해조류 파래로부터 지질 추출에 미치는 전처리 방법의 영향 (Effect of Pretreatment Method on Lipid Extraction from Enteromorpha intestinalis)

  • In this study, we investigate the effect of pretreatment method on lipid extraction from Enteromorpha intestinalis using physical, thermo-chemical, and enzymatic process such as ultrasonication, high temperature treatment, freezing, microwave irradiation, osmotic shock, pH shock, homogenizing, and enzymatic treatment. In pretreatment with separated lipid extraction, the high extraction yield was obtained by high temperature treatment ($121^{\circ}C$ for 5 min) with 0.1 N HCl, which is 1.4 times higher than that of control. In pretreatment with direct lipid extraction, the high extraction yields were obtained by 0.1 N HCl pretreatment, microwave irradiation (700W, 1 min with twice), and 10% NaCl pretreatment, which is 1.45 times higher than that of control. In the result of enzymatic pretreatment with 17 kinds of enzymes, Cellic CTec II showed the high extraction yield of 5.3%, and which is 1.9 times higher than that of control. Moreover, the extraction yield was increased by the increase of enzyme amounts. In 10% enzyme amount, about 5.8% yield was obtained.

Bleaching of Hardwood Kraft Pulp by Xylanase Pretreatment

  • This study was carried out to investigate the effect of xylanase pretreatment of the unbleached hardwood kraft pulp during the conventional Chlorine-Extraction- Hypochlorite (CEH) bleaching on pulp property. Optimum bleaching condition was evaluated by using Novozym produced from the fungus Humicola insolens. Also the effect of chelating agent prior to enzyme treatment was analyzed. The kappa number of enzymatic bleached pulp at the enzyme charge 10 IU/ml was slightly similar to that of bleached pulp without enzyme. By enzyme treatment, the chlorine charge in conventional CEH bleaching process of hardwood KP could be reduced by 17%, while no adverse effect on pulp yield and strength was. The optimum condition for enzyme pretreatment was 10 IU/ml xylanase charge, 3 to 4 hrs treatment, and 2% pulp consistency. In sugar composition in the enzyme pretreated pulp, arabinose and mannose were not much different, but more xylose was retained. This high content of hemicellulose in pulp seems to play an important role in pulp properties. The pulp pretreatment by chelating agent prior to enzyme treatment could improve the enzyme activity and enhance the bleaching effect at 0.2% diethylenetriamine pentaacetic acid (DTPA) charges.

Effect of Naringin Pretreatment on Bioavailability of Verapamil in Rabbits

  • The aim of present study is to investigate the effect of naringin on the pharmacokinetics of verapamil and its major metabolite, norverapamil in rabbits. The pharmacokinetic parameters of verapamil and norverapamil were determined after administering verapamil (9 mg/kg) orally to rabbits in the pretreated with naringin (1.5, 7.5, and 15 mg/kg). Naringin pretreatment significantly altered the pharmacokinetic parameters of verapamil. Compared with the control group (given verapamil alone), the $K_a,\;C_{max}$ and AUC of verapamil were significantly (p<0.05 or p<0.01) increased in the pretreatment of naringin, However there were no significant change in $T_{max}\;and\;t_{1/2}$ of verapamil. Consequently, pretreatment of naringin significantly (p<0.05, p<0.01) increased the AB% of verapamil significantly in a dose dependent manner (p<0.05 or p<0.01 ), and elevated the RB% of verapamil by 1.26- to 1.69-fold. the MR of verapamil were significantly (p<0.05) increased in the pretreatment of naringin, implying that pretreatment of naringin may effectively inhibit the CYP3A4-mediated metabolism of verapamil. In conclusion, pretreatment of naringin enhanced the oral bioavailability of verapamil. Based on these results, the verapamil dosage should be adjusted when given with naringin or a naringin-containing dietary supplement.

흰쥐에 있어서 주정중독이 Toluene 대사에 미치는 영향 (Effect of Ethanol Pretreatment on the Toluene Metabolism in Toluene-treated Rats)

  • To evaluate an effect of ethanol pretreatment on the toluene metabolism, toluene (50% in olive oil) was given three times at 0.2 ml/100g body weight at the interval of one day to the rats fed with 5% ethanol during two months. The ethanol pretreated rats were not identified particular liver injury by the histopathologic findings. In case of toluene treatment, the ethanol pretreatment to the rats led to more increased concentration of urinary hippuric acid than those treated with only toluene. The ethanol pretreatment to the rats led to the increased activities of hepatic aniline hydroxylase and these enzyme activities were higher both in toluene treated and those pretreated with ethanol, but no differences were found in two groups. Ethanol pretreated rats showed the more increased activities of benzylalcohol dehydrogenase than control group. Moreover, the ethanol pretreatment to the toluene treated rats led to significantly more increased activities of benzylalcohol dehydrogenase compared with those treated with toluene only. Furthermore, the alcohol pretreatment to the toluene treated rats also led to somewhat higher activities of benzaldehyde dehydrogenase than those treated with toluene. In conclusion, these results indicate that the chronic pretreatment of ethanol at not so much liver damage as normal may rather activate the toluene metabolism.

흰쥐 대동맥에서 fluoxetine의 혈관 이완 효과 (Vasorelaxant effect of fluoxetine in isolated rat aorta)

  • The vasorelaxant effect of serotonin reuptake inhibitor fluoxetine was investigated in rat isolated thoracic aorta. Fluoxetine induced a concentration-dependent relaxation in aorta precontracted with phenylephrine (PE) and KCl. These relaxations were suppressed by removal of the endothelium (-E) or pretreatment of nitric oxide synthase inhibitors, N(G)-nitro-L-arginine (L-NNA) and N(omega)-nitro-Larginine methyl ester (L-NAME), guanylate cyclase inhibitors, methylene blue (MB) and 1H-[1,2,4]oxadiazolo [4,3-a]quinoxalin-1-one (ODQ), and $Ca^{2+}$ channel blockers, nifedipine and verapamil, in PE-precontracted +E rings. However, fluoxetine-induced relaxations were not suppressed by pretreatment of $K^{+}$ channel blockers, tetrabutylammonium and glibenclamide, in PE-precontracted endothelium intact (+E) rings. The fluoxetine-induced relaxations were not suppressed by removal of the endothelium or pretreatment of LNNA and MB in KCl-precontracted +E rings. Also, fluoxetine inhibited PE-induced sustained contraction in +E rings. These inhibitory effects of fluoxetine on contractions could be reversed by removal of the endothelium or pretreatment of L-NNA, L-NAME, MB, ODQ, nifedipine and verapamil, but not by pretreatment of etrabutylammonium and glibenclamide. These findings suggest that the vasorelaxant effect of fluoxetine is modulated by intracellular $Ca^{2+}$ with an involvement of endothelial NO-cGMP pathway and also may be related to the inhibition of $Ca^{2+}$ entry through voltage-gated channel.

방사선 피폭 마우스에서 홍삼, 백삼 및 diethyldithoicarbamate의 효과 (Effect of Red Ginseng, White Ginseng and Diethyldithoicarbamate in Irradiated Mice)

  • Studies were performed to determine the effect of red ginseng and white ginseng on jejunal crypt survival, endogenous spleen colony formation, and apoptosis of jejunal crypt cells in irradiated mice. The radioprotective effect of ginseng was compared with the effect of diethyldithocarbamate(D). Jejunal crypts were protected from irradiation by pretreatment of red ginseng (50 mg/kg B.W., I.P. at 36 and 12 hours before irradiation) and white ginseng (50 mg/kg B.W., I.P. at 36 and 12 hours before irradiation). Red ginseng administration before irradiation and both pretreatment and posttreatment (50 mg/kg B.W., I.P. at 30 minutes after irradiation) of white ginseng resulted in an increase of the formation of endogenous spleen colony. the frequency of radiation-induced apoptosis in intestinal crypt cells was also reduced by both pretreatment and posttreatment of red ginseng, and pretreatment of white ginseng. The radioprotective effect of DDC (1000 mg/kg B.W., I.P. at 30 minutes before irradiation) on jejunal crypt survival and apoptosis was similar to those of ginseng treatment. Treatment with DDC showed no significant modifying effects on formation of endogenous spleen colony. These results indicated that ginseng might be a useful radioprotector. Further studies are needed to characterize effective radioprotective components and mechanism of ginseng.

Biofilter pretreatment for the control of microfiltration membrane fouling

  • A pilot scale biofilter pretreatment-microfiltration system (BF-MF) was operated to investigate the effect of biofilter treatment in fouling reduction of microfiltration. Biofiltration was expected to reduce the membrane fouling by removal of turbidity and metal oxides. The hollow-fiber MF module with a nominal pore size of 0.1$\mu$m and a surface area of 8m$^2$ was submerged in a filtration tank and microfiltration was operated at a constant flux of 0.5 m/d. Biofiltration using polypropylene pellets was performed at a high filtration velocity of 320 m/d. Two experimental setups composed of MF and BF/MF, i.e., without and with biofilter pretreatment, were compared. Throughout the experimental period of 9 months, biofilter pretreatment was effective to reduce the membrane fouling, which was proved by the result of time variations of trans-membrane pressure and backwash conditions. The turbidity removal rate by biofiltration varied between 40% to 80% due to the periodic washing for biofilter contactor and raw water turbidity. In addition to turbidity, metals, especially Mn, Fe and Al were removed effectively with average removal rates of 89.2%, 67.8% and 64.9%, respectively. Further analysis of foulants on the used membranes revealed that turbidity and metal removal by biofiltration was the major effect of biofiltration pretreatment against microfiltration fouling.

Pro-Oxidantive Effect of Dehydroepiandrosterone on Indomethacin-Induced Acute Gastritis in Rats

  • This study examined whether or not a pretreatment with dehydroepiandrosterone (DHEA) has an effect on indomethacin-induced gastric mucosal damage. The DHEA group, male Sprague-Dawley rats, was administrated with DHEA orally at a dose of 4 mg/day for one week before inducing gastritis with indomethacin (50 mg/kg, p.o.). Histological assay, lipid peroxidation assay, superoxide dismutase (SOD), glutathione peroxidase (GPx) and Catalase activities were determined. Interestingly, it was found that the DHEA pretreatment attenuated the gastric lesion area induced by indomethacin. Rather, the pretreatment with high dose of DHEA led to submucosal edema, leukocyte infiltration in submucosa and mucosal necrosis. The levels of MDA in the DHEA pretreatment were also higher than those in the rats given with vehicle pretreatment. This suggests that the DHEA pretreatment deteriorates severe inflammation in indomethacin-induced gastritis. DHEA supplementation significantly increased SOD activity in the gastric mucosa. However, the catalase and GPx activities were not altered by DHEA. The co-administration of DHEA with an indomethacin might not offer a protective effect against the acute gastritis induced by indomethacin.

병아리 공장(空腸)의 교감신경지배(交感神經支配)에 관한 연구(硏究) (Studies on Sympathetic Innervation of the Jejunum in the Chick)

  • It has been generally understood that the intestinal tracts are under the control of the autonomic nerves; the parasympathetics are excitatory and the sympathetics inhibitory. However, it is recently reported that the actions of these autonomic nerves in the newborn animals are shown to be different from those in the adult animals in some species. In order to elucidate the role of sympathetic innervation to the intestinal tracts, the effects of periarterial nerve stimulation were studied in the periarterial sympathetics-jejunum preparations of the chick and the effects of some autonomic drugs on the isolated muscle strips were also studied. The results obtained were as follows: 1. The periarterial stimulation in the periarterial sympathetics-jejunum preparation elicited the responses of three patterns; 1) contrcation followed by relaxation 2) contraction only 3) relaxation only. The excitatory response was most effective in the stimulus frequencies of 40 cps, whereas the inhibitory response was maximal in the stimulus frequencies of 30 cycle per second. 2. The excitatory response to the periarterial stimulation was not affected by the pretreatment with phenoxybenzamine, dibenamine, propranolol and atropine, whereas the inhibitory response was completely blocked by the pretreatment with phenoxybenzamine and propranolol. 3. In the periarterial syrnpathetics-jejunum preparation treated with reserpine, the periarterial stimulation evoked only contraction, and the contraction was not affected by the pretreatment with phenoxybenzamine, propranolol and atropine. 4. The administration of norepinephrine evoked a relaxation in the isolated jejunum muscle strips and the effect was completely blocked by the pretreatment with phenoxybenzamine. 5. The administration of isoproterenol produced a relaxation in the isolated jejunum muscle strips and the effect was not affected by pretreatment with phenoxybenzamine, whereas the effect was completely blocked by the pretratment with propranolol. 6) The administration of acetylcholine produced a marked contraction in the isolated jejunum muscle strips and the effect was completely abolished by the pretreatment of atropine. These experimental evidences indicate that the inhibitory response to the periarterial stimulation is due to adrenergic fibers and the excitatory response is due to neither adrenergic nor cholinergic component.

줄풀을 활용한 면직물의 염색 (Dyeing of Cotton Fabrics Using Zizania latifolia Turcz. Extracts)

  • This study examined the pretreatment effect, appropriate dyeing conditions, color fastness and functionality of cotton fabrics dyed with Zizania latifolia Turcz. Dye uptake was substantially increased by pretreatment and chitosan pretreatment was much more effective than tannic acid pretreatment. Optimal dyeing conditions were colorant concentration of 100%, dyeing temperature of $100^{\circ}C$, dyeing time of 80 minutes and dyebath pH of 5.5. Color fastness of chitosan pretreatment and dyed cotton to washing, rubbing, perspiration and light was 4, 5, 3-4(acidic), 3(alkaline) and 3 respectively. Color fastness of tannic acid pretreatment and dyed cotton to washing, rubbing, perspiration and light was 4, 5, 3-4(acidic), 4-5(alkaline) and 4 respectively. Deodorization rates of ammonia(NH3) were 99.9% in both cases. UV protection rate were 98.3% of UV-A and 98.5% of UV-B in case of chitosan pretreatment. UV protection rate were 98.2% of UV-A and 98.5% of UV-B in case of tannic acid pretreatment. UV protection factor(UPF) was 50+ in both cases. Reduction rate of Staphyloccus aureus were 99.9% in both cases. Therefore Zizania latifolia Turcz. could be used a new functional colorant.
