Fenofibrate reduces adiposity in pregnant and virgin rats but through different mechanisms

  • Fenofibrate has been proven to reduce adiposity. Since gestation produces an increase in white adipose tissue (WAT) mass, we comparatively studied this drug-effect in virgin and pregnant rats. Fenofibrate reduced lumbar WAT weight in both pregnant and virgin rats. Fenofibrate treatment did not modify plasma free fatty acid (FFA) concentration in virgin rats, it greatly increased it in pregnant animals. Remarkable differences between the two groups were obtained for two proteins related to fatty acid oxidation and esterification and storing. Respectively, the mRNA levels of carnitine palmitoyltransferase I (CPT-I) were increased by the fenofibrate only in the virgin rats and a similar finding was observed for the expression of phosphoenolpyruvate carboxykinase (PEPCK). These findings indicate that fenofibrate reduces adiposity in pregnant and virgin rats through different mechanisms: a) in virgin rats, by promoting fatty acid oxidation; and b) in pregnant rats, by enhancing fatty acid output.

  • The reproductive effect of $Telazol^{\circledR}$ was studied in pregnant rats. Two different doses (20 or 40 mg/kg/day) were injected on days one of 3, 5 or 8 of pregnancy and 3 consecutive days of 3~5 or 5~7 in pregnant rats. On days 8 or 21, the rats were anesthetized, the cesarean section was performed, the uterus was carefully examined, and the numbers of corpora lutea, implantation sites and viable fetuses, fetal and placental weights and pre-and post-implantation losses in rats were surveyed. Simultaneously the concentrations of sodium and potassium of the plasma, uterine and amniotic fluids were determined. And gestation period in pregnant rats, postnatal offspring survival and body weight gains were evaluated. Administration of $Telazol^{\circledR}$ did not exert a bad influence upon body weight gains in pregnant rats. Reproductive indices were normal. The concentrations of sodium and potassium of the plasma, uterine and amniotic fluids in pregnant rats remained unchanged. Gestation period of rats, postnatal offspring survival and body weight gains were normal. It is concluded that $Telazol^{\circledR}$ is devoided of significant adverse reproductive effects in pregnant rats.

Transforming growth factor-beta and liver injury in an arginine vasopressin-induced pregnant rat model

  • Objective: Approximately 30% of preeclamptic pregnancies exhibit abnormal liver function tests. We assessed liver injury-associated enzyme levels and circulating transforming growth factor beta (TGF-β) levels in an arginine vasopressin (AVP)-induced pregnant Sprague-Dawley rat model. Methods: Pregnant and non-pregnant Sprague-Dawley rats (n=24) received AVP (150 ng/hr) subcutaneously via mini-osmotic pumps for 18 days. Blood pressure was measured, urine samples were collected, and all animals were euthanized via isoflurane. Blood was collected to measure circulating levels of TGF-β1-3 isomers and liver injury enzymes in pregnant AVP (PAVP), pregnant saline (PS), non-pregnant AVP (NAVP), and non-pregnant saline (NS) rats. Results: The PAVP group showed significantly higher systolic and diastolic blood pressure than both saline-treated groups. The weight per pup was significantly lower in the AVP-treated group than in the saline group (p<0.05). Circulating TGF-β1-3 isomer levels were significantly higher in the PAVP rats than in the NS rats. However, similar TGF-β1 and TGF-β3 levels were noted in the PS and PAVP rats, while TGF-β2 levels were significantly higher in the PAVP rats. Circulating liver-type arginase-1 and 5'-nucleotidase levels were higher in the PAVP rats than in the saline group. Conclusion: This is the first study to demonstrate higher levels of TGF-β2, arginase, and 5'-nucleotidase activity in PAVP than in PS rats. AVP may cause vasoconstriction and increase peripheral resistance and blood pressure, thereby elevating TGF-β and inducing the preeclampsia-associated inflammatory response. Future studies should explore the mechanisms through which AVP dysregulates liver injury enzymes and TGF-β in pregnant rats.

PMSG를 투여한 미성숙랫드에 발생한 자궁축농증 발생례 (Pyometra in the PMSG-treated Immature Female Rats)

  • Nine immature 30-day-old female rats were injected sc at 0800 hr with pregnant mare serum gonadotrophin(PMSG) to induce ovulation and mating. Fifty-six hours later the animals were placed with mature male rats overnight (one female and one male). Five of 9 immature female rats treated with PMSG were pregnant and allowed to maintain the pregnancy to term. Three of 5 pregnant rats were failed to maintain pregnancy to term. Two of 5 pregnant rats seemed to be developed normally and increased abdominal enlargement as pregnancy progresses, but did not occurred parturition on day of 43 or 48 of pregnancy, respectively. On day 44 or 49, pregnant rats were killed and examined uterus and ovaries. There was no fetus but approximately 50∼60ml. of mucopurulent fluids were accumulated in the uterine cavity and 40 or 42 corpora lutea persisted in the ovaries. Pyometra was developed after coitus in PMSG-treated immature female rat.

Valproic Acid Exposure of Pregnant Rats During Organogenesis Disturbs Pancreas Development in Insulin Synthesis and Secretion of the Offspring

  • Valproic acid (VPA) plays a role in histone modifications that eventually inhibit the activity of histone deacetylase (HDAC), and will affect the expressions of genes Pdx1, Nkx6.1, and Ngn3 during pancreatic organogenesis. This experiment was designed to study the effect of VPA exposure in pregnant rats on the activity of HDAC that controls the expression of genes regulating the development of beta cells in the pancreas to synthesize and secrete insulin. This study used 30 pregnant Sprague-Dawley rats, divided into 4 groups, as follows: (1) a control group of pregnant rats without VPA administration, (2) pregnant rats administered with 250 mg VPA on day 10 of pregnancy, (3) pregnant rats administered with 250 mg VPA on day 13 of pregnancy, and (4) pregnant rats administered with 250 mg VPA on day 16 of pregnancy. Eighty-four newborn rats born to control rats and rats administered with VPA on days 10, 13, and 16 of pregnancy were used to measure serum glucose, insulin, DNA, RNA, and ratio of RNA/DNA concentrations in the pancreas and to observe the microscopical condition of the pancreas at the ages of 4 to 32 weeks postpartum with 4-week intervals. The results showed that at the age of 32 weeks, the offspring of pregnant rats administered with 250 mg VPA on days 10, 13, and 16 of pregnancy had higher serum glucose concentrations and lower serum insulin concentrations, followed by decreased concentrations of RNA, and the ratio of RNA/DNA in the pancreas. Microscopical observations showed that the pancreas of the rats born to pregnant rats administered with VPA during pregnancy had low immunoreaction to insulin. The exposure of pregnant rats to VPA during pregnancy disturbs organogenesis of the pancreas of the embryos that eventually disturb the insulin production in the beta cells indicated by the decreased insulin secretion during postnatal life.

익모초(益母草)와 도인(桃仁)이 임신 흰쥐에 미치는 영향 (Effect of Leonuri Sibirici Herba and Pruni Persicae Semen On Pregnant Rats)

  • Objectives : The present study has been undertaken to investigate the effects of Leonuri Sibirici Herba and Pruni Persicae Semen on pregnant rats. Method : In this experiment, the pregnant rats were administered by water extracts of Leonuri Sibirici Herba and Pruni Persicae Semen. The levels of weights, ALT, AST, ALP, BUN, creatinine, progesterone, Na and K in serum and reproductive indices of the rats were measured after treatment. Results : 1. The levels of body weight gains were not significantly changed in comparison with Control group in Leonuri Sibirici Herba group and Pruni Persicae Semen group. 2. In the levels of reproductive indices of the rats, the number of corpora lutea, implantation, viable fetuses, pre-implantation loss, post-implantation loss, fetal weight and placental wight were not significantly changed in comparison with Control group in Leonuri Sibirici Herba group and Pruni Persicae Semen group. 3. The levels of BUN, creatine, ALT, AST and ALP were not significantly changed in comparison with Control group in Leonuri Sibirici Herba group and Pruni Persicae Semen group. 4. The level of progesterone was not significantly changed in comparison with Control group in Leonuri Sibirici Herba group and Pruni Persicae Semen group. 5. The levels of Na and K were not significantly changed in comparison with Control group in Leonuri Sibirici Herba group and Pruni Persicae Semen group. Conclusion : Reviewing these experimetal results, it appeared that Leonuri Sibirici Herba and Pruni Persicae Semen had not toxicity on pregnant rats.

Streptozotocin 유발 당뇨 흰쥐의 태자 간 발생 및 호흡에 미치는 Taheebo의 영향 (The Effects of Taheebo on the Streptozotocin-Induced Diabetes in the Rats during Pregnancy and in the Fetal Life)

  • The effects of Taheebo on the diabetic-piegnant rats and their fetus was investigated. It has been reported that diabetic condition of the pregnant rats can affect the process of liver formation and damage the respiratory function in the fetus. Therefore we investigated the effects of Taheebo on the prevention of liver damage and respiratory failure in the fetus and those results were compared with that of dexamethasone (DXM). In pregnant rats, streptozotocin(STZ, 45 mg/kg, 0.01 M citrate buffer) was injected into the pregnant rats on the third day of pregnancy. Methanol extracts of Taheebo(500 mg/kg p.o.) was administered once daily during pregnancy. DXM (10 $\mu\textrm{g}$/g i.p.) was injected into the pregnant rats in 16th and 18th days of pregnancy. Body weights were measured and fetal number and abortion rate in pregnancy rats were determined. Lecithin/sphingomyelin ratio in amniotic fluid and malondialdehyde, glycogen, triglyceride, protein and cholesterol levels in the liver homogenate were determined. Also blood glucose level was analyzed. Body weights of maternal rats were increased in the all groups except the DXM group. Fetal number of the Taheebo treated group was similar to the control group, and a significant increase in the body weights of fetus was observed in the STZ treated group and the Taheebo treated group compared with the control group. Blood glucose of fetus produced hypoglycemia in the control group and hyperglycemia in the diabetic-pregnant rats. The protein and cholesterol levels in fetus liver were significantly increased in the DXM treated group compared with the control group. Triglyceride content was significantly increased in all groups compared with the control group. Liver malondialdehyde level of fetus in the STZ treated group was similar to the control group. Glycogen level was significantly increased in the all groups compared with the control group. Methanol extract of Taheebo showed hypoglycemic effect on the pregnant rats. However, we could not observe any hypoglycemic effect on the fetus. There's no difference between the control and Taheebo treated group in terms of the levels of triglyceride, cholesterol, protein and glycogen in the fetus liver. Further study to identify the effect of Taheebo on the fetus is under investigation.

  • The effect of corticosteroid on the diabetic pregnant rats and their fetuses was investigated. Streptozotocin (STB) was injected into the pregnant rats on the fifth day of pregnancy. Dexamethasone (DXM) was injected into the pregnant rats on the 17th, 18th, 19th and 20th days of pregnancy In prenatal rats, the body weight, an abortion rate, number of fetus, the ratio of lecithin/sphingomyelin (L/S) and the levels of blood glucose and phosphatidylglycerol (PG) were determined. In the postnatal rats, the body weight, the levels of blood glucose, fetal number, stillbirth rate, an organ weight and the levels of hepatic glycogen, protein and triglyceride were determined. The body weight of fetuses was lower in the DXM group and higher in the STZ group than the those of control group. Blood glucose of fetuses produced hypoglycemia in the STZ group compared with the control group. A significant increase in the abortion and stillbirth rates was observed in STZ group. The levels of glycogen, protein and triglyceride in fetus liver and the weight of pancreas were significantly increased in the 572 and STZ+DXM groups compared with the control group. The L/S ratio and the level of PG in the amniotic (quid were significantly decreased in STZ group compared with the control group, whereas those of the STZ+DXM group were similar to the control group. It has been observed that corticosteroid administration on the STZ-induced diabetic rats during final stage of pregnancy can prevent the respiratory depression syndrome of neonatal rats.

안존이천탕 추출물이 흰쥐의 모체 및 태자에 미치는 영향 (The Toxicological Effects of Ahnjon-Yichun-Tang in Pregnant Rats and Fetuses)

  • The objective of this study was to characterize the adverse effects of Ahnjon-Yichun-Tang during early pregnancy. Following successful mating, female Sprague-Dawley rats were given Ahnjon-Yichun-Tang(AYT) extract by oral administration daily with dose of 150mg (n=10), 300mg(n=10), 450mg(n=10) for 20 days of pregnancy. The rats in Control group(n=10) were orally administrated with Saline. All pregnant rats of Ahnjon-Yichun-Tang-treated and Control groups were sacrificed on day 20 of pregnancy. The pregnancy outcome was determined and the internal and reproductive organs of pregnant rat were observed. The fetuses were examined for the presence of various developmental toxic endpoints and stained with alcian blue and alizarin red S, and observed skeletal malformations. The results obtained in this study represent that there is no significant changes between Control and Ahnjon-Yichun-Tang-treated groups in body weight, organ weight, blood chemistry values, hematological values and pregnancy indexes of pregnant rat. The skeletal malformation of fetus was not observed as well. These results suggest that oral administration of Ahnjon-Yichun-Tang does not produce either maternal or developmental toxicity.
