Microbial β-Galactosidase of Pediococcus pentosaceus ID-7: Isolation, Cloning, and Molecular Characterization

  • Pediococcus pentosaceus ID-7 was isolated from kimchi, a Korean fermented food, and it showed high activity for lactose hydrolysis. The ${\beta}$-galactosidase of P. pentosaceus ID-7 belongs to the GH2 group, which is composed of two distinct proteins. The heterodimeric LacLM type of ${\beta}$-galactosidase found in P. pentosaceus ID-7 consists of two genes partially overlapped, lacL and lacM encoding LacL (72.2 kDa) and LacM (35.4 kDa). In this study, Escherichia coli MM294 was used for the production of LacL, LacM, and LacLM. These three types of recombinant proteins were expressed, purified, and characterized. The specific activities of LacLM and LacL were 339 and 31 U/mg, respectively. However, activity was not detected with LacM alone. The optimal pH of LacLM and LacL was pH 7.5 and pH 7.0, and the optimal temperature of LacLM and LacL was $40^{\circ}C$ and $50^{\circ}C$, respectively. The optimal temperature changes indicate that LacLM is able to achieve higher activity at a relatively lower temperature. LacLM was strongly activated by $Mg^{2+}$, $Mn^{2+}$, and $Zn^{2+}$, which was not true for LacL. Consistent with this, EDTA strongly inactivated LacLM and LacL, but the presence of reducing agents did not dramatically alter the activity. Taken together, multiple alignment of amino acid sequences and phylogenetic analysis results of LacL and LacM of P. pentosaceus ID-7 suggest the evolution of LacL into LacLM and that the use of divalent metal ions results in higher activity.

2'-Spirocyclopropyl-carbocyclic Nucleoside as a Novel Scaffold for Potent Anti-HCV Agents

  • The discovery of 2'-spirocyclopropyl-ribocytidine (J. Med. Chem. 2010, 53, 8150-8160) as a potent inhibitor of RNA synthesis by NS5B ($IC_{50}=7.3{\mu}M$), the RNA polymerase encoded by hepatitis C Virus (HCV), has led to the synthesis and biological evaluation of several carbocyclic versions of 2'-spiropropyl-nucleosides. The cyclopentenol intermediate 7 was successfully constructed via ring-closing metathesis (RCM) from divinyl 6. Spirocyclopropanation of enone 8 was effected by using (2-chloroethyl)-dimethylsulfonium iodide and potassium tert-butoxide to form the desired intermediate 9. The synthesized nucleoside analogues 21-24 were assayed for their ability to inhibit HCV RNA replication in a subgenomic replicon Huh7 cell line. Among them, the cytosine nucleoside analogue 22 exhibited significant anti-HCV activity ($EC_{50}= 8.2{\mu}M$).

Racemic Synthesis of Novel 6'-Methylene-5'-norcarbocyclic Purine Phosphonic Acid Analogues via Mitsunobu Reaction

  • Novel 5'-norcarbocyclic adenine and guanine phosphonic acid analogues with 6'-electronegative moiety such as unsaturated C-C bond were designed and synthesized from commercially available 2-methylene-propane-1,3-diol (4). Regioselective Mitsunobu reaction successfully proceeded from an allylic functional group (${\pm}$)-12b at low reaction temperature in polar cosolvent (DMF/1,4-dioxane) to give purine phosphonate analogues (${\pm}$)-13 and (${\pm}$)-20. The purine nucleoside phosphonate and phosphonic acid analogues were subjected to antiviral screening against HIV-1. Guanine analogue (${\pm}$)-23 shows significant anti-HIV activity in PBM cell lines ($EC_{50}=8.1\;{\mu}M$).

  • The purpose of this study is to define the role of a T. Q. C. engineer in the plant engineering activities. A "plant engineering" moves to complete a new project through a series of stages such as: 1. Research & Development stage. 2. Economic evaluation & engineering design stage. 3. Erection & start up stage. 4. Final production & control stage. This suggests that the plant engineering project should be carried out with a wide variety of tasks, as well as a central control. For a successful completion of the project, a T. Q. C. engineer is required, in practice, either as a central activity creator or as a key advisor in each task.

  • Novel 2'(${\beta}$)-fluoro-3'(${\alpha}$)-hydroxy-threose nucleosides (iso-FMAU) as antiviral agents were designed and racemically synthesized from Solketal. Condensation successfully proceeded from a glycosyl donor 9 under $Vorbr{\ddot{u}}ggen$ conditions yielded the nucleoside analogues. Ammonolysis and hydrolysis of isopropylidene protection group gave the desired nucleoside analogues 12, 15, 18, and 19. The antiviral activities of the synthesized compounds were evaluated against the HIV-1, HSV-1, HSV-2 and HCMV. Compound 12 displayed some anti-HCMV activity ($EC_{50}=24.7{\mu}g/ml$) without exhibiting any cytotoxicity up to $100{\mu}M$.

  • The objectives of this paper are to introduce the PI(Public Involvement) in railway project and case study about public participation in decision making process for Ko-Yang light rail line development, besides to emphasize the importance of information considering the difference of resident location, regional character and so on are investigated before and after public participation. As the results, it was clarified about PI activities styles that consensus is not achieved in Ko-Yang public participation, the psychological factor such as the knowledge and satisfaction, receptivity about light rail project strongly is affected to making decision.

  • This study analyzed the pattern of preschool children's abilities based on Project Spectrum and compared the relationship between intelligence domains with working styles of children. Generally, children showed both strengths and weaknesses in specific Spectrum domains, but some children had only strengths and others had only weaknesses without strengths. It appeared that working style does not relate to children's strong domains, but individual disposition was reflected by characteristics of the task. During both individual and group spectrum activities, children with strengths were more "easily engaged in activity", "confident", and "focused" than those without strong intelligence domains. During individual activities, the "reflective" domain showed children with strengths in intelligence domains are more reflective than those without strengths.

Design of ESN(Educational Sensor Network) for interpretation of the data

  • This paper has focused on the development of an educational sensor network (ESN) based on wireless sensor networks(WSN) and pervasive monitoring systems for students' activity during scientific experiments. A number of WSN systems have been proposed with integrated wireless transmission, mounted sensor boards and local processing. However, there is no trail to employ WSN on the educational field. In this paper, to facilitate research and development using wireless sensor network and multi-sensor data fusion, the educational sensor network (ESN) hardware development platform is presented. The ESN project is conducted over one semester time period (Spring Semesters). It involves approximately twenty middle school students who enrolled a gifted program in Kongju National University. Though under prepared, these students are in general highly motivated to learning specially when presented with the ESN project. An ESN project such as this is expected to provide an excellent means for teaching and learning scientific and mathematical principles.

Performance Analysis of Local Optimization Algorithms in Resource-Constrained Project Scheduling Problem (자원제약 프로젝트 스케쥴링 문제에 적용 가능한 부분 최적화 방법들의 성능 분석)

  • The objective of this paper is to define local optimization algorithms (LOA) to solve Resource-Constrained Project Scheduling Problem (RCPSP) and analyze the performance of these algorithms. By representing solutions with activity list, three primitive LOAs, i.e. forward and backward improvement-based, exchange-based, and relocation-based LOAs are defined. Also, combined LOAs integrating two primitive LOAs are developed. From the experiments with standard test set J120 generated using ProGen, the FBI-based LOA demonstrates to be an efficient algorithm. Moreover, algorithms combined with FBI-based LOA and other LOA generate good solutions in general. Among the considered algorithms, the combined algorithm of FBI-based and exchangebased shows best performance in terms of solution quality and computation time.

The Productivity Improvement for Steel Framing Work Efficiency by Work Sampling and 5-minute Rating Technique

  • This study presents the results of our analysis and recommendations for process and productivity improvements. The project studied consists of a 5-story research building, with a structure of steel frames supporting concrete slabs. The observations focused on the analysis of the overall erection and framing process. The methods used for the analysis consisted in intensive visits on site, where construction processes were observed in term of resources, activities, durations, materials' handling procedures, and technology used. Back to the office, authors used the information captured to model the different trades' activities, using work sampling and 5-minute rating technique. The work sampling provides insight into the activity, hence allowing for process improvements. The productivity of various trades is strongly dependent on the organization of the work process and work site conditions. Improving the productivity of the entire project or company is not possible until everyone is committed to improvement.
