Identity of the 155 ATPase with the Eukarvotic Mg"'-Arpase and Its Immunoreactivitv to the 26S protease Complex from Chick Skeletal Muscle (15S ATPase와 진핵세포에 존재하는 Mg2+-ATPase의 동질성 및 계 골격근에서 분리된 26S 단백질 분해효소와의 면역반응에 관한 연구)

  • Partial internal amino acid sequences of the 15S ATPase from chick skeletal muscle were determined and found to be identical to the corresponding regions of the Mg2+_ATPase from Xenopus laevis oocytes, that is a close homolog of N-ethvlmaleimide-sensitive factor (called NSF) in hamster and Sec18p in yeast, both of which are believed to plaN an essential role in vesicle fusion in secretory process. Thus, the 15S Arpase in chick skeletal muscle maw also belong to a protein family of the "vesicle fusion proteins". Unlike the Mg2'-Afpase with an isoelectric point (pl) of 5.5, however, the 15S Arpase was separated into four spots with pls of 4.9,6.4 and 6.9 upon analysis by twoiimensional gel electrophoresis. In addition, the anti-15S ATPase IgG was found to be capable of interacting with the 265 protease complex upon analysis by immunoprecipitation. Moreover, immunoblot analysis revealed that the anti-155 Arpase IgG recognizes three subunits, ko of which show the same mobilities as the 510-kDa subunit 4 and 48-kDa subunit 7 of the 26S protease complex that are known to contain a highly consented ATP-binding motif. These results surest that a common antigenic site, likely the consensus nucleotide-binding site, exists in the 15S ATPase and the 26S pretense complex and hence both the enzymes maw also be related ATPases.d ATPases.

Changes in Species Diversity and Spatiotemporally Fluctuation across Human-modified Ecosystems (도시화로 인한 생태계에서 종 다양성의 변화와 공간시기적 변동)

  • The changes of plant communities at Angol valley and Baetgol valley in Geoje-do were studied. Both ${\alpha}$-diversity and ${\gamma}$-diversity decreased with the sizes and degrees of habitat fragmentations. The mean number of winner species decreased with habitat fragmentation. All Quercus species, including Quercus acutissima, significantly declined in importance, while the many introduced species, including Trifolium pretense, all increased in importance. As the proportional change in adult survival rate increased, the absolute value of the bias in the elasticity prediction also increased from 2003 to 2011 at undisturbed forests. However, the bias was low for decreases in disturbed populations. Moran's I values showed overall decreases for habitat fragmentation and for the periods of habitat conversion. Eventually, plant communities, due to urbanization in Goeje-do, might have led to decreased chances of common species when the environment was disturbed.

Antibacterial Activities of Ginkgo Biloba Leaves Extracts Against Isolated Bacteria from Museums (박물관에서 분리된 세균에 대한 은행잎 추출물의 항균활성)

  • The textile remains have been affected largely by environmental factors including microorganisms because they were composed of organic compounds to be easy to damage. So, we selected 8 strains of the 131 isolated strains from museum environments and textile remains by high pretense activity, and identified them for measuring the antibacterial activity of Gingko biloba extracts. They were identified Genus Arthrobacter spp. 3 strains (Arthrobacter nicotiannae A12, Arthrobacter sp B12, Arthrobacter oxidans B13), Cenus Bacillus spp. 2 strains (Bacillus licheniformis D9, Bacillus cereus D33), Genus Pseudomonas spp. 2 strains (Pseudomonas putida A24, Pseufomonas fluorescene C21) and a Genus Staphylococcus sp. 1 strain (Staphylococcus pasteuri D3) as closest strains through the blast search of NCBI. Though antibacterial activity of the extracts of Gingko biloba leaves as MIC was lower than that of other pharmaceutical antibiotics. However the extracts was crude extracts, the extracts might have good antibacterial against most of the isolates from museum. Especially, the antifungal activity of Gingko biloba is known previously, the extracts of Gingko biloba leaves has possibility of usage as a good natural material for conservation of remains.

A study of apoptosis induction of Euonymus alatus (Thunb.) Sieb via mitochondrial pathway prooxidant in leiomyomal smooth muscle cells (귀전우(鬼箭羽)의 인간 자궁근종 세포에서 미토콘드리아 경로를 통한 산화제로서 apoptosis 유도작용에 관한 연구)

  • Purpose : 귀전우(Euonymus alatus, EA)는 현재까지 항종양활성을 나타낸다고 보고되었지만 그 작용 메커니즘에 대해서는 아직 밝혀지지 않은 채 남아 있다. 본 연구에서는, 자궁근종세포(ULSMC)에서 EA의 분자적 수준에서의 작용메커니즘을 연구${\cdot}$검토하고자 하였다. Methods : EA의 열수추출액이 자궁근종세포(ULSMC)와 caspase-3 pretense의 활성도에 미치는 영향을 측정하였다. Results : 우리는 자궁근종에서 EA 유도 세포독성의 메커니즘을 검토하였는바, 근종 세포들은 20-200g/ml 농도의 EA추출물에 6시간 배양될 때, caspase-3가 활성화되고, 그때 세포들은 apoptosis를 유발하게 되었다. EA에 의한 apoptosis의 유도가 진행되었으며, cytochrome- c의 세포질분획에서 양적증가가 caspase-3의 활성보다도 우세하였다. GSH합성의 저해제인 5mM buthionine용액에 전처리는 EA유도 apoptosis를 용이하게 하지만 pan-caspase inhibitor인 Z-VAD-fmk용액 전 처리는 부분적으로 apoptosis유도를 억제하였다. 한편, EA는 건강한 지원자들로 부터 채취한 말초혈액 단핵세포들에 있어서는 독성의 효과는 없었다. Conclusion : 이들 결과들은 EA가 prooxidant로 작용을 하고 그리고 caspase-3 activation과 mitochondrial pathway를 경유하는 apoptosis를 유발한다는 것을 나타낸다. EA의 탕제약제로서 열수추출액이 항산화활성뿐만 아니라, 종양세포에 대한 세포독성효과를 나타낸다고 보고된 바, 이에 향후 근종치료에 대한 임상연구가 필요할 것으로 보인다.

Trypsin Inhibitor from Streptomyces sp. ( Part 1) Isolation of microorganism and purification of the inhibitor (Streptomyces 속 균주가 생성하는 Trypsin Inhibitor (제1보) 균의 분리 및 저해물질의 정제)

  • One strain of Streptomyces sp. (AS-707) isolated from soil was found to produce a biologically active substance that showed a strong inhibitory activity against proteolytic enzymes viz. trypsin, papain, $\alpha$-chymotrypsin, Azotobacter protease, and Bacillus pretense. The substance was separated from culture filtrate by ion exchange column chromatography using Amberlite IRC-50 and CM-cellulose column chromatography. It was found that the recovery yield was 26% as activity basis. The substance was stable in wide pH range from 2.0 to 12.0 at 37$^{\circ}C$, but it was unstable in alkaline pH values at 6$0^{\circ}C$. The activity was thermostable to give 90% activity compared to the intact sample when it was treated at pH5.6 at 10$0^{\circ}C$ for 2 hours.

  • Alkalophilic coryneform bacteria TU-19 isolated from soil extracellularly produced at least three proteases (Protease I, II, and III). Investigating the cultural conditions related to the enzyme production of this bacterial cell, the optimum pH and temperature were 10.0 and $30^{\circ}C$, respectively. In order to purify these enzymes from the 2 day culture broth ammonium sulfate fractionation, gel filtration and QAE-Sephadex column chromatography were performed step by step. And then these three proteases were purified to near homogeneity by judging from SDS-PAGE pattern, and had the molecular weights of 120, 80, and 45 kilodaltons, respectively. The optimum pH and temperature for the enzyme activity of Protease I and II were 10.5 and $45^{\circ}C$, respectively, and Protease II were 11.0 and $50^{\circ}C$. And the enzymes were completely inhibited by PMSF suggesting serine protease, but not affected by pCMB. 1,10-phenanthroline, IAA, and EDTA.

Protective Effects of Sanyakbojungbangam-tang Ethanol Extracts on Cisplatin-induced Apoptosis in ECV304 Cells (혈관내피세포에서 cisplatin에 의한 세포고사에 대한 산약보정방암탕 에탄을 추출물의 방어효과)

  • This study was designed to investigate the protective effect of Sanyakbojungbangam-tang Ethanol Extracts (SB Et-OH) on the cisplatin-induced apoptosis of human endothelial cell line ECV304 cells. After cells were treated with cisplatin, MTT assay was performed for cell viability test. To explore the mechanism of cytotoxicity, we used the several measures of apoptosis to determine whether this processes was involved in cisplatin-induced cell damage in ECV304 cells. Also, cells were treated with SB Et-OH and then, followed by the addition of cisplatin. Cisplatin decreased the viability of ECV304 cells in a dose-dependent manner and increased the caspase-3 enzyme activity ECV304 cells treated cisplatin were revealed as apoptosis characterized by nuclear staining. SB Et-OH protected ECV304 cells from cisplatin-induced nuclear fragmentation and chromatin condensation. Also, SB Et-OH inhibited the activation of caspase-3 pretense and the cleavage of poly(ADP-ribose) polymerase (PARP) in cisplatin-treated ECV304 cells. According to above results, SB Et-OH may protect ECV304 cells from the apoptosis induced by cisplatin.

General Pharmacology of LB71350, a New HIV-1 Pretense Inhibitor

  • Safety evaluation of LB71350, a new HIV-1 protease inhibitor, was performed in mice, rats and dogs. For the general behavior of mice, LB71350 at an oral dose of 200 mg/kg did not show any significant effects on muscle tone and locomotor activity. In terms of central nervous system, at oral doses of 200 mg/kg and 1000 mg/kg, LB71350 inhibited acetic acid-induced pain response approximately 41% and 83% of control. respectively. At oral doses of 200 mg/kg and 500 mg/kg, it reduced the rectal body temperature in rats. Pentylenetetrazole-induced seizure in mice was slightly potentiated by oral administration of LB71350 at doses ranging from 200 mg/kg to 1000 mg/Ag. Single or five day treatment of LB71350 doubled the hexobarbital- induced sleeping time in mice at oral doses ranging from 50 mg/kg to 500 mg/kg. It did not cause any effects on gastric secretion and acidity in rat at oral doses of 200 mg/kg and 1000 mg/kg and also it did not change intestinal motility in mice up to 1000 mg/kg. Blood coagulation indices such as prothrombin time (PT), activated partial thromboplastin time (aPTT), and thrombin time (TT) in rats were not affected by the treatment of LB71350 up to 500 mg/kg. LB71350 caused no significant effects on the cardiac output, stroke volume, heart rate, and mean blood pressure when infused intravenously to the anesthetized rats and dogs. Taken together, LB71350 at high oral doses caused significant pharmacological effects on the central nervous system and the hexobarbital-induced sleeping time.

Effects of Sensory Integration Therapy with Sibling on Play Level and Time for Children with Disabilities

  • Purpose : There is a lack of research on this field in the Republic of Korea, especially those that have seen the effect of interaction between siblings through sensory integration therapy (SIT). Therefore, this study sought to find out the effectiveness of SIT with siblings through the Revised Knox Preschool Play Scale (RKPPS) and playtime. Methods : The experimental group consisted of 10 disabled children, and 10 normal siblings of the disabilities joined the program as a helper. In addition, 10 children with disabilities conducted the intervention as a control group. The difference between the experimental group and the control group is whether they participate with their sibling or not during the intervention period. The present study was conducted a SIT for 40 minutes per week for the experimental and the control group and then had 10 minutes of an interview with the parents of children with disabilities. A total of 10 programs were implemented for 10 weeks. Results : The experimental group showed statistically significant differences in space management, material management, pretense/symbolic, participation, and total scores. The control group showed significant differences between pre and post results in the participation and the total scores. The experimental group and the control group showed significant differences in the pre-post comparison results. The comparison of post-intervention between both groups of the RKPPS and playtime results showed a statistically significant increase in the experimental group. Conclusion : The sibling SIT showed better play level and time than the individual therapy. However, comparisons before and after the intervention in the level of play showed significant results only in participation and total scores in the control group. Clinically, it is recommended to make good use of sibling relationships when applying SIT, and if that is not possible, continuous observation is needed that children who received treatment become familiar with the environment in which they can be treated.

  • This experiment was conducted to find out the changes in microflora of submerged soil uncultivated rice plant by application of CNP herbicide (2, 4, 6 - Trichlorophenyl-4-Nitrophenyl ether) under conditions applied with compost, rice straw, glucose or without organic material. The soil, sandy loam textured was incubated in green house for 66 days. Sampling and analysis of microorganisms were carried out during submergence periods. The results were summarilized as follows. 1. Number of aerobic total bacteria was increased by application of CNP herbicide during submerbed 50 days, afterthen, could not seen the difference. The application of rice straw increased number of aerobic bacteria regardless of CNP herbicide application or not, but glucose tended to decrease. 2. Number of Fungi was constantly maintained at $8-20{\times}10^3$ levels during period of submergence regardless of application of CNP herbicide and organic materials or not. 3. The CNP herbicide application tended to decrease the number of actinomycetes, particularly, in the treatments without organic substances and rice straw were remarkably decreased. 4. Anaerobic-and gram-negative bacteria populations were not showed any difference by application of herbicide and organic materials. 5. The ratios of aerobic bacteria to fungi and aerobic bacteria to actinomycetes appeared high values by application of herbicide and of organic substances. 6. At 66 days after submergence, the ratio of chromogenic actinomycetes to the total number of actinomycetes was lowered in application of herbicide. On the other hand, the percentage of both pretense-positive and cellulase-positive actinomycetes to the total isolates were higher in the treatment with herbicide than An without herbicide, particularly. The ratios of pretense-positive actinomycetes were high in the rice straw application regardless of herbicide application or not, but cellulase-positive actinomycetes was not remarkably difference.
