Production of pullulan by Aureobasidium pullulans HP-2001 with continuous culture

  • In batch culture of A. pullulans HP-2001, maximal production of pullulan was 57.90 g/l at 96 hr. The maximal production of pullulan with 76.55 g/l occurred when substituted solution was 10% (w/v) sucrose, 0.25% (w/v) yeast extract and mineral salts. The maximal production of pullulan was 75.00 g/l with a dilution rate of $0.015\;h^{-1}$. Productivity of pullulan in the continuous culture fed with the fresh medium was higher than that fed with the fresh medium without nitrogen source.

Seaweed Cultivation in Indonesia: Recent Status

  • Indonesia is well-known as biggest producer of seaweed especially for Eucheuma and Gracilaria and also has huge potential resources and capability to develop seaweed cultivation and product. There are several provinces which have potential resources and have been contributing on seaweed production. The next challenge about seaweed production is using integrated system on brackishwater and marine aquaculture. Furthermore, about 2,000,000 ton of potential seaweed production is not explored yet. This article also tries to figure out some related aspects which are technical, economical and forecasting aspect. There is a disease which named "ice-ice" is one of the main problem and giving a new challenge in developing of problem solving for seaweed cultivation method. Economical parameters are also main important key to find out the feasibility of seaweed cultivation industry. In addition, the seaweed cultivation and production in Indonesia also have potential performance on biofuel resources as a part for solving the world problem on energy demand.

  • Since FMS(Flexible Manufacturing System) such as semiconductor production systems have the characteristic that each device has to be commonly used in several stages, it is difficult to find an optimal solution. In this paper, we proposed the new algorithm which can get the optimal ratio of sharing resources. We will implement the proposed algorithm to semiconductor production systems. We introduce the optimal algorithm, which is modeled and analyzed by ExSpect, a petri net based simulation tool. When there exist conflicts of sharing resources, the scheduling method is adopted, which gives a priority to the most preceded process. The suggested algorithm can be used not only in semiconductor production systems but also in various FMS.

A Resource Scheduling for Supply Chain Model

  • This paper presents an optimization formulation for resource scheduling in Critical Resource Diagramming (CRD) of production scheduling networks. A CRD network schedules units of resources against points of needs in a production network rather than the conventional approach of scheduling tasks against resource availability. This resource scheduling approach provides more effective tracking of utilization of production resources as they are assigned or 'moved' from one point of need to another. Using CRD, criticality indices can be developed for resource types in a way similar to the criticality of activities in Critical Path Method (CPM). In our supply chain model, upstreams may choose either normal operation or expedited operation in resource scheduling. Their decisions affect downstream's resource scheduling. The suggested optimization formulation models resources as CRD elements in a production two-stage supply to minimize the total operation cost

Leaf and Stem Production of Perilla frutescens by Fertilizing of Sta-Green

  • This study was conducted to determine feasibility of production system of Perilla frutescens leaf-stem by fertilizing of Sta-Green in pots. Germination rate of Perilla frutescens seeds collected in 2002 was $7\%$, also germination rate of seeds collected in 2003 was $62\%$, while germination rate of seeds collected in 2004 was above $93\%$. Seed germination rate of Perilla frutescens collected in 2004 were higher than seed gathering in 2002. Especially, plant growth and yield of Perilla frutescens grown in pot(The pots was filled with soil mixtures of Sta-Green and Peat Moss mixed with 40:60 ratio.) was the highest. These results indicate that leaf and stem production of Perilla frutescens can be improved by fertilizing of Sta-Green in pots.

  • In this study, we investigated the immune-enhancing activity of water extracts from Hydrangea macrophylla subsp. serrata (WE-HML). WE-HML increased cell viability and production of immunomodulators, which contributed to activating phagocytic activity in RAW264.7 cells. Inhibition of JNK and NF-κB reduced the production of immunomodulators by WE-HML. ROS inhibition suppressed the production of immunomodulators, and the activation of JNK and NF-κB signaling by WE-HML. TLR4 inhibition attenuated the production of immunomodulators, and activation of JNK and NF-κB signaling by WE-HML. In the immunosuppressed mouse model, WE-HML increased the spleen index, the levels of the cytokines, the numbers of white blood cells, lymphocytes, and neutrophils. However, WE-HML inhibited LPS-mediated overproduction of pro-inflammatory mediators in RAW264.7 cells, which indicated that WE-HML may have anti-inflammatory activity under excessive inflammatory conditions. Taken together, WE-HML may be considered to have immune-enhancing activity and expected to be used as a potential immune-enhancing agent.

  • This study aims to make detail examination of smallholder dairy farming systems in the Nongpho Dairy Cooperative. Forty-three dairy farms were selected from three geographical areas i.e. irrigated area, municipality area, and factory area. Within each area some number of sample farms were selected from each of the three levels of farm and animal crowdedness (very crowded, crowded, and not crowded farms). Detail data were collected during 1996 to 1997, they were socio-economic conditions of the sample farms and farmers, dairy production systems and management of resources (animals, bam, feeds, stocking rates, herd structure, animal body conditions, milk yield and milk quality, manure and farm wastes management, and other related items). Detail information useful for the improvement of farm production efficiency were discussed. It was very clear that much improvement of smallholder dairy production can be achieved if the recommendations given by this study were implemented.

  • Asia is not only the primary region for global pig production but also the largest consumer of pork worldwide. Although the pig production in Asia has made great progress in the past, it still is confronted with numerous challenges. These challenges include: inadequate land and feed resources, a substantial number of small-scale pig farms, escalating pressure to ensure environmental conservation, control of devastating infectious diseases, as well as coping with high temperatures and high humidity. To solve these problems, important investments of human and financial capital are required to promote large-scale production systems, exploit alternative feed resources, implement precision feeding, and focus on preventive medicine and vaccines as alternatives to antibiotics, improve pig breeding, and increase manure recycling. Implementation of these techniques and management practices will facilitate development of more environmentally-friendly and economically sustainable pig production systems in Asia, ultimately providing consumers with healthy pork products around the world.

Optimal Fermentation Conditions for Enhanced Glutathione Production by Saccharomyces cerevisiae FF-8

  • The influence of feedstock amino acids, salt, carbon and nitrogen sources on glutathione production by Saccharomyces cerevisiae FF -8 was investigated. Glucose, yeast extract, KH$_2$PO$_4$, and L-cysteine were found to be suitable feedstock. Highest glutathione production was obtained after cultivation with shaking for 72 h in a medium containing glucose 3.0% (w/v), yeast extract 3.0%, KH$_2$PO$_4$ 0.06% and L-cysteine 0.06%. The glutathione concentration achieved using this medium increased 2.27-fold to 204 mg/l compared to YM basal medium.

Screening of Inhibition Activity of LPS-induced NO Production by Ethanol Extracts from Jeju Island Native Plants and Algae

  • Herbal medicines have been used as a basic means of clinical trial throughout history, and traditional medicines are targeted to seek functional components. To discover new cosmetic or food ingredients among numerous natural resources from Jeju island, we screened for inhibition activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Although NO formation plays an important role to relax vascular muscles or eliminate tumors, NO produced excessively in inflammatory condition can cause metabolic diseases or inflammatory dysfunctions. Among 52 natural resources ethanol extracts, 5 extracts inhibited NO production over 25% compared to only LPS-treated control at the concentration of $100{\mu}g/mL$. In further study, we try to investigate other bio-activities and the phytochemicals of 5 different extracts as useful ingredients for cosmetics or functional foods.
