N-Methylhydrasteine hydroxylactam and 1-methoxyberberine chloride, both alkaloids, extracted from Corydalis longipes have been assayed for their activities against two powdery mildews. The spore germination of Erysiphe cichoracearum on detached leaf of balsam(Impatiens balsaminia) following pre- and post-inoculation treatments by their mixture has shown high efficacy against the pathogen at 100, 200 and 300 ${\mu}g/ml$. The mixture was also effective at both pre- and post-inoculation treatments at 500, 1000, 1500 ${\mu}g/ml$ doses against E. pisi causing pea powdery mildew in pea(Pisum sativum) under field conditions. The significant efficacy of the mixture of two compounds against spore germination on detached leaves of balsam and also under field conditions in pea warrants its inclusion in trials against some other diseases under field conditions.
The inoculation effect of the highly nitrogen fixing strains of R. japonicum were tested in the 3 kinds of soil with different cultural history onto Gycine max cv. Jang-yeob. The nodulation and nitrogen fixation activity in 3 test soils all showed the great increase in inoculated group compared to the non-inoculated group. The plant dry weight of the in-oculated groups were increased about 10% than that of the non-inoculated groups. The numerical index of the increase in total nitrogen fixation activity were 238% in the pre-cultivated, 266% in the immatured and 157% in the matured soil and these results suggested the clear effect of inoculation. Among the strains tested, R. japonicum R214 and Rl38 showed the excellent inoculation effect.
Volatile fatty acids (VFAs) production from marine brown algae, Saccharina japonica, was investigated in anaerobic dark fermentation. In order to evaluate the VFAs productivity, various experimental parameters (i.e., physicochemical pre-treatment, microorganism inoculation ratio, substrate concentration, and pH) were evaluated. According to the physicochemical pre-treatment methods, the maximum concentrations of VFAs were obtained in the order of sulfuric acid, subcritical water and subcritical water with lipid-extraction. Also, we investigated the operating parameters such as microorganism inoculation ratio (MV/M = 10 to 30), the substrate concentration (18.0 to 72.0 g/L) and pH (6.0 to 7.0) in sulfuric acid pre-treatment method. When the substrate concentrations were 18.0, 36.0, 54.0 and 72.0 g/L at $35^{\circ}C$, microorganism inoculation ratio 15, pH 7.0 for 372 hours, the maximum concentrations of VFAs were respectively 9.8, 13.9, 18.6 and 22.3 g/L. The change in VFAs concentrations was detected that acetic- and propionic acids increased according to increasing pH, while the butyric acid increased with decreasing pH. The VFAs obtained from concentration and separation process may be used as basic chemistry materials and bio-fuel, and they will expect to produce alternative energy of fossil fuel.
Resistance inductions on the leaves of cucumber plant by an arbuscular mycorrhiza Glomus intraradices were investigated. In addition, the infection structures were observed at the penetration sites on the leaves of plant inoculated with Colletotrichum orbiculare using a fluorescence microscope. The severity of anthracnose disease caused by Colletotrichum orbiculare was significantly decreased on the leaves of cucumber plant colonized with G intraradices compared with those of non-treated control plants. As a positive control, pre-treatment with DL-3-aminobutyric acid (BABA) caused a remarkable reduction of the disease severity on the pathogen-inoculated leaves. There were no significant differences in the frequency of either germination or appressorium formation of the plant pathogen between mycorrhiza colonized and non-treated plants. It was also the same on the BABA pre-treated plants. However, the frequency of callose formation was significantly high on the leaves of G intraradices colonized plants compared to those of non-treated control plants at 5 days after challenge inoculation. On the leaves of BABA treated plants callose formation was not significantly high than those of non-treated, although the disease severity was more strongly suppressed. It was suggested that the resistance induced by colonization with G. intraradices might be related to the enhancement of callose formation at the penetrate sites on the leaves invaded by the pathogen, whereas resistance by BABA did not.
Gerber (Gerbera hybrida) is a valuable ornamental species grown as a potted plant and cut flowers. However, genetic variability within the gerbera genus is very limited. So it is absolutely needed to introduce and widen genetic resources into gerbera lines by genetic transformation. For the purpose, 18 Korean gerbera lines were screened to establish Agrobacterium-mediated genetic transformation procedure. In an experiment to select Korean gerbera lines which are amenable to Agrobacterium-inoculation, 12 lines turned out to be positive in Agrobacterium-inoculation. More callus were produced from BA 2ppm, Zeatin 2ppm, IAA 0.2ppm in pre-culture and regeneration medium (2X media) but there was no difference in the frequency of GUS expression rate. In another experiment to find out optimal condition for highly efficient Agrobacterium-inoculation, petiole and leaf explants have been treated with four different pre-culture periods, two different co-culture periods and two different Agrobacterium strains. As a result, high GUS expression has been showed from petiole and leaf explants treated no pre-culture period with LBA4404 Agrobacterium tumerfaciens, 5 day co-culture period and dipping treatment.

Crude extracts and active principles from medicinal plants have shown potential role in controlling plant diseases in glasshouses as well as in fields as one of the safest and ecofriendly methods. The effect of nor-securinine (an alkaloid) isolated from Phyllanthus amarus has been seen against spore germination of some fungi (Alternaria brassicae, A. solani, Curvularia pennisetti, Curvularia sp., Erysiphe pisi, Helminthosporium frumentacei) as well as pea powdery mildew (Erysiphe pisi) under glasshouse conditions. The sensitivity of fungi to nor-securinine varied considerably. Nor-securinine was effective against most of the fungi. H. frumentacei was more sensitive even at the lowest concentration ($1,000\;{\mu}g/ml$). Likewise conidia of E. pisi were also inhibited in partially or completely appressorium formation. Pre-inoculation treatment showed greater efficacy than post-inoculation in inhibiting powdery mildew development on pea plants in a glasshouse. Maximum inhibition occurred at $2000\;{\mu}g/ml$.

Polycyclic aromatic hydrocarbons (PAHs) are a kind of toxic environmental pollutants and has been accumulated usually in marine sediments. Due to their potential hazardous to human, removal of PAHs from environments has been great concern. In the present study, the effect of microbial inoculation and the supplementation of mixed form cyclodextrin (M-CD) was assessed in the pre-sterilized or nonsterilized microcosms for optimizing operational conditions for ex situ bioremediation of sediments contaminated by PAHs. Activity of electron transport system (ETSA) was increased by the addition of M-CD regardless of inoculation of microorganisms in microcosms without sterilization. The degradation rate of PAHs in sterilized microcosms was app. 9-20% by the inoculation of single strain and 24-37% by the inoculation of microbial consortium supplemented with 1% M-CD, respectively. The degradation was not observed in microcosms without sterilization under the same conditions. The proportion of inoculated microorganisms also decreased in nonsterilized microcosms. Signals of inoculated bacteria were decreased to detection limit after 2 days in the microcosms without M-CD. In conclusion, microbial inoculation with appropriate carbon sources and removal of natural flora and grazers are required for the efficient ex situ bioremediation of sediments contaminated by PAHs in bioslurry reactor.
Pre-infection behavior of Fusarium circinatum on stems of pine species was investigated with scanning electron microscopy. Two-year-old stems of Pinus densiflora and p. rigida were inoculated with the fungal conidial suspension and subjected to $25^{\circ}C$ for up to 16 hr. Most microconidia germinated 12 hr after inoculation on pine stems. Conidia produced germ tubes from either one or both ends of microconidia. Germ tubes grew over the stem surface and appeared to enter host tissues through natural openings on pine stems. Surface cracks in the cork were entrance sites of germ tubes of F. circinatum. In addition, host cell wall cracks were often found at the tip of germ tubes. The cuticle appeared to be eroded either at the tip of germ tubes or around germlings. Germ tubes also produced appressoria-like structures, exhibiting swollen tips of germ tubes on the stem surface. There seems to be no significant differences in the pre-infection behavior of F. circinatum on stems between the two pine species.
Reproduction of the soybean cyst nematode (SCN), Heterodera glycines Ichinohe on the susceptible soybean cultivar, Lee 74, was significantly reduced by pre-, post- and simultaneous treatments of acetylsalicylic acid (ASA, aspirin). The control efficiencies were 60%, 64% and 87% for pre-, post- and simultaneous treatments, respectively. ASA had no significant effect on the survival of 2nd stage juveniles and their penetration into the soybean root tissues, but significantly inhibited the early stage nematode growth in the roots. Syncytia were formed 2∼3 days after inoculation in the susceptible soybean without ASA treatment, characterized by dense cytoplasm and increased cellular organelles such as mitochondria and endoplasmic reticulum. The nematode stylet was penetrated into the syncytial cell, and feeding tube was formed at the nematode stylet was penetrated into the syncytial cell, and feeding tube was formed at the nematode stylet entry. However, in the ASA treatments, syncytium was not formed or degenerated, depending on the root tissues. In the pre-treatments of ASA, nematode stylets did not penetrate into cells, showing callose-like cell wall thickening formed at the nematode probing sites, or retracted from the infected cells. The stylet penetration sites of syncytial cells appeared to be sealed off with fibrillar materials. With post-treatment of ASA, syncytia formed by the nematode were degenerated, characterized by degradation of syncytial cytoplasm.
Objective: To develop the fermentation quality and chemical composition of alfalfa (Medicago sativa Lam.) silage, plants were inoculated with different lactic acid bacteria (LAB) strains at field 24 hours before harvest. Methods: The treatment groups were as follow: silage without additive as a control and inoculated with each strains of Lactobacillus brevis (LS-55-2-2), Leuconostoc citerum (L. citerum; L-70-6-1), Lactobacillus bifermentans (L. bifermentans; LS-65-2-1), Lactobacillus plantarum (L. plantarum; LS-3-3) and L. plantarum (LS-72-2). All the silages were stored at 25℃. Parameters such as pH, microorganism and volatile fatty acid contents, crude protein, neutral detergent fiber, acid detergent fiber, net gas, metabolizable energy, organic matter digestibility, dry matter intake and relative feed value were measured to determine fermentation quality, chemical compositions and relative feed value of alfalfa silages. Results: Significant differences were found among the control and treated groups in terms of pH and microorganism contents at all opening times and crude protein, net gas, metabolizable energy and organic matter digestibility of final silage. The pH values ranged from 4.70 to 5.52 for all treatments and control silage had the highest value of overall treatments at T75d silages. Volatile fatty acid of silages was not influenced significantly by inoculations. However, lactic acid content of L. bifermentans (LS-65-2-1) was higher than the other treatments. The highest metabolizable energy and organic matter digestibility were recorded from L. citerum (L-70-6-1) inoculation. In addition, no significant differences were found among treatments in terms of neutral detergent fiber, acid detergent fiber, dry matter intake and relative feed value. Conclusion: Among the treated LAB isolates, L. bifermentans came into prominence especially in terms of organic acid composition and quality characters of silages.
