죽상동맥경화는 대동맥의 만성염증에 의해 주로 발병되는 폐쇄동맥질환이다. 혈관평활근세포의 증식 및 이동은 죽상동맥경화 발병의 주된 병리적 반응이다. 본 연구에서는 죽상동맥경화 발병기전을 유도하는 표적 염증반응 물질의 탐색 및 이들에 의한 신호전달 기전을 연구하였다. 혈관평활근세포의 증식 및 이동은 prostaglandin $F_{2{\alpha}}$ ($PGF_{2{\alpha}}$)에 의해 의미 있게 증가하였으나 tumor necrosis factor ${\alpha}$ ($TNF{\alpha}$)에 의해서는 증가하지 않았다. Prostacyclin $I_2$ ($PGI_2$)는 혈관평활근세포의 증식은 촉진시켰으나 이동은 오히려 억제하였다. prostaglandin $D_2$ ($PGD_2$) 및 prostaglandin $E_2$ ($PGE_2$)는 혈관평활근세포의 증식을 촉진시켰으나 이동에는 영향을 미치지 않았다. $PGF_{2{\alpha}}$는 용량 의존적으로 혈관평활근세포의 증식 및 이동을 촉진시켰고 EC50는 약 $0.1{\mu}M$로 관찰되었다. 혈관평활근세포에서 phospholipase $C-{\beta}3$ ($PLC-{\beta}3$) 아형의 발현은 매우 높았으나 $PLC-{\beta}1$, $PLC-{\beta}2$, 및 $PLC-{\beta}4$의 발현은 관찰되지 않았다. U73122 처리를 통해 PLC의 활성을 억제하면 $PGF_{2{\alpha}}$에 의한 혈관평활근세포의 이동이 억제되었다. 또한 $PLC-{\beta}3$의 발현을 억제하면 $PGF_{2{\alpha}}$에 의한 혈관평활근세포의 증식 및 이동이 억제되었다. 이러한 결과들을 바탕으로 $PGF_{2{\alpha}}$ 는 혈관평활근세포의 증식 및 이동에 중요한 역할을 수행하고, 여기에는 $PLC-{\beta}3$가 필수적인 역할을 담당하고 있음을 제안한다.
The corpus luteum (CL) is a temporary endocrine gland whose main function is to secrete progesterone to support pregnancy. On the other hand, the cyclic bovine CL has also been shown to be a site of prostaglandin $F_{2{\alpha}}$ ($PGF_{2{\alpha}}$) production. Although there is general agreement that endometrial $PGF_{2{\alpha}}$ is an essential luteolysin in cattle, luteal $PGF_{2{\alpha}}$ seems to play a luteotropic role as an autocrine and/or paracrine factor, especially for the development and maintenance of the CL. This supposition is based on evidence that some of the prerequisites for autocrine/paracrine mechanisms are present, including local production of $PGF_{2{\alpha}}$ and the existence of specific binding sites within the CL. The purpose of this paper is to review the regulation of luteal $PGF_{2{\alpha}}$ secretion, its action on CL as an autocrine and/or paracrine factor and the receptivity of bovine CL to. $PGF_{2{\alpha}}$.
Although the overproduction of prostaglandin $E_2$ ($PGE_2$) in intestinal epithelial cells has been considered to be highly correlated with the colorectal carcinogenesis, the precise mechanism of action remains poorly elucidated. Accumulating evidence suggests that the PGE receptor (EP)-mediated signal transduction pathway might play an important role in this process. In the present study, we investigated the mechanism of action underlying $PGE_2$-mediated cell proliferation and the effect of resveratrol on the proliferation of human colon cancer cells in terms of the modulating $PGE_2$-mediated signaling pathway. $PGE_2$ stimulated the proliferation of several human colon cancer cells and activated growth-stimulatory signal transduction, including Akt and ERK. $PGE_2$ also increased the phosphorylation of GSK-$3{\beta}$, the translocation of ${\beta}$-catenin into the nucleus, and the expressions of c-myc and cyclin D1. Resveratrol, a cancer chemopreventive phytochemical, however, inhibited $PGE_2$-induced growth stimulation and also suppressed $PGE_2$-mediated signal transduction, as well as ${\beta}$-catenin/T cell factor-mediated transcription in human colon cancer cells. These findings present an additional mechanism through which resveratrol affects the regulation of human colon cancer cell growth.
The inhibitors of prostaglandin $E_2\;(PGE_2)$ production and cyclooxygenase-2 (COX-2) activity have been considered as potential anti-inflammatory agents. In this study, we evaluated 9 compounds isolated from 5 Korean phytomedicinal plants (Spirea prunifolia, Paeonia suffruticosa, Salvia miltiorrhiza, Scutellaria baicalensis, and Artemisia capillaris) for the inhibition of $PGE_2$production and COX-2 expession in lipopolysaccharide (LPS)-stimulated human macrophages U937 cells. As a result, several compound such as prunioside A, penta-O-galloyl-beta-D-glucose, tanshinone IIA, baicalin, baicalein, wogonin, scopolatin, scoparone and decursinol showed potent inhibition of $PGE_2$production (50-70% inhibition at the test concentration of $10\;{\mu}M$). In addition, these compounds were also considered as potential inhibitors of COX-2 activity (45-73% inhibition at the test concentration of $10\;{\mu}M$). These active compound mediating COX-2 inhibitory activities are warranted for further elucidation of active principles for development of anti-inflammatory agents and these properties may contribute to the anti-atopic dermatitis activity.
The efficiency of a short-term treatment with an intravaginal device containing progesterone (CIDR) combined with pregnant mares' serum gonadotrophin (PMSG) and prostaglandin analogue ($PGF_{2\alpha}$) was evaluated for the induction of estrus, initiation of cyclic activity, and fertility in postpartum suckled yak cows. Seventy-five postpartum suckled yak cows were assigned to three treatments: (1) insertion of a CIDR intravaginal progesterone (1.9 g) (day 0), an administration of $PGF_{2\alpha}$ (0.2 mg i.m.) on day 6 and PMSG (1,000 IU i.m.) at the time of CIDR withdrawal on day 7 (CPP group, n=28); (2) an administration of $PGF_{2\alpha}$ (0.2 mg i.m.) on day 6 and PMSG (1,000 IU i.m.) on day 7 (PP group, n=21); (3) untreated animals served as the control (CG group, n=26). Seven yak bulls were placed in pastures with the cows for natural mating. Estrus rate in the CPP group (28/28) was higher (p<0.01) than in the PP group (6/21) and in the CG group (0/26) within 96 h after the end of treatment. The first service conception rate in the CPP group (21/28) was higher (p<0.01) compared with in the PP group (2/9) as judged by serum $P_4$ concentration $\geq$2.35 ng/ml on day 21 after breeding. It is concluded that a short-term progesterone treatment combined with PMSG and prostaglandin increased the proportion of yak cows that exhibited behavioral estrus with more synchronized estrus response and satisfactory conception rate in postpartum suckled yak cows.

Biotin is a water-soluble vitamin used as a skin conditioning agent and promotes the formation of intercellular lipid layers through increased lipid synthesis, which improves the skin's natural barrier function. The anti-inflammatory effects of biotin have been investigated using in vitro assay models, such as MTT assay, measurements of concentrations of nitric oxide(NO), prostaglandin E2(PGE$_2$), and inhibition rate of 5-lipoxygenase(5-LOX). In comparison with biotin, other plant extracts were tested at the same time which were kudzu vine extract, sage extract, paeonia extract, and dipotassium glycyrrhetinate. Nitric oxide is a signal molecule with functions such as neurotransmission, local vascular relaxation, and anti-inflammation in many physiological and pathological processes. NO can cause apoptosis and necrosis of target cells such as keratinocytes and is generated from L-arginine by nitric oxide synthase (NOS). Prostanoids, including prostaglandins and thromboxanes, are generated by the phospholipase $A_2$/cyclooxygenase(COX) pathway, and leukotrienes are generated by the 5-lipoxygenase pathway from arachidonic acid. Prostaglandin E2 recently have been shown to be beneficial in the resolution of tissue injury and inflammation, also has been implicated as an immunosuppressive agent and plasma levels of PGE$_2$ are elevated in patients sustaining thermal injury. Lipoxygenase metabolites from arachidonic acid have been implicated in inflammation, anti-inflammatory activity of the raw materials was evaluated in vitro by the offered inhibition of lipoxygenase.
프로스타글란딘 D2 (PGD2)와 E2 (PGE2)는 COX-2로부터 유래되는 주요 프로스타노이드로서, 슈도모나스에 의한 폐감염이 발생하였을 경우 폐에서 합성되어 슈도모나스 세균감염을 조절할 수 있음을 밝힌바 있음. 본 연구에서는 두 프로스타노이드의 생성 비율을 조절하는 기전을 연구하고자함. 마크로파아지에 의해 PGD2/PGE2 비율이 결정되는 지 조사하기 위해, 마우스의 허파로부터 마크로파아지를 분리하고 LPS로 처리할 경우, COX-2, PGD2합성 효소인 L-PGDS, PGE2의 합성효소인 mPGES-1 등의 발현이 두 프로스타노이드의 생성 비율에 미치는 영향을 조사하였음. 또한 이 효소들의 발현이 일차 허파 마크로파아지에 특이적인지의 여부를 조사하기 위해, 허파 마트로파이지 세포주인 MH-S와 비교 조사하였음. COX-2가 프로스타글린딘 비율에 미치는 영향을 조사하기 위해, COX-2 특이적 siRNA릉 이용하여 COX-2의 발현을 억제하고 L-PGDS, mPGES-1 등의 발현을 조사하였음. 결과에 따르면, 일차 허파 마트로파아지는 MH-S과는 달리 많은 양의 PGE2를 생성하나, 두 세포간 COX-2, L-PGDS, mPGES-1의 발현에는 큰 차이가 없었음. 이는 이들 효소 외에 다른 인자들이 두 프로스타노이드의 비율을 결정하는데 관여함을 제시함. LPS의 처리에 의해 폐염증을 발생시키고 허파에서의 PGD2/PGE2 비율을 조사한 결과, LPS에 의해 폐염증이 발생할 경우 LPS를 처리한 일차 허파 마크로파아지와 유사하게 PGE2의 발현이 PGD2에 비해 상당히 높았음. 결과적으로 본 연구의 결과는, 허파에서의 PGD2/PGE2 비율은, COX-2, L-PGDS, mPGES-1 등 PGD2나 PGE2의 합성에 직접적인 영향을 주는 효소에 의해 결정되지 않으며, 허파마크로파아지의 PGD2/PGE2 비율을 반영할 가능성을 제시함.
