Preservative Effect of Soybean Sprouts Pre-soaked and Cultivated in the Solution of Natural Antimicrobial Mixture (천연 항균복합제재용액을 침지 및 재배용수로 처리한 콩나물의 선도유지 효과)

  • The antimicrobial effect of mixed solution of botanical antimicrobial agent-citrus product (BAAC) and such a natural additive as aloe, ginseng or Prunus mume extract on the spoilage microorganisms of soybean sprouts were investigated by paper disk method. The mixture (BAAG) of BAAC and ginseng extract showed the remarkable antimicrobial activity in the result of the experiment. Therefore, we used BAAG as pre-soaking and cultivating solution of soybean sprouts. Total bacterial and Escherichia coli cell count of soybean sprouts pre-soaked and cultivated in the BAAG-diluted solution (50 ppm) showed 2.5 ${\times}$ 10 CFU/mL and 1.3 ${\times}$ 10 cfu/mL in comparison with 3.5 ${\times}$ 10$^3$ cfu/mL and 5.8 ${\times}$ 10$^3$ cfu/mL of the control, respectively. BAAG-treated soybean sprouts also showed no slimy brown product and undesirable odor characterized in the control. It was confirmed that BAAG could be a proper pre-soaking and cultivating solution of soybean sprouts.

The Effects of Thiamin on the Fruiting of Lentinula edodes (표고버섯 자실체 형성에 미치는 티아민의 영향)

  • The investigation was carried out to identify the active constituent in yeast extract for fruit body formation of Lentinula edodes. The result suggests that free thiamin which is hewn as the active substance for the fruiting of L edodes, was detected but thiamin mono, di, three phosphates were not detected in the yeast extract produced by Difco Co.. Therefore, the thiamin content of the yeast extract was determined, the yeast extract was fractionated to five portion by the post-column fluorescence method. The content of thiamin in yeast extract( 1g) was 0.436mg as thiamin hydrochloride. It was found that 76% of the total thiamin(0.332mg) was contained in fraction II. About 20% of the total thiamin(0.087mg) was present in fraction I, but not in fractions III, IV and V. In accordance with the contents of thiamin in the fractions, the fruit body formation was the highest by the treatment of fraction II(100%) and followed by fraction I (60%), V(50%), III(30%). Thiamin did not influence for the vegetative mycelial growth of L. edodes, but be used for fruit body formation.

Effects of Supercritical Carbon Dioxide on Sterilization and Enzyme Inactivation in Dongchimi (초임계 이산화탄소의 동치미 살균 및 효소 불활 효과)

  • We investigated how supercritical $CO_2$ affected sterilization and enzyme inactivation in dongchimi, a conventional Korean fermented food. The growth of bacteria, including lactic acid bacteria, in dongchimi juice tended to decrease with increased pressure and temperature during treatment with supercritical $CO_2$. D values were affected by pressure more than by temperature. The lowest total number of cells and D values of lactic acid bacteria were observed after treatment with supercritical $CO_2$ at 25 MPa and $25^{\circ}C$; these conditions also reduced polygalacturonase activity in radishes by approximately 40.3%. Supercritical $CO_2$ can be used as an alternative method of sterilization and enzyme inactivation, minimizing sensory loss and textural changes in vegetable materials.

Quality change of sliced ginseng depending on different drying methods (건조방법에 따른 홍삼 절편의 품질 변화)

  • This study investigated quality change of ginseng depending on far-infrared (FIR) drying methods. The ginseng was cut into 2 mm slices and measured moisture content (MC), color, total polyphenol content (TPC), total flavonoid content (TFC) and DPPH radical scavenging activity (DPPH). The color values (L, a, and b) were decreased after far-infrared (FIR) drying. As increase in drying time, color was changed. Considering quality parameters including MC, TPC, TFC, and DPPH, the optimum condition for drying was set at $65^{\circ}C$ for 60 min. Furthermore, we compared color and functional compounds depending on drying process including FIR, freeze-, vacuum, and hot-air drying. The color values of ginseng were decreased when dried using vacuum and hot-air. While the color values of ginseng were increased after freeze drying. Comparison with another drying method, FIR drying presented the highest color difference. FIR and vacuum-dried ginseng indicate higher contents of TPC and TFC than freeze- and hot air-dried ginseng. And hot-air-dried ginseng present the highest DPPH when compared to another drying methods. On the other hand, freeze dried ginseng show the lowest TPC, TFC, and DPPH.

  • Protected Flora are natural heritage that involve a wide variety of valuable natural values and cultural values. Over time, "Cultural Properties Preservation Law" has been changing the definition and the criteria of Protected Flora. In addition to the academic value and the rarity of the plant, historical, ethnic and landscape values have become increasingly important. For that reason the declaration and management processes of the Protected Flora need to be improved. However, the current statute has focused on the morphological characteristics of the plants which have a limit to encompass various values of the plant. It also does not correspond to the definition of Protected Flora and its declaration criteria stated on the "Cultural Properties Preservation Law." For this reason, the main purpose of this study is to develop new types of Protected Flora so that various Protected Flora can be discovered in the future.

Effects of Amount of Slaking Water on Physical and Chemical Properties of Handmade Hydrated Lime used for Preservation of Architectural Heritage (소화에 사용되는 물의 양이 건축 문화재 보존용 수제 소석회의 물리 및 화학적 특성에 미치는 영향)

  • Handmade hydrated lime has been used for preservation and repair of architectural heritage in Korea. However, the effect of the amount of water used for slaking quicklime on the physical and chemical properties of the hydrated lime, which is the result of the slaking process, has not been clearly understood. In this study, particle size distribution, chemical composition and crystalline phases of the hydrated lime are investigated by varying the amount of water used for the slaking. In addition, temperature history during the slaking process is examined. For this, various experiments, such as laser diffraction analysis, X-ray fluorescence, X-ray diffraction, thermogravimetric analysis, and temperature recording using a thermocouple, were performed. When the quicklime came into contact with water, its temperature reached $100^{\circ}C$ within 10 min due to sudden exothermic reaction of calcium oxide, and this temperature was maintained for about 30 min. The water to lime ratio influenced the cooling rate during the slaking process; that is, the more water was used, the longer it took to reach an ambient temperature. The amount of water for the slaking did not have a noticeable effect on the contents of major components of the hydrated lime such as calcium hydroxide and calcium carbonate, but when slaked with more amount of water, average particle size of the lime tended to decrease. The experimental results in this study can be used as references for developing guidelines on the safety or appropriate amount of water in the lime slaking process.

  • Architectural cultural properties suffer a lot of damage due to various environmental factors. In order to preserve damaged cultural properties, preventive preservation and long-term preservation management are becoming more important. Therefore, research on a scientific non-destructive testing method applicable to regular inspection is required. For related research, DangGan with a high flag-pole shape was selected as the subject of study among various cultural properties. Among the preserved DangGans, a basic study was conducted on the analysis technique to evaluate the structural stability by selecting Treasure No. 49 Naju SeokDangGan. An idealized model was presented and a multi-degree of freedom equation of motion was derived. In addition, an equation for estimating the critical stiffness value for each joint position is presented.

Effect of a dual trigger on oocyte maturation in young women with decreased ovarian reserve for the purpose of elective oocyte cryopreservation

  • Objective: The aim of this study was to determine whether co-administration of a gonadotropin-releasing hormone (GnRH) agonist and human chorionic gonadotropin (hCG) for final oocyte maturation improved mature oocyte cryopreservation outcomes in young women with decreased ovarian reserve (DOR) compared with hCG alone. Methods: Between January 2016 and August 2019, controlled ovarian stimulation (COS) cycles in women (aged ≤35 years, anti-Müllerian hormone [AMH] <1.2 ng/mL) who underwent elective oocyte cryopreservation for fertility preservation were retrospectively analyzed. Results: A total of 76 COS cycles were triggered with a GnRH agonist and hCG (the dual group) or hCG alone (the hCG group). The mean age and serum AMH levels were comparable between the two groups. The duration of stimulation, total dose of follicle-stimulating hormone used, and total number of oocytes retrieved were similar. However, the number of mature oocytes retrieved and the oocyte maturation rate were significantly higher in the dual group than in the hCG group (p=0.010 and p<0.001). After controlling for confounders, the dual-trigger method remained a significant factor related to the number of mature oocytes retrieved (p=0.016). Conclusion: We showed improved mature oocyte collection and maturation rate with the dual triggering of oocyte maturation in young women with DOR. A dual trigger appears to be more beneficial than hCG alone in terms of mature oocyte cryopreservation for young women with DOR.

A Study on the Email Management of Undergraduate Students (대학생의 이메일 관리 행태 연구)

  • This study examines email records management patterns of undergraduate students and their perceptions of university email policy. The research collected data from 317 undergraduate students using a survey based on the five essential elements of email records management: common elements, email creation, email management, email preservation, and perception. Also, an in-depth interview was conducted with 6 undergraduate students, according to the method of university email service: Google, Microsoft, and in-house webmail. As a result, undergraduate students used email as an address book, cloud storage, file storage, and collaboration tool, and they had difficulties in systematically managing the email records according to the records lifecycle. Also, there was a tendency to prefer using an external email platform that can use additional functions to in-house webmail that has limitations in mobile service and preservation of attachments. Thus, this study suggests ways to help undergraduate students manage their email records, including maintaining graduates' email accounts, providing sufficient email storage, and providing email records management training and guidelines for undergraduate students.

Effects of chlorine dioxide gas fumigation on the storage quality of astringent persimmon (Diospyros kaki T.) Cheongdobansi

  • Because of their short harvest season, large quantities of persimmons must often be processed within a limited time. Therefore, new methods to extend their storage life are required. This study examined the effects of chlorine dioxide (ClO2) gas fumigation for various treatment periods on the storage quality of astringent persimmons Cheongdobansi under low-temperature conditions. The conditions consisted of continuous treatment with ClO2, treatment for 2 weeks with ClO2, and no treatment, all of which are stored at low temperatures. Control samples (storage 0 days) without any treatment were prepared and all experiments were conducted for 10 weeks at two-week intervals. The ClO2 gas treatment maintained the moisture content, color value, hardness, soluble tannin content, and sensory characteristics. However, ClO2 gas treatment did not affect the soluble solids, pH, and total sugar content. In particular, continuous treatment with ClO2 maintained the storage quality after 6-8 weeks of storage, particularly the hardness and weakness (sensory evaluation). The results suggest the potential of continuous treatment with ClO2 as a highly effective method for maintaining the freshness of Cheongdobansi.
