicochemical properties of C127 mammalian cell-derved r-HBsAg are similar to those of p-HBsAg, suggesting that the r-HBsAg can be used in developing a new preventive vaccine against hepatitis B.

  • Some effects of dietary conditions on the metabolism of nucleic acid and protein in organs of the Albino Rats have been studied. The young rats to be examined were fed on the control diet and the diets deprived of one component among protein, carbohydrate, and lipid, such as protein free diet (PF: 432 kcal/100g) carbohydrate free diet (CF: 432kcal/100g), and lipid free diet (LF: 392kcal/100g) for three, seven, and fifteen days, respectively. The contents of DNA and RNA in the liver and the brain, and also those of protein-nitrogen(PN) and nonprotein-nitrogen (NPN) in the live, the brain, and the serum have been measured. The results are as followe: 1. The contents of DNA per gram of liver were increased by feeding on protein free diet. It is concluded that the critical factor for the result is not the increase in the rates of DNA syntheses, but the decrease in the turnover rates of DNA. 2. The metabolism of DNA in the liver showed the normal status by feeding on carbohydrate free diet. On the other hand, the rates of DNA syntheses were increased by feeding on lipid free diet. 3. The rates of DNA syntheses in the brain were decreased by feeding on the unbalanced diet, such as protein free, carbohydrete free, and lipid free diet. 4. In the liver and the brain, the rates of DNA syntheses were decreased by feeding on protein free diet. But the rates showed the normal status by feeding on the carbohydrate free diet, and also showed the similar metabolism to that in the case of the control group by feeding on lipid free diet. 5. In the liver, the rates of protein syntheses were decreased, whereas the contents of nonprotein-nitrogen were increased by feeding on protein free diet. 6. In the liver and the brain, the protein syntheses showed the more increasing rates than the rates in the case of the control diet by feeding on lipid free diet. 7. In the serum, the contents of protein did not change in a short period, also the insufficient feeling on protein was examined. It is clear that in the liver the rates of protein syntheses are decreased and the rates of protein catabolism are increased, since the rates of nucleic acid syntheses are decreased by feeding on the protein free diet. On the other hand, it is considered that in the brain the turnover rates of protein does not have correlation with the rates of nucleic acid syntheses, also these are decreased by feeding on protein free diet. And also it is believed that the phenomena of homeostasis for carrying the normal metabolism of nucleic acid and protein in the liver and the brain are operated in a short period as possible, by feeding on carbohydrate free and lipid free diets.

Ionizing Radiation Effect on the Carbohydrate Moiety of Chicken Ovomucoid (계란 ovomucoid의 탄수화물 부분에 미치는 이온화방사선의 영향)

  • Radiation effects on carbohydrate moiety of chicken ovomucoid, a protease inhibitor as a typical allergenic glycoprotein of egg white, was observed. The trypsin inhibitory activity of chicken ovomucoid decreased exponentially and the inactivation was more significant irradiated in $N_2$ than in $O_2$. From the protein blotting, radiation caused protein degradation in $O_2$ and protein aggregation also in $N_2$. The patterns of carbohydrate blotting were also similar with that of protein blotting. Sugar chains in low molecular weight fraction (MW<5,000) were released by radiation and those in $O_2$ were higher than in $N_2$. From the HPLC patterns of the degradation of sugar chains, all peaks of oligosaccharides have the tendency to decrease with the increase of radiation dose and more remarkable in $O_2$ than in $N_2$. These results suggest that ionising radiation could cause the overall conformational changes of ovomucoid by the degradation and release of oligosaccharides.

Influence of Refeeding of Protein, Carbohydrate and Fat on Hepatic Insulin-Like Growth Factor-I mRNA Level in Fasted Chicks

  • The influence of refeeding either protein, carbohydrate or fat on hepatic insulin-like growth factor-I (IGF-I) mRNA level in chicks which had been fasted for 2 days was examined. The hepatic IGF-I mRNA was measured by ribonuclease protection assay. Fasting reduced hepatic IGF-I mRNA levels to less than half of those in the fed control. When chicks were refed either a control, protein or carbohydrate diet, IGF-I mRNA levels significantly increased to those in the fed control until 2 hours of refeeding. Refeeding of fat did not alter hepatic IGF-I mRNA levels. The significant correlation between liver weight and hepatic IGF-I gene expression suggests that when chicks are refed after 2-d fasting, the acute increase in hepatic IGF-I gene expression brought about after refeeding may be partly regulated by the increase in liver protein metabolism.

Composition and Yield of Korean Breast Milk and Maternal Intakes of Foods and Nutrients (수유부의 식품 및 영양소 섭취와 유즙 생성량 및 조성)

  • This study was conducted to investigate the relationship between lactational capacity and intakes of energy and energy-yielding nutrients. Food consumption, intakes of carbohydrate, protein, fat and energy and quantity and proximate composition of milk of 11 lactating Korean mothers were determined at 1, 2 and 3 months postpartum longitudinally. Food consumption was estimated using a 24-hour recall method ; intakes of energy and energy-yielding nutrients were calculated according to the Food Composition Table. Daily milk production was estimated using a 72-hour test-weighing method ; protein, fat, lactose and energy concentrations were analyzed. Average intakes of energy and protein were 1974$\pm$386㎉/day and 67.0$\pm$12.3㎉/day, these were 73% and 74% of the Korean Recommended Allowances, respectively. Average milk yield was 720.1$\pm$123.3ml/day containing energy 59.6$\pm$9.5㎉/dl, protein 1.1$\pm$0.1g/dl, fat 2.7$\pm$0.8g/dl, and lactose 6.3$\pm$0.3g/dl. No relationship existed between the intakes of carbohydrate, protein, fat and energy and the quantity and proximate composition of the milk. However, the intakes of energy, carbohydrate and vegetable protein were inversely related to the concentrations of energy and lactose in the milk. This result indicates that lactational capacity may be affected by the other factors excluding intakes of energy and energy-yielding nutrients and a high intake of energy may not guarantee optimal lactational capacity.
