  • From June 28th to July 17th in 1985, this study was performed to obtain the information of the fresh corn forage production through the spraying hydrophonics in summer. The experiments were conducted in vinyl house on Kyungpook University's Experimental Station. The results obtained were as follows: It was suggested that the treatment of soaking for 24 hrs and followed by the pretreatment for 48 hrs was better than other treatments. Seed cost for the production of 1 kg fresh forage was the most inexpensive at the seeding rate of 450 g per $30{\times}60cm$ tray than other seeding rates. Application of 1000 times diluted Hyponex solution (1000X Hyponex) or 300 times diluted Yogen solution (300X Yogen) one time per day resulted in the heavier fresh weight than the other treatments. Treatments of 300X Yogen and 500X Yogen gave the better fresh weight in one application per two days and per three days, respectively. But, this method was not useful in summer because the lowest seed cost for the production 1kg fresh forage was 73Won, and collection of the fresh forage from the mountain range was easy.

Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat (흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과)

  • This study was performed to investigate the ameliorating effect of a hangover beverage mixture (SBJ) that contains Dendropanax morbifera Lev. and several medicinal plant extracts, on hepatoprotection and alcohol-metabolizing enzymes in alcohol-induced hangover in both in vitro and in vivo models. In human hepatoma cell line, HepG2, 300 mM of ethanol-induced hepatotoxicity was significantly improved by pretreatment of SBJ by dose-dependent manner. In the in vivo study, administration of alcohol to rats raised to the concentration of blood alcohol and lactate dehydrogenase (LDH). Blood alcohol and LDH levels in SBJ-treated rats significantly decreased at 0.5 h and 8 h after acute ethanol administration (40%, 4.6 g/kg body weight) as compared to alcohol-treated rats. Hepatic alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were significantly higher in SBJ-treated rats than in alcohol-treated rats. SBJ supplementation reduced formation of malondialdehyde (MDA), and inhibited reductions of hepatic superoxide dismutase (SOD), hepatic glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) levels, compared with rats administered alcohol. Plasma catalase (CAT), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels showed unaltered resulted in all experimental groups compared with the control group. These results suggest that SBJ exhibit hepatoprotective properties by enhancing ADH, ALDH activity and stimulating the antioxidant defense system in alcohol-induced hangover.

The Signal Transduciton of Ginsenosides, Active Ingredients of Panax ginseng, in Xenopus oocyte: A Model System for Ginseng Study

  • Recently, we have provided evidence that ginsenosides, the active components of Panax ginseng, utilize pertussis toxin (PTX)-insensitive $G{\alpha}_{q/11}-phospholipase\;C-{\beta}3(PLC-{\beta}3)$ signal transduction pathway for the enhancement of $Ca^{2+}-activated\;Cl^{-}$ current in the Xenopus oocyte (British J. Pharmacol. 132, 641-647, 2001; JBC 276, 48797-48802, 2001). Other investigators have shown that stimulation of receptors linked to $G{\alpha}-PLC$ pathway inhibits the activity of G proteincoupled inwardly rectifying $K^+$ (GIRK) channel. In the present study, we sought to determine whether ginsenosides influenced the activity of GIRK 1 and GIRK 4 (GIRK 1/4) channels expressed in the Xenopus oocyte, and if so, the underlying signal transduction mechanism. In oocyte injected with GIRK 1/4 channel cRNAs, bath-applied ginsenosides inhibited high potassium (HK) solution-elicited GIRK current $(EC_{50}:4.9{\pm}4.3\;{\mu}g/ml).$ Pretreatment of the oocyte with PTX reduced the HK solution-elicited GIRK current by $49\%,$ but it did not alter the inhibitory ginsenoside effect on GIRK current. Prior intraoocyte injection of cRNA(s) coding $G{\alpha}_q,\;G{\alpha}_{11}\;or\;G{\alpha}_q/G{\alpha}_{11},\;but\;not\;G{\alpha}_{i2}\;or\;G{\alpha}_{oA}$ attenuated the inhibitory ginsenoside effect. Injection of cRNAs coding $G{\beta}_{1{\gamma}2}$ also attenuated the ginsenoside effect. Similarly, injection of the cRNAs coding regulators of G protein signaling 1, 2 and 4 (RGS1, RGS2 and RGS4), which interact with $G{\alpha}_i\;and/or\;G{\alpha}_{q/11}$ and stimulates the hydrolysis of GTP to GDP in active GTP-bound $G{\alpha}$ subunit, resulted in a significant reduction of ginsenoside effect on GIRK current. Preincubation of GIRK channel-expressing oocyte in PLC inhibitor (U73122) or protein kinase C (PKC) inhibitor (staurosporine or chelerythrine) blocked the inhibitory ginsenoside effect on GIRK current. On the other hand, intraoocyte injection of BAPTA, a free $Ca^{2+}$ chelator, had no significant effect on the ginsenoside action. Taken together, these results suggest that ginsenosides inhibit the activity of GIRK 1/4 channel expressed in the Xenopus oocyte through a PTX-insensitive and $G{\alpha}_{q/11}$-,PLC-and PKC-mediated signal transduction pathway.

Enhanced Tolerance of Chinese Cabbage Seedlings Mediated by Bacillus aryabhattai H26-2 and B. siamensis H30-3 against High Temperature Stress and Fungal Infections

  • Two rhizobacteria Bacillus aryabhattai H26-2 and B. siamensis H30-3 were evaluated whether they are involved in stress tolerance against drought and high temperature as well as fungal infections in Chinese cabbage plants. Chinese cabbage seedlings cv. Ryeokgwang (spring cultivar) has shown better growth compared to cv. Buram-3-ho (autumn cultivar) under high temperature conditions in a greenhouse, whilst there was no difference in drought stress tolerance of the two cultivars. In vitro growth of B. aryabhattai H26-2 and B. siamensis H30-3 were differentially regulated under PEG 6000-induced drought stress at different growing temperatures (30, 40 and $50^{\circ}C$). Pretreatment with B. aryabhattai H26-2 and B. siamensis H30-3 enhanced the tolerance of Chinese cabbage seedlings to high temperature, but not to drought stress. It turns out that only B. siamensis H30-3 showed in vitro antifungal activities and in planta crop protection against two fungal pathogens Alternaria brassicicola and Colletotrichum higginsianum causing black spots and anthracnose on Chinese cabbage plants cv. Ryeokgwang, respectively. B. siamensis H30-3 brings several genes involved in production of cyclic lipopeptides in its genome and secreted hydrolytic enzymes like chitinase, protease and cellulase. B. siamensis H30-3 was found to produce siderophore, a high affinity iron-chelating compound. Expressions of BrChi1 and BrGST1 genes were up-regulated in Chinese cabbage leaves by B. siamensis H30-3. These findings suggest that integration of B. aryabhattai H26-2 and B. siamensis H30-3 in Chinese cabbage production system may increase productivity through improved plant growth under high temperature and crop protection against fungal pathogens.

Comparative assessment of urban stormwater low impact strategies equipped with pre-treatment zones (침강지 시설이 조성된 LID 시설의 환경적 영향평가)

  • Recently, Low impact development techniques, a form of nature-based solutions (NBS), were seen cost-efficient alternatives that can be utilized as alternatives for conventional stormwater management practices. This study evaluated the effectiveness of an infiltration trench (IT) and a small constructed wetland (SCW) in treating urban stormwater runoff. Long-term monitoring data were observed to assess the seasonal performance and cite the advantages and disadvantages of utilizing the facilities. Analyses revealed that the IT has reduced performance during the summer season due to higher runoff volumes that exceeded the facility's storage volume capacity and caused the facility to overflow. On the other hand, the pollutant removal efficiency of the SCW was impacted by the winter season as a result of dormant biological activities. Sediment data also indicated that fine and medium sand particles mostly constituted the trapped sediments in the pretreatment and media zones. Sediments in SCW exhibited a lower COD and TN load due to the phytoremediation and microbiological degradation capabilities of the system. This study presented brief comparison LID facilities equipped with pre-treatment zones. The identified factors that can potentially affect the performance of the systems were also beneficial in establishing metrics on the utilization of similar types of nature-based stormwater management practices.

A Study of the Salt Rejection from the Surface of Marine Waste using Ultra Fine Bubble (초미세기포를 이용한 해양쓰레기 표면 내 염분제거에 관한 연구)

  • Nano bubble water is used in various washing processes, including cleaning of solar panels, salt rejection of roads, and cleaning precision parts of machines. High cleaning efficiency and water conservation are obtained by applying nano bubbles during pretreatment of the marine waste cleaning system. This study compared the salt rejection of nano bubble water, and it was revealed that marine waste was produced by wood immersed in 200,000 mg/L NaCl solution. Using tap water and nano bubble water for washing, comparisons of the surface salt concentrations of wood were determined according to the nozzle, orifice diameter, pump speed and washing time. Decreased surface salt concentration was observed on the wood surface with increasing washing time. Water consumption was optimal between 5- and 10-seconds washing time. Increasing orifice diameter of the nozzle reduced the spraying pressure, with consequent increase in the wood surface salt concentration, thereby establishing the importance of orifice diameter of the nozzle. Compared to levels obtained with tap water, salt concentration of the wood surface after washing with nano bubble water was 2.2% lower with sector nozzle, and 30.9% lower with circular nozzle. In the washing experiment using nano bubble water, the salt concentration on the wood surface was about 9.5 mg/L lower when washed with sector nozzle than the circular nozzle.

Systemically administered neurotensin receptor agonist produces antinociception through activation of spinally projecting serotonergic neurons in the rostral ventromedial medulla

  • Background: Supraspinal delivery of neurotensin (NTS), which may contribute to the effect of a systemically administered agonist, has been reported to be either pronociceptive or antinociceptive. Here, we evaluated the effects of systemically administered NTSR1 agonist in a rat model of neuropathic pain and elucidated the underlying supraspinal mechanism. Methods: Neuropathic pain was induced by L5 and L6 spinal nerve ligation in male Sprague-Dawley rats. The effects of intraperitoneally administered NTSR1 agonist PD 149163 was assessed using von Frey filaments. To examine the role of 5-HT neurotransmission, a serotonin (5-HT) receptor antagonist dihydroergocristine was pretreated intrathecally, and spinal microdialysis studies were performed to measure the change in extracellular level of 5-HT in response to PD 149163 administration. To investigate the supraspinal mechanism, NTSR1 antagonist 48692 was microinjected into the rostral ventromedial medulla (RVM) prior to systemic PD 149163. Additionally, the effect of intrathecal DHE on intra-RVM PD 149163 was assessed. Results: Intraperitoneally administered PD 149163 exhibited a dose-dependent attenuation of mechanical allodynia. This effect was partially reversed by intrathecal pretreatment with dihydroergocristine and was accompanied by an increased extracellular level of 5-HT in the spinal cord. The PD 149163-produced antinociception was also blocked by intra-RVM SB 48692. Direct injection of PD 149163 into the RVM mimicked the maximum effect of the same drug delivered intraperitoneally, which was reversed by intrathecal dihydroergocristine. Conclusions: These observations indicate that systemically administered NTSR1 agonist produces antinociception through the NTSR1 in the RVM, activating descending serotonergic projection to release 5-HT into the spinal dorsal horn.

Effect of Temperature Conditions and Chemical Treatments on Seed Germination of Pseudolysimachion kiusianum var. diamantiacum (Nakai) T.Yamaz. (봉래꼬리풀의 종자 발아에 미치는 온도 조건과 화학적 처리의 영향)

  • The germination characteristics of Pseudolysimachion kiusianum var. diamantiacum (Nakai) T.Yamaz., a rare and endemic plant designated by the Korea Forest Service, were investigated according to light conditions, temperature, and pretreatment. As a result of the germination experiment according to light conditions, it was determined that P. kiusianum is a photoblastic seed that does not germinate at all in dark conditions. The optimum germination temperature of the seeds was found to be 20-25℃, considering the final germination rate and germination time. Three growth regulators (IAA, GA3, kinetin) and two inorganic salts (KNO3, KCl) were pretreated to improve the germination rate of P. kiusianum seeds. The growth regulators IAA and kinetin had no significant effect on improving the germination rate of P. kiusianum seeds. On the other hand, GA3 significantly increased the final germination percentage and germination rate regardless of the concentration, especially the treatment of more than 500 mg·L-1 at 20℃ was more than 4 times more effective than the untreated. The inorganic salts KNO3 and KCl had no significant effect on the seeds of P. kiusianum at low concentrations, but at higher concentrations (40 mM and 300 mM, respectively), they improved the germination rate and germination age by 2 times compared to the untreated. The results of this study will be useful for the mass propagation of P. kiusianum, which has the potential to be utilized as a native plant for restoration.

Experimental Study on the Adhesion and Performance Evaluation of Joints for Modified Polyethylene Coated Steel Pipes (개질 폴리에틸렌 코팅 강관의 부착 및 체결부 성능 평가 연구)

  • In this study, as part of the development of a monitoring system for the efficient maintenance of steel pipes, an experimental study was conducted to evaluate the performance of steel pipes treated with modified polyethylene coating. In the case of the conventional mechanical pre-coating method, there was a deterioration in polyethylene adhesion during expansion testing, which led to the application of a chemical pre-treatment process using a calcium-mixed phosphate zinc film to resolve this issue. SEM and EDX analyses showed that the densest structure was observed at a Zn/Ca ratio of 1.0, and improved heat resistance compared to the conventional method was confirmed. Additionally, to prevent coating detachment during expansion, an evaluation of adhesion and elongation was conducted on steel pipes with modified polyethylene coating, incorporating materials such as elastomers based on maleic anhydride grafting, metal oxides, blocking agents, and slip agents. Experimental results showed that the specimen (S4) containing all modified materials exhibited more than a 25% performance improvement compared to the specimen (S2) containing only metal oxides. Lastly, the development and performance evaluation of wedge-shaped socketing and pressing wheels, which are part of the pipe fixing accessories, were conducted to prevent surface coating damage on the completed pipes.

Influence of Intracerebroventricular Haloperidol on the Renal Function of the Rabbit (가토신장기능에 미치는 측뇌실내 Haloperidol의 영향)

  • In an effort to provide evidence as to the regulatory role of the central dopaminergic system on the renal function, the effects of centrally administered dopamine and its specific antagonist haloperidol were investigated. Haloperidol (HA) given intracerebroventricularly (i.c.v.) induced antidiuresis in doses of 15 and $50{\mu}g/kg$. With $15{\mu}g/kg$ sodium reabsorption in the tubules was increased, while with $50{\mu}g/kg$ free-water reabsorption was increased. However, a marked diuresis with increased sodium and potassium was observed with $150{\mu}g/kg$. Hemodynamic changes were not evident, indicating that the diuresis is of tubular origin. Dopamine (DA), on the other hand, produced antidiuresis when given i.c.v. in a dose-related fashion. With smaller doses of 5 and $15{\mu}g/kg$ the antidiuresis was related to increased reabsorption of sodium in the tubules, but higher doses of 50 and $150{\mu}g/kg$ the decreases in renal blood flow and glomerular filtration rate were evident in addition to the tubular action. After pretreatment with $150{\mu}g/kg$ HA, the effects of $15{\mu}g/kg$ DA was abolished, but the antidiuretic actions of 50 and $150{\mu}g/kg$ were not blocked, and the natriuretic diuretic action of HA was overcome and became inconspicuous. These observations indicate that the central dopaminergic system influences the renal function by producing antidiuresis, and HA elicits diuresis and natriuresis by competitively antagonizing DA specifically on the central dopaminegic receptors. The antidiuresis observed with smaller doses of HA can be best explained by the facts that there are more than two types of DA-receptors in the brain and that the presynaptic autoreceptors on the dopaminergic neurones which affect the dopamine release at the synapse are more sensitive than the postsynaptic receptors. Overall, these data provide an evidence indicating that the central dopaminergic system plays a role in the regulation of renal function in the rabbit.
