In an attempt to improve the quality stability of dried layer (Porphyra yezoensis Ueda), during longterm storage and distribution, a 6 months storage trial at different temperature and packaging condition was conducted. Dried layer samples were prepared by conventional procedure with different washing condition and $5^{\circ}C$ and $30^{\circ}C$ were employed as storage temperature. The moisture sorption isotherm was drawn at $5^{\circ}C$ and $25^{\circ}C$ and BET equation was prepared from the moisture sorption isotherm. The monolayer moisture contents of dried layer were 7.4% at $5^{\circ}C$ and 6.4% at $25^{\circ}C$, respectively. It was also found that the changes in comprehensive quality of dried layer during storage was much less at low temperature and the degree of washing before drying process as far as its moisture contents are lower than monolayer moisture contents. With regard to the packing methods, vaccum packing was superior to air containing packaging for the maintainance of quality during storage.
The community structure of intertidal benthic marine algae were studied seasonally at Odongdo, southern coast of Korea, from June 1982 to May 1983. Algal coverage in $50{\times}50\;cm$ quadrat were recorded for each species by line transect method. The vertical zonation investigated by line transects is recognized into three groups : Upper, middle, and lower zones. The representative species are Gelidium divaricatum, Enteromorpha linza, Porphyra yezoensis, Scytosiphon lomentaria, Blidingia nana, Ectocarpus confervoides in the upper, Ulva pertusa, Chondria crassicaulis in the middle, and Sargassum sagamianum, S. thunbergii, Undaria pinnatifida, Gelidium amansii and various other red algae in the lower zone. The number of algal species and coverage were generally highest in April and lowest in August. Species which appear dominant at least once a year were all of the spring type and the others were autumn type. According to the cluster analysis by similarity index community coefficient(SICC) among 5 transects, the algal communities are divided into two groups, i. e. open-sea group and inland-sea group.
The chemical and rheological properties of deproteinated porphyrans from laver Porphyra yezoensis were investigated to obtain basic data for the production of food materials with biological functionality. Deproteinated porphyran was prepared by acid extraction (pH 4.0, $80^{\circ}C$, 4 hr) and successive hydrolysis with 0.5% Alcalase and 0.5% Flavourzyme. The porphyran constituted 10.7% of the dry laver and consisted of 0.6% protein, 14.8% ester sulfate, 3.2% 6-O-methyl galactose, 16.0% 3,6-anhydro-L-galactose, and 67.3% galactose. The effects of concentration and temperature on the apparent viscosity were examined by applying the power law and Arrhenius equations. The porphyran solution showed the typical behavior of a pseudoplastic liquid and the flow behavior index decreased with increasing concentration. The activation energy of the deproteinated porphyran solution at a 1,000 L/s shear rate also increased from $1.4954{\times}10^{4}\;to\;1.9544{\times}10^{4}\;J/kg$ mol with the concentration.

BACKGROUND/OBJECTIVES: Increased mass of adipose tissue in obese persons is caused by excessive adipogenesis, which is elaborately controlled by an array of transcription factors. Inhibition of adipogenesis by diverse plant-derived substances has been explored. The aim of the current study was to examine the effects of the aqueous methanol extract of laver (Porphyra yezoensis) on adipogenesis and apoptosis in 3T3-L1 adipocytes and to investigate the mechanism underlying the effect of the laver extract. MATERIALS/METHODS: 3T3-L1 cells were treated with various concentrations of laver extract in differentiation medium. Lipid accumulation, expression of adipogenic proteins, including CCAAT enhancer-binding protein ${\alpha}$, peroxisome proliferator-activated receptor ${\gamma}$, fatty acid binding protein 4, and fatty acid synthase, cell viability, apoptosis, and the total content and the ratio of reduced to oxidized forms of glutathione (GSH/GSSG) were analyzed. RESULTS: Treatment with laver extract resulted in a significant decrease in lipid accumulation in 3T3-L1 adipocytes, which showed correlation with a reduction in expression of adipogenic proteins. Treatment with laver extract also resulted in a decrease in the viability of preadipocytes and an increase in the apoptosis of mature adipocytes. Treatment with laver extract led to exacerbated depletion of cellular glutathione and abolished the transient increase in GSH/GSSG ratio during adipogenesis in 3T3-L1 adipocytes. CONCLUSION: Results of our study demonstrated that treatment with the laver extract caused inhibition of adipogenesis, a decrease in proliferation of preadipocytes, and an increase in the apoptosis of mature adipocytes. It appears that these effects were caused by increasing oxidative stress, as demonstrated by the depletion and oxidation of the cellular glutathione pool in the extract-treated adipocytes. Our results suggest that a prooxidant role of laver extract is associated with its antiadipogenic and proapoptotic effects.

The chemical properties of porphyran from dried laver (Porphyra yezoensis) and decolored laver were measured. Dried laver contained 12.5% porphyran while decolored laver contained 12.3%. The chemical components of porphyran extracted from dried and decolored laver were examined. The content of total sugar was 64.5% and 63.7% on dry base, respectively, while that of sulfate was 17.6% and 16.9%, respectively. The content of 3,6-anhydrogalactose was 18.8% and 18.1%, respectively.

The purpose of this study was to process extraction and separation for polyglucose porpyran from pophyra yezoensis as high yield. The acidic porphyran solution was extracted with 0.1~1N of ${\alpha},{\beta}$-dichloromaleic acid or ${\alpha}$-chloromaleic acid of 2% solution as organic acid instead of inorganic acid at $60^{\circ}C$ and then porphyran was collection of high yield to acidic solution and it was neutralization treated with 0.1N of $d-Ca(OH)_2$ solution and the mixture was conversion to porphyran salt form and treated with a shell powder of ouster and then added of ethanol as precipitator. It recovery porphyran contained of violet purple laver coloring matter was obtained as a crystalline and used for the next step without future purification to prepare of porphyran laver jam. so, The resulting porphyran and porphyran jam was characterized by it component and physical properties.

Organic light-emitting materials in marine macrophytes from various coastal environments were identified. Twentyeight species from the solvent fractions were examined and identified as candidates for bioorganic light-emitting materials using photoluminescence (PL) spectra and gas chromatography-mass spectrometry. We selected 16 solvent fractions from a total of 1,221 prepared from Ishige okamurae, Sargassum confusum, Grateloupia elliptica, Chondracanthus intermedius, Porphyra yezoensis, Meristotheca papulosa, Gelidium amansii, and Scytosiphon lomentaria. The maximum light-emitting PL spectra appeared at various colors, mainly between blue and green, based on chromaticity coordinates, from solvent fractions of M. papulosa, G. amansii, G. elliptica, P. yezoensis, S. lomentaria, I. okamurae, and C. intermedius. These results will contribute to the development of novel organic light-emitting materials.
Marine algal flora and community structure of medicinal seaweeds were examined at Kkotji of Taean Peninsula, Korea from May 2005 to January 2006. Seventy-nine seaweeds including 42 medicinal algae and one marine plant were identified. Sargassum thunbergii was the representative alga occurred at all seasons and shore levels. The dominant medicinal seaweeds were perennial S. thunbergii, Neorhodomela aculeata, and Corallina pilulifera, and ephemeral Monostroma grevillei, Porphyra yezoensis, and Ulva pertusa. Their vertical distribution were N. aculeata – P. yezoensis, M. grevillei, and U. pertusa – C. pilulifera from high to low intertidal zone. The average biomass of medicinal seaweeds varied from 34.17 g m–2 in spring to 56.41 g m–2 in summer. At Kkotji shore, the opportunistic species (Enteromorpha, Ulva, and Cladophora) and turf-forming algae (Caulacanthus okamurae and Gelidium divaricatum) were easily observed. Such fast growing ESG II (ecological state group) was 87.50% and slow growing perennial algae, ESG I was only 12.15%. Also, diversity index (H’) and dominance index (DI) indicate that the seaweed community of Kkotji is unstable. Therefore, Kkotji rocky shore should be more protected from human activities such as turbulence and eutrophication in order to maintain species diversity and abundance of medicinal seaweeds.
Pythium species (Pythiales, Oomycetes) are well known as the algal pathogen that causes red rot disease in Pyropia / Porphyra species (Bangiales, Rhodophyta). Accurate species identification of the pathogen is important to finding a scientific solution for the disease and to clarify the host-parasite relationship. In Korea, only Pythium porphyrae has been reported from Pyropia species, with identifications based on culture and genetic analysis of the nuclear internal transcribed spacer (ITS) region. Recent fungal DNA barcoding studies have shown the low taxonomic resolution of the ITS region and suggested the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene as an alternative molecular marker to identify Pythium species. In this study, we applied an analysis of both the ITS and cox1 regions to clarify the taxonomic relationships of Korean Pythium species. From the results, the two closely related Pythium species (P. chondricola and P. porphyrae) showed the same ITS sequence, while the cox1 marker successfully discriminated P. chondricola from P. porphyrae. This is the first report of the presence of P. chondricola from the infected blade of Pyropia yezoensis in Asia. This finding of the algal pathogen provides important information for identifying and determining the distribution of Pythium species. Further studies are also needed to confirm whether P. chondricola and P. porphyrae are coexisting as algal pathogens of Pyropia species in Korea.
Studies have been made of the species composition and community structure of benthic marine algae at intertidal zone and subtidal zone of Youngdo at Busan. In this area, a total of 69 marine algal species including 10 Chlorophyta, 17 Phaeophyta and 42 Rhodophyta was identified. Dominant species in coverage were Enteromorpha linza, Ulva pertusa, Gelidium divaricatum, Hildenbrandtia rubra and Chondrus ocellatus throughout the year. Enteromorpha linza, Ulva pertusa, Porphyra yezoensis, Gelidium divaricatum, Corallina pilulifera, Grateloupia elliptica and Chondrus ocellatus vertically distributed in intertidal zone, while Ecklonia stolonifera, Gelidium amansii, Prionitis cornea, Gracilaria textorii, Acrosorium polyneurum and Phycodrys fimbriata in subtidal zone. The vertical distribution of subtidal zone were characterized by Ecklonia stolonifera at 1 to 5 m depths. During a year, biomass and length of Ecklonia stolonifera gradually decreased from March to January, but increased from February. Zoosporangial sori were observed from September to December. In conclusion, number of species in this area was remarkably reduced as compared to the previous data at Busan and it’s vicinity.
