A natural population of Adonis multiflora, a spring ephemeral herb growing in temperate deciduous forests, was studied to determine the seed production characteristics. Plant size, flowering time, and seed number were monitored from February 2009 to May 2011 in main growing season (i.e., from March through May). The biomass rates of the shoot and the root in the A. multiflora population were 22-24% and 76-78%, respectively, and the biomass of the root was proportional to that of the shoot. The flowering rate was 60% in the plants with 1 to 2 g of shoot biomass, and 100% in the plants with >2 g of shoot biomass. In the plants with root biomass between 4 and 6 g, the flowering rate was 43% and, in the plants with the root biomass over 8 g, it was 100%. The shoot biomass was a better predictor of the flower production probability than the root biomass. The number of flowers and seeds was closely correlated to shoot biomass at 1% significance level. The size of the plant that produced seed excessively instead of the shoot biomass in one year typically decreased in the next year and vice versa. The flowering time and its duration were closely related to the number of faithful seeds but not to that of total seeds. The number of faithful seeds was proportionate to flowering duration and inversely proportionate to flowering time (year day, YD). In a plant, the number of faithful seeds noticeably decreased with the inflorescence (i.e., order of flower in a plant), and this difference between the two successive flowers was significant at the 1% level between the first and the third flower in 2009 and 2011 but not between the third and the fourth. However, the number of total seeds was mostly similar in the first through the fourth flower for all three years.
To characterize the culture medium for the biosurfactant production by Bacillus pumilus IJ-1, the influences of various carbon, nitrogen and mineral sources were assessed. As a result, the highest biosurfactant production was observed after 96 h cultivation containing 0.5% (w/v) tryptone. The strain was able to grow and produce biosurfactant at 0-10% (w/v) NaCl, in the pH range of 5-10, and at $20-45^{\circ}C$. Optimal culture conditions for the biosurfactant production were at $20^{\circ}C$ and pH 9.0 after 72 h incubation and the surface tension of biosurfactant was 27.0 dyne/cm.
Macrophages generated in vitro using macrophage-colony stimulating factor (M-CSF) and interleukin (IL)-6 from bone marrow cells (BM-Mp) are defective in antigen presenting cell (APC) function as shown by their ability to induce the proliferation of anti-CD3 mAb-primed syngeneic T cells. However, they do express major histocompatibility (MHC) class I and II molecules. accessory molecules and intracellular adhesion molecules. Here we demonstrate that the defective APC function of macrophages is mainly due to production of $TGF-{\beta}1$ by BM-Mp. Methods: Microarray analysis showed that $TGF-{\beta}1$ was highly expressed in BM-Mp, compared to a macrophage cell line, B6D. which exerted efficient APC function. Production of $TGF-{\beta}1$ by BM-Mp was confirmed by neutralization experiments of $TGF-{\beta}1$ as well as by real time-polymerase chain reaction (PCR). Results: Addition of $anti-TGF-{\beta}1$ monoclonal antibody to cultures of BM-Mp and anti-CD3 mAb-primed syngeneic T cells efficiently induced the proliferation of syngeneic T cells. Conversely, the APC function of B6D cells was almost completely suppressed by addition of $TGF-{\beta}1$. Quantitative real time-PCR analysis also confirmed the enhanced expression of $TGF-{\beta}1$ in BM-Mp. Conclusion: The defective APC function of macrophages generated in vitro with M-CSF and IL-6 was mainly due to the production of $TGF-{\beta}1$ by macrophages.
A stockperson has a significant influence on the productivity and welfare of his animals depending on his stockmanship competence. In this study, stockmanship competence (SC) is defined as the capacity of the stockperson to ensure the welfare of his animals by providing his animals' needs. The study was conducted to evaluate the SC of backyard goat raisers and examine its relationship to goat productivity and economic profitability. There were 101 respondents for this study who have all undertaken farmer livestock school on integrated goat management (FLS-IGM). Interview was conducted in Region I, Philippines on September 3 to 30, 2012 and March 4 to 17, 2013. Data on SC, goat productivity and farmer's income were gathered. Questions regarding SC were formulated based on the Philippine Recommendations for Goat Production and from other scientific literature. Housing, feeding, breeding and health and husbandry management were the indicators used in computing stockmanship competence index score (SCIS). Pearson correlation using Statistical Package for Social Science (SPSS) was carried out to analyse the relationship between SCIS, productivity and income. Based from the results of the study, a majority of the respondents raised native and upgraded goats. The computed mean SCIS before and after undergoing FLS-IGM were 38.52% and 75.81% respectively, a percentage difference of 65.23%. Both index scores resulted in significant differences in productivity and income. The median mature weight and mortality rate of goats before FLS-IGM was 14 kg and 30% respectively. After FLS-IGM, median mature weight was 19 kg and mortality rate decreased from 30% to 11.11%. Likewise, fewer goat diseases were observed by farmers who were able to undergo FLS-IGM. With regard to income, there was a 127.34% difference on the median net income derived by farmers. Result implies that improved SC could lead not only to increased productivity and income of backyard goat raisers but also to better animal welfare.

Commercial application of laccase is often hampered by insufficient enzyme stocks, with very low yields obtained from natural sources. This study aimed to improve laccase production by mutation of a Coriolopsis gallica strain and to determine the biological properties of the mutant. The high-yield laccase strain C. gallica TCK was treated with N-methyl-N-nitro-N-nitrosoguanidine and ultraviolet light. Among the mutants isolated, T906 was found to be a high-production strain of laccases. The mutant strain T906 was stabilized via dozens of passages, and the selected ones were further processed for optimization of metallic ion, inducers, and nutritional requirements, which resulted in the optimized liquid fermentation medium MF9. The incubation temperature and pH were optimized to be 30℃ and 4.5, respectively. The mutant strain T906 showed 3-times higher laccase activity than the original strain TCK under optimized conditions, and the maximum laccase production (303 U/ml) was accomplished after 13 days. The extracellular laccase isoenzyme 1 was purified and characterized from the two strains, respectively, and their cDNA sequence was determined. Of note, the laccase isoenzyme 1 transcription levels were overtly increased in T906 mycelia compared with values obtained for strain TCK. These findings provide a basis for C. gallica modification for the production of high laccase amounts.
High prices of raw materials used as media for the mushroom cultivation increased the cost of commercial production of oyster mushroom (Pleurotus ostreatus). In this study, garlic peels (Allium sativum f. pekinese) as an agricultural by-product were investigated to replace the saw dust for the bottle cultivation of oyster mushroom. Mycelial growth of oyster mushroom were examined by the extracts made from the sawdust, rice bran and garlic peels. The mycelial growth was very poor in the agar media containing the extracts of sawdust or garlic peels, but was good when those of the rice bran were added. In the polypropylene bottle experiment, the sawdust medium which ammended with minerals vitamin was essential for the production of the mushroom fruitbodies. The rice bran was considered to stimulate the mycelial growth, but not the development of basidiocarps. The garlic peel was not a factor to stimulate the production of mushroom fruitbodies, but a raw material ammended with the rice bran produced much amounts of mushroom. In this work, garlic peels ($10{\sim}70%$ v/v) added to the mixture of sawdust and rice bran (4 : 1, v/v) was considered to help the productions of mushroom fruitbodies. Based on the result, the replacement of expensive saw dust with inexpensive garlic peels was a good example to reduce production cost of the bottle cultivation of mushroom.

Allow systematic education, support, and development of a meister system for fostering sewing manpower. 3. Undertake efforts to improve the sewing work environment. 4. Establishment of the system for realizing the actual cost of labor. 5. Establishment of a quota system to secure domestic sewing production. 6. Construction of Smart DB to connect work orders. 7. Construction of a smart factory using technology such as automated systems of production suitable for the 4th Industrial Age. 8. Enforcement of specialized strategies to encourage fashion sewing companies, not only Seoul but also in other urban areas.
Optimal culture conditions were characterized for production of crude biosurfactant of Bacillus subtilis JK-1. During incubation of B. subtilis JK-1, the bacterial growth pattern, changes of the surface tension at variable temperatures, pH and NaCl concentrations in bacterial culture medium were studied. The strain was able to grow and produce biosurfactant at $15-45^{\circ}C$, in the pH range of 6-10, and at 0-10% (w/v) NaCl. In case, culture broth pH was gradually changed to neutral or weak alkaline. Optimal culture conditions for crude biosurfactant production were at $35^{\circ}C$ and pH 7.0 after 48 h incubation and the surface tension of biosurfactant was 24.0 mN/m. Besides, as the concentration of NaCl was increased from 0 to 10% (w/v), the growth was decreased, pH of the culture broth was converted from weak alkaline to acidic, and the surface tension rised.

This study aimed to examine the influence of adding an oxygen vector, n-dodecane, on hyaluronic acid (HA) production by batch culture of Streptococcus zooepidemicus. Owing to the high viscosity of culture broth, microbial HA production during 8-16 h was limited by the oxygen transfer coefficient $K_La$, which could be enhanced by adding n-dodecane. With the addition of n-dodecane to the culture medium to a final concentration of 5% (v/v), the average value of $K_La$ during 8-16 h was increased to $36{\pm}2h^{-1}$, which was 3.6 times that of the control without n-dodecane addition. With the increased $K_La$ and dissolved oxygen (DO) by adding 5% (v/v) of n-dodecane, a 30% increase of HA production was observed compared with the control. Furthermore, the comparison of the oxygen mass transfer in the absence and presence of n-dodecane was conducted with two proposed mathematical models. The use of n-dodecane as an oxygen vector, as described in this paper, provides an efficient alternative for the optimization of other aerobic biopolymer productions, where $K_La$ is usually a limiting factor.
