Isolation and Characterization of an Alkaline Protease Produced by Bacillus subtilis JK-1 (알칼리성 Pretease를 생산하는 Bacillus subtilis JK-1의 분리 동정 및 효소 특성)

  • A bacterium producing the alkaline pretense was isolated from Chungkookjoug, and was identified as Bacillus subtilis JK-1 based on morphological, physiological and biochemical characteristics, as well as phylogenetic analysis using 16S rRNA gene sequence. The optimum pH and temperature of the pretense activity were pH 9.0 and $55^{\circ}C$, respectively. This enzyme was stable at the temperatures $40{\sim}80^{\circ}C$. The maximum alkaline pretense production was obtained when 1.0% (w/v) xylose, 1.0% (w/v) yeast extract and 0.3% (w/v) $CuSO_4$ were used as carbon source, nitrogen source and mineral source. Under the optimal condition, growth of the isolate was reached at stationary phase after 12 hr followed by incubation, the alkaline pretense production reached a maximum level with $16{\sim}20$ hr cultivation.

Effects of Pretense Treatment of Soy Milk on Acid Production by Lactic Acid Bacteria and Quality of Soy Yogurt (두유(豆乳)의 단백질분해효소 처리가 젖산균의 산생성(酸生成)과 대두요구르트의 품질에 미치는 영향)

  • The soy milk prepared from soy protein concentrate was treated with pretense of various concentrations. Growth and acid production by Lactobacillus acidophilus or L. bulgaricus in soy milk containing partially hydrolyzed protein were investigated. Sensory evaluation of soy yogurt beverages prepared from pretense treated soy milk was also performed. The treatment of soy milk with pretense markedly enhanced acid production by lactic acid bacteria, particularly by L. acidophilus, whereas pH and number of viable cells were not .affected by pretense treatment. Protease treatment of 15 minutes greatly enhanced acid production by lactic acid bacteria, but further treatment up to three hours did not affect the acidity markedly. The sensory evaluation showed that overall acceptability of soy yogurt beverages was slightly improved when soy milk was treated with pretense of 0.1%. The amount of non-protein nitrogen was considerably increased by pretense treatment of 15 minutes and it reached the maximum value by treatment of two hours.

  • V parahaemolyticus possessed an extracellular alkaline pretense activity during the stationary growth phase. Various factors such as nitrogen sources, the concentration of NaCl and metal ions were investigated for optimizing the production of alkaline pretense from V. parahaemolyticus ATCC 17802. Among the nitrogen sources tested skim milk showed the distinct increase of the activity and the activity was the highest at 2% in final concentration after 60 hours incubation. The addition of NaCl and metal ions did not increase the alkaline pretense activity. CoC$_2$, CuC1$_2$, and HgCl rather highly inhibited alkaline protease production.

  • Distribution of the proteolytic activities of crude pretense extracted from the viscera of ten kinds of fish was examined. Their proteolytic activities on proteinous substrates (azocasein, hemoglobin, and casein) from the viscera of anchovy, bastard flatfish, mackerel and red sea bream were higher than those of other fishes, and the crude pretenses were further fractoinated with acetone or ammonium sulfate. Optimum concentrations for pretenses fractionation were $0\~55\%$ for acetone and $30\~70\%$ for ammonium sulfate. The fractionated viscera pretense of mackerel showed the highest proteolytic activity among four kinds of fishes. Activities of cathepsin D- and pepsin-like enzymes at pH 3.0, cathepsin L-, B-, H- and G-like enzyme at pH 6,0, and Hypsin- and chymotrypsin- like enzymes (pH 8.0) were detected in the fractionated viscera pretense, whereas activities of cathepsin L- and chymoeypsin-like enzyme were observed in commercial pretenses. Proteolytic activities of Alcalase, Protamex, and Aroase AP-10 for azocasein were slightly higher than the fractionated viscera pretenses, but their amidolytic activities at pH 6.0 and 8.0 toward synthetic substrates were lower than counterpart. The fractionated pretenses from fish viscera would be utilized as commercial pretenses.

Effect of Earthworm Pretense on Dyeing Properties of Protein Fibers

  • An earthworm protease, Lumbricus rubellus, was used to improve the dyeing properties of protein fibers such as wool and silk. The optimal condition for the activity of the earthworm pretense was about $40^{\circ}C$ at pH 7. The wool and silk were treated with the protease extracted from an earthworm and the K/S values of the dyed wool and silk were measured using a spectrophotometer in order to compare the dye uptake. The pretense treatment enhanced the dyeing properties of protein fibers without severe changes in mechanical properties. The surface appearances of pretense-treated fibers were observed by microscopy analysis.

  • We describe here the cDNA sequence and mRNA expression of a novel serine pretense from the firefly, Pyrocoelia rufa. The 771 bp cDNA encodes for 257 amino acid residues. The deduced protein of P. rufa serine pretense gene contains the catalytic triad and six-conserved cysteine residues. Alignment of the deduced protein of P. rufa serine pretense gene showed 47.4% protein sequence identity to known coleopteran insect Rhyzopertha dominica midgut trpsin-like enzyme. Northern blot analysis revealed that the P. rufa serine pretense is specifically expressed in the midgut of P. rufa larvae.

  • In order to develope a new pretense applicable to industries, a bacterium which produces a remarkable amount of extracellular pretense were isolated from soil. About 10 bacterial strains producing pretense were isolated from samples of soil, and strain PANH765 showed the highest activity of pretense production among them. The strain was identified as Bacillus subtilis according to the Bergey's Manual of Systematic Bacteriology based on its morphological, cultural and physiological characteristics. B. subtilis PANH765 showed the maximal production of pro-tease in the medium containing 2.0% glucose, 1.0% yeast extract, 0.2% ammonium nitrate, 0.02% ferrous sulfate and 0.02% dipotassium hydrogen phosphate. Under the optimal condition with temperature of 3$0^{\circ}C$, initial pH of 7.0 and shaking speed of 150 rpm, the pretense production reached a maximum level with 36 hr cultivation (6.34 U).

  • We investigated the optimal condition for the production of extracellular pretense(a cuticle-degrading pretense) from entomopathogenic fungus Beauveria bassiana(ATCC7159) in liquid medium by adding of gelatin, bovine serum albumin(BSA), casein and polypeptone. The optimal induction medium for production of extracellular pretenses is composed of 0.5% polypeptone, trace elements and 50 mM potassium phosphate(pH 6.0). In this condition, the production of extracellular pretenses increased rapidly after the 24hrs, peaking at the third day and there was little inductive effect in culture broth more than pH 7.0. The pretenses were inhibited by phenyl methyl sulfonyl fluoride(PMSF). High activity of pretense was showed both range of pH 8.5 and 11.5 and also detected by three different portions of slice gel derived from non-denaturing isoelectricfocusing gel. At least three different extracellular pretenses are produced in optimal production medium when polypeptone is used as the sole carbon and nitrogen source.

Purification and Characterization of Protease from Bacillus subtilis PANH765 (Bacillus subtilis PANH765가 생산하는 Protease의 정제 및 특성)

  • Pretense produced by Bacillus subtilis PANH765 was purified from culture supernatant by using ammonium sulfate fractionation DEAE-cellulose ion exchange chromatography, and gel filtration with Sephacryl S 200 HR and Sepharose CL-6B. DEAE-cellulose ion exchange column chromatography, separated the pretense into one fraction. This fraction was further purified using Sephacryl S 200 HR and Sepharose CL-6B gel titration. The molecular mass of pretense was estimated to be 35.0 kDa by the SDS-PAGE and gel filtration using Sepharose CL-6B. The results indicated that the purified pretense are monomeric proteins. Specific activity and purification folds of pretense were 657 U/mg and 4.35, respectively. The optimum temperature, optimum pit stable at a temperature range and pH ranges for the purified protease were 65$^{\circ}C$, 7.05, 50 ∼ 75$^{\circ}C$ and 6.0 ∼ 7.5, respectively. The pretense activity was decreased by the presence of PMSF and DFP, which the protease activity was increased by the presence of Na$\^$+/, K$\^$+/, Mg$\^$2+/ and NH$_4$$\^$+/ ions.

  • Effects of silkworm powder addition on rheological properties of dough and quality characteristics of bread were investigated. Protein content of silkworm powder was 53,98%, much higher than 12.46% of wheat powder, Crude fiber, fat, and pretense contents of silkworm powder were higher than those of wheat flour, Acid analysis revealed glutamic acid content was highest 4,046.16 mg thus, significant depreciation of breadmaking was expected due to weakened gluten structure or dough. Addition of silkworm powder(optimum at 2%) with pretense inactivated by heat treatment resulted in significant improvement of volume and bread quality, with external and internal scores close to those of the control.
