Comparative Analysis of Expressed Sequence Tags from Flammulina velutipes at Different Developmental Stages

  • Flammulina velutipes is a popular edible basidiomycete mushroom found in East Asia and is commonly known as winter mushroom. Mushroom development showing dramatic morphological changes by different environmental factors is scientifically and commercially interesting. To create a genetic database and isolate genes regulated during mushroom development, cDNA libraries were constructed from three developmental stages of mycelium, primordium, and fruit body in F. velutipes. We generated a total of 5,431 expressed sequence tags (ESTs) from randomly selected clones from the three cDNA libraries. Of these, 3,332 different unique genes (unigenes) were consistent with 2,442 (73%) singlets and 890 (27%) contigs. This corresponds to a redundancy of 39%. Using a homology search in the gene ontology database, the EST unigenes were classified into the three categories of molecular function (28%), biological process (29%), and cellular component (6%). Comparative analysis found great variations in the unigene expression pattern among the three different unigene sets generated from the cDNA libraries of mycelium, primordium, and fruit body. The 19-34% of total unigenes were unique to each unigene set and only 3% were shared among all three unigene sets. The unique and common representation in F. velutipes unigenes from the three different cDNA libraries suggests great differential gene expression profiles during the different developmental stages of F. velutipes mushroom.

Microspore-derived Embryo Formation and Morphological Changes during the Isolated Microspore Culture of Radish (Raphanus sativus L.)

  • Raphanus sativus L. cv. Taebaek, a efficiently microspore-derived embryo (MDE)-forming cultivar, and 'Chungwoon', a non-MDE-forming cultivar were selected as donor plants for isolated microspore culture. Radish flower bud of 2.0 (small, S), 4.0 (medium, M), and 6.0 (large, L) ${\pm}$ 0.5 mm in length were isolated to determine the temporal relationship between flower bud size and MED yield. Anatomical observations revealed no difference in the structure of the flower buds between the two cultivars. In both cultivars, the stigmas were much longer than the floral leaf in M-sized flower buds. The MDE yields for 'Taebaek' per petri dish were 6.6 and 1.3 for M- and L-sized of flower buds, respectively, but MDE formation was not induced in the S flower buds. On the other hand, 'Chungwoon' failed to form MDEs in all flower buds. The microspore density of 'Taebaek' was 1.3 times more than that of 'Chungwoon' for M sized flower buds. Of the M-sized buds from 'Taebaek' and 'Chungwoon', 92.1 and 81.6%, respectively, were in the late uninucleate microspore stage, which is characterized by the highest frequency of MDE formation. Anatomical observations of MDE formation revealed that the microspores were able to divide to form a primordium from which cell division took place continuously in the 'Teabeak' cultivar. However, the microspores of 'Chungwoon' failed to progress beyond the primodium stage, resulting in lack of MDE formation. By contrast, after the formation of the primordium, various developmental stages of embyos from microspore were observed in the 'Taebaek' cultivar. These results can be used to determine MDE forming potentials of radish cultivars.

  • Ultrastructural study of the development of the atrioventricular (AV) node was studied by electron microscopy in human fetus ranging from 30 mm to 260 mm crown rump length, and compared with human adult. By 30 mm fetus, the right AV nodal primordium was located below the attachment of the right venous valve. The left AV nodal primordium was observed below the attachment of septum primum. The cytoplasm of the nodal primordia contained few mitochondria, and myofibrils. These cells were apposed to each other with occasional desmosomes. In 40 mm fetus, the AV node cells were poorly organized myofibrils, while working myocardial cells were well organized myofibrils with sarcomere. At 70 mm fetus, intercalated discs were developed in the working myocardial cells. At 100 mm fetus, the nodal cells contained a relatively clear cytoplasm with a few groups of myofibrils and mitochondria. By $140\sim200$ mm fetuses, the nodal cells were an increasing number of myofibrils and mitochondria and these were scattered throughout the cytoplasm. At 260 mm fetus, the nodal cells were small and contained a clear cytoplasm with sparse and poorly organized myofibrils and mitochondria. All major ultrastructural features which characterize the adult AV nodal cells were found in this stage. The working myocardial cells were larger and had a more compact cytoarchitecture than nodal cells. Zonula adherens or fasciae adherens type junction were not found between nodal cells, but they frequently observed between nodal and working myocardial cells.

Effect of LED Light on Primordium Formation, Morphological Properties, Ergosterol Content and Antioxidant Activity of Fruit Body in Pleurotus eryngii (LED광원이 큰느타리버섯 자실체의 발생, 생육, 에르고스테롤 함량 및 항산화활성에 미치는 영향)

  • Light wavelength is the major factor of fruit body development associated with mushroom cultivation, but its wavelength range in Pleurotus eryngii is poorly understood. Using four kinds of light emitting diode (LED) including blue (475 nm), green (525 nm), yellowed (590 nm) and red (660 nm), we investigated to elucidate suitable light wavelength during primordium formation and fruit body development of P. eryngii on bottle cultivation. Primordia formation did not occur in blue light and red light. The morphological properties of fruit body in fluorescent lamp and blue light irradiation were showed thicker and larger pileus than those in other LEDs. However, length of stipe in fluorescent lamp and blue light was shorter than that of other LEDs. The DPPH radical was high in blue light, green light, and yellow light except for red light, and the polyphenol was high in four kinds of LED sources. And ergosterol was the highest in the green light. Thus, the high-quality mushroom production of P. eryngii is possible to green light condition considering productivity and functional materials.

Enhancement of ${\beta}$-Glucan Content in the Cultivation of Cauliflower Mushroom (Sparassis latifolia) by Elicitation

  • The effectiveness of three kinds of enzymes (chitinase, ${\beta}$-glucuronidase, and lysing enzyme complex), employed as elicitors to enhance the ${\beta}$-glucan content in the sawdust-based cultivation of cauliflower mushroom (Sparassis latifolia), was examined. The elicitors were applied to the cauliflower mushroom after primordium formation, by spraying the enzyme solutions at three different levels on the sawdust-based medium. Mycelial growth was fully accomplished by the treatments, but the metabolic process during the growth of fruiting bodies was affected. The application of a lysing enzyme resulted in an increase in the ${\beta}$-glucan concentration by up to 31% compared to that of the control. However, the treatment resulted in a decrease in mushroom yield, which necessitated the need to evaluate its economic efficiency. Although we still need to develop a more efficient way for using elicitors to enhance functional metabolites in mushroom cultivation, the results indicate that the elicitation technique can be applied in the cultivation of medicinal/edible mushrooms.

Embryonic Effects of Radiation on ICR Mice Depending Developmental Stages (ICR 마우스의 주요기관 형성기에 있어서 기형 발생의 시기특이성에 관한 실험적 검토)

  • The ICR pregnant mice were irradiated at 1.5Gy in every 6 hours in the period of organogenesis in order to classify the stage specificity of the embryonic effects of radiation and the stage of development differentiation of the primordium of each major organ. Intrauterine death, fetal body weight and external malformation in live fetuses were observed on day 18 of gestation. There was no statistically significant difference in the intrauterine mortality at any stage organogenesis. The fetal body weight of the mice irradiated in the intermediate stage of organogenesis showed significantly lower. There were specific highly sensitive stages in the incidences of each external malformation, that is exencephalia, open eyelid, cleft palate, anomalies of extremities and anomalies of the tail. At these stage, the primordia of the major organs are established in ICR mice.

  • The portion of Cuscuta japonica haustorium which lies external to the host tissues, the upper haustorium, was investigated at the light- and electron-microscopic levels. The haustorium lightly contacted with the host was formed by the expansion of the epidermis and cortex of the stem at the contact side, and to have a group of meristematic cells within the haustorial cortex. When such a haustorium was closely contacted with the host, the meristematic region transformed into a primordial structure of the endophyte (endophyte primordium, EP) which may penetrate into the host tissues. EP consisted of the three kinds of cell group: dividing cells at the adaxial or proximal side; large, elongate cells (idioblasts) at the middle portion,; compressed cells at the abaxial or basal side. the idioblasts were characterized by the presence of large nucleus, dense cytoplasm, several small vacuoles, and abundant cell organelles including the multilamellar structures and cytosegresomes, and thus suggested to have a high metabolic activity. The features of the EP were discussed in relation to the possibility of the penetrating into host tissues.

Turion as Dormant Structure in Spirodela polyrhiza (개구리밥 휴면구조 잠아의 구조적 특성)

  • Hydrophytes such as Spirodela polyrhiza form dormant turions to withstand cold winters. The turion is an anatomically distinct structure from which a vegetative frond arises later during germination. The turions sink to the bottom of the pond when temperatures drop and remain there throughout the winter. In the spring, they float to the surface and germinate into a new frond from the turion primordium. Unlike fronds, turions are known to possess small aerenchyma, starch grains, and relatively dense cytoplasm. These features allow the turions to survive the cold winter season at the bottom of the pond. Spirodela polyrhiza has been investigated previously to a great extent, especially in its physiological, biochemical and ecological attributes. However, a little is known about the structural features of the frond and turion during turion development. Thus, the aim of the present study was to reveal the structural characteristics of the frond and turion with regard to tissue differentiation, aerenchyma development, starch distribution, and ultrastructure, with the use of electron microscopy. A moderate degree of mesophyll tissue differentiation was found in the frond, whereas the turion did not exhibit such differentiation. Within the frond tissue, approximately $37{\sim}45%$ of the cellular volume was occupied by a large aerenchyma, but only $9{\sim}15%$ was taken up by the aerenchyma in the turion. The turion cells, especially those of the turion primordium, were derived from frond cells, and contained cytoplasm. Their cytoplasm was densely packed with plastids, mitochondria, endoplasmic reticulum, Golgi bodies, and microtubules. Plasmodesmata were also well developed within these cells. The most striking feature observed was the distribution of starch grains within the plastids of turion cells. Before the turion sank to the bottom of the pond, a considerable amount of starch accumulated in the plastid stroma. The starch grains dissolved when temperatures rose in the spring, and this promptly provided the nutrients which the primordium needed for turion germination. The turion therefore, was an appropriate dormant structure for free-floating, reduced hydrophytes like Spirodela polyhriza due to its small aerenchyma and large starch grains that aided in the purpose of sinking below the surface of the water to survive cold winters. The new fronds that arose from such turions grew rapidly in the spring, beginning the new life cycle.

Studies on Favorable Light Condition for Artificial Cultivation of Grifola frondosa (잎새버섯 재배에 적합한 광조건 연구)

  • To elucidate optimum light conditions for artificial cultivation of Grifola frondosa, the effects of light quality (blue, green, white) and light intensity (200, 500, 800, 1200 lux) on primordium formation, morphological properties and yield of fruiting bodies of G. frondosa using bag cultivation method were tested. Among three light sources, white light source ($400{\sim}620\;nm$) had a higher mushroom yield (242 g/bag) and a shorter cultivation period (52 days) than those of the others. In particular, blue light source ($400{\sim}560\;nm$) induced the morphology of wide and deep colored pilei in G. frondosa fruiting body. The experimental results on the appropriate light intensity indicated that 500 lux light was the most effective on mushroom production, whereas primordium formation was effective at 200 lux.

Growth characters and harvest time for the artificial cultivation of Mycoleptodonoides aitchisonii (침버섯 인공재배 생육 특성과 수확 최적 시기)

  • Mycoleptodonoides aitchisonii has been used a culinary material and traditional medicine for a long time in worldwide and recently the researches to find biological effects have been increased such as dopamine activation, preventive effect against phytopathogens, inhibitive effect against erythrocyte coagulation, anti-oxidative effect, anti-tumorigenic effect, etc. However it is hard to cultivate Mycoleptodonoides aitchisonii it is impossible to be mass-produced and in order to solve the problem in this study we found the appropriate cultivation period and the harvest point for it. For life cycle (from primordium formation to harvest) the morphology, weight, and quantity of polysaccharide of Mycoleptodonoides aitchisonii were measured using with 5 bodies per a day and it could be divided for 4 stages; primordium formation, growth, needle maturation, and aging. And then from the results the Zeide nonlinear growth curve could be gotten. At 13th day after cultivation there is the relation between the change of media weight and body weight and at 14th day after cultivation the rate of polysaccharide in the body was 11 %. However in the case of O2 insufficient supply the malformation of them was observed.
