Synthesis of New Boron Derived Compounds; Anticancer, Antioxidant and Antimicrobial Effect in Vitro Glioblastoma Tumor Model

  • Objective : The aim of our study is to investigate the cytotoxic, antioxidant, and antimicrobial effects of newly synthesized boron compounds in U87MG glioblastoma cell treatment. Methods : We synthesized boron glycine monoester (BGM) and boron glycine diester (BGD) structures containing boron atoms and determined their cytotoxic activities on glioblastoma by the MTT method. The inhibitory concentration 50 (IC50) value was calculated with GraphPad Prism 5.0 program. The IC50 values were administered 48 hours on U87MG glioblastoma cell. Catalase (CAT), acid phosphatase (ACP) and alkaline phosphatase (ALP) enzyme activity, malondialdehyde (MDA), total glutathione (GSH), and total protein levels were detected using spectrophotometric methods. We determined the antimicrobial activities of BGM and BGD with the disc diffusion method. Results : After 48 hours of BGM and BGD application to U87MG glioblastoma cells, we found the IC50 value as 6.6 mM and 26 mM, respectively. CAT and ACP enzyme activities were decreased in BGM and BGD groups. MDA which is a metabolite of lipid peroxidation was increased in both boron compounds groups. GSH level was reduced especially in BGD group. BGM and BGD have been found to be antimicrobial effects. Conclusion : Boron compounds, especially the BGM, can provide a new therapeutic approach for the treatment of glioblastoma with their anticancer, antioxidant, and antimicrobial effects.

A Study on COVID-19 Symptoms according to Sasang Constitution (사상체질에 따른 COVID-19 감염증상에 대한 연구)

  • Objectives This study aims to identify differences in clinical manifestations of COVID-19 between different Sasang constitution. Methods Subjects were recruited from August 29, 2022, to July 11, 2023. COVID-19 clinical symptoms were self-reported via questionnaires. Sasang constitutional diagnosis was performed using the K-PRISM, and Sasang constitutional specialist. Results A total of 66 subjects were recruited for the study, and the Sasang constitutional distribution of the subjects was 19 soyangin, 25 taeeumin, and 22 soeumin. For most of the COVID-19 clinical manifestations, the study found that soyangin experienced symptoms at a higher rate than other constitutions. Among the symptoms observed in the study, sore throat, pantalgia, and cough were severe in all subjects regardless of constitution. Soeumin was more likely to experience abdominal pain, chest pain, and diarrhea, while soyangin was more likely to experience chest pain, nausea/vomiting, diarrhea, and cutaneous symptoms compared to other constitutions. Taeeumin had more sputm, nasal congestion, and skin symptoms, but fewer digestive symptoms. There were differences in the onset and duration of symptoms by constitution. Conclusions This study is an important contribution to our understanding of the differences in response to the COVID-19 virus among different Sasang constitutions. Symptomatic differences between constitutions may have important implications for prevention and treatment strategies for infectious diseases, and personalized treatment and management based on these differences may be needed in the future.

A Study for the Syndrome Differentiation of Retroverted Uterus Using Ultrasound (초음파 자궁형상진단을 이용한 자궁후굴증의 변증 진단 연구)

  • Objectives: We have studied the syndrome differentiation of ultrasound of retroverted uterus(RU) as the inspection in traditional Korean medicine. Methods: Eighty-four patients, who visited H clinic for infertility treatment, were recruited and evaluated by questionnaires and ultrasound, from April, 2012 to May, 2012. We used ultrasound(Logiq C3; GE Health care) for examination of RU. For statistics, we used Fisher's extract test to evaluate the RU and symptoms, and Chi-square test to evaluated the RU and syndrome differentiations, and Student T-test for the relationship between RU and each score of syndrome differentiation, using PASW Statistics(version 18.0.0) and GraphPad Prism(version 5.01). Results: 1. RU was correlated with moderate or severe dysmenorrhea and premenstrual lower abdominal pain. 2. In syndrome differentiation, blood stasis questionnaire was correlated with RU. 3. RU was related with dysmenorrhea, mass of low abdomen, pressure pain, clod of menstrual blood, dyspareunia. Conclusions: The ultrasound of RU might be related with the syndrome of blood stasis.

Knowledge and Awareness about Breast Cancer and its Early Symptoms among Medical and Non-Medical Students of Southern Punjab, Pakistan

  • Breast cancer is the leading cause of morbidity and mortality globally but has an even more significant impact in developing countries. Pakistan has the highest prevalence among Asian countries. A general lack of public awareness regarding the disease often results in late diagnosis and poor treatment outcomes. The literacy rate of the Southern Punjab (Pakistan) is low compared to its Northern part. It is therefore vital that university students and especially medical students develop a sound knowledge about the disease so that they can spread awareness to others who may be less educated. This study therefore considers current knowledge and understanding about the early signs of breast cancer amongst a study group of medical and non-medical university students of the Southern Punjab, Pakistan. A cross-sectional descriptive analysis of the university students was carried out using a self-administered questionnaire to assess their awareness of breast cancer from March to May 2014. A total of 566 students participated in this study, out of which 326 were non-medical and 240 were from a medical discipline. Statistical analysis was carried out using Graph Pad Prism Version 5 with a significance level set at p<0.05. The mean age of the non medical and medical participants was 23 (SD 2.1) and 22 (SD 1.3) years, respectively. Less than 35% students were aware of the early warning signs of the breast cancer development. Knowledge of medical students about risk factors was significantly better than the non medical ones, but on the whole was insufficient. Our study indicated that knowledge regarding breast cancer was generally insufficient amongst the majority of the university students (75% non-medical and 55% medical) of Southern Punjab, Pakistan. This study highlights the need to formulate an awareness campaign and to organize conferences to promote breast cancer awareness among students in this region.

AN EXPERIMENTAL STUDY ON ADHESION PATTERN, ADHESION STRENGTH AND FRACTURE PATTERN OF THE ADHESIVE CAST GOLD INLAY (접착형 구조 금 인레이의 접착 형태, 강도 및 파절 양태에 관한 실험적 연구)

  • Zinc Phosphate Cement hand been used for about more than 100 years in luting of cast gold inlay. But many scientists had been trying to develop the new form of luting agent because the ZPC hand shown the lack of adhesiveness on the tooth structure and the toxicity to the pulp tissue. Recently many researches about the surface treatment of the cast body are being done to increase the adhesion of cement to it. The conventional Class I gold inlays were fabricated in the 20 permanent molars. After the internal surface of the cast body was sandblasted with $Al_2O_3$ particles and was tin-plated, the inlays were cemented with adhesive cement [G I cement and resin cement(Super-Bond & $Panavia_{EX}$)] and the evaluation on the adhesion pattern, adhesive strength and the fracture pattern of the adhesive cast gold inlay was compared to that of the cast gold inlay cemented conventionally with ZPC. The results were as follows : 1. The surface roughness of the cast body was increased significantly after sandblasting with the $Al_2O_3$ particles and the tin oxide layer, which was consisted of round particles, came into being. 2. The bond strength was in the order of Super-Bond, ZPC, Fuji I, $Panavia_{EX}$ group. The group cemented with Super-Bond showed statistically greater strength than the other groups(p<0.05). 3. The group cemented with ZPC was fallen apart by principal adhesion failure and that with Fuji I was by complete adhesion failure. But the group with Super-Bond showed pricncipal cohesive failure pattern and in the group with $Panavia_{EX}$, complete cohesive fracture pattern was shown and small protion of tooth structure was fractured out with cast body and the fractured surface showed the figure just as the enamel prism. 4. Various gaps were shown at the pulpal side regardless of little gap at the side walls of the cavity in all groups. Only the Super-Bond was attached to the tooth structure and the other cements were detached from both the tooth and the cast body.

Horizontal Effects of 10-mm Inferior Oblique Recession versus 14-mm Inferior Oblique Recession

  • Purpose: The aim of this study was to investigate the quantitative effect of inferior oblique (IO) 10- and 14-mm recession on postoperative horizontal deviation. Methods: Patients (22 men and 18 women) who underwent IO recession were divided into two groups for comparison studies: group 1 (10-mm IO recession, 15 patients) and group 2 (14-mm IO recession, 25 patients). Preoperative and postoperative horizontal deviations were measured, and the resulting horizontal deviations from the 10- and 14-mm IO recession surgeries were compared. The effects of superior oblique underaction, IO overaction, and combined exodeviation on postoperative horizontal deviation were analyzed. Results: Although group 1 did not show a significant horizontal deviation change after surgery ($1.9{\pm}4.5$ prism diopters [PD], p = 0.452), group 2 had a meaningful horizontal change after 14-mm recession ($2.2{\pm}3.8PD$, p = 0.022). Both groups showed a significant esodrift in horizontal deviation (group 1, p = 0.017; group 2, p = 0.030) in patients with exodeviation over 8 PD. The mean change in horizontal deviation was $6.0{\pm}5.4PD$ for group 1 and $9.0{\pm}5.0PD$ for group 2. Although the amount of superior oblique underaction did not affect the extent of change in horizontal deviation, patients with severe IO overaction showed a significant change in horizontal deviation after 14-mm IO recession. Conclusions: Fourteen-millimeter IO recession could make a statistically significant change in horizontal deviation after surgery. In addition, esodrift should be considered after IO recession in patients with a preoperative exodeviation greater than 8 PD or severe IO overaction.

Protective effect of Capsosiphon fulvescens on oxidative stress-stimulated neurodegenerative dysfunction of PC12 cells and zebrafish larva models

  • Reactive oxygen species (ROS) at high concentrations induce oxidative stress, an imbalanced redox state that is a prevalent cause of neurodegenerative disorders. This study aimed to investigate the protective effect of Capsosiphon fulvescens (CF) extract on oxidative stress-induced impairment of cognitive function in models of neurodegenerative diseases. CF was extracted with subcritical water and several solvents and H2O2 (0.25 mM) or aluminum chloride (AlCl3; 25 µM) as an inducer of ROS was treated in PC12 neuronal cells and zebrafish larvae. All statistical analyses were performed using one-way analysis of variance and Dunnett's test using GraphPad Prism. H2O2 and AlCl3 were found to significantly induce ROS production in PC12 neuronal cells and zebrafish larvae. In addition, they strongly affected intracellular Ca2+ levels, antioxidant enzyme activity, brain-derived neurotrophic factor (BDNF) and tropomyosin receptor kinase B (TrkB) signaling, acetylcholinesterase (AChE) activity, and hallmarks of Alzheimer's disease. However, treatment of H2O2-induced PC12 cells or AlCl3-induced zebrafish larvae with CF subcritical water extract at 90℃ and CF water extract effectively regulated excessive ROS production, intracellular Ca2+ levels, and mRNA expression of superoxide dismutase, glutathione peroxide, glycogen synthase kinase-3 beta, β-amyloid, tau, AChE, BDNF, and TrkB. Our study suggested that CF extracts can be a potential source of nutraceuticals that can improve the impairment of cognitive function and synaptic plasticity by regulating ROS generation in neurodegenerative diseases.

Outcome and Prognosis in Critically III Children Receiving Continuous Renal Replacement Therapy (소아 중환자에서 지속적 신대체요법의 치료 결과와 예후)

  • Purpose : Continuous renal replacement therapy(CRRT) has been the first choice for the treatment of acute renal failure in critically ill children not only in western countries but also in Korea. However, there are very few studies that have analyzed the outcome and prognosis of this modality in Korean children. We performed this study to evaluate the factors associated with the outcome and prognosis of patients treated with CRRT. Methods : We retrospectively reviewed the medical records of 32 children who had received CRRT at Severance hospital from 2003 to 2006. The mean age was 7.5 years(range 4 days-16 years) and the mean body weight was 25.8 kg (range 3.2-63 kg). Results : Eleven(34.4%) of the 32 patients survived. Bone marrow transplantation and malignancy were the most common causes of death and underlying disease leading to the need for CRRT Mean patient weight, age, duration of CRRT, number of organ failures, urine output, estimated glomerular filtration rate(eGFR), C-reactive protein, and blood urea level did not differ significantly between survivors and nonsurvivors. (1) Pediatric risk of mortality(PRISM) III score at CRRT initiation($9.8{\pm}5.3$ vs. $26.7{\pm}7.6$, P<0.0001), (2) maximum pressor number ($2.1{\pm}1.2$ vs. $3.0{\pm}1.0$, P=0.038), and (3) the degree of fluid overload($5.2{\pm}6.0$ vs. $15.0{\pm}8.9$, P=0.002) were significantly lower in survivers than in nonsurvivors. Multivariate analysis revealed that fluid overload was the only independent factor reducing survival rate. Conclusion : CRRT was successfully applied to the treatment of acute renal failure in a wide range of critically ill children. To improve survival, we suggest the early initiation of CRRT to prevent the systemic worsening and progression of fluid overload in critically ill children with acute renal failure. Nutrient Recovery from Sludge Fermentation Effluent in Upflow Phosphate Crystallization Process (상향류 인 결정화공정을 이용한 슬러지 발효 유출수로 부터의 영양소 회수)

  • The nutrient recovery in phosphate crystallization process was investigated by using laboratory scale uptlow reactors, adopting sequencing batch type configuration. The industrial waste lime was used as potential cation source with magnesium salt($MgCl_2$) as control. The research was focused on its successful application in a novel integrated sludge treatment process, which is comprised of a high performance fermenter followed by a crystallization reactor. In the struvite precipitation test using synthetic wastewater first, which has the similar characteristics with the real fermentation effluent, the considerable nutrient removal(about 60%) in both ammonia and phosphate was observed within $0.5{\sim}1$ hr of retention time. The results also revealed that a minor amount(<5%) of ammonia stripping naturally occurred due to the alkaline(pH 9) characteristic in feed substrate. Stripping of $CO_2$ by air did not increase the struvite precipitation rate but it led to increased ammonia removal. In the second experiment using the fermentation effluent, the optimal dosage of magnesium salt for struvite precipitation was 0.86 g Mg $g^{-1}$ P, similar to the mass ratio of the struvite. The optimal dosage of waste lime was 0.3 g $L^{-1}$, resulting in 80% of $NH_4-N$ and 41% of $PO_4-P$ removal, at about 3 hrs of retention time. In the microscopic analysis, amorphous crystals were mainly observed in the settled solids with waste lime but prism-like crystals were observed with magnesium salt. Based on mass balance analysis for an integrated sludge treatment process(fermenter followed by crystallization reactor) for full-scale application(treatment capacity Q=158,880 $m^3\;d^{-1}$), nutrient recycle loading from the crystallization reactor effluent to the main liquid stream would be significantly reduced(0.13 g N and 0.19 g P per $m^3$ of wastewater, respectively). The results of the experiment reveal therefore that the reuse of waste lime, already an industrial waste, in a nutrient recovery system has various advantages such as higher economical benefits and sustainable treatment of the industrial waste.

Case of Anterior Segment Ischemia after Two Vertical Rectus Muscles Surgery (두 개의 수직근수술 후 발생한 전안부허혈 1예)

  • Purpose: To report a case of anterior segment ischemia after superior and inferior rectus muscle surgery in a patient with superior rectus muscle enlargement. This is the first report in the Republic of Korea of anterior segment ischemia after two rectus muscles surgery. Case summary: An 80-year-old male was referred to our clinic with a 3 years history of diplopia. The patient had 30 prism diopters right eye hypertropia in the primary position and downgaze limitation. The orbital computed tomography scan revealed enlargement of the right superior rectus muscle. He underwent right superior rectus recession and inferior rectus resection. On postoperative day 2, slit lamp examination revealed diffuse corneal edema, Descemet's membrane folding, an aqueous flare and a dilated pupil. Treatment with 45 mg oral steroid was initiated, and 1% prednisolone acetate and 0.5% Levofloxacin eye drops were administered. At postoperative 1 month, the patient was orthophoric in the primary position, and there was no corneal edema, pupil abnormality or aqueous flare. Conclusions: Base on the present case, the possibility of anterior segment ischemia should be considered after even two muscles surgery, and older patients with vertical muscle surgery should be considered more carefully.
