Protection of rainbow trout (Oncorhynchus mykiss) against infectious hematopoietic necrosis virus (IHNV) by immunization with G gene's cytoplasmic and transmembrane region-deleted single-cycle IHNV

  • Single-cycle viruses generated by reverse genetic technology are replication-incompetent viruses due to the elimination of gene(s) essential for viral replication, which provides a way to overcome the safety problem in attenuated viruses. Infectious hematopoietic necrosis virus (IHNV) is a major pathogen causing severe damage in cultured salmonid species. In the present study, we generated a single-cycle IHNV lacking the transmembrane and cytoplasmic domain in the G gene (rIHNV-GΔTM) and evaluated the prophylactic potential of rIHNV-GΔTM in rainbow trout (Oncorhynchus mykiss). To produce rIHNV-GΔTM, IHNV G protein-expressing Epithelioma papulosum cyprini (EPC) cells were established. However, as the efficiency of rIHNV-GΔTM production in EPC cell clones was not high, fish were immunized with a low-tittered single-cycle virus (1.5 × 102 PFU/fish). Despite the low dose, the single-cycle IHNV induced significant protection in rainbow trout against IHNV infection, suggesting high immunogenicity of rIHNV-GΔTM. No significant difference in serum ELISA titers against IHNV between the rIHNV-GΔTM immunized group and the control group suggests that the immunized dose of rIHNV-GΔTM might be too low to induce significant humoral adaptive immune responses in rainbow trout. The involvement of adaptive cellular immunity or innate immunity in the present significantly higher protection by the immunization with rIHNV-GΔTM should be further investigated to know the protection mechanism.

  • This paper presents the application of Convolutional Neural Networks (CNNs) and Region of Interest (ROI) techniques for concrete crack analysis. Surfaces of concrete structures, such as beams, etc., are exposed to fatigue stress and cyclic loads, typically resulting in the initiation of cracks at a microscopic level on the structure's surface. Early detection enables preventative measures to mitigate potential damage and failures. Conventional manual inspections often yield subpar results, especially for large-scale infrastructure where access is challenging and detecting cracks can be difficult. This paper presents data collection, edge segmentation and ROI techniques application, and analysis of concrete cracks using Convolutional Neural Networks. This paper aims to achieve the following objectives: Firstly, achieving improved accuracy in crack detection using image-based technology compared to traditional manual inspection methods. Secondly, developing an algorithm that utilizes enhanced Sobel edge segmentation and ROI techniques. The algorithm provides automated crack detection capabilities for non-destructive testing.

Imipramine enhances neuroprotective effect of PEP-1-Catalase against ischemic neuronal damage

  • The protein transduction domains have been reported to have potential to deliver the exogenous molecules, including proteins, to living cells. However, poor transduction of proteins limits therapeutic application. In this study, we examined whether imipramine could stimulate the transduction efficiency of PEP-1 fused proteins into astrocytes. PEP-1-catalase (PEP-1-CAT) was transduced into astrocytes in a time- and dose-dependent manner, reducing cellular toxicity induced by $H_2O_2$. Additionally, the group of PEP-1-CAT + imipramine showed enhancement of transduction efficiency and therefore increased cellular viability than that of PEP-1-CAT alone. In the gerbil ischemia models, PEP-1-CAT displayed significant neuroprotection in the CA1 region of the hippocampus. Interestingly, PEP-1-CAT + imipramine prevented neuronal cell death and lipid peroxidation more markedly than PEP-1-CAT alone. Therefore, our results suggest that imipramine can be used as a drug to enhance the transduction of PEP-1 fusion proteins to cells or animals and their efficacies against various disorders.

  • As the scales & density of the Korean national infrastructures have been increased, they will be identified as rich and attractive potential targets for intensified North Korea's attack in the rear region and terrorism attack. In addition, due to changes in security environment such as drone threats and lack of security forces under the 52-hour workweek law, I think that it is the proper time point to reevaluate the effectiveness and appropriateness of the current physical protection system and its shift to a new system. In this study, the direction and improvement of the perimeter physical protection systems of the national infrastructures are to be studied from the viewpoints of its concepts of operations and design methodology, focusing on the nuclear power plant. The reason why we focus on nuclear power plants is because they cause wide-range and long-term damages caused by radioactive materials disperal and pollution, along with short-term damage caused by the interruption of electricity generation in the event of damage to nuclear power plants. With the aim of extracting improvement directions, as we will comprehensively review domestic research trends and domestic·overseas related laws, and consider Korea's specificity, we try to reframe the concept of operation - systematization, mobilization and flexibility -, and establish criteria on system change. In order to improve the technical performance of the new perimeter physical protection system, we study on high-fidelity·multi-methodology based integrated design methodology, breaking from individual silo-type design methods, and I suggest improvement of government procurement, its expansion to export business and other national infrastructure.

A Quantitative Approach to the influence on the South Korean Air Transportation System in the Event of Volcanic Ash Dispersal (화산재에 따른 국내항공교통의 영향에 대한 정량화 방안)

  • There has been a growing interest on the effect of volcanic eruption on the aviation safety, air travel and economy especially after the eruption of Eyjafjallajokull in Iceland. Since volcanic eruption is influential on a large geographic region, the effect usually extends to other neighboring countries. Korea also has an active volcano named Mountain Baekdu. Hence, the need to estimate in advance the quantitative impact of the potential eruption of Mt. Baekdu on South Korean air transportation system. However, previous studies with quantitative estimation were confined to the calculation of the direct economic loss from shut down of the airports, grounding of airlines, and trade deficits caused by the eruption. Therefore, this paper introduces a new approach to assess more accurate impact simultaneously considering volcanic ash dispersal and aviation routes. This approach is then applied to a virtual scenario to predict the damage to air traffic. With further development, this method can help estimate the damage in the air transportation industry in more accurate and faster ways. Prediction outcomes can also be utilized in setting up the emergency response plan for the air transportation industry and contribute to the creation of more proactive and predictive measures in the future.

A Comparative Study of Ship Collision Legislation in Korea and China (한국과 중국의 선박충돌법제의 비교법적 연구)

  • The increasing trade volume between Korea and China has rapidly expanded the maritime transport between the two countries. However, safety, particularly considering the ship collisions in the Yellow Sea and East Sea waters, has not been fully ensured. These collision accidents in that region endanger traffic safety and the marine environment, moreover, it has the potential to cause legal complexity because Korea and China haver domestic legislation, that are considerably different in some aspects. International conventions and domestic legislation in China provide detailed laws with respect to ship collisions, however, the theory of ship collision infringement still needs to be improved, enriched, and developed. Because these laws are not very clear on the resolution of disputes resulting from ship collisions, we focused on the final judgments by the Supreme Court of China (SPC), and the judicial judgments set by the Maritime Court of China. This study aimed to explore the domestic legislation applicable to disputes related to ship collisions in China, and comparatively investigate the legal provisions of Korea and China on the issue of ship collisions, particularly on the aspect of damage compensation, fault ration, and liability apportionment.

Episode Analysis of the Habit and Phase Changes of Snow Crystals in the Wintertime Yeongdong Region (겨울철 영동지역 눈 결정 습성과 성상 변화 에피소드 분석)

  • The Yeongdong region has suffered from severe snowstorms and the relevant damage such as traffic accidents on slippery roads, and the collapse of greenhouses and temporary buildings. While a lot of research on snowfall has been conducted, the detailed study of snow crystals' phase and habit through intensive observations and the relevant microphysical analysis is still lacking. Therefore, a snowflake camera, PARSIVEL, and intensive radiosonde soundings were utilized to investigate phase and habit changes in solid precipitation. Two remarkable episodes of phase and habit changes were selected such as 19 March 2022 and 15 February 2023. Both events occurred in the synoptic condition of the High in the north and the Low passing by the south, which was accompanied by rapid temperature cooling below 2.5 km. During the events of a short period between 3 to 6 hours, the temperature at 850 hPa decreased by about 4 to 6℃. This cooling led to a change in the main habit of snow particles from riming to aggregate, identified with both MASC and PARSIVEL. Meanwhile, the LDAPS model analyses do not successively represent the rapid cooling and short-term variations of solid precipitation, probably by virtue of overestimating low-level equivalent potential temperature during these periods. The underlying causes of these the low-level temperature variations within 6 hours, still remain unclear. It might be associated with mesoscale orographic phenomenon due to the mountains and East Sea effects, which certainly needs an intensive and comprehensive observation campaign.

  • Large-scale earthquakes are occurring globally, especially in the South Asian crust, which is experiencing a state of tension in the aftermath of the 2011 East Japan Earthquake. Uncertainty and fear regarding the possibility of further seismic activity in the near future have been on the rise in the region. The National Disaster Management Research Institute has previously studied and analyzed the overflow characteristics of a tsunami and the rate of flood forecasting through tsunami numerical simulations of the East Sea of South Korea. However, there is currently a significant lack of research on the Southern Coast tsunamis compared to the East Coast. On the Southern Coast, the tidal difference is between 1~4 m, and the impact of the tides is hard to ignore. Therefore, it is necessary to analyze the impact of the tide propagation characteristics on the tsunami. Occurrence regions that may cradle tsunamis that affect the southern coast region are the Ryukyu Island and Nankai Trough, which are active seafloor fault zones. The Southern Coast has not experienced direct damage from tsunamis before, but since the possibility is always present, further research is required to prepare precautionary measures in the face of a potential event. Therefore, this study numerically simulated a hypothetical tsunami scenario that could impact the southern coast of South Korea. In addition, the tidal wave propagation characteristics that emerge at the shore due to tide and tsunami interactions will be analyzed. This study will be used to prepare for tsunamis that might occur on the southern coast through tsunami hazard and risk analysis.

Neuroprotective effects of herbal mixture HT070 on global cerebral ischemia in rats

  • Objectives : HT070 is a mixture of herbal extracts from root of Scutellaria baicalensis and stem bark of Eleutherococcus senticosus , which have long been used for stroke therapy in traditional Korean Medicine. The purpose of this study was to investigate the neuroprotective effects of HT070 on global cerebral ischemia and its potential mechanisms.Methods : Transient global cerebral ischemia was produced by 10 min of four-vessel occlusion (4-VO) in male Wistar rats. HT070 was administered orally at a dosage of 200 mg/kg twice at 0 and 90 min after reperfusion. Hippocampal neuronal damage was measured 7 days after reperfusion. To explore the potential mechanisms, we used hydrogen peroxide (H2O2)-induced rat pheochromocytoma (PC12) cells as an in vitro model. PC12 cells were pretreated with HT070 for 1 h and then exposed to 100 μM H2O2 for 6 h in the presence of HT070. Cell viability was measured by MTT assay and the mRNA expression of Bax, Bcl-2, iNOS and COX-2 were measured by quantitative RT-PCR.Results : Oral administration of HT070 at a dose of 200 mg/kg significantly reduced neuronal death in the hippocampal CA1 region by 13.4% as compared to the vehicle-treated group. HT070 increased cell viability, reversed the down-regulated Bcl-2 mRNA level, and suppressed the up-regulated mRNA expressions of Bax, iNOS, and COX-2 in H2O2-treated PC12 cells.Conclusions : HT070 protects against delayed neuronal death after global cerebral ischemia and its neuroprotection properties might be attributed to the inhibition of mitochondrial apoptosis and ROS-generating enzymes.

Cordycepin Enhanced Therapeutic Potential of Gemcitabine against Cholangiocarcinoma via Downregulating Cancer Stem-Like Properties

  • Cordycepin, a valuable bioactive component isolated from Cordyceps militaris, has been reported to possess anti-cancer potential and the property to enhance the effects of chemotherapeutic agents in various types of cancers. However, the ability of cordycepin to chemosensitize cholangiocarcinoma (CCA) cells to gemcitabine has not yet been evaluated. The current study was performed to evaluate the above, and the mechanisms associated with it. The study analyzed the effects of cordycepin in combination with gemcitabine on the cancer stem-like properties of the CCA SNU478 cell line, including its anti-apoptotic, migratory, and antioxidant effects. In addition, the combination of cordycepin and gemcitabine was evaluated in the CCA xenograft model. The cordycepin treatment significantly decreased SNU478 cell viability and, in combination with gemcitabine, additively reduced cell viability. The cordycepin and gemcitabine co-treatment significantly increased the Annexin V+ population and downregulated B-cell lymphoma 2 (Bcl-2) expression, suggesting that the decreased cell viability in the cordycepin+gemcitabine group may result from an increase in apoptotic death. In addition, the cordycepin and gemcitabine co-treatment significantly reduced the migratory ability of SNU478 cells in the wound healing and trans-well migration assays. It was observed that the cordycepin and gemcitabine cotreatment reduced the CD44highCD133high population in SNU478 cells and the expression level of sex determining region Y-box 2 (Sox-2), indicating the downregulation of the cancer stem-like population. Cordycepin also enhanced oxidative damage mediated by gemcitabine in MitoSOX staining associated with the upregulated Kelch like ECH Associated Protein 1 (Keap1)/nuclear factor erythroid 2-related factor 2 (Nrf2) expression ratio. In the SNU478 xenograft model, co-administration of cordycepin and gemcitabine additively delayed tumor growth. These results indicate that cordycepin potentiates the chemotherapeutic property of gemcitabine against CCA, which results from the downregulation of its cancer-stem-like properties. Hence, the combination therapy of cordycepin and gemcitabine may be a promising therapeutic strategy in the treatment of CCA.
