Protective Effects of Ginseng Coffee against Hydrogen Peroxide-induced Oxidative Damage in L6 Muscle Cells

  • This study investigated the antioxidant effects of ginseng coffee in L6 muscle cells. Ginseng coffee was prepared by coating and digesting coffee beans with ginseng concentrate. The ginseng coffee water extract potently protected against hydrogen peroxide-induced L6 cell death and adenosine triphosphate reduction in a dose-dependent manner; in fact, these cytoprotective effects were significantly greater than those of normal coffee. However, ginseng coffee did not exhibit significant radical scavenging or catalase-like activity. These results suggest that ginseng coffee might act as a cytoprotective agent in muscles, but that the protective effects are not due to a direct radical-reduction property but rather to another intracellular signaling factor.

2(3)-tert-Butyl-4-hydroxyanisole의 cis-Dichlorodiammineplatinum 신장독성에 대한 억제효과 (Protective Effects of 2(3)-tert-Butyl-4-hydroxyanisole on cis-Dichlorodiammineplatinum Nephrotoxicity in Rats)

  • cis-Dichlorodiammineplatinum(cisplatin) is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. So, to evaluate the effects of 2(3)-tert-butyl-4-hydroxyanisole(BHA) on cisplatin nephrotoxicity in rats, both compounds were given intraperitoneally. Remarkable protective effects of BHA against nephrotoxicity of cisplatin were observed when BHA was administered to rats 1hr after cisplatin injection. On the other hand pretreatment with BHA 1hr prior to cisplatin did not reduce weight loss, blood urea nitrogen and creatinine levels. Hepatotoxicity induced by combination treatment of cisplatin and BHA was evaluated by measuring serum glutamic pyruvate transaminase and serum glutamic oxalate transaminase. Combination treatment did not affect the levels of SGPT and sGOT except 1hr pretreatment. The present results indicate that BHA may provide protection against cisplatin nephrotoxicity, when it is given 1hr after cisplatin.

Schisandra Chinensis Inhibits Oxidative DNA Damage and Lipid Peroxidation Via Antioxidant Activity

  • Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties

Kidney protective potential of lactoferrin: pharmacological insights and therapeutic advances

  • Kidney disease is becoming a global public health issue. Acute kidney injury (AKI) and chronic kidney disease (CKD) have serious adverse health outcomes. However, there is no effective therapy to treat these diseases. Lactoferrin (LF), a multi-functional glycoprotein, is protective against various pathophysiological conditions in various disease models. LF shows protective effects against AKI and CKD. LF reduces markers related to inflammation, oxidative stress, apoptosis, and kidney fibrosis, and induces autophagy and mitochondrial biogenesis in the kidney. Although there are no clinical trials of LF to treat kidney disease, several clinical trials and studies on LF-based drug development are ongoing. In this review, we discussed the possible kidney protective mechanisms of LF, as well as the pharmacological and therapeutic advances. The evidence suggests that LF may become a potent pharmacological agent to treat kidney diseases.

이진탕가미방(二陳湯加味方)의 항산화 효과를 통한 역류성 식도염 억제효과 (Protective Effects of Yijin-tang-gamibang Aqueous Extracts on Reflux Esophagitis Mediated by Antioxidant Defense Systems)

  • The object of this study was to observe the protective effects of Yijin-tang-gamibang (YJGMB), Yijin-tang has been traditionally used in the Korean Medicine for treating various digestive diseases. The study showed that it is effective on reflux esophagitis induced by pylorus and forestomach ligation in rats. Three different dosages of YJGMB extracts, 200, 100 and 50 mg/kg, were orally pretreated, once a day for 28 days before pylorus and forestomach ligation. Seven groups, each of 8 rats per group were used in the present study. The results were compared with omeprazole, antioxidant and proton pump inhibitor, 30 and 10 mg/kg treated group. YJGMB 200 mg/kg were showed similar protective effects as compared with 30 mg/kg of omeprazole but more favorable effects were observed in 200, 100 and 50 mg/kg of all YJGMB treated rats as compared with omeprazole 10 mg/kg in the present study. In addition, YJGMB 200 mg/kg were showed more favorable antioxidant effects as compared with 30 mg/kg of omeprazole in the present study. Detail mechanism studies should be conduced in future with the screening of the biological active chemical compounds in herbs.

Protective Effect of Fucoidan Extract from Ecklonia cava on Hydrogen Peroxide-Induced Neurotoxicity

  • We evaluated the antioxidant activity and neuronal cell-protective effect of fucoidan extract from Ecklonia cava (FEC) on hydrogen peroxide ($H_2O_2$)-induced cytotoxicity in PC-12 and MC-IXC cells to assess its protective effect against oxidative stress. Antioxidant activities were examined using the ABTS radical scavenging activity and malondialdehyde-inhibitory effect, and the results showed that FEC had significant antioxidant activity. Intracellular ROS contents and neuronal cell viability were investigated using the DCF-DA assay and MTT reduction assay. FEC also showed remarkable neuronal cell-protective effect compared with vitamin C as a positive control for both $H_2O_2$-treated PC-12 and MC-IXC cells. Based on the neuronal cell-protective effects, mitochondrial function was analyzed in PC-12 cells, and FEC significantly restored mitochondrial damage by increasing the mitochondrial membrane potential (${\Delta}{\Psi}m$) and ATP levels and regulating mitochondrial-mediated proteins (p-AMPK and BAX). Finally, the inhibitory effects against acetylcholinesterase (AChE), which is a critical hydrolyzing enzyme of the neurotransmitter acetylcholine in the cholinergic system, were investigated ($IC_{50}$ value = 1.3 mg/ml) and showed a mixed (competitive and noncompetitive) pattern of inhibition. Our findings suggest that FEC may be used as a potential material for alleviating oxidative stress-induced neuronal damage by regulating mitochondrial function and AChE inhibition.

가족 기능과 융합형 심리적응 보호요인의 관한 연구 (A study of family function and convergent adaptation hearing protection factor)

  • 본 연구의 목적은 가족의 기능적 결손이라는 위험상황에서 위험요인, 보호요인, 심리적응 간의 관계를 검증하고, 심리적응 수준이 높은 청소년을 보호해주는 보호요인을 파악하고자 한다. 이를 위해 한국국청소년정책연구원(2008)에서 조사한 한국청소년패널조사 중2 패널 5차년도 자료를 가지고 기술통계 분석과 로지스틱 회귀 분석을 실시하였다. 연구결과 청소년의 보호요인인 자아존중감과 학교친구애착에 따라 기능적 결손을 경험한 청소년의 심리적응 수준이 다르게 나타났다. 따라서 사회복지실천 현장에서 청소년의 위험요인에 초점을 맞추기보다는 위험요인을 완화시키는 과정이나 보호요인에 초점을 맞춤으로써 위험요인에 노출된 청소년을 바라보는 시각을 전환시킬 수 있으며 이들에 대한 개입도 달라질 것으로 판단된다.

PC12 세포의 허혈모델에 있어 광두근 분획물의 항산화효과연구 (Protective Effect of Sophorae Subprostratae Radix and Each Fractions on PC12 cell Damage Induced by Hypoxia/Reperfusion)

  • This research was performed to investigate protective effect of Sophorae subprostratae Radix and each fractions against ischemic damage using PC12 cells. To observe the protective effect of Sophorae subprostratae Radix on ischemia damage, vibility and changes in activities of Superoxide dismutase (SOD), Glutathione Peroxidase (GPx), Catalase and Production of Malondialdehyde (MDA) were observed after treating PC12 cells with Sophorae subprostratae Radix during ischemic insult. Groups were divided into five groups: no treated (Normal), hypoxia chamber for 48hrs followed by 6h at normoxic chamber (H/R), Sop horae subprostratae Radix total phase treated group with H/R (Total), Sophorae subprostratae Radix water phase treated group with H/R (Water), Sophorae subprostratae Radix BuOH phase treated group with H/R (BuOH), Sophorae subprostratae Radix alkaloid phase treated group with H/R (Alkaloid). The results showed that (1) in hypoxiajreperfusion model using PC12 cell, the Sophorae subprostratae Radix has the protective effect against ischemia in the dose of 0.2 ㎍/㎖, 2 ㎍/㎖ and 20 ㎍/㎖, (2) Sophorae subprostratae Radix increased the activities of glutathione peroxidase and catalase. (3) the activity of Superoxide Diamutase(SOD) increased by ischemic damage, which might represent the self protection. This study suggests that Sophorae subprostratae Radix has neuroprotective effect against neuronal damage following hypoxiajreperfusion cell culture model using PC12 cell and dose dependency effects. In conclusion, Sophorae subprostratae Radix has protective effects against ischemic oxidative damage at the early stage of ischemia.

$H_2O_2$에 의한 배양심근세포고사에 미치는 사물탕의 방어효과 (Protective Effects of Samul-tang on ${H_2O_2}-induced$ Cell Apoptosis in Cultured Cardiomyoblast Cells)

  • Objectives : This study was designed to investigate the protective mechanisms of Samul-tang (SMT) on $H_2O_2$-induced toxicity in H9c2 cardiomyoblast cells. Methods : The cultured cells were pretreated with SMT and exposed to $H_2O_2$. The cell damage was assessed by using MTT assay. Also, we used Hoechst staining, Western blotting analysis. Results : SMT significantly reduced both $H_2O_2$-induced cell death and chromatin fragmentation. The decrease of Bcl2 expression by $H_2O_2$ was inhibited by SMT. In addition, the increase of Bax expression was also inhibited by SMT. In particular, Fas expression, which is generally recognized as cell death inducing signal by Fas/FasL interaction, was markedly decreased by $H_2O_2$ in a time-dependent manner, whereas this decrease was completely prevented by SMT. The cotreatment of SMT and $H_2O_2$ in H9c2 cells also induced the phosphorylation of ERK in a time-dependent manner. Moreover, PD098059, a specific inhibitor of ERKl/2, attenuated the protective effect of SMT on $H_2O_2$-induced toxicity in H9c2 cardiomyoblast cells. Furthermore, the protective effect of SMT was significantly blocked by treatment of SB203580, a specific inhibitor of p38. Conclusions : Taken together, this study suggests that the protective effects of the water extract of SMT against oxidative damages may be mediated by the modulation of Bel2 and Bax expression via the regulation of ERK and p38 signaling pathway.

Protective efficacy of vaccination with Neospora caninum multiple recombinant antigens against experimental Neospora caninum infection

  • Protective efficacy of vaccination with Neospora caninum multiple recombinant antigens against N. caninum infection was evaluated in vitro and in vivo. Two major immunodominant surface antigens (NcSAG1 and NcSRS2) and two dense granule proteins (NcDG1 and NcDG2) of N. caninum tachyzoites were expressed in E. coli, respectively. An in vitro neutralization assay using polyclonal antisera raised against each recombinant antigen showed inhibitory effects on the invasion of N. caninum tachyzoites into host cells. Separate groups of gerbils were immunized with the purified recombinant proteins singly or in combinations and animals were then challenged with N. caninum. Following these experimental challenges, the protective efficacy of each vaccination was determined by assessing animal survival rate. All experimental groups showed protective effects of different degrees against experimental infection. The highest protection efficacy was observed for combined vaccination with NcSRS2 and NcDG1. Our results indicate that combined vaccination with the N. caninum recombinant antigens, NcSRS2 and NcDG1, induces the highest protective effect against N. caninum infection in vitro and in vivo.
