Effects of CO2 and Climate on water use efficiency and their linkage with the climate change

  • Gross Primary production (GPP) and evapotranspiration (ET) are the two critical components of carbon and water cycle respectively, linking the terrestrial surface and ecosystem with the atmosphere. The ratio between GPP to ET is called ecosystem water use efficiency (EWUE) and its quantification at the forest site helps to understand the impact of climate change due to large scale anthropogenic activities such as deforestation and irrigation. This study was conducted at the FLUXNET forest site CN-Qia (2003-2005) using Community land model (CLM 5.0). We simulated carbon and water fluxes including GPP, ecosystem respiration (ER), and ET using climatic variables as forcing dataset for 30 years (1981-2010). Model results were validated with the FLUXNET tower observations. The correlation showed better performance with values of 0.65, 0.77, and 0.63 for GPP, ER, and ET, respectively. The model underestimated the results with minimum bias of -0.04, -1.67, and -0.40 for GPP, ER, and ET, respectively. Effect of climate 'CLIM' and '$CO_2$' were analyzed based on EWUE and its trend was evaluated in the study period. The positive trend of EWUE was observed in the whole period from 1981-2010, and the trend showed further increase when simulated with rising $CO_2$. The time period were divided into two parts, from 1981-2000 and from 2001 to 2010, to identify the warming effect on EWUE. The first period showed the similar increasing trend of EWUE, but the second period showed slightly decreasing trend. This might be associated with the increase in ET in the wet temperate forest site due to increase in climate warming. Water use efficiency defined by transpiration (TR) (TWUE), and inherent-TR based WUE (IT-WUE) were also discussed. This research provides the evidence to climate warming and emphasized the importance of long term planning for management of water resources and evaporative demand in irrigation, deforestation and other anthropogenic activities.

  • Microalgae are primary producers of aquatic ecosystems, securing biodiversity and health of the ecosystem and contributing to reducing the impact of climate change through carbon dioxide fixation. Also, they are useful biomass that can be used as biological resources for producing valuable industrial products. However, harvesting process, which is the separation of microalgal biomass from mixed liquor, is an important bottleneck in use of valorization of microalgae as a bioresource accounting for 20 to 30% of the total production cost. This study investigates the applicability of sewage sludge-derived extracellular polymeric substance (EPS) as bioflucculant for harvesting microalgae. We compared the flocculation characteristics of microalgae using EPSs extracted from sewage sludge by three methods. The flocculation efficiency of microalgae is closely related to the carbohydrate and protein concentrations of EPS. Heat-extracted EPS contains the highest carbohydrate and protein concentrations and can be a best-suited bioflocculant for microalgae recovery with 87.2% flocculation efficiency. Injection of bioflocculant improved the flocculation efficiency of all three different algal strains, Chlorella Vulgaris, Chlamydomonas Asymmetrica, Scenedesmus sp., however the improvement was more significant when it was used for flocculation of Chlamydomonas Asymmetrica with flagella.

A Study on the Optimum Operating Condition of Acid Fermenter for the BNR Performance Improvement (BNR 효율개선을 위한 산 발효조 최적운전 조건에 관한 연구)

  • The most problems of domestic sewage for BNR(Biological Nutrients Removal) process are deficiency of carbon source and low C/N ratio. Primary sludge fermentation is seemed to be one of the best solutions producing biodegradable organic substrates. Soluble organic materials from sludge fermentation are mainly SCFAs(Short-Chain Fatty Acids) with 2~5 carbon atoms. In this research, it was attempted to apply $A_2/O$ process with the side-stream acid fermenter to improve the nutrients removal efficiency. The result showed that proper SCFAs production is about 3.000mg/L with SRT of 4~5 days. SCFAs yield of approximately 0.10~0.16 mg SCFAs(as COD) per mg of primary sludge(as COD) were achieved. The ratio of acetic. propionic. butyric and valerie acid were 1, 0.7, 0.5 and 0.6. Significant improvements of nutrients removal over 70% in BNR process were observed. thus will reduce the demand for chemical dosing to increase nutrients removal efficiency. When the fermentate was entered $A_2/O$ process, the ratio of phosphate release to substrate uptake amounts to $0.34gPO_4-Pg^{-1}COD$.

A Study on Characteristics of Solubilization and Biogas Production for Sewage Sludge using Thermal Pretreatment (열적가용화공정을 이용한 하수슬러지의 가용화 특성 및 바이오가스 생산량 평가)

  • In this study, an applicability of thermal pretreatment for primary and secondary sludge, which are generated in a sewage treatment plant, was evaluated. The efficiency and charateristics was investigated with each sludge after pretreatment under the condition of $100{\sim}220^{\circ}C$ for 30 minutes. As the result, it was found that $SCOD_{Cr}$, $NH_4{^+}$, VFAs concentrations increased as the pre-treatment temperature increased. For COD solubilization, it was also highly dependent on an increase of temperature resulting in acceleration on hydrolysis and acid fermentation. In the BMP (Biochemical Methane Potential) experiment, for the primary sludge, it showed the higher biogas production rate at a temperature of $220^{\circ}C$, however, the effect was insignificant (5.6%). Whereas, for the secondary sludge, the increase on biogass production rate was 38.8% ($180^{\circ}C$) and this means that the secondary sludge is more suitable for an applicability of thermal pretreatment.

Evaluation of carbon flux in vegetative bay based on ecosystem production and CO2 exchange driven by coastal autotrophs

  • Studies on carbon flux in the oceans have been highlighted in recent years due to increasing awareness about climate change, but the coastal ecosystem remains one of the unexplored fields in this regard. In this study, the dynamics of carbon flux in a vegetative coastal ecosystem were examined by an evaluation of net and gross ecosystem production (NEP and GEP) and $CO_2$ exchange rates (net ecosystem exchange, NEE). To estimate NEP and GEP, community production and respiration were measured along different habitat types (eelgrass and macroalgal beds, shallow and deep sedimentary, and deep rocky shore) at Gwangyang Bay, Korea from 20 June to 20 July 2007. Vegetative areas showed significantly higher ecosystem production than the other habitat types. Specifically, eelgrass beds had the highest daily GEP ($6.97{\pm}0.02g\;C\;m^{-2}\;d^{-1}$), with a large amount of biomass and high productivity of eelgrass, whereas the outer macroalgal vegetation had the lowest GEP ($0.97{\pm}0.04g\;C\;m^{-2}\;d^{-1}$). In addition, macroalgal vegetation showed the highest daily NEP ($3.31{\pm}0.45g\;C\;m^{-2}\;d^{-1}$) due to its highest P : R ratio (2.33). Furthermore, the eelgrass beds acted as a $CO_2$ sink through the air-seawater interface according to NEE data, with a carbon sink rate of $0.63mg\;C\;m^{-2}\;d^{-1}$. Overall, ecosystem production was found to be extremely high in the vegetated systems (eelgrass and macroalgal beds), which occupy a relatively small area compared to the unvegetated systems according to our conceptual diagram of a carbon-flux box model. These results indicate that the vegetative ecosystems showed significantly high capturing efficiency of inorganic carbon through coastal primary production.

Construction of a High-Quality Yeast Two-Hybrid Library and Its Application in Identification of Interacting Proteins with Brn1 in Curvularia lunata

  • Curvularia lunata is an important maize foliar fungal pathogen that distributes widely in maize growing area in China, and several key pathogenic factors have been isolated. An yeast two-hybrid (Y2H) library is a very useful platform to further unravel novel pathogenic factors in C. lunata. To construct a high-quality full length-expression cDNA library from the C. lunata for application to pathogenesis-related protein-protein interaction screening, total RNA was extracted. The SMART (Switching Mechanism At 5' end of the RNA Transcript) technique was used for cDNA synthesis. Double-stranded cDNA was ligated into the pGADT7-Rec vector with Herring Testes Carrier DNA using homologous recombination method. The ligation mixture was transformed into competent yeast AH109 cells to construct the primary cDNA library. Eventually, a high qualitative library was successfully established according to an evaluation on quality. The transformation efficiency was about $6.39{\times}10^5$ transformants/$3{\mu}g$ pGADT7-Rec. The titer of the primary cDNA library was $2.5{\times}10^8cfu/mL$. The numbers for the cDNA library was $2.46{\times}10^5$. Randomly picked clones show that the recombination rate was 88.24%. Gel electrophoresis results indicated that the fragments ranged from 0.4 kb to 3.0 kb. Melanin synthesis protein Brn1 (1,3,8-hydroxynaphthalene reductase) was used as a "bait" to test the sufficiency of the Y2H library. As a result, a cDNA clone encoding VelB protein that was known to be involved in the regulation of diverse cellular processes, including control of secondary metabolism containing melanin and toxin production in many filamentous fungi was identified. Further study on the exact role of the VelB gene is underway.

  • Crud is a corrosion deposit that forms in equipments and piping of nuclear reactor's primary systems. When crud circulates through the reactor's primary system coolant and adheres to the surface of the nuclear fuel cladding tube, it can lead to the Axial Offset Anomaly (AOA) phenomenon. This occurrence is known to potentially reduce the output of a nuclear power plant or to necessitate an early shutdown. Consequently, worldwide nuclear power plants have employed ultrasonic cleaning methods since 2000 to mitigate crud deposition, ensuring stable operation and economic efficiency. This paper details the system configuration of ultrasonic nuclear fuel cleaning equipment, outlining the function of each component. The objective is to contribute to the local domestic production of ultrasonic nuclear fuel cleaning equipment. Additionally, the paper introduces a method for accurately measuring the weight of removed crud, a crucial factor in assessing cleaning effectiveness and providing input data for the BOA code used in core safety evaluations. Accurate measurement of highly radioactive filters containing crud is essential, and weighing them underwater is a common practice. However, the buoyancy effect during underwater weighing may lead to an overestimation of the collected crud's weight. To address this issue, the paper proposes a formula correcting for buoyancy errors, enhancing measurement accuracy. This improved weight measurement method, accounting for buoyancy effects in water, is expected to facilitate the quantitative assessment of filter weights generated during chemical decontamination and system operations in nuclear power plants.

  • Objectives : The purpose of this study is to review and summarize about occupational medicine and to applicate the occupational medicine to oriental medicine. Methods : We investigated several books, articles and related internet sites to know occupational medicine's present conditions in Korea and other countries, and researched the plan on oriental medical participation in the industrial health. Results and Conclusion : The conclusions are as follows, 1. Recent years, industrialization has changed working environment and health condition in workplace, so industrial health service has became important part. 2. Occupational medicine has the purpose of protection. prevention, fitness, health management, medical treatment. rehabilitation, and primary medical service. The range of occupational medicine is enlarged mental, social good health and social, economical production capacity recently. 3. After research each country's occupational medicine, we can know the purpose is same, but the system and work contents are different one another. 4. Oriental medical participation in the industrial health has various benefits, satisfaction of workers, efficiency of cost, and increasing rate of medical treatment through the western and oriental medical cooperation.

Performance Enhancement of Gas-Liquid Mixed Plasma Discharge System using High Speed Agitation (고속 교반을 이용한 기-액 혼합 플라즈마방전 시스템의 성능 향상)

  • Dielectric Barrier Discharge (DBD) plasma is a new technique for use in environmental pollutant degradation, which is characterized by the production of hydroxyl radicals as the primary degradation species. Due to the short lifetime of the chemically active species generated during the plasma reaction, the dissolution of the plasma gas has a significant effect on the reaction performance. The plasma reaction performance can be enhanced by combining the basic plasma reactor with a homogenizer system in which the bubbles are destroyed and turned into micro-bubbles. For this purpose, the improvement of the dissolution of plasma gas was evaluated by measuring the RNO (N-dimethyl-4-nitrosoaniline, an indicator of the generation of OH radicals). Experiments were conducted to evaluate the effects of the diameter, rotation speed, and height of the homogenizer, pore size, and number of the diffuser and the applied voltage on the plasma reaction. The results showed that the RNO removal efficiency of the plasma reactor combined with a homogenizer is two times higher than that of the conventional one. The optimum rotor size and rotation speed of the homogenizer were 15.1 mm, and 19,700 rpm, respectively. Except for the lowest pore size distribution of $10-16{\mu}m$, the pore size of the diffuser showed little effect on RNO removal.

  • Fuel consumption in fisheries is a primary concern due to environmental effects and costs to fishermen. Much research has been carried out to reduce the fuel consumption related to fishing operations. The fuel consumption of fishing gear during fishing operation is generally related to hydrodynamic resistance on the gear. This research demonstrates a new approach using numerical methods to reduce fuel consumption. The results from the simulation were verified with results that mirrored the model experiments. By designing the fishing gear using drawing software, the whole and partial resistance force on the gear can be calculated as a result of simulations. The simulation results will suggest suitable materials or gear structure for reducing the hydrodynamic forces on the gear while maintaining the performance of the gear. Furthermore, the efficiency of low energy used trawl as economic point of view will be dealt. This research will helpful to reduce the GHG emissions from fishing operations and lead to reduce fishing costs due to fuel savings.
