Antimicrobial Effect of Portulaca oleracea Extracts on Food-Borne Pathogens

  • This study was performed to investigate the antimicrobial effects of Portulaca oleracea extracts against food-borne pathogens. First, the Portulaca oleracea was extracted with methanol at room temperature, and then further fractionated by using petroleum ether, chloroform, ethyl acetate and methanol, respectively. The antimicrobial activity of the Portulaca oleracea extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Portulaca oleracea showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. There was also a synergistic effect of the combined extracts of Portulaca oleracea and Indigofera kirilowii as compared to each extract alone. Finally, the growth inhibition curve of ethyl acetate extracts of Portulaca oleracea against Staph­ylococcus aureus and Shigella dysenteriae was determined The ethyl acetate extract of Portulaca oleracea showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Portulaca oleracea, retarded the growth of S. aureus by more than 24 hand Shigella dysenteriae up to 12 h at $37^{\circ}C$.

  • Cigarette smoking is the potential risk factor for lung cancer and chronic pulmonary disease, as well as inflammatory bowel disease and reproductive malfunction. Nicotine and tar have been im plicated as a major factor in the pathogenesis of the diseases. Nicotine increases heart rate and blood pressure due to stimulation sympathetic neurotransmission and tar also accounts for the severe damage of peridontal diseases and osteoporosis in postmenopausal women. Portulaca oleracea, which contains significant amount of K+, noradrenaline and dopamine as well as various nutrients, has been used for many medicinal purposes and one of which is the detoxification of insect or snake toxins. The purpose of this study was to investigate the action of Portulaca oleracea extracts on the reduction of harmful materials of tabacco. The reduction percentages were measured in the presence and absence of each solvent extract of Portulaca oleracea using reversed C18 column of HPLC. Nicotine reduction effects were obtained from aqueous, methanol and chloroform extracts of Portulaca oleracea as 89%, 55% and 51%, respectively. The results suggest that the polar extracts of Portulaca oleracea affects the reduction of nicotine which is responsible for many diseases.

  • This study was performed to investigate the lipolytic effects of Portulaca oleracea L. extract in 3T3-L1 adipocytes. The Portulaca oleracea L. was extracted with extrusion method using twin-screw extruder under $58{\sim}60rpm$ screw speed, $4{\sim}5kg/hr$ feed rate, $140^{\circ}C$ extrusion temperature. The lipolytic action of Portulaca oleracea L. extract was estimated by measuring the amount of glycerol and free fatty acids (FFA) released from 3T3-L1 adipocytes and by measuring the cellular lipid content in 3T3-L1 adipocytes. The hormone sensitive lipase (HSL) mRNA level was analyzed using quantitative real-time PCR. The Portulaca oleracea L. extract at 1 to $100{\mu}g/ml$ suppressed lipid accumulation. The release of glycerol and FFA into the medium, and the mRNA level of HSL were significantly increased by the addition of Portulaca oleracea L. extract at dose-dependent manner. In conclusion, the Portulaca oleracea L. extract was suggested to have the lipolytic effect through release of lipolytic products (FFA and glycerol) of triacylglyceride to the culture medium and suppression of lipid accumulation via up-regulation of HSL gene expression in 3T3-L1 adipocytes.

염산-에탄올에 의해 유발된 흰쥐 위염에 대한 마치현의 항산화 작용 (Anti-oxidant Effects of Portulaca oleracea L. on HCl-ethanol Induced Gastritis in Rats)

  • Objectives : The purpose of this study is to evaluate whether or not a pretreatment with Portulaca oleracea has an antioxidant effect in HCl-ethanol induced gastric mucosal damage. Methods : We elucidated the level of reactive oxygen species (ROS), lipid peroxidation, and two important constituents of antioxidant defense such as superoxide dismutase (SOD), glutathione (GSH) in these effects. Results : The oral administration of crude extract from P. oleracea attenuated the gastritic lesion area, submucosal edema and hemorrhage, and mucosal necrosis induced by HCl-ethanol. The MDA levels of control group were higher than those in the rats given the P. oleracea pretreatment. While the GSH levels of control were decreased, the GSH activity on the gastric mucosal layer maintain normal level in rats given the Portulaca oleracea pretreatment before HCl-ethanol induced gastritis significantly increased. However, the SOD activites were not altered by P. oleracea. Conclusions : The administration of Portulaca oleracea have a protective antioxidant effect against the gastric lesion induced by HCl-ethanol and may therefore be a promising drug for gastritis and gastric ulcer.

Anti-Asthmatic Effects of Portulaca Oleracea and its Constituents, a Review

  • Objectives: The medicinal plants are believed to enhance the natural resistance of the body to infections. Some of the main constituents of the plant and derived materials such as, proteins, lectins and polysaccharides have anti-inflammatory effects. Portulaca oleracea (P. oleracea) were used traditionally for dietary, food additive, spice and various medicinal purposes. This review article is focus on the anti-asthmatic effects of P. oleracea and its constituents. Methods: Various databases, such as the PubMed, Scopus, and Google Scholar, were searched the keywords including "Portulaca oleracea", "Quercetin", "Anti-inflammatory", "Antioxidant", "Cytokines", "Smooth muscle ", and " Relaxant effects " until the end of Jul 2018. Results: P. oleracea extracts and its constituents increased $IFN-{\gamma}$, IL-2, $IFN{\gamma}/IL-4$ and IL- 10/IL-4 ratio, but decreased secretion of $TNF-{\alpha}$, IL-4 and chemokines in both in vitro and in vivo studies. P. oleracea extracts and quercetin also signifcantly decreased production of NO, stimulated ${\beta}$-adrenoceptor and/or blocking muscarinic receptors in tracheal smooth muscles. Conclusion: P. oleracea extracts and quercetin showed relatively potent anti-asthmatic effects due to decreased production of NO, inflammatory cytokines and chemokines, reduced oxidant while enhanced antioxidant markers, and also showed potent relaxant effects on tracheal smooth muscles via stimulatory on ${\beta}$-adrenoceptor or/and blocking muscarinic receptors.

고지방식이로 유도된 비만 Mice에서 쇠비름 에탄올 추출물의 고지혈증 개선효과 (Antihyperlipidemic Effect of Diet Containing Portulaca oleracea L. Ethanol Extract in High Fat Diet-Induced Obese Mice)

  • 쇠비름 에탄올 추출물의 고지혈증 개선 효과를 알아보기 위하여 C57BL/6J mice에게 9주 동안 고지방식이를 섭취시켰다. 마지막 4주 동안은 고지방식이에 쇠비름 에탄올 저농도투여군(HFD+POE 75) 및 쇠비름 에탄올 고농도투여군(HFD+POE 125)으로 나누어 하루에 한 번씩 경구투여 하였으며 고지방식이군(HFD)은 증류수를 투여하였다. HFD+POE 75군 및 HFD+POE 125군의 체중변화량은 유의적인 차이를 나타나지는 않았으나 식이효율은 HFD군에 비해 유의적으로 감소하였다. 혈중 ALT 함량은 HFD군에 비해 HFD+POE 125군에서 유의적으로 감소하였으나 AST 함량은 그룹 간 차이를 나타내지 않았다. 실험식이 4주 때에 혈청 중 총콜레스테롤, 중성지질 및 LDL-콜레스테롤 함량은 쇠비름 에탄올 추출물 경구투여로 유의적으로 감소하였으나 HDL-콜레스테롤 함량은 유의적 차이를 나타내지 않았다. 심순환기 질환의 위험성을 나타내는 지표로 알려진 동맥경화지수와 심혈관위험지수는 고농도의 쇠비름 에탄올 추출물 경구투여로 HFD군에 비해 유의적으로 감소하였다. 이상의 결과 고지방식이에 쇠비름 에탄올 추출물 경구투여로 혈중 및 간장 중의 총콜레스테롤 및 LDL-콜레스테롤, 중성지질이 저하되었는데 이는 쇠비름 에탄올 추출물이 고지혈증 개선 효과가 있는 것으로 사료되나 쇠비름 에탄올 추출물의 어떤 성분에 의한 지질 저하효과를 나타내는지와 어떠한 기전에 의해 지질 저하효과를 나타내는지 보다 자세한 연구가 요구된다.

마치현 에틸아세테이트 분획물의 뇌세포 보호효과 (Neuroprotective Effect of Ethyl Acetate Fraction of Portulaca oleracea L.)

  • Portulaca oleracea L. is known to have many biological benefits such as anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of P. oleracea L. against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. P. oleracea L. 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, P. oleracea L. treatment with ERK inhibitor (PD98059) and c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea L. ethyl acetate fraction induced HO-1 expression and P. oleracea L. ethyl acetate fraction also increased ERK and JNK phosphorylation. Furthermore, we found that treatment of P. oleracea L. caused the nuclear accumulation of Nrf2. In conclusion, the ethyl acetate fraction of 70% ethanol extract of P. oleracea L. significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2, ERK and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that P. oleracea L. ethyl acetate fraction is good source for taking active compounds and may be a potential therapeutic agent for brain disorder that induced by oxidative stress and neuronal damage.

Cactus의 광합성과 질소동화작용에 관한 연구 - 한국산 쇠비름(Portulaca oleracea L.) - (A Study on Photosynthesis and Nitrogen Assimilation in Cactus -Portulaca oleracea L.-)

  • Crassulacean acid metaholism (CAM) was investigated in leaves and stems of the succulent $C_4$dicot Portulaca oleracea L. Under 14-hour days, stem tissues showed much greater fluctuation of acidity than leaf tissues. But leaf and stem tissues showed almost same CAM-like pattern of acid fluctuation under 8-hour days. Stem tissues of R oleracea grown under the naturai environment showed high CAM activity, but no CAM activity was seen in leaves of those plants. In the naturally growing plants, the rapid acidification was seen in intact stems at dawn, but defoliated stems showed only a gradual increase. RuBP carlboxylase activity was very high at 2:00 P.M. in both leaves and stems. However, its activity at 1:00 A.M. and 5:30 AM. was hardly detected. particularly, activity of PEP carboxylase in leaves was very high in the early morning, though that in stem tissues was little. These results indicate that $CO_2$ passed through open stomata at dawn may be assimilated by PEP carboxylase in leaves, and then $C_4$ products move to stems. The levels of nitrate concentration and of nitrate reductase were higher in stems than in leaves. The levels were also higher in the light than in the dark. It would be suggested that considerable amount of nitrate absorbed from roots ho assimilated in stems, and nitrate transferred to leaves via stem tissues be reduced there. Portulaca oleracea Seed Oil Exerts Cytotoxic Effects on Human Liver Cancer (HepG2) and Human Lung Cancer (A-549) Cell Lines

  • Portulaca oleracea (Family: Portulacaceae), is well known for its anti-inflammatory, antioxidative, anti-bacterial, and anti-tumor activities. However, cytotoxic effects of seed oil of Portulaca oleracea against human liver cancer (HepG2) and human lung cancer (A-549) cell lines have not been studied previously. Therefore, the present study was designed to investigate the cytotoxic effects of Portulaca oleracea seed oil on HepG2 and A-549 cell lines. Both cell lines were exposed to various concentrations of Portulaca oleracea seed oil for 24h. After the exposure, percentage cell viability was studied by (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) (MTT), neutral red uptake (NRU) assays, and cellular morphology by phase contrast inverted microscopy. The results showed a concentration-dependent significant reduction in the percentage cell viability and an alteration in the cellular morphology of HepG2 and A-549 cells. The percentage cell viability was recorded as 73%, 63%, and 54% by MTT assay and 76%, 61%, and 50% by NRU assay at 250, 500, and $1000{\mu}g/ml$, respectively in HepG2 cells. Percentage cell viability was recorded as 82%, 72%, and 64% by MTT assay and 83%, 68%, and 56% by NRU assay at 250, 500, and $1000{\mu}g/ml$, respectively in A-549 cells. The 100 $100{\mu}g/ml$ and lower concentrations were found to be non cytotoxic to A-549 cells, whereas decrease of 14% and 12% were recorded by MTT and NRU assay, respectively in HepG2 cells. Both HepG2 and A-549 cell lines exposed to 250, 500, and $1000{\mu}g/ml$ of Portulaca oleracea seed oil lost their normal morphology, cell adhesion capacity, become rounded, and appeared smaller in size. The data from this study showed that exposure to seed oil of Portulaca oleracea resulted in significant cytotoxicity and inhibition of growth of the human liver cancer (HepG2) and human lung cancer (A-549) cell lines.

Portulaca oleracea의 생리활성과 항균활성 (Biological And Antimicrobial Activity of Portulaca oleracea)

  • 쇠비름 추출물로부터 항산화효과, 피부상재균 및 Helicobacter pylori 억제효과, tyrosinase 억제효과를 살펴보았다. 페놀함량을 측정한 결과 물 추출물에서는 3.05 ${\mu}g/ml$, ethanol 추출물에서는 80% ethanol 추출물에서 6.33 ${\mu}g/ml$로 가장 높은 페놀함량을 나타내었다. 항산화효과는 DPPH, ABTS, Antioxidant Protection Factor 모두 페놀함량이 가장 높은 80% ethanol 추출물에서 항산화력이 강한것으로 나타났다. tyrosinase 저해활성 역시 80% ethanol 추출물에서 38.71%로 가장 저해가 높은 것으로 나타났다. 쇠비름 추출물의 tyrosinase 억제효과는 물 추출물에서 20.24%, 80% ethanol 추출물에서 38.71%의 억제효과를 나타냈다. 또한 Helicobacter pylori, Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli 및 Streptococcus mutans에 대한 항균활성을 평가한 결과 Helicobacter pylori는 200 ${\mu}g/ml$ 농도에서 13mm의 저해환을 보였고, Staphylococcus epidermidis는 40mm의 저해환을, Staphylococcus aureus, Escherichia. coli, Streptococcus. mutans는 각각 15, 17, 20 mm의 저해환을 나타내어 200 ${\mu}g/ml$의 농도 이상에서는 모두 생육억제효과가 나타나는 것을 확인 할 수 있었다.
