마이크로웨이브 추출조건에 따른 참취 추출물의 총 폴리페놀 함량 및 항산화작용의 변화 (Changes of Total Polyphenol Content and Antioxidant Activity of Aster scaber Thunb Extracts with Different Microwave-Assisted Extraction Conditions)

  • 마이크로웨이브 추출방법과 환류 냉각 추출방법을 비교한 결과, 물과 에탄올의 혼합용매로 추출한 경우 마이크로웨이브 추출 방법에 의하여 추출시간을 단축시키면서 환류 냉각 추출 방법에서와 같은 수준의 가용성 고형분 및 총 폴리페놀 함량을 갖는 참취 추출물을 얻을 수 있었다. 마이크로웨이브 추출 시 최적 마이크로웨이브 에너지는 120∼150 W였고 추출시간은 4∼8분이 적당하였다. 추출에 사용한 용매들 가운데 에탄올, 메탄을 보다 물 그리고 물과 에탄을 또는 메탄을 혼합용매를 사용한 추출물의 가용성 고형분, 총 폴리페놀 함량 및 항산화 효과가 높은 것으로 나타났다.

  • 나비류 추출물은 전통적으로 다양한 활성을 보유하여 의약용으로 사용되어졌다. 5종의 나비(산제비나비[Papilio maackii], 호랑나비[Papilio xuthus], 배추흰나비[Pieris rapae], 남방호랑나비[Eurema hecabe], 왕오색나비[Sasakia charonda])를 이용하여 물, dimethly sulfoxide (DMSO), ehtanol 및 methanol로 추출한 추출물로 항산화 활성 및 Cox-2 promoter assay를 수행하였다. 그 결과 산제비나비의 추출물이 전반적으로 높은 항산화활성을 나타내었으며, Cox-2 promoter assay에서는 호랑나비의 DMSO 분획이 가장 저해활성이 높은 것으로 나타났다. 활성이 높은 추출물을 대상으로 분리 및 정제를 통하여 유용한 식의약 소재로의 기초연구가 더 필요하다.

Privacy-Preservation Using Group Signature for Incentive Mechanisms in Mobile Crowd Sensing

  • Recently, concomitant with a surge in numbers of Internet of Things (IoT) devices with various sensors, mobile crowdsensing (MCS) has provided a new business model for IoT. For example, a person can share road traffic pictures taken with their smartphone via a cloud computing system and the MCS data can provide benefits to other consumers. In this service model, to encourage people to actively engage in sensing activities and to voluntarily share their sensing data, providing appropriate incentives is very important. However, the sensing data from personal devices can be sensitive to privacy, and thus the privacy issue can suppress data sharing. Therefore, the development of an appropriate privacy protection system is essential for successful MCS. In this study, we address this problem due to the conflicting objectives of privacy preservation and incentive payment. We propose a privacy-preserving mechanism that protects identity and location privacy of sensing users through an on-demand incentive payment and group signatures methods. Subsequently, we apply the proposed mechanism to one example of MCS-an intelligent parking system-and demonstrate the feasibility and efficiency of our mechanism through emulation.

Ameliorative Effect of Chitosan Complex on Miniature Pig Sperm Cryopreservation

  • Cryopreservation is mainly used for preservation of boar sperm. However, this method stresses the sperm by reactive oxygen species (ROS), and the conception rate and the litter size are not more efficient than the liquid preservation of spermatozoa. Therefore, we use chitosan which is a natural product derived antioxidant compound. We used GnHA (chitosan+hyaluronic acid) and GnHG (chitosan hydrogel) as chitosan complexes to cryopreserve boar sperm for improve sperm metabolism and function. Sperm parameter (sperm motility, progressive motility, path velocity, straight-line velocity, curvilinear velocity) is measured by computer-assisted sperm analysis (CASA) using frozen sperm with GnHA or GnHG (0, 0.25, 0.5, 1 mg/mL), respectively. Also, lipid peroxidation analysis using malondialdehyde (MDA) is performed to confirm the antioxidative effect of chitosan in frozen spermatozoa. CASA analysis showed GnHA and GnHG are effective against cryopreserved boar sperm. And antioxidant effect is measured by lipid peroxidation analysis. GnHA and GnHG, which is chitosan complex are effective for boar sperm cryopreservation by antioxidant effect.

Bone remodeling of the fibula segment as a form of neocondyle after free vascularized bone transfer: a report of two cases

  • The temporomandibular joint is a unique structure composed of the joint capsule, articular disc, mandibular condyles, glenoid fossa of the temporal bone, surrounding ligaments, and associated muscles. The condyle is one of the major components of a functional temporomandibular joint. Reconstruction of large mandibular defects involving the condyle is a surgical challenge for oral and maxillofacial surgeons. To restore large mandibular defects, there are different options for free flap method such as fibula, scapula, and iliac crest. Currently, the vascularized fibula free flap is the gold standard for reconstruction of complex mandibular defects involving the condyle. In the present report, neocondyle regeneration after mandible reconstruction including the condyle head with fibula free flap was evaluated. In this report, two patients were evaluated periodically, and remodeling of the distal end of the free fibula was observed in both cases after condylectomy or mandibulectomy. With preservation of the articular disc, trapezoidal shaping of the neocondyle, and elastic guidance of occlusion, neocondyle bone regeneration occured without ankylosis. Preservation of the articular disc and maintenance of proper occlusion are critical factors in regeneration of the neocondyle after mandible reconstruction.

Isolation and characterization of a lytic Salmonella Typhimurium-specific phage as a potential biofilm control agent

  • This study aimed to characterize a lytic Salmonella Typhimurium-specific (ST) phage and its biofilm control capability against S. Typhimurium biofilm on polypropylene surface. ST phage was isolated, propagated, and purified from water used in a slaughterhouse. The morphology of ST phage was observed via transmission electron microscopy. Its bactericidal effect was evaluated by determining bacterial concentrations after the phage treatment at various multiplicities of infection (MOIs) of 0.01, 1.0, and 100. Once the biofilm was formed on the polypropylene tube after incubation at 37℃ for 48 h, the phage was treated and its antibiofilm capability was determined using crystal violet staining and plate count method. The phage was isolated and purified at a final concentration of ~11 log PFU/mL. It was identified as a myophage with an icosahedral head (~104 nm) and contractile tail (~90-115 nm). ST phage could significantly decrease S. Typhimurium population by ~2.8 log CFU/mL at an MOI of 100. After incubation for 48 h, biofilm formation on polypropylene surface was confirmed with a bacterial population of ~6.9 log CFU/cm2. After 1 h treatment with ST phage, the bacterial population in the biofilm was reduced by 2.8 log CFU/cm2. Therefore, these results suggest that lytic ST phage as a promising biofilm control agent for eradicating S. Typhimurium biofilm formed on food contact surfaces.

Non-destructive quality prediction of domestic, commercial red pepper powder using hyperspectral imaging

  • We analyzed the major quality characteristics of red pepper powders from various regions and predicted these characteristics nondestructively using shortwave infrared hyperspectral imaging (HSI) technology. We conducted partial least squares regression analysis on 70% (n=71) of the acquired hyperspectral data of the red pepper powders to examine the major quality characteristics. Rc2 values of ≥0.8 were obtained for the ASTA color value (0.9263) and capsaicinoid content (0.8310). The developed quality prediction model was validated using the remaining 30% (n=35) of the hyperspectral data; the highest accuracy was achieved for the ASTA color value (Rp2=0.8488), and similar validity levels were achieved for the capsaicinoid and moisture contents. To increase the accuracy of the quality prediction model, we conducted spectrum preprocessing using SNV, MSC, SG-1, and SG-2, and the model's accuracy was verified. The results indicated that the accuracy of the model was most significantly improved by the MSC method, and the prediction accuracy for the ASTA color value was the highest for all the spectrum preprocessing methods. Our findings suggest that the quality characteristics of red pepper powders, even powders that do not conform to specific variables such as particle size and moisture content, can be predicted via HSI.

Effects of storage temperature on quality characteristics of texturized vegetable protein

  • This study evaluated the impact of storage temperature on the quality characteristics of texturized vegetable protein (TVP). TVP was prepared by mixing defatted Daewon soybean flour at 80℃, gluten, and corn starch in a 5:3:2 ratio, which was then extruded at a screw speed of 250 rpm and a barrel temperature of 190℃ with moisture addition at 9 rpm. Subsequently, the extruded TVP was vacuum-sealed in polyethylene packaging and stored at -20℃, 0℃, and 4℃ for 9 days. Texture analysis revealed that the curing rate followed 4℃ > 0℃ > -20℃ sequence. No significant color variation was observed across the storage conditions, although water content increased at all temperatures. Notable changes were detected in moisture absorption capacity (%) and solid leaching (%), following the order of -20℃ > 0℃ > 4℃. The turbidity of the solution released during cooking varied, with the highest to the lowest sequence being -20℃ > 4℃ > 0℃, while pH levels remained neutral. Regarding free amino acids, sweetness and textural quality improved with storage across all temperatures, whereas bitterness components diminished at 4℃. The study suggests that refrigerated storage at 4℃ is a viable method for distributing TVP, which was previously distributed only in a frozen and dry state.

Influence of temperature, time, and moisture content on rheology of tomatoes and pepper purees

  • This study explored how temperature, time, and moisture content affect the rheological properties (apparent viscosity, flow behavior index, and consistency coefficient) of stored tomato and pepper purees. These purees were prepared with moisture contents of 85%, 90%, and 95% (w/v) using the hot-break method and tested over 6 days at 2-day intervals and temperatures of 5℃, 10℃, and 15℃. Results displayed distinct ranges for apparent viscosity, consistency coefficient, and flow behavior indices: tomato puree (2,519.9-4,324.6 mPa·s, 258.0-550.6 mPa·Sn, 1.80-0.48) and pepper puree (2,105.6-4,562.0 mPa·s, 268.4-580.4 mPa·Sn, 0.22-0.48). The temperature and storage time had significant (p≤0.05) effects, but moisture content did not affect these properties. Flow behavior and consistency coefficients demonstrated relative variation with apparent viscosity, indicating pseudoplastic behavior. Optimal processing and storage conditions were identified within specific ranges: 13.21-14.42℃ for 2 days with 92.22-94.23% (w/v) moisture content for pepper, and 8.42-11.77℃ for 2-6 days with 85% (w/v) moisture for tomato.

삼요드티로닌을 포함한 폐보존액을 이용한 20시간 폐보존 - 새로운 폐 보존액의 개발 II - (Successful 20 hours Canine Allograft Preservation with new Solution Containing Triiodothyronine - Development of new lung preservation solution II -)

  • 배경: 폐장의 허혈-재관류 손상은 폐이식에서 발생하는 조기 이식장기 실패의 주요 원인의 하나로 알려져 있다. 최근 갑상선 호르몬의 활성형인 삼요드티로닌 (T3)이 심장을 비롯한 여러 장기의 허혈 손상을 줄여주는 효과가 있음을 시사하는 보고가 있다. 본 연구에서는 이러한 T3의 허혈-재관류 손상에 대한 효과가 폐장의 허혈 손상에도 효과가 있을 것으로 기대하고 덱스트란을 주성분으로 하는 세포외액성 폐보존액에 T3를 추가한 새로운 폐보존액을 제조하여 그 효과를 검증하고자 하였다. 대상 및 방법: 12마리의 황견을 6마리씩 두 군으로 나누어 제 1군에서는 새로 개발한 폐보존액을 사용하고, 제 2군에서는 유로콜린스 용액을 사용하여 폐를 적출 하였다. 적출한 폐장은 각각의 보존액에 담그어 섭씨 4도에서 20시간 보관한 후, 각 군에 6마리씩 총 12례의 좌측 폐이식을 시행하였다. 이식된 좌측폐만의 기능을 관찰하기 위해서 폐이식 후 재관류를 15분시킨 후에 우측 주폐동맥과 우측 주기관지를 결찰하고, 2시간 동안 혈역학적 변수와 가스분석을 시행하고, 측정이 종료된 후 바로 희생하여 폐조직 일부를 떼어내어 조직검사와 수분 함량 및 MDA양을 측정 비교하였다. 결과: 동맥혈 산소분압은 제 1군에서 재관류 후 60분, 90분 120분에 각각 147$\pm$25 mmHg, 148$\pm$22 mmHg, 159$\pm$21 mmHg, 제 2군에서는 각각 133$\pm$26 mmHg, 132$\pm$29 mmHg, 135$\pm$30 mmHg로 제 1군에서 조금 높은 경향을 보였으나 통계적으로 유의성은 없었다. 각 시간에서의 최대 흡기압은 제 1군에서 14.0$\pm$0.5 cmH2O, 14.2$\pm$0.6 cmH2O, 15.7$\pm$0.8 cmH2O, 제 2군에서는 17.8$\pm$2.0 cmH2O, 18.0$\pm$1.9 cmH2O, 19.3$\pm$2.7 cmH2O으로 제 1군에서 조금 낮은 경향을 보였으나 통계적 유의성은 없었다. 동맥혈 이산화탄소 분압은 제 1군에서 각 시간에 27.9$\pm$2.2 mmHg, 27.7$\pm$2.4 mmHg, 28.0$\pm$3.0 mmHg, 제 2군에서는 36.8$\pm$6.0 mmHg, 43.2$\pm$8.1 mmHg, 53.1$\pm$17.4 mmHg로 제 1군에서 유의하게 낮았다 (p<0.05). 폐혈관 저항 및 조직 MDA양은 두 군간에 유의한 차이가 없었다. 폐 조직 수분 함량은 제 1군에서 유의하게 낮았고, 조직학 검사상 폐조직 손상의 정도도 제 1군에서 적었다. 결론: 이상의 결과에서 T3를 포함한 새로 개발한 폐보존액이 유로콜린스 용액과 비교하여 폐보존능이 우수함을 확인할 수 있었고, 이는 T3가 폐이식시 폐장의 효과적 보존에 유용한 역할을 함을 시사하였다.
