The Effect of Porphyra tenera Extract on Serum Lipid Content in Ovariectomized Rats (김 추출물이 갱년기 장애 유도 흰쥐의 혈 중 지질 변화에 미치는 영향)

  • The aim of this study is to evaluate the effect of Porphyra tenera (PT) extract on serum lipids content in ovariectomized rats. From day 3 until 42 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovarjectomized rats supplemented with PT at 50 mg/kg bw/day (OVX-PT50), 200 mg/kg bw/day (OVX-PT200). The PT ethanol extract were orally administrated at 1 m1 per day. Body weight gain, food intake and food efficiency ratio were not significantly different between groups. Although total-cholesterol was increased in OVX-control, supplementation with the PT decreased the level of cholesterol in serum. Especially, it was significantly decreased in OVX-PI50 (p<0.05). Triglyceride was significantly decreased to almost all of groups supplemented with PT (p<0.05). The levels of HDL-cholesterol was higher in PT extract group than OVX-control. These results suggest that supplementation with the PT extract positively influenced on lipid concentration in serum. Consequently, we expect that PT extract decrease risk of cardiovascular diseases by postmenopausal.

Effects of Porphyra tenera Extracts on Formation of Collagen Cross-link in Ovariectomized Rats (김 추출물이 갱년기 유도 흰쥐의 Collagen 가교형성에 미치는 영향)

  • The purpose of this study was to investigate the effects of Porphyra tenera (PT) extracts on formation of collagen cross-link in ovariectomized rats. From day 3 until 42 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with PT at 50 mg/kg bw/day (OVX-PT5O), 200 mg/kgbw/day (OVX-PT200). The PT ethanol extracts were orally administrated 1 mL per day. Body weight gain, food intake and food efficiency ratio were significantly different among the groups. The change of collagen content was studied in lung, bone, cartilage and skin of ovariectomized rats. The effects of PT extracts on the amount of collagen were examined by measuring the hydroxyproline, which is a specific amino acid existing in collagen. Pyridinoline is pyridinium cross-link formed in the mature form of collagen from lysine and hydroxylysine residues. Pyridinoline content was analyzed by HPLC. Pyridinoline content in bone collagen was decreased by ovariectomy but supplementation with the PT extracts was similarly increased to Sham. These results suggest that the PT supplementation could decrease bone loss in postmenopausal women.

Anti-proliferating Effects of Porphyra tenera Fractions on Several Cancer Cell Lines in uitro (김 분획물의 in vitro에서의 항발암효과)

  • This study was performed to investigate the effects of Porphyra tenera (PT) on cytotoxicity and quinone reductase (QR) activity in the cancer cells. PT was extracted with methanol and further fractionated into five different types: hexane (PTMH), ethyl-ether (PTMEE), ethylacetate (PTMEA) butanol (PTMB) and aquous (PTMA) partition layers. We determined the cytotoxic effect of these layers on C6, HepG2, MCF-7, and HT-29 cell lines by MTT assay. Among the various fractions, hexane (PTMH) of PT showed the strongest cytotoxic effect on C6, HepG2 and MCF-7 cell lines. PTMH displayed very low level of cytotoxicity at the lower concentration levels and at 300 $\mu$g/mL. PTMH resulted in 87.5$\%$ growth inhibition on C6 cell 70 $\%$ on the HepG2 cell and 89$\%$ on the MCF-7 cell, which were significantly high compared to other fractions. A 400 $\mu$g/mL PTMH concentration level, 99$\%$, 94.5$\%$ and 99$\%$ of cell growth inhibition were resulted on the same cell lines. On HT-29 cell line, both hexane (PTMH) and aqueous (PTMA) fraction of PT showed cytotoxic effects, but the Percentage was not as high as previous results tested on other cell lines such as C6 HepG2 and MCF-7 cell lines. Also, we observed quinone reductase (QR) inducing-effects in all fractions of PT on HepG2 cells. The QR inducing effects of the PTMH on HepG2 cells at 150 $\mu$g/mL concentration was 6.6 times higher than the control. Although further studies are needed, the present work suggests that PT was a potential to be used as a chemopreventive.
