Cold Shock Response of an Antarctic Streptomyces Strain Showing Demulsifying Ability

  • The hydrophobic spores of Streptomyces sp. AA8321 isolated from the Antarctic coast displayed demulsification ability. The aerial spores demulsified an emulsion of kerosene/$0.2\%$ Triton X-100 (2:1, v/v) to $50\%$ and $95\%$ within 1 min contact at the concentrations of $5.0{\times}10^7$ and $1.0{\times}10^8$ spores/ml, respectively. A cold shock protein (csp) gene was cloned from the hydrophobic spore- producing Streptomyces sp. AA8321 using PCR. It encoded a low molecular protein with 68 amino acids showing very low homology with previously reported csp genes. Only the sequence of the first six amino acids was just the same and yet others were different. RNA blot analysis indicated that the csp gene was induced by cold shock, i.e., transferring from $30^{\circ}C$ to $10^{\circ}C$, and this cold shock response proposed that the isolated gene be a new type of csp gene.

Development of University Liberal Education Curriculum for Creating Video Content -Understanding and Production of Video Content- (영상콘텐츠 제작을 위한 대학 교양 커리큘럼 개발 -영상콘텐츠의 이해와 제작-)

  • Today, the media environment has changed dramatically with the development of Internet transmission technology, computer performance, and various media production technologies. In particular, video content is widely used every day through mobile device or computer connected to the Internet. In addition, the ability to produce video content today is more important than ever, as the use of video content is also increasing in the area of information delivery and communication over the Internet. In this study, it was developed a liberal education curriculum in universities for the production of video content. The developed curriculum was composed of a combination of an online curriculum for theory and an offline curriculum for practicum. In the online curriculum, a 54 DOOR contents were developed to allow students to learn the theoretical knowledge and basic making techniques related to the production of video content. In an offline 15-week practicum curriculum, it covered the practical training contents of video content production that should be acquired every week. This developed liberal education curriculum aims to give non-major students in universities the opportunity to learn the technology of producing video content and to foster the ability to communicate in the media environment by using the video content produced.

Screening and Characterization of Thermotolerant Alcohol-producing Yeast

  • Two strains of yeast (RA-74-2 and RA-912) showing superior fermenting ability at a high temperature were isolated from soils and wastewaters by an enrichment culture method. Based on the morphological and physiological charateristics, the two strains were identified as Saccharomyces cerevisiae and Kluyveromyces marxianus, respectively. RA-74-2 was able to grow upto $43^{\circ}C$ and sustain similar fermenting ability in the temperatures range from 30 to $40^{\circ}C$. In addition, the sugar- and ethanol-tolerance of RA-74-2 were 30% (w/v) glucose and 10% (v/v) ethanol, which appeared to be higher than those of nine other industrial yeast strains currently being used in the alcohol factories. The thermotolerant ethanol fermenting yeast RA-912 showed identical growth in the temperatures range from 35 to $45^{\circ}C$ and was resistant to various heavy metals. The quality and quantity of byproducts of the isolated yeast strains in fermentation broth after fermentation at $40^{\circ}C$ and $45^{\circ}C$ were similiar with those obtained at $30^{\circ}C$. These results show that RA-74-2 can be adopted for the ethanol fermentation process where the expenses for cooling system is significant, and suggest that RA-912 may be applied in either SSF(simultaneous saccharification and fermentation) or Flash-fermentation process and RA-912 may be used as a gene donor for the development of thermotolerant ethanol-fermenting yeasts.

Math Creative Problem Solving Ability Test for Identification of the Mathematically Gifted

  • The purpose of this study was to develop math creative problem solving test in order to identify the mathematically gifted on the basis of their math creative problem solving ability and evaluate the goodness of the test in terms of its reliability and validity of measuring creativity in math problem solving on the basis of fluency in producing valid solutions. Ten open math problems were developed requiring math thinking abilities such as intuitive insight, organization of information, inductive and deductive reasoning, generalization and application, and reflective thinking. The 10 open math test items were administered to 2,029 Grade 5 students who were recommended by their teachers as candidates for gifted education programs. Fluency, the number of valid solutions, in each problem was scored by math teachers. Their responses were analyzed by BIGSTEPTS based on Rasch's 1-parameter item-response model. The item analyses revealed that the problems were good in reliability, validity, difficulty, and discrimination power even when creativity was scored with the single criteria of fluency. This also confirmed that the open problems which are less-defined, less-structured and non-entrenched were good in measuring math creativity of the candidates for math gifted education programs. In addition, it discriminated applicants for two different gifted educational institutions and between male and female students as well.

  • The universities introduce and manage education program to improve the problem-solving ability directly associated with respective professional fields as breaking from lecture-concentrated management for the acquisition of knowledge. In this paper, we design and implement a system which standardizes forms of productions induced from capstone design and manages programs integrally according to the predefined schedules. The proposed system intends the successful management of capstone design which is most representative education program for improving practical business ability. Additionally, the system gives practical assistance for applying employment and entering higher educational institution through automatic producing portfolio which comprises total processes for capstone design.

  • Recently, in the case that provides electronic services using Internet, we need the non-repudiation service that supplies a technological evidence about actions between a sender and a receiver that violate the promised protocol. Also, this service offers legal evidences while producing controversy. In this paper, we propose a protocol that improves the efficiency and offers the fairness of non-repudiation service by the extension of ability of TTP (Trusted Third Party). The proposed protocol adds a Time Check function and an Alert Message to extend the ability of TTP. Through the computer simulation, we prove that the proposed protocol has better efficiency than previous protocols.

Morphological and Physiological Properties of Interspecific Electrofusants, Bacteriocin Producer, from Lactobacillus sp. JC-7 and Lactobacillus acidophilus 88 (Lactobacillus sp. JC-7과 Lactobacillus acidophilus 88간의 Bacteriocin 생산 세포융합주들의 형태 및 생리학적 성질에 관한 연구)

  • Interspecific fusants were made from the cells of two strains of Lactobacillus genus, a streptomycin resistant Lactobacillus sp. JC-7 and a kanamycin resistant L. acidophilus 88. The morphological and physiological properties of the fusants were examined by determining bacteriocin productivity, acid-producing activity, ability of carbohydrates utilization and three important enzyme activities. The fusants produced a bacteriocin against indicator strains and fusant No. 1, 4 exhibited a larger inhibition zone compared to that of L. acidophilus 88. $\beta$-Galactosidase, phospho-$\beta$-galactosidase, lipase activities and resistance to NaCl of Lactobacillus sp. JC-7 were better than those of L. acidophilus 88. Fusant No. 3 and 7 exhibited excellent lipase activities. Protease activity and acid productivity of L. acidophilus 88 were better than those of Lactobacillus sp. JC-7. Proteasse activities of all fusants were higher than those of parental strains, and expecially fusant No. 5 and 7 exhibited excellent proteolysis ability.

Embryo sexing methods in bovine and its application in animal breed

  • The ability to determine the sex of bovine embryos before the transfer is advantageous in livestock management, especially in dairy production, where female calves are preferred in milk industry. The milk production of female and male cattle benefits both the dairy and beef industries. Pre-implantation sexing of embryos also helps with embryo transfer success. There are two approaches for sexing bovine embryos in farm animals: invasive and non-invasive. A non-invasive method of embryo sexing retains the embryo's autonomy and, as a result, is less likely to impair the embryo's ability to move and implant successfully. There are lists of non-invasive embryo sexing such as; Detection of H-Y antigens, X-linked enzymes, and sexing based on embryo cleavage and development. Since it protects the embryo's autonomy, the non-invasive procedure is considered to be the safest. Invasive methods affect an embryo's integrity and are likely to damage the embryo's chances of successful transformation. There are different types of invasive methods such as polymerase chain reaction, detection of male chromatin Y chromosome-specific DNA probes, Loop-mediated isothermal amplification (LAMP), cytological karyotyping, and immunofluorescence (FISH). The PCR approach is highly sensitive, precise, and effective as compared to invasive methods of farm animal embryonic sexing. Invasive procedures, such as cytological karyotyping, have high accuracy but are impractical in the field due to embryonic effectiveness concerns. This technology can be applicable especially in the dairy and beef industry by producing female and male animals respectively. Enhancing selection accuracy and decreasing the multiple ovulation embryo transfer costs.

  • Entrepreneurs are valuable assets for any country. Rather than being confronted with new obstacles, they should be fostered and nurtured. Entrepreneurial firms have the power to influence how we live and work, in addition to producing jobs and contributing to economic progress. Entrepreneurs have the ability to change the world by creating diversified skill sets and profitable businesses that are vital to the advancement of our economies. How does one go about being a successful business, though? More resources are believed to be needed to create entrepreneurial environments in higher education that foster progressive ideas and innovation while also providing students with the practical knowledge and skills they need to navigate the troublesome, difficult, and uncertain situations that come with owning a business. This article will outline the scales of components from which to measure variables impacting universities' ability to encourage entrepreneurial behavior among students, based on the aforementioned significance of universities. This article is based on a survey of 507 students from different universities and backgrounds in Vietnam. The research methods used are Cronbach's alpha test and Structural Equation Modeling. From the research results, it can be seen that the university's environment plays a significant role in fostering entrepreneurial behavior among students.

A Study on the Cleaning Efficiency using the d-Limonene Oil Extracted in Wasted Mandarin Peels (폐감률피에서 추출한 limonene 오일의 세정성에 관한 연구)

  • The object of this research is to conform of practicable possibility and recycling of producing junk after citrus fruits is processed. With extracting d-limonene oil that have 70~90% a component of oil out of junk citrus peel, making certain the about 12000ppm concentration of it. Limonene derived from citrus in jeju using conventional synthetic detergents can be replaced with the development of environmentally friendly natural detergent investigated the possibility. Mostly due to ocean dumping, disposal and cause environmental problems by recycling natural citrus cleaner alternative to the research conducted on the possibility. Cleaning efficiency with temperature did not affect the largest concentrations were able to identify the difference between cleaning efficiency. At least 10% of the d-limonene oil could be from the cleaning performance, increasing the concentration of the cleaning efficiency was increased in size. Ultrasonic is very high removal efficiency under the conditions shown in the cause of pure self-generated ultrasonic cleaning power as co-effects of d-limonene oil appears to chemical cleaning effect of ultrasonic cavitation occurs in the physical cleaning effect due to a combination of synergistic stability is maximized by low concentrations of d-limonene oil in a short time showed an excellent cleaning ability. Having the ability of cleaning at the same time, considering the side recycling in the junk citrus peels reflects possibility of basic materials utility eco-friendly in the skin soap, bath soap, cosmetics etc, through ability of exclusion a contaminant in based cleaning effect(EC) it can prospect substitution effect environmentally in the pre existence synthetic detergents.
