Isolation and Characterization of Antioxidative Peptides from Enzymatic Hydrolysates of Yellowfin Sole Skin Gelatin (가자미피 젤라틴 가수분해물로부터 항산화성 펩티드의 분리${\cdot}$정제 및 특성)

  • To develop a natural antioxidative peptide, the gelatin was extracted from fish (Yellowfin sole) skin by hot $water(50^{\circ}C)$ extraction method and hydrolyzed with Alcalase, pronase and collagenase through a continuous 3-step membrane reactor. Each step enzymatic hydrolysates were determined the antioxidative activity and their synergistic effects, compared with $\alpha-tocopherol$ and butylated hydroxytoluene (BHT). Also, we tried to investigate the antioxidative disposition of peptide which was successfully separated by gel filtration, ion-exchange chromatography, and HPIC in cultured rat hepatocytes intoxicated with tert-butyl hydroperoxide (TBHP). Second step enzymatic hydrolysate (SSEH) among all hydrolysates and $\alpha-tocoperol$ was showed the strongest antioxidative activity. The optimum concentration of antioxidative activity for SSEH was $1\%(w/w)$ in linoleic acid. The synergistic effects were increased in using the hydrolysate with tocopherol and BHT. In the presence of the peptide isolated from SSEH, supplemented hepatocytes exposed to TBHP showed that delayed cell killing and decreased significantly the lipid peroxidation, compared with hepatocytes not cultured with isolated peptide.

  • We investigated the immunological function of cheese whey protein concentrate (CWPC), which is a by-product of cheese production, using mitogenic activity in murine splenocytes as an index. A fraction isolated by gel filtration and anion exchange chromatography of CWPC showed high mitogenic activity, comparable to the activity of lipopolysaccharide (LPS). The fraction was detected as a single band on SDS-PAGE. It contained calcium, inorganic phosphorus, and carbo-hydrate, indicating the active component to be a glycophosphopeptide (GPP) Since pronase digestion of GPP did not reduce its mitogenic activity, carbohydrate rather than peptide may be important in the activity, When applied on an anti-${\beta}$-caseinophosphopeptide (${\beta}$-CPP ) antibody affinity column, the GPP was separated into two components, one with affinity to ${\beta}$-CPP and the other without such affinity. Both the components contained N-linked oligosaccharide chains and had the mitogenic activity. These results demonstrate that cheese whey contains a GPP having strong mitogenic activity

  • In order to utilize the protein source from a fish proessing by-product, cod was hydrolyzed with various enzymes such as tuna pyloric caeca crude enzyme (TPCCE), a-chymotrypsin, trypsin, papain and pronase E. The TPCCE hydrolysate acquired the highest sensory properties on taste, odor and color. The resultant cod rfame protein hydrolysate (CFPH) which was hydrolyzed with TPCCE, was separated through a series of ultrafiltration membranes with molecular weight cut-off (MWCO) of 30, 10, 5 and 1 kDa, and four types of permeates in cluding 30 K (permeate from 30 kDa membrane), 10 K (permeate from 10 kDa membrane), 5 K (permeate from 5 kDa membrane) and 1 K (permeate from 1 kDa membrane) were obtained. The natural sauces were prepared with 30 K, 10 K, 5 K and 1 K hydrolysate, and the sauce prepared with 1 K hydrolysate was the best score in sensory evaluations. In addition the mixed sauce prepared with 1 K hydrolysate and commercial soy sauce was similar to commercial sauce in sensory properties. These results suggest that the mixed sauce would be utilized as the substitute of acid-hydrolysis sauce.

Molecular and functional characterization of the adiponectin (AdipoQ) gene in goat skeletal muscle satellite cells

  • Objective: It is commonly accepted that adiponectin binds to its two receptors to regulate fatty acid metabolism in adipocytes. To better understand their functions in the regulation of intramuscular adipogenesis in goats, we cloned the three genes (adiponectin [AdipoQ], adiponectin receptor 1 [AdipoR1], and AdipoR2) encoding these proteins and detected their mRNA distribution in different tissues. We also determined the role of AdipoQ in the adipogenic differentiation of goat skeletal muscle satellite cells (SMSCs). Methods: SMSCs were isolated using 1 mg/mL Pronase E from the longissimus dorsi muscles of 3-day-old female Nanjiang brown goats. Adipogenic differentiation was induced in satellite cells by transferring the cells to Dulbecco's modified Eagle's medium supplemented with an isobutylmethylxanthine, dexamethasone and insulin cocktail. The pEGFP-N1-AD plasmid was transfected into SMSCs using Lipofectamine 2000. Expression of adiponectin in tissues and SMSCs was detected by quantitative polymerase chain reaction and immunocytochemical staining. Results: The three genes were predominantly expressed in adipose and skeletal muscle tissues. According to fluorescence and immunocytochemical analyses, adiponectin protein expression was only observed in the cytoplasm, suggesting that adiponectin is localized to the cytoplasm of goat SMSCs. In SMSCs overexpressing the AdipoQ gene, adiponectin promoted SMSC differentiation into adipocytes and significantly (p<0.05) up-regulated expression of AdipoR2, acetyl-CoA carboxylase, fatty-acid synthase, and sterol regulatory element-binding protein-1, though expression of CCAAT/enhancer-binding $protein-{\alpha}$, peroxisome proliferator-activated receptor ${\gamma}$, and AdipoR1 did not change significantly. Conclusion: Adiponectin induced SMSC differentiation into adipocytes, indicating that adiponectin may promote intramuscular adipogenesis in goat SMSC.

Purification and Characterization of Antibacterial Compound Produced by Bacillus subtilis MJP1 (Bacillus subtilis MJP1이 생산하는 항세균 물질의 분리.정제 및 특성규명)

  • Antibacterial compound from Bacillus subtilis MJP1 was purified using C18 Sep-Pak cartridge, ion exchange chromatography, and gel filtration chromatography. The purified antibacterial compound showed antibacterial activity against Listeria monocytogenes, Bacillus subtilis, Staphylococcus aureus subsp. aureus, and Enterococcus faecalis. The purified antibacterial compound was found to be stable at $100^{\circ}C$ for 5 min and in the pH range of 3.0~9.0, but it was unstable at pH 10.0. It was inactivated by proteinase K and pronase E, and heat treatment at $121^{\circ}C$ for 15 min, but it was stable with lipase and $\alpha$-amylase treatment, which indicated its proteineous nature. Ultra performance liquid chromatography and electrospray ionization tandem mass spectrometry analysis were used to identify the purified antibacterial compound and confirmed the existence of two peptides (3356.54 Da, 3400.5244 Da).

Purification and Characterization of an Antimicrobial Substance from Bacillus subtilis HH28 Antagonistic to Bacillus cereus (Bacillus cereus를 억제하는 Bacillus subtilis HH28의 항균물질 정제와 특성규명)

  • A bacterium producing antimicrobial substance was isolated from cheonggukjang. The bacterium was identified as a strain of Bacillus subtilis by 16S rDNA sequencing and designated as Bacillus subtilis HH28. The antimicrobial substance produced from Bacillus subtilis HH28 was purified by 0-80% ammonium sulfate precipitation, DEAE-sepharose FF column chromatography, and Sephacryl S-200 HR gel chromatography. The molecular weight of the purified antimicrobial substance was estimated to be approximately 3,500 Da using Tricine sodium dodecyl sulfate-polyacrylamide gel electrophoresis and direct detection analysis. Antimicrobial substance from B. subtilis HH28 not only inhibited B. cereus, but also Listeria monocytogenes and Vibrio parahaemolyticus. The purified antimicrobial substance was stable at $40-80^{\circ}C$, and between pH 2 and 8. Antimicrobial activity of the purified substance was completely destroyed by treatment of protease, proteinase K, and pronase E, indicating that it is proteinaceous.

  • To develop novel anti-aging peptides from the germinated black rice, we treated with bromelain, papain and Pronase E. And we investigated the effects of the germinated black rice peptide (GBRP) as anti-aging cosmetic ingredients, and compared with the non-germinated black rice protein (NBRP). We investigated the effects on in vitro inhibition of matrix-metalloprotease (MMP), proliferation of human skin fibroblasts, stimulation of collagen synthesis and expression of UVA-induced MMPs in human skin fibroblasts, UVA induced MMP-1 expression and collagen contents in human skin fibroblasts were analyzed by enzyme-linked immunosorbent assay (ELISA). As a result, the molecular weight distributions of GBRP and NBRP were determined by gel permeation chromatography to be approximately 900 and 10,000 daltons. GBRP increased skin cell proliferation about 40% and reduced UVA-induced MMP-1 expression about 50%. Also the collagen protein level of cells, which were cultured with GBRP, was increased about 25%. These results suggest that the geminated plant seed peptides can be novel anti-aging ingredients for cosmetics.

The Color Measurement and Sensory Evaluation for the Accelerated Fish Sauce Products (속양(速釀) 어장유(魚醬油)에 대한 색(色) 측정(測定) 및 관능검사(官能檢査))

  • The color distribution of fish sauces was studied by using the change in log absorbance per 100nm as a parameter of color tone. A linear relationship was found between the logarithm of absorbance (log A) and wavelength at 450 nm to 650 nm in the color of fish sauces. Change in log A per 100nm $({\Delta}A)$ in the color of the fish sauce products was in the range of 0.55 to 0.59. Absorbance at 450 nm (A 450) of the fish sauce treated with soy sauce koji was high in comparison with A 450 of the fish sauce treated by pronase and control. The color of fish sauces treated with soy sauce koji was dark reddish orange, and was similar to the color of soy sauce. A 450 of every fish sauce increased with the progress of fermentation but ${\Delta}A$of the fish sauces slightly increased at the beginning of fermentation and then decreased at the end of fermentation. In the results of sensory evaluation for the flavor of fish sauce products, the fish sauce product treated with soy sauce koji that 20% salt was added at the first stage was the molt desirable in the strength of flavor, tastefulness and after taste, and the fish sauce was also the best in acceptability.

Study of Macrophage Stimulating Activity of the Polysaccharide Isolated from Leaves of Carthamus tinctorius L. (홍화(Carthamus tinctorius L.)잎으로부터 분리한 다당류의 Macrophage 활성화에 대한 연구)

  • In the screening of Korean traditional tea sources for the cellular lysosomal enzyme activity of peritoneal macrophage from mice, CT-0, a cold-water extract from Carthamus tinctorius L., showed the highest macro-phage-stimulating activity. CT-1-IIa-2-1, a purified macrophage-stimulation polysaccharide was obtained by a series of purification steps such as anion exchage chromatography with DEAE-Toyopearl 650M, gel permeation chromatography with Sepharose CL-6B, Sephacryl S-200, and HPLC with Superdex G-75. The molecular weight of homogeneous purified polysaccharide was estimated about 68 kDa. CT-1-IIa-2-1 consisted of xylose 27.44%, arabinose 16.14%, mannose 15.92% and glucose 14.47%. To measure acute toxicity, dose of 50, 100, 500, and 1000 mg/kg were intraperitoneally injected to ICR mice. The LD$\_$50/ was about 397 mg/kg.

Studies on the Production of Antibody to Mouse Zona Pellucida and its Effect on the Fertilization of Mouse Eggs (생쥐卵子의 透明帶에 대한 抗體의 生産과 이 抗體가 受精에 미치는 影響)

  • 本 試驗은 受精初期段階에서 생쥐卵子 透明帶의 機能을 糾明하고 種特異的인 精子受容體의 存在 및 構造理解를 위한 基礎硏究로서 ICR 생쥐의 透明帶를 免疫原으로 하여 토끼에서 抗透明帶抗體를 生産하고 免疫分析法 및 生物學的 檢査法으로 生産된 抗體가 생쥐 透明帶에 特異性이 있음을 確認한 후 이 抗體가생쥐 未受精卵의 體外受精에 미치는 影響과 또 이 抗體를 생쥐에 受動免疫하였을 때 受精에 미치는 影響을 調査하였던 바 다음과 같은 結果를 얻었다. 1. 處理區로서 抗血淸을, 對照區로서 對照血淸을 卵子培養液에 各各 0.3~10%로 含有한 培養液에서 생쥐의 未受精卵을 2時間 培養한 後 觀察하였을 때 對照區 卵子들의 透明帶가 正常的인 形態를 나타내었던 反面에, 處理區의 卵子들은 모두 透明帶 表面에 뚜렷한 沈澱層을 나타내었다. 이어 對照區와 處理區의 卵子를 0.1% pronase가 含有된 培養液으로 옮긴 후 觀察하였을 때, 對照區 卵子들의 透明帶가 5~7分만에 完全히 融解한 反面에, 處理區에서는 2~3時間까지 殘存하였다. 2. 處理區와 對照區 卵子를 FITC-goat anti-rabbit IgG가 含有된 培養液에서 養液한 후 螢光顯微鏡下에서 觀察하였을 때 對照區에서 對照血淸을 10% 含有한 培養液에서 培養한 일부 卵子들의 透明帶에서 희미한 螢光이 나타났으나 血淸稀釋度가 增加함에 따라 전혀 螢光을 나타내지 않았던 反面에, 處理區에서는 抗血淸이 1 : 1,000으로 含有된 培養液에서 培養한 卵子의 透明帶에서도 강한 螢光이 나타났다. 3. 帶抗體를 抗原으로 하여 間接酵素免疫分析法을 實施하였을 때 抗血淸과 對照血淸이 PBS에 1 : 200까지 稀釋되었을 때의 O.D.價는 별다른 差異를 나타내지 않았으나 血淸의 稀釋度가 增加함에 따라 O.D.價가 有意한 差異를 나타냄으로써 透明帶에 特異性을 갖는 抗體임을 確認할 수 있었다. 4. 抗血淸과 對照血淸이 各各 0.3~10%까지 含有된 培養液으로 생쥐의 未受精卵을 豫備培養한 다음 體外受精을 實施하였을 때의 受精率은 對照區에서 77.7%~87.7%이었던 反面에, 處理區에서는 0~29.8%로서 抗透明帶抗體가 생쥐卵子의 體外受精을 完全히 또는 顯著히 抑制하였다. 그러나 抗血淸과 對照血淸이 各各 2.5~10%까지 含有된 培養液으로 透明帶를 除去한 未受精卵의 體外受精을 하였을 때의 受精率은 處理區와 對照區에서 各各 93.0~98.3% 및 93.3~94.3%로서 受精抑制現象을 나타내지 않았다. 5. 抗血淸과 正常血淸, 그리고 兩 血淸으로부터 protein A-affinity chromatography에 의해 分離한 immunoglubulin G를 各各 생쥐에 受動免疫하였을 때, 對照區로서 對照血淸을 0.1~0.3ml 또는 control IgG를 1~3ml 投與했을 때의 各各의 受精率이 80.4~90.2%, 82.2~94.4%이었던 反面에, 處理區로서 抗血淸을 0.3ml 또는 immune IgG를 3mg 投與한 생쥐에서는 完全한 受精抑制現象이 確認되었다.
