Screening for Fermentative Microorganisms that Grow on Brown Rice with High Amylase and Pretense Activities (Amylase와 Protease의 활성이 높은 현미 발효 미생물의 선별)

  • Brown rice contains rice bran and germ with higher nutritional value and dietary fiber content compared with the polished rice. However, brown rice has a limitation of poor digestion. fermented brown rice could be better nutritional source and improve digestibility. Therefore, we tried to select good fermentative microorganisms which have nutritional values with high amylase and protease activities, and probiotic effects. Nineteen micro-organisms, including eight Bacillus strains isolated from Chongkukjang and 11 lactic acid bacteria, were screened for the fermentation ability and enzyme production. The liquid broths containing 2.5%(w/v) of raw brown rice powder as a sole nutritional source were used for culture media. Among the strains tested, all of the Bacillus strains and two lactic acid bacteria (Leuconostoc gelidum and Pediococcus pentosaceus) showed increase in cell population and enzyme activities. The viable cell counts of all the Bacillus strains and two lactic acid bacteria exceeded $10^7 CFU/mL$. The maximal enzyme activities produced by Bacillus sp. Bl, Bacillus sp. B2, Bacillus sp. B11, L. gelidum and P. pentosaceus were 17.85, 17.50, 17.10, 17.10 and 3.24 U/mL for amylase and 22.48, 22.04, 23.76, 12.13, and 3.4 U/mL for pretense, respectively. Therefore, the results of this study demonstrated that the above strains could be potential starters for the fermentation of raw brown rice.

Expression, purification and characterization of ubiquitin-specific pretense 1 for hydrolysis of ubiquitin-fused human growth hormone expressed in recombinant Escherichia coli

  • This research was focused on expression, purification and characterization of ubiquitin-specific protease 1 (UBP1) expressed in recombinant Escherichia coli. Various systems were constructed by fusing polycationic fusion tails or fusion partners to the C- or N-terminus of the product protein. In particular, UBP1 containing 6 histidine residues at the N-terminal end showed best results in terms of expression level and purification efficiency. The N-terminal $6{\times}His-tagged$ UBP1 was overproduced in recombinant E. coli using high cell density cultivation technology and purified using immobilized metal affinity chromatography. The molecular weight of UBP1 was found to be 83,500 daltons. The optimum temperature and pH for the enzyme reaction when ubiquitin-human growth hormone (hGH) was used as a substrate were $40^{\circ}C$ and pH 8.0, respectively.

Effect of Injection Application of Pig Slurry on Ammonia and Nitrous Oxide Emission from Timothy (Phleum pretense L.) Sward

  • The objective of this study was to determine the effect of injection application of pig slurry on ammonia ($NH_3$) and nitrous oxide ($N_2O$) emission from timothy (Phleum pretense L.) sward. The three treatments were applied: 1) only water as a control, 2) pig slurry application by broadcasting, 3) pig slurry application by injection. The pig slurry was applied at a rate of $200kg\;N\;ha^{-1}$. Total $NH_3$ and $N_2O$ emission, expressed as a cumulative amount throughout the measurement time (40 days), was $2.68kg\;NH_3-N\;ha^{-1}$ and $6.58g\;N_2O-N\;ha^{-1}$, respectively, in the control. The injection application of pig slurry decreased total $NH_3$ and $N_2O$ emission by 39.8% and 33.3%, respectively, compared to broadcasting application of pig slurry. The present study clearly showed that injection application exhibited positive roles in reducing N losses through $NH_3$ and $N_2O$ emission.

Separating and purifying polysaccharide, polypeptide from sea cucumber and their functions (해삼으로부터 다당류와 폴리펩타이드의 분리, 정제 및 기능성)

  • For thestudying and utilizing the thorn sea cucumber synthetically, the method preparing polysaccharide and polypeptide from sea cucumber was studied. The comparatively rational craft for preparing crude polysaccharide is fresh raw materials deal with oar form, $60\%$ of the ethanol precipitate after hydrolyzed by pretense and vibrated with ultrasonic wave auxiliary. The purification of polysaccharide and removal of protein fromcrude polysaccharide were made through precipitation method using the acetate. The polypeptide is obtained by concentrating in vacuum, freeze-drying after mixing precipitations of two times. Polysaccharide acute poisoning experiment indicate there is no bad reaction when LD50>5000g/kg, there strain rate of liver tumor H11 approach present generally acknowledged tumor treat medicine cyclophosphamide, and there is no side effect. The fatigue resistance function of polypeptide experiment also indicates that the fatigue resistance ability of mouse which fed on added sea cucumber polypeptide has a great improvement.

  • Assuming that Asp225 is the substrate specificity determinant of Achromobacter pretense I (APl) which is lysine-specific serine protease, the 225th residue was substituted for other amino acids with a hope that the substrate specificity of a mutant API is altered. Furthermore, to maturate preform of mutant API autocatalytically, Lys(-1) was also replaced by Met, Asp, or Glu. However, all the mutants were not expressed, or accumulated as inactive precursor proteins. This result implicats that Asp225 plays a critical rol in restricted substrate specificity as a lysylendopeptidase but the substrate specificity of API is not determined only by the nature of residue 225.

  • A nuluk, a Korean traditional koji for brewing, was made with wheat flour and Aspergillus usamii mot. shirousamii S1 which had strong abilities in producing amylase and protease. The cultural conditions for the production of saccharogenic and proteolytic enzymes were tested. The productivities of saccharogenic and dextrogenic enzymes were improved when nuluk was made with unsteamed wheat flour as compared with steamed one, but those of proteolytic enzyme and organic acid were reduced. The addition of water containing 0.5% of hydrochloric acid was unfavorable for the production of saccharogenic, dextrogenic and proteolytic enzymes. The optimum ratios of water added to wheat flour for the production of saccharogenic enzyme and proteolytic enzyme were 32% and 28%, respectively on the basis of wheat flour. The optimum temperatures for the production of saccharogenic enzyme and proteolytic enzyme were 36$^{\circ}C$ and 28$^{\circ}C$, respectively. The activity of saccharogenic enzyme reached its maximum after 120 hours of cultivation at 36$^{\circ}C$, but that of proteolytic enzyme 96 hours. The productivity of saccharogenic enzyme was enhanced when the nuluk was molded after 24 hours of precultivation but that of proteolytic enzyme was reduced as compared with no molding.

Changes of Microbiological Distribution in Food Waste for Animal Feed (동물 사료화를 위한 음식물 쓰레기의 미생물 분포 변화)

  • This study investigated the microbiological changes and distribution of bacteria producing enzyme in order to change food wastes to animal feed during 6 days at room temperature. Food wastes were divided as follows: one is untreated food wastes containing 80% water content and another wastes containing 40% water content adiusted by wheat bran. During the fermentation of food wastes, Lactobacillus sp. and Streptococcus sp. were grown the top position among investigated microorganisms in both of food wastes. Numbers of total microorganisms were much more than untreated wastes during fermentation of flood wastes with 40% water content. But, Streptococcus sp. and Enterobacteriaceae from 4 to 6 days were not detected in treated wastes with 40% water contents. This fact indicated that the adiustment of water content in food waste was effective in fermentation. However, the numbers of microorganisms producing pretense were low in both treated and untreated food wastes. These results suggested that flood wastes should be treated as food wastes with 40% water content to maintain a lot of fermentative microorganisms such as Lactobacillus sp., Streptococcus sp. and yeasts together with pretense to make final animal feed.

Production of a Keratinolytic Protease by a Feather-Degrading Bacterium, Bacillus megaterium F7-1 (우모분해세균 Bacillus megaterium F7-1에 의한 Keratinolytic Protease의 생산)

  • Bacillus megaterium F7-1 producing keratinolytic protease was isolated from decayed chicken feather. The optimal culture conditions for the production of keratinolytic protease by B. megaterium F7-1 were investigated. The composition of optimal medium for the keratinolytic protease was 0.2% glucose, 0.8% skim milk, 0.05% NaCl, 0.01 % $(K_2HPO_4$, 0.02%, $(KH_2PO_4$ and 0.01 % $MgCl_2$. Especially, skim milk was found to be the most effective compound in keratinolytic protease production. The optimal temperature and initial pH were 6.5 and $25^{\circ}C$, respectively. The keratinolytic protease production under optimal condition reached a maximum of 269 U/ml after 5 days of cultivation. B. megaterium F7-1 degraded 98% of the feather used in the optimized medium within 6 days.

Effects of Mud Flat Bacteria Origin Protease Supplementation on Egg Quality, Nutrient Digestibility and Total Protein Concentration of Serum in Laying Hens (산란계에 갯벌 미생물 유래 단백질 분해 효소제의 급여가 계란 품질, 영양소 소화율 및 혈청내 총 단백질 함량에 미치는 영향)

  • This study was conducted to investigate the effects of mud flat bacteria origin pretense supplementation on e99 qualify, nutrient digestibility and total protein concentration of serum in laying hens. A total of two hundred fifty two laying hens were randomly allocated into three treatments with seven replications for eight weeks. Dietary treatments included 1) CON (basal diet), 2) PRO1 (basal diet + 0.05% pretense) and 3) PRO2 (basal diet + 0.1% pretense). During the entire experimental peripd, hen-day egg production was not affected by treatments (P>0.05). Difference of yolk height was increased in PRO1 treatment compared with CON treatment (P<0.05). Difference of egg weight was increased in PRO2 treatment compared with CON and PRO1 treatments (P<0.05). Shell quality, yolk color unit, haugh unit and egg yolk index were not affected by treatments (P>0.05). DM digestibility was improved in CON and PRO2 treatments compared with PRO1 treatment (P<0.05). N digestibility was improved in PRO2 treatment compared with CON treatment (P<0.05). Total protein concentration in serum were not affected by treatments (P>0.05). In conclusion, mud flat bacteria origin pretense was effective for improving egg weight, yolk height and nutrient digestibility in laying hens.
