Avian influenza is a serious threat to both public health and the poultry industry worldwide. This respiratory virus can be combated by eliciting robust immune responses at the site of infection through mucosal immunization. Recombinant probiotics, specifically lactic acid bacteria, are safe and effective carriers for mucosal vaccines. In this study, we engineered recombinant fusion protein by fusing the hemagglutinin 1 (HA1) subunit of the A/Aquatic bird/Korea/W81/2005 (H5N2) with the Bacillus subtilis poly γ-glutamic acid synthetase A (pgsA) at the surface of Lactobacillus casei (pgsA-HA1/L. casei). Using subcellular fractionation and flow cytometry we confirmed the surface localization of this fusion protein. Mucosal administration of pgsA-HA1/L. casei in mice resulted in significant levels of HA1-specific serum IgG, mucosal IgA and neutralizing antibodies against the H5N2 virus. Additionally, pgsA-HA1/L. casei-induced systemic and local cell-mediated immune responses specific to HA1, as evidenced by an increased number of IFN-γ and IL-4 secreting cells in the spleens and higher levels of IL-4 in the local lymphocyte supernatants. Finally, mice inoculated with pgsA-HA1/L. casei were protected against a 10LD50 dose of the homologous mouse-adapted H5N2 virus. These results suggest that mucosal immunization with L. casei displaying HA1 on its surface could be a potential strategy for developing a mucosal vaccine against other H5 subtype viruses.

The poultry industry faces disposal difficulty in waste, but recent advancements in material science and sustainability have enabled the innovative transformation of waste into valuable resources. In this study, eggshell (EC) was added as a bio-filler to gum Arabic (GA) to fabricate a GA-EC bio-composite film. Bio-composites containing 0.5 g (GA-EC0.5) and 1.0 g (GA-EC1.0) EC dispersed in 30 mL of 15% GA solution were fabricated and characterized using standard analytical techniques. The GA-EC0.5 composites showed significantly higher moisture content, transparency, water solubility, and water vapor permeability but lower tensile strength and thermal stability than GA-EC1.0. Following a post-harvest wrapping of tomato fruits with the GA-EC composite films and storage at 25±2℃ for 20 days, significant (p>0.05) reductions in weight loss, pH, lycopene content, and activities of polyphenol oxidase and pectin methylesterase compared to unwrapped fruits were recorded. Adding EC to GA has enabled the fabrication of composite films with improved mechanical, barrier, and thermal properties with potential application in the post-harvest storage of tomato fruits.
Woody breast (WB) is one of muscle myopathy found in chicken breast, characterized with enlarged size and extremely stiff texture. The WB condition is one of the most prevalent quality issues in the modern poultry industry. WB has been shown to be heritable, but no effective detection method of WB severity in live birds exists for the selection purpose. The objective of this study was to determine potential of a non-invasive, portable digital palpation device as WB detection method that can be used for the selection to estimate the heritability of WB. The physical and functional properties of WB was also investigated in comparison with normal breast (NB). Two hundred ten breast muscles were obtained from a local processing plant one day after harvest and sorted based on WB scoring (1 for NB and 2 and 3 for WB). The samples were subjected to physical and physicochemical analyses, determining biomechanical properties (muscle tone, stiffness, elasticity, relaxation, and creep), pH, color, cooking yield, and texture (firmness and compression energy were used for raw meat and shear force and energy for cooked meat). The least squares means of the following variables were significantly different between WB and NB (p < 0.01): stiffness (603.4 vs 565.8; N/m), and elasticity (1.40 vs 1.55). However, relaxation and creep were not significantly different (p > 0.05). These results collectively showed that biomechanical properties of WB differ from NB. The degree of muscle stiffness in WB can be considered as a trait to be selected. The WB score showed strong negative correlations with cooking yield (-0.77) and cooked L*(-0.74), which means that as the breast becomes harder, the cooking yield decreases, and the color becomes darker after cooking. The WB score showed high correlations with physical and functional characteristics and exhibited strong correlations with the biomechanical properties measured by the device. Therefore, the results indicated that the digital palpation device has potential to detect the WB severity (degree of stiffness) of breast muscle.

This study was to investigate effects of different phytogenic feed additives (PFA) in grower finishing pigs with stressed by high stocking density. A total of 84 growing pigs ([Landrace × Yorkshire] × Duroc) with initial body weight (BW) of 28.23 ± 0.21 kg were used for 10 weeks (4 replicate pens with 3 pigs per pen). The dietary treatment consisted of basal diets in animal welfare density (positive control [PC]), basal diet in high stocking density (negative control [NC]), NC + 0.04% bitter citrus extract (PT1), NC + 0.01% microencapsulated blend of thymol & carvacrol (PT2), NC + 0.10% mixture of 40% bitter citrus extract and 10% microencapsulated blend of thymol and carvacrol (PT3), NC + 0.04% premixture of grape seed and grape marc extract, green tea and hops (PT4), and NC + 0.10% fenugreek seed powder (PT5). The reduction of space allowance significantly decreased (p < 0.05) growth performance (average daily gain, average daily feed intake, feed efficiency) and nutrient digestibility (dry matter, crude protein). Also, the fecal score of NC group increased (p < 0.05) compared with other groups. In blood profiles, lymphocyte decreased (p < 0.05), and neutrophil, cortisol, TNF- α increased (p < 0.05) when pigs were in high stocking density. Basic behaviors (feed intake, standing, lying) were inactive (p < 0.05) and singularity behavior (biting) were increased (p < 0.05) under high stocking density. However, PFA groups alleviated the negative effects such as reducing growth performance, nutrient digestibility, increasing stress indicators in blood and animal behavior. In conclusion, PFA groups improved the health of pigs with stressed by high stocking density and PT3 is the most effective.
Importance: Infectious bursal disease (IBD) is an important viral poultry disease that vaccination can control. Objective: This study examined the immune protection of immune-complex (Vaccine A) and attenuated live (Vaccine B) IBD vaccines in specific-pathogen-free (SPF) chickens against a novel Malaysian variant IBD virus (vaIBDV) challenge. Methods: One-day-old (n =75) SPF chickens were divided randomly into the following three groups of 25 chicks each: Control, Vaccine A, and Vaccine B groups. The vaIBDV strain, UPM1432/2019, was used for the challenge at 21 and 28days post-vaccination (dpv). Five birds from unchallenged and challenged groups were sacrificed seven days post-challenge, and blood, bursa, spleen, and cloacal swabs were collected. The IBD antibodies (Abs), lymphoid lesions, and viral load were determined. Results: The UPM1432/2019 virus induced bursal damage in vaccinated SPF chickens despite Ab titers. The mean Ab titers of the Vaccine A challenged group were significantly lower (p < 0.002) than in the unchallenged group at 28 dpv. The bursal indices of the vaccinated unchallenged groups did not differ significantly from those of the vaccinated challenged groups (p = 0.94). Microscopically, the bursae of the challenged groups showed significant atrophy. The bursal lesion score was higher (p < 0.05) in the control and Vaccine B challenged groups than the Vaccine A challenged group. The challenged group had a higher viral load than the vaccinated groups (p < 0.001). Conclusions and Relevance: Neither vaccine fully protected against a vaIBDV challenge, highlighting the limitations of current vaccines and the need for further research.
The present study was conducted to compare the disease resistance to fowl typhoid among White Leghorn (WL) known as possessing greater genetic resistance to Salmonella, Rhode Island Red (RIR) susceptible to the disease and their mating crossbreed, WL ${\times}$RIR. Also, it was carried out to compare fowl typhoid-resistance among Hyline white${\times}$Hyline brown(HwHb), Hw${\times}$lsa brown(Hwlb), Isa white${\times}$lb(Iwlb), Iw${\times}$ Hb(IwHb) and Ib ${\times}$ Iw(IbIw) in order to select crossbreed chickens which have disease resistance to fowl typhoid and simultaneously produce the brown eggs which our domestic consummers prefer There was no death of WL chickens challenged with Sal. gallinarum at each 2, 5 and 8 wk of age, while average 86.2 to 64.1% of RIR chickens tested died at the same challenge age during the test period of 2 weeks. Their maiting crossbreed, WL ${\times}$ RIR were intermediate between the two breeds. weight gains of tested chickens were significantly higher in WL and WL ${\times}$RIR without difference between them than RIR. In recovery rate of tested strain of Sal. gallinarum from braun, heart, fiver, spleen and cecum of chickens challenged, WL ${\times}$RIR fell between the other two breeds, with somewhat higher rate then WL but much lower then RIR. In the disease resistance determination test using HwHb, Hwlb, Iwlb, IwHb and IbIw$.$ crossbreed chickens experimentally infected with Sal. gallinarum, it was recognized that all of crossbreeds tested developed marked resistance to fowl typhoid, based on the results judged by the measurement parameters of mortality, weight gain, recovery rate of challenged strain of bacteria, and positive rate of agglutination antibody, and HwHb had a tittle higher disease resistance then ethers.
This experiment was conducted to study the effect of early-life feed restriction with diet dilution on compensatory growth, feed efficiency and abdominal fat pad deposition in broilers. In this study, the chicks were randomly assigned to five treatments. Twenty chicks were assigned to each floor pen, and each dietary treatment was replicated with three pens. Birds In control group (C) were full-fed a starter diet throughout the experimental period, and all birds in four dietary treatments (T1-T4) were fed as starter diet diluted with 50% rice hulls. Birds in T1 were fed with a diluted starter diet ad libitum from 7- to 74-d. In T2,the feeding program was 1-d withdrawal alternating with 3-d feed and in T3 1-d withdrawal alternating with 2-d feed. The feeding Program in T4 was alternate days withdrawal and feeding. The feeding with the diluted starter diet (T1) did not significantly affect to growth rate as compared to the birds of C. When periods of 24 h feed withdrawal were Imposed in conjunction with the diluted diets, birds were under weight at 49 d. As the diluted diet treatment was combined with feed withdrawal (T2-T4), there were further less growth. During the 22 to 49 d Period,T1 birds had greater weight gain compared to other treatments (P<0.05) . Birds consumed less feed from 7- to 49-d when the rice hull dilution was used, and this effect was increased by imposition of feed withdrawal (P<0.07) . If rice hull was excluded from the calculation of feed intake (assumed indigestible) then Intake of the starter diet was markedly less for restricted vs. control birds. After 22 d and from 7 to 49 d, restricted birds had superior feed conversion (P<0.05) compared to control birds. Abdominal fat pad deposition and mortality were not influenced by early feed restriction by diluted diet (P<0.05) .
Two experiments were conducted to studs the effects of early-life feed restriction, ages and duration on compensatory growth, feed efficiency, and abdominal fat pad deposition in broilers. For treatments 1, 2, and 3, bards were fed diets containing 0.75 kcal ME/gB $W^{o.67}$/d for 3 d(T1), 1.5 ㎉ ME/gB $W^{o.67}$/d for 5 d (T2), and 2.25 local ME/gB $W^{0.67}$/d for 7 d (T3), all imposed beginning at 7 d of age. The selected value seems appropriate for maintenance feeding because the feed restriction at T1 resulted no growth in virtually. Although the birds previously subjected with restricted feeding significantly (P<0.05) enhanced growth partly (in 22~49 d), the weight gain over the entire growth period (7-49 d) was not influenced by the restricted feeding Programs. Whereas, the restricted feedings had effects with significant (P<0.75) increase in feed intake not only in 22~49 d, but in overall period (7-49 d). The feed conversion for the birds previously subjected with restricted feeding was significantly (P<0.05) improved after 21 d. The birds with restricted feedings (T1,T2,T3) showed better (P<0.05) feed conversion ratio than C both in 22-49 d and in 7-49 d. The restricted feeding programs did not show effects in reducing for abdominal fat pad deposition or mortality In experiment 2, the birds of C were full-fed in overall period. But for treatments 1, 2, 3, 4, 5, and 6 (T1-T6),the birds were fed diets containing 0.75 ㎉ ME/gB $W^{0.67}$/d for 3 or 5 d, starting at either 4, 7 or 10 d of age. The selected value seems appropriate for maintenance feeding because the feed restriction resulted no growth in virtually; which was same as experiment.
This study was conducted to determine the effect of aluminum sulfate[Al$_2$(S0$_4$)$_3$$.$14H$_2$O], commonly referred to as ALUM, addition to broiler litter on 3 and 6 week old broiler performance and the nitrogen content of the litter at 6 weeks of age. The two treatment groups were 134 identical diets with the same protein levels, but one group(T$_1$) had ALUM added as a top dressing to the litter at a rate of 200 g ALUM / kg of rice bran, while T$_2$ did not have ALUM added to the litter. Feed consumption for T$_1$ was higher in the 22 to 42 day-old and 0 to 42 day-old periods (P<0.05). Body weight in T$_1$ was also higher in the 0 to 42 day -old period (P<0.0l). There was no difference, however, in the feed : gain ratio between T$_1$ and T$_2$ During the first 5 weeks, T$_1$ had significantly less(P<0.05 or 0.01) ammonia emission from the litter than T$_2$ but at 6 weeks there was no difference in ammonia concentration between the two groups. At 6 weeks, T$_1$ had a lower litter pH than Ta (P<0.05) and total Kjeldahl nitrogen(TKN) was higher far T$_1$ than T$_2$ (P<0.05). However,71 did not show any difference from T$_2$ in the content of NH4-N and NO3-N. In summary, the addition of ALUM to broiler litter improved broiler performance at 6 weeks, while increasing nitrogen content from the litter used as the nitrogen fertilizer although ammonia emission was increased in T$_1$ at 6 weeks.
Two experiments were conducted to compare the effects of the natural and synthetic commercial pigments on the laying Performance, Pigmentation and fatty acid contents in e99 yolk of laying hens. The experimental diets were formulated to have isocalories and isonitrogen. In experiment I, the diet does not contain the com in which the xanthophylls are free, and in experiment II, the diet contained 54% of corn in which the level of xanthophylls are 19.34 ppm/kg (calculated levels). In the experiment 1, 480 ISA Brown laying hens were divided into eight groups. Each group has 60 birds fed the 0% of corn with 8 types of pigment levels for 4 weeks. in the experiment 2, 600 ISA Brown laying hens were divided into ten groups. Each group has 60 birds fed the 54% of com diet with 10 types of pigment levels for 4 weeks. Feed intake, e99 Production, egg weight and feed efficiency did not have significant difference in experiments I and II. Albumen height and haugh unit did not have significant difference in both experiments. In order to approach the yolk pigmentation to 12∼13 of Roche color fan, addition level of natural red pigment was 25∼30 ppm. In the case of synthetic red pigment, the level was 15∼20 ppm. In this experimental condition, the pigmenting effect of the synthetic pigment was better than that of the natural pigment. In the experiment 2, the pigmenting effect of mixing pigments were investigated between TM2 mixed with natural red pigment, and TM6 mixed with synthesis red pigment. The pigmenting effect of synthetic red pigment was greater than that of the natural red pigment. However, the pigmenting effect of natural pigment was greater than that of synthetic pigments when the levels of synthetic and natural red pigments in diet are higher in TM3 and TM8. The fatty acid content in yolk was not affected by pigment addition.
