Statistical Optimization for Improved Production of Cyclosporin A in Solid-State Fermentation

  • This work evaluates the effect of different amino acids on production of Cyclosporin (CyA) production in solid-state fermentation that was previously optimized for different fermentation parameters by one factor at-a-time for the maximum production of CyA by Tolypocladium inflatum MTCC557. Based on the Plackett-Burman design, glycerol, ammonium sulfate, $FeCl_3$, and inoculum size were selected for further optimization by response surface methodology (RSM). After identifying effective nutrients, RSM was used to develop mathematical model equations, study responses, and establish the optimum concentrations of the key nutrients for higher CyA production. It was observed that supplementation of medium containing (% w/w) glycerol, 1.53; ammonium sulfate, 0.95; $FeCl_3$, 0.18; and inoculum size 6.4 ml/5g yielded a maximum of 7,106 mg/kg as compared with 6,480 mg CyA/kg substrate using one factor at-a-time. In the second step, the effect of amino acids on the production of CyA was studied. Addition of $_L$-valine and $_L$-leucine in combination after 20 h of fermentation resulted in maximum production of 8,166 mg/kg.

  • The fourth industrial revolution encourages manufacturing industry to pursue a new paradigm shift to meet customers' diverse demands by managing the production process efficiently. However, it is not easy to manage efficiently a variety of tasks of all the processes including materials management, production management, process control, sales management, and inventory management. Especially, to set up an efficient production schedule and maintain appropriate inventory is crucial for tailored response to customers' needs. This paper deals with the optimized inventory policy in a steel company that produces granule products under supply contracts of three targeted on-time delivery rates. For efficient inventory management, products are classified into three groups A, B and C, and three differentiated production cycles and safety factors are assumed for the targeted on-time delivery rates of the groups. To derive the optimized inventory policy, we experimented eight cases of combined safety stock and data analysis methods in terms of key performance metrics such as mean inventory level and sold-out rate. Through simulation experiments based on real data we find that the proposed optimized inventory policy reduces inventory level by about 9%, and increases surplus production capacity rate, which is usually used for the production of products in Group C, from 43.4% to 46.3%, compared with the existing inventory policy.

Hydrogen Fermentation of the Galactose-Glucose Mixture (갈락토스-글루코스 혼합당 수소 발효)

  • Galactose, an isomer of glucose with an opposite hydroxyl group at the 4-carbon, is a major fermentable sugar in various promising feedstock for hydrogen production including red algal biomass. In this study, hydrogen production characteristics of galactose-glucose mixture were investigated using batch fermentation experiments with heat-treated digester sludge as inoclua. Galactose showed a hydogen yield compatible with glucose. However, more complicated metabolic steps for galactose utilization caused a slower hydrogen production rate. The existence of glucose aggravated the hydrogen production rate, which would result from the regulation of galactose-utilizing enzymes by glucose. Hydrogen produciton rate at galactose to glucose ratio of 8:2 or 6:4 was 67% of the production rate for galactose and 33% for glucose, which could need approximately 1.5 and 3 times longer hydraulic retention time than galacgtose only condition and glucose only condition, respectively, in continuous fermentation. Hydrogen production rate, Hydrogen yield, and organic acid production at galactose to glucose ratio of 8:2 or 6:4 were 0.14 mL H2/mL/hr, 0.78 mol $H_2$/mol sugar, and 11.89 g COD/L, respectively. Galactose-rich biomass could be usable for hydogen fermenation, however, the fermentation time should be allowed enough.

  • In manfacturing systems, to overcome the inherent complexities and uncertainties, due-dates are usually decided by some simple rules. This study proposes a computational procedure for estimating production lead time for computerized manufacturing systems. Based on the current system status stored in the computers, the procedure estimates production lead time for an incoming order. The result is more accurate than the ones produced by old rules, and yet the procedure is fast in computation speed enough to use in real time. Simulations are used to prove these.

  • This paper develops a standard time table using STD(Standard Tine Data) Technique in multi-product and one-unit production systems. STD leads to greater consistency between similar jobs. The constant-time and special elements will always be the same even though they pertain to different part numbers. Actually, the standards will very based on the size of the variable elements that are in control of the time. A real-field example is presented.

Effects of Meteorological and Oceanographic Properties on Variability of Laver Production at Nakdong River Estuary, South Coast of Korea (낙동강 하구 해양환경 및 기상 요인이 김P(orphyra yezoensis) 생산량 변화에 미치는 영향)

  • To understand the effects of marine environmental and meteorological parameters on laver Porphyra yezoensis production at Nakdong River Estuary, we analyzed marine environmental (water temperature, salinity, nutrients, etc.) and meteorological properties (air temperature, wind speed, precipitation, sunshine hours) with yearly and monthly variations in laver production over 10 years (2003-2013). Air and water temperature, wind speed, sunshine hours and precipitation were major factors affecting yearly variability in laver production at the Nakdong River Estuary. Lower air and water temperatures together with higher levels of nutrients and sunshine and stronger wind speeds resulted in higher laver harvests. Salinity and nitrogen did not show clear correlations with laver production, mainly due to the plentiful supply of nitrogen from river discharge and the low frequency of environmental measurements, which resulted in low statistical confidence. However, environmental factors affecting monthly laver production were related to the life cycle (culturing stage) of Porphyra yezoensis and were somewhat different from factors affecting annual laver production. In November, a young laver needs lower water temperatures for rapid growth, while a mature laver needs much stronger winds and more sunshine, as well as lower temperatures for massive production and effective photosynthesis, mostly in December and January. However, in spring (March), more stable environments with fewer fluctuations in air temperature are needed to sustain the production of newly deployed culture-nets ($2^{nd}$ time culture). These results indicate that rapid changes in weather and marine environments caused by global climate change will negatively affect laver production and, thus, to sustain the yield of and predict future variability in laver production at the Nakdong River estuary, environmental variation around laver culturing farms needs to be monitored with high resolution in space and time.

Determining Appropriate Production Conditions in Cellular Manufacturing Systems (셀생산(生産)의 효율적(效率的)인 운용(運用)을 위한 시뮤레이션 연구(硏究))

  • Although there are numerous studies that address the problem of optimal machine grouping and part family classification for cellular manufacturing, little research has been reported that studies the conditions where cellular manufacturing is appropriate. This paper, in order to evaluate and compare the job shop with the GT cellular shop, the performance of those shops were simulated by using SIMAN. We tested the effect of independent variables including changes of product demands, intercell flow level, group setup time, processing time variability, variety of material handling systems, and job properties (ratio of processing time and material handling time). And also performance measures (dependent variables), such as machine utilization, mean flow time, average waiting time, and throughput rate, are discussed. Job shop model and GT cellular shop written in SIMAN simulation language were used in this study. These systems have sixteen machines which are aggregated as five machine stations using the macro feature of SIMAN. The results of this research help to better understand the effect of production factors on the performance of cellular manufacturing systems and to identify some of the necessary conditions required to make these systems perform better than traditional job shops. Therefore, this research represents one more step towards the characterization of shops which may benefit from cellular manufacturing.

A Study on Cycle Time and Power Saving Effect of a Hydraulic Hybrid Injection Molding Machine using a Servo Motor (서보모터를 이용한 유압 하이브리드식 사출성형기의 공정시간 및 절전효과에 관한 연구)

  • The cycle time and power saving effect of a hydraulic hybrid injection molding machine using a servo motor are considered in this paper. In order to verify control characteristics, such as pressure and speed, experiments were performed with the hydraulic hybrid injection molding machine, clamping force of 110 ton. The power consumption and production cycle time of a conventional hydraulic injection molding machine were measured to compare its performances with the hydraulic hybrid injection molding machine. An injection molding machine with a clamping force of 1300 ton was used as the conventional machine, the hybrid machine was implemented by replacing its induction motors with servo motors. In the remodeled hybrid machine, experiments were performed to investigate how the displacement of the mold clamping pump affects the power consumption and production cycle time. The results showed that the production cycle time of the hybrid injection molding is similar to a conventional hydraulic injection molding machine but with a significant energy saving of about 40%.

Effects of Hyeolbuchukeo-tang(Xiefuzhuyu-tang) on NO Production in Aortic Vascular Smooth Muscle Cells (혈부축어탕이 대동맥 평활근 세포에서 NO 생성에 미치는 영향)

  • Objectives : Nitric oxide (NO) plays an important role in normal and pathophysiological cells as a messenger molecule, neurotransmitter, microbiological agent, or dilator of blood vessels and arteriosclerosis, respectively. This study was undertaken to understand the mechanism of NO production and effect of Hyeolbuchukeo-tang (Xiefuzhuyu-tang) on NO production in cultured vascular smooth muscle cell (VSMC). Methods and Results : VSMC was isolated from aorta and cultured. Cultured primary cells were identified as VSMC with anti--smooth muscle actin antibody. A large amount of NO was produced in cultured VSMC treated with $IFN-{\gamma}$ plus TNF in a time- and dose-dependent manner. $TNF-{\alpha}$ was a more efficient stimulator than $IFN-{\gamma}$ in NO production of cultured VSMC. iNOS protein wasdetected within 3 hrs and it increased up to 12 hrs in a time-dependent manner. However, accumulated NO in cytokine-treated VSMC was not detected within 3 hrs. NO production in cytokine-treated VSMC showed the dose- and time-dependent manner, and increased up to 48 hrs. The activated VSMC produced a large amount of NO (about 60 uM). Hyeolbuchukeo-tang (Xiefuzhuyu-tang) alone did not induceNO production, but it potentiated the effect of $TNF-{\alpha}$ on NO production and increased NO production by about 20%. Hyeolbuchukeo-tang (Xiefuzhuyu-tang) did not affect the transcriptional activity of iNOS gene, but increased the accumulation of iNOS. These results indicate that Hyeolbuchukeo-tang (Xiefuzhuyu-tang) could modulate the translational level of iNOS. PKC did not modulate NO production, but calcium ionophore A23187 decreased NO production. However, Hyeolbuchukeo-tang (Xiefuzhuyu-tang) elevated the decreased NO production in A23187-treated VSMC by modulating the stability of iNOS transcripts. Half-life of the synthesized transcripts appeared to have about 6 hrs. PDTC, an $NF-{\kappa}B$ inhibitor, blocked the accumulation of iNOS mRNA, indicating that $NF-{\kappa}B$ served as an important modulator in the transcriptional regulation of iNOS. As Hyeolbuchukeo-tang (Xiefuzhuyu-tang) potentiated the effect of the $TNF-{\alpha}$ on NO production but had no additional effect on PDTC-modulated NO production, it is suggested that Hyeolbuchukeo-tang (Xiefuzhuyu-tang) enhances the $TNF-{\alpha}-mediated$ NO production of VSMC by modulating the iNOS activity and the stability of iNOS transcripts in activated VSMC having the elevated intracellular calcium ion. Conclusions : This study suggests that Hyeolbuchukeo-tang (Xiefuzhuyu-tang) has a potential capacity for preventing and treating diseases of the circulation system, including arteriosclerosis.
