탁주료중의 단백질분해효소에 관한 연구 (Studies on the proteinase in Takjoo mashes during the process of brewing)

  • The mash of Takjoo, Korean flour wine, is fermented through two brewing processes ; the primary brewing process to saccharify and the main one to produce ethyl alcohol. The activities of acid proteinase (pH3), weak acid proteinase (pH 6), and alkaline proteinase (Ph 80 on the processes are determined with time by the Folin phenol method as a strength of casein digestion. Hydrogen ion concentration, the content of total organic acids, protein, free amino acids and oligopeptides, which effect the activities of proteinase, are also measured. The results are briefly summarized as follows : 1. In general, the activities of acid proteinase and weak acid proteinase in the mesh of primary brewing process are stronger than those in main brewing process. 2. The activities of acid proteinase are remarkably stronger than those of weak acid proteinase in both processes. It reveals that they decrease slowly through the fermentation. Activities of alkaline proteinase are weaker than others. 3. As the raw materials are mixtured, the total amount of organic acids is equivalent to 0.150 mg/ml acetic acid in the mesh of primary brewing process and 0.02 mg/ml acetic acid in the main one. They increase gradually with time. 4. Hydrogen ion concnetration shows 3.9 in the mesh of main brewing process and 3.28 in the primary one. They increase to the maximum in 60-72 hrs., and decrease since 108 hrs. 5. The content of crude protein shows 66.90mg/ml in the mesh of main brewing process, while shows 64.29mg/ml in the mesh of primary one. they decrease slowly with time. it seems that a small content of crude protein, as a substrate, converts into amino acids and soluble nitrogen compounds by proteinase. 6. The content of free amino acids and oligopeptides shows 0.36 mg/ml in the mesh of primary brewing process and 0.24mg/ml in the main brewing process. It is evident that the reason they increase continuously through the fermentation is the effect of proteinase. 7. According to the results, the strong activities of proteinase in primary brewing process has been derived from the decrease of hydrogen ion concentration due to the production of organic acids.

에탄올에 의하여 유도된 마우스 췌장 선포세포의 염증성 손상에서 췌장분비 효소의 활성 및 세포 재생관련 유전자들의 발현에 미치는 EGCG의 영향 (Effects of (-)-Epigallocatechin-3-gallate on the Release of Pancreatic Enzymes and Expression of Regenerating Genes in Ethanol-injured Murine Pancreatic Primary Acinar Cells)

  • 본 연구에서는 마우스 췌장의 선포세포(primary acinar cell)에서 에탄올에 의해 유도된 염증성 손상에 미치는 녹차의 생리활성 물질인 EGCG의 효과를 췌장분비 효소의 활성과 선포세포에서의 기능 회복 관련 마크로 알려진 유전자들(AMPK, RKIP 및 Reg1)의 발현 조절 측면에서 조사하였다. 대조군에 비해 에탄올이 처리된 세포에서는 ${\alpha}$-amylase와 chymotrypsin의 활성이 증가되었지만, EGCG 처리에 의하여 그들 활성이 유의적으로 감소하였다. 세포 생존 및 보호와 관련 있는 AMPK 인산화 수준은 에탄올 단독 처리시 그 발현이 감소하였지만, EGCG 처리에 의하여 유의적으로 회복되었다. 세포 사멸과 세포독성 조절과 연관이 있는 RKIP 또한 그 발현 경향이 AMPK인산화 변화의 정도와 유사하였다. 또한 베타세포 재생에 관여하는 Reg1 발현은 에탄올이 처리된 선포세포에 EGCG를 처리하였을 경우 그 발현량이 유의적으로 증가하였다. 이상의 결과들을 종합하여, 항산화성 폴리페놀인 EGCG는 알코올성 췌장염으로 인한 세포의 손상이나 당뇨병 예방과 회복에 치료적 효과를 가질 수 있다고 제안한다.

조기개입 프로그램에 참여한 문제음주자의 경험 (Experiences of Problem drinkers Participation in Early Intervention Program for Moderate Alcohol Drinking)

  • 연구목적: 이 연구는 문제음주자 관점에서 조기개입 프로그램의 효과를 조명함으로써 탐색적 이해를 보여주는데 있다. 연구방법: 이 연구는 질적 기술 연구 설계가 사용되었다. 프로그램에 참여한 13명의 문제음주자와 개별 면담을 통해 연구자료를 수집하였으며, 질적 내용 분석으로 분석하였다. 연구결과: 조기개입 프로그램에 참여한 문제음주자는 알코올 정보, 알코올의 효과에 대한 기대, 자기효능감과 행동 변화 측면에서 4개의 주제 및 9개의 하위주제가 도출되었다. 이들은 직장 내에서 운영된 조기개입 프로그램에 참여하였기 때문에 개인별로 동기 수준에 차이가 있었으나, 프로그램의 필요성은 대체로 수용하였다. 프로그램 진행자를 신뢰하는 참여자일수록 음주행동 변화를 긍정적으로 생각하였다. 결론: 조기개입 프로그램에 참여한 문제음주자는 충분한 정보를 습득함으로써 음주 폐해에 대한 인식과 변화 동기의 증진 및 행위기술인 자기효능감의 획득을 통하여 문제음주 행동 변화에 유용한 접근임을 보여주었다. 이러한 결과는 직장 등 다양한 일차건강관리 현장에서 알코올중독 예방을 위한 조기개입을 적용하기 위한 기초자료로 활용될 수 있을 것이다.

펙틴의 정제 및 분석 (Purification and Analysis of Pectins)

  • Pectins present in the primary cell walls and middle lamellae of plant cell walls are extracted by water, cheating agents, acid or alkali solutions. However, some neutral contaminating components are extracted in conjunction with pectins during the extraction process. Thus, the accurate characterization of physi-cochemical properties of pectins necessitates to get rid of the impurities. In this review, dialysis, alcohol precipitation, ion exchange chromatography and metal precipitation were compared as procedures to purify the pectin extracts. In addition, the chemical methods to analyze pectins are discussed in terms of three major chemical constituents, i.e., anhydrogalacturonic acid, methoxyl groups and neutral sugars.

Synthesis and Photopolymerization of Vinyl Ether and Epoxy-Functionalized Silicones

  • The reactive precursors, vinyl ethers, and epoxy-silicones, were synthesized. The vinyl ether monomers were prepared from primary alcohol and ethyl vinyl ether with mercury (II) acetate. The epoxy-functionalized silicones have been achieved by the controlled, rhodium-catalyzed, chemoselective hydrosilation of vinyl ether with siloxanes or silane. It was shown that the hydrosilation proceeds exclusively at the vinyl ether group of alkenyl vinyl ether without participation at the alkenyl group. The photoinduced cationic polymerization of these monomers was studied and found to be all highly reactive.

  • The acetone-butanol fermentation by C. acetobutylicum has gained increasing attention for the following reasons. First, the finite supply of petrochemical resources, combined with increasing concern over global environmental effects and the unstable nature of the price of petroleum has renewed interest in the development of fermentation technology that allows utilzation of biomass wastes for the production of alcohol. Second, it serves as excellent model system for understading the regulation and molecular biology of tightly regulated complex primary metabolism, and for applications of metabolic engineering. In this review various aspects of acetone-butanol fermentation by C. acetobutylicm including strain and fermentation characteristics, enzyme regulation, and solvent formation mechanism, and product recovery and summarized.

한국인의 평생건강관리 프로그램 (Lifetime Health Maintenance Program for Korean)

  • Lifetime Health Maintenance Program(LHMP) for Koreans is the Korean guide to clinical preventive services which include screening for chronic diseases, counseling and immunization. The Korean Task Force on the LHMP vigorously reviews evidence for interventions to prevent over 50 different illnesses and conditions which are important in Korea. The problems addressed in this program are common ones seen every day by primary care physicians in Korea: cardiovascular, infectious diseases, gastrointestinal and endocrine diseases, cancers, alcohol and smoking, and many others. Primary care clinicians have a key role in screening for many of these problems and immunizing against others. Of equal importance, however, is the clinician's role in counseling patients to change unhealthful behaviors related to diet, smoking, exercise, injuries, and sexually transmitted diseases. The recommendations are grouped by age, sex, and other risk factors.

Lifestyle Components and Primary Breast Cancer Prevention

  • Breast cancer primary prevention is a high research priority due to the high psychological and economic costs. The disease is a multistep process and several risk factors have been recognized. Over the past three decades numerous studies have investigated the association of lifestyle with breast cancer, showing independent effects of various factors. We report here a summary of the present state of knowledge on the role of lifestyle patterns, such as physical activity, diet, smoking, hormone therapy, and experience of psychological stress in the modulation of breast cancer in women, and discuss commonly accepted biological mechanisms hypothesized as responsible for the associations. The findings indicate that regular physical activity of moderate to vigorous intensity is probably linked with the decreased breast cancer risk among postmenopausal females and suggestive for a decrease of the risk in premenopausal women. In contrast, the consumption of high-fat diet, alcohol intake, and use of combined estrogen and synthetic progestagen hormonal therapy may increase the risk. Epidemiological findings dealing with a role of smoking and experience of psychological stress are conflicting.

TEMPO와 NaBr 의 농도 및 온도가 옥수수 전분의 선택적 산화반응에 미치는 영향 (The Effects of TEMPO, NaBr and Temperature on the Selective Oxidation of Primary Alcohol Groups in Corn Starch)

  • 이 연구에서는 TEMPO와 NaBr의 농도 및 온도가 옥수수 전분 내 1차 알코올기의 선택적 산화 반응에 미치는 영향을 조사하였다. TEMPO와 NaBr 및 온도의 수준이 증가함에 따라 반응 시간은 감소되는 경향을 나타내었다. TEMPO, NaBr 및 온도의 수준이 각각 1.7 mM/100 mM AGU, 100 mM/100 mM AGU 및 $14^{\circ}C$까지는 이들의 수준이 증가함에 따라 반응 시간이 급격히 단축되었으나, 이 이후의 수준부터는 단축정도가 적었다. 수율은 NaBr의 수준이 증가함에 따라 감소하였고, TEMPO와 온도는 수율에 큰 영향을 주지 않는 것으로 나타났다. 선택성은 NaBr의 농도와 온도가 각각 100 mM/100 mM AGU와 $6^{\circ}C$까지는 유의적인 차이를 보이지 않았으며, 이 이후의 수준부터 NaBr 및 온도가 증가함에 따라 유의적으로 감소하는 것으로 나타났다.
