포장돈육 가공공정 중 돈육과 작업자 손과의 Listeria monocytogenes의 교차오염 전이율 (Transfer Rate of Cross Contamination of Listeria monocytogenes between Pork Meat and Workers' Hands during Pork Meat Processing)

  • 본 연구에서는 포장돈육 가공공정에서 교차오염의 정도를 알아보기 위하여 실제 돈육 가공공정에서 돈육과 빈번하게 접촉하는 작업자의 손(polyethylene glove 착용, 면장갑 착용, 맨손) 및 작업도구(칼, 도마)를 대상으로 각각에 대하여 Listeria monocytogenes의 상호 교차오염 전이율을 분석하였다. 오염된 돈육에서 작업자로의 전이율은 면장갑이 100.00%로 가장 높았고, 다음이 맨손 2.513%, PEG 1.511% 순으로 나타났다. 특히, 면장갑의 경우 면장갑으로 전이된 균이 다시 맨손으로의 재오염 전이(0.08%)가 이루어지는 것으로 나타났으며, 오염된 돈육에서 칼과 도마로의 전이율은 0.352-3.791%이다. 반대로 오염된 작업자로부터 돈육 및 기구로의 전이율은 0.001-0.141%로 전체적으로 오염된 돈육으로부터의 작업자의 손과 기구로의 전이보다 상당히 낮은 수준으로 전이되었다. 따라서 PEG 착용이 작업공정에서의 교차오염을 줄이거나 예방하는 효과를 얻을 수 있는 것으로 파악되었다. 축산물 작업공정에서의 교차오염은 원료육에서 손 또는 장갑, 도마, 칼 등으로 오염이 전이되고 이들이 다시 돈육을 오염시킬 수 있으므로, 안전한 식품생산을 위해서는 이러한 교차오염의 연속성을 차단하는 것이 무엇보다 중요함을 확인하였다.

돈육의 위생화와 품질보존을 위한 감마선 조사 (Irradiation of Pork Meat for Improvement of Hygienic and Keeping Quality)

  • 이상의 결과로 돼지고기는 비교적 미생물 오염도가 낮아 1kGy 조사로 저온성 미생물을 제외한 오염미생물의 생육을 검출한계 이하로 감소시킬 수 있었으며 모든 오염미생물의 살균을 위해서는 3kGy의 선량이 요구되었다. 감마선조사와 저장기간에 따른 일반성분 변화의 경우 수분함량에 있어서만이 저장기간에 따라 다소 감소되었으며, pH 및 산도는 감마선 조사에 의한 영향은 없었으나 저장기간이 경과함에 따라 pH는 증가하고 산도는 감소하는 경향을 보였다. 총 아미노산 조성에서는 가장 많이 검출된 것이 glutamic acid였고, 저장기간에 따른 함량변화는 거의 없었으며 감마선 조사에 의한 영향도 매우 미비하였다. 무기질 함량의 경우 주요 무기질성분으로 potassium, phosphorus, sodium, magnesium 순이었고 감마선 조사에 의한 조성변화는 없었다.

서울지역 식육판매업소의 미생물 오염도 조사 (A survey of the microbial contamination level in butcher's shops in Seoul, Korea)

  • This survey was conducted to evaluate the microbial contamination level of butcher's shops in Seoul, Korea. For microbial inspections, a total of 584 samples (146 cotton work gloves, 146 utensils and equipments, 154 beef samples, 138 pork samples) were collected from butcher's shops. E. coli and pathogenic bacteria such as Staphylococcus aureus, Yersinia enterocolitica, Listeria monocytogenes, Salmonella spp., and E. coli O157:H7 were tested in the samples. As a result, the level of aerobic plate count (APC) ranged ${\leq}10^4\;CFU/cm^2$ from utensils and equipments, $10^1{\sim}10^9$ CFU/glove from cotton work gloves and ${\leq}10^6$ CFU/g from meat. The APC level of E. coli ranged ${\leq}10^1\;CFU/cm^2$ from utensils and equipments, ${\leq}10^5$ CFU/glove from cotton work gloves, and ${\leq}10^3$ CFU/g from meat, respectively. Staphylococcus aureus was detected in 2 beef samples, 1 pork sample, and 10 used cotton work gloves. Yersinia enterocolitica was detected in 3 beef samples, 1 pork sample, and 3 used cotton work gloves. Listeria monocytogenes was detected in 2 used cotton work gloves. In order to improve the sanitation status of butcher's shops, application of HACCP (Hazard Analysis Critical Control Point) or SSOP (Sanitation Standard Operating Procedure), regular hygiene education, and continuous monitoring for microorganisms will be required.

원료돈육의 오염원 추적 및 위생기준 설정에 관한 연구 (Study on Pursuit of Contamination Sources and Establishment of Sanitary Standard from Raw Pork Meat)

  • The aims of this study were to examine contamination sources and provide the basic data in establishment of sanitary standard for raw pork meat. From \"Random sampling(I)\", initial total plate counts of post-slaughter samples for the group A, B and C were 1.5${\times}10^4$cfu/$textrm{cm}^2$, 5.5${\times}10^5$cfu/$textrm{cm}^2$ and 1.8${\times}10^4$cfu/$textrm{cm}^2$, respectively, and of post-prechilling samples for the group A, B and C were 1.0${\times}10^4$cfu/$textrm{cm}^2$, 4.6${\times}10^5$cfu/$textrm{cm}^2$ and 2.5${\times}10^4$cfu/$textrm{cm}^2$, respectively. Initial total plate counts of post-transportation samples for the group D, E and F did not increased, as did the group A, B and C. From \"Normal sampling(II)\", initial total plate counts of post-slaughter, post-prechilling, post-transportation and post-2 days preservation samples were 7.3${\times}10^4$cfu/$textrm{cm}^2$, 9.6${\times}10^4$cfu/$textrm{cm}^2$, 2.0${\times}10^5$cfu/$textrm{cm}^2$ and 2.5${\times}10^5$cfu/$textrm{cm}^2$, respectively. From \"Clean sampling(III)\", initial total plate counts of post-slaughter, post-prechilling, post-transportation and post-2 days preservation samples were decreased to 7.0${\times}10^2$cfu/$textrm{cm}^2$, 7.5${\times}10^2$cfu/$textrm{cm}^2$, 8.5${\times}10^2$cfu/$textrm{cm}^2$ and 5.5${\times}10^3$cfu/$textrm{cm}^2$, respectively, compared with "Normal sampling(II)". No E. coli O157:H7, Staphylococcus aureus and Salmonella were detected at each sampling step. Consequently, a slaughter method like "Clean sampling(III)" showed a better sanitary effect to low total plate counts of 10$^2$∼10$^3$times, compared with "Normal sampling(II)". The one of contamination sources for raw pork meat was at a slaughtering step, and "Clean sampling" method may be considered as the one of sanitary standards.

몬테카를로 시뮬레이션을 이용한 돈육 생산공정에서의 Listeria monocytogenes 및 Yersinia enterocolitica의 오염수준 예측 (Predicting the Contamination of Listeria Monocytogenes and Yersinia enterocolitica in Pork Production Using Monte Carlo Simulation)

  • 본 연구는 돈육의 생산공정에서 발생 가능성이 높은 병원성 미생물인 L. monocytogenes와 Y. enterocolitica의 돈육 생산공정에서의 위해를 평가하기 위하여 확률적 접근방법을 통하여 오염수준을 예측하여 보았다. 몬테카를로 simulation을 이용하여 simulation을 한 결과 이분도체, 가공한 부분육, 저장 후 출고 전의 부분육에서의 L. monocytogenes의 오염 평균값은 각각 -4.59, -4.46, -4.45 $log_{10}CFU/cm^2$이었으며 Y. enterocolitica의 경우는 오염 평균값이 각각 -3.44, -4.00, -3.97 $log_{10}CFU/cm^2$의 값을 나타내었다. Sensitivity analysis 결과 생산된 돈육의 L. monocytogenes 및 Y. enterocolitica 오염 정도에 영향이 가장 큰 요인은 장비에서의 이들 병원성 미생물의 prevalence였다. 최종 세척의 경우는 세척의 효과가 클수록 돈육의 오염수준이 감소하는 것으로 나타났으며, 육가공장에서의 부분육 저장 온도는 $5^{\circ}C$ 이하로만 유지한다면 L. monocytogenes 및 Y. enterocolitica의 오염을 억제하기에 충분한 것으로 분석되었다.

Fate and Risk Comparison of Foodborne Pathogens in Raw Chicken, Pork, and Beef Meat at Various Temperatures

  • The objectives of this study were to investigate the behavior characteristics of pathogenic E. coli, Salmonella Typhimurium, Campylobacter jejuni, and Listeria monocytogenes in various kinds of meat (beef, chicken, and pork) and to compare their risk using FDA-iRISK. The growth of S. Typhimurium in chicken and pathogenic E. coli in pork and beef was well supported and posed a high risk. A similar trend was observed in the risk comparison results using the iRISK. When comparing total disability adjusted life years (DALY) per year based on the kinds of meat, chicken was the highest (88.2), followed by pork (58.5) and beef for "yukhoe" (18.8). When comparing scenarios grouped by bacteria, The highest total DALYs per year was observed with pathogenic E. coli (121), followed by S. Typhimurium (44.8) and L. monocytogenes (1.67E-3). These results indicate that the risk of combining meat and foodborne pathogens varies under the same distribution environment. Thus, strict management and supervision are required to store and deliver raw meat to prevent cross-contamination among the raw meats at the processing plant and retail market.

Assessment of the Microbial Level for Livestock Products in Retail Meat Shops Implementing HACCP System

  • This study aimed to examine the microbial contamination levels in livestock products at retail stores. Beef, pork, and chicken samples from raw materials and final products were obtained between January and December 2015. All homogenized meat samples (25 g) were tested for the aerobic plate count (APC), coliform count (CC), and Escherichia coli count (E. coli). The highest APCs in meat samples, by month, at retail shops were obtained in September, followed by July, May, and October (p<0.001). However, APC was the highest in summer and the lowest in winter (p<0.001). Average APCs for beef, pork, and chicken samples were 2.90, 3.19, and 3.79 Log CFU/g, respectively (p<0.05). A comparison between different months revealed that, CC levels in meat samples ranged from 0 to 1.13 CFU/g, and the highest CC was obtained in August (p<0.001). By season, the highest CC was found in the summer, followed by autumn, and spring (p<0.001). All meat samples were negative for E. coli. The average log10APC and CC for all samples was 3.10 and 0.37 Log CFU/g, respectively. Furthermore, there was a direct correlation between the season and coliform presence (p<0.001). There was also a positive correlation between the APC and CC (r = 0.517, p<0.001). The microbiological APCs for livestock products were in most cases below 106 CFU/g.

돈육 생산공정에서의 정량적 위해 평가에 fuzzy 연산의 적용 (Application of Fuzzy Math Simulation to Quantitative Risk Assessment in Pork Production)

  • 돈육 가공 공정에 대한 QRA에 Monte Carlo simulation이 적용된 바 있는데, 새로운 방법으로 fuzzy 연산을 적용하여 Monte Carlo simulation과 비교 분석하였다. Carcass단계에 대한 오염 예측치인 fuzzy 값과 Monte Carlo simulation 확률분포 값의 기술통계량인 평균값은 각각 -4.393 log $CFU/cm^2$, -4.589 log $CFU/cm^2$ 로 나타났으며, processing 단계에서는 -4.185 log $CFU/cm^2$, -4.466 log $CFU/cm^2$으로 두 가지 접근 방법들이 비슷한 경향을 보였다. Fuzzy 값은 Monte Carlo simulation 확률분포 값을 포함하는 것으로 나타났다. 한편 최근 국내에서는 위해 평가에 대한 연구가 많이 이루어지고 있는데 대부분 데이터 분석은 Monte Carlo simulation에만 의존하고 있고, 다른 접근 방법에 대한 연구는 미진한 실정이다. 따라서 본 연구는 위해 평가를 위한 방법적 도구들을 개발하는데 새로운 접근 방향을 제시하였다 또한 향후 fuzzy 연산법은 데이터가 불충분한 위해 평가의 초기 단계에서 유용하게 사용될 수 있는 방법이 될 것이다.

Comparison of Quality of Bologna Sausage Manufactured by Electron Beam or X-Ray Irradiated Ground Pork

  • Ground lean pork was irradiated by an electron beam or X-rays to compare the effects of two types of radiation generated by a linear accelerator on the quality of Bologna sausage as a model meat product. Raw ground lean pork was vacuum packaged at a thickness of 1.5 cm and irradiated at doses of 2, 4, 6, 8, or 10 kGy by an electron beam (2.5 MeV) or X-rays (5 MeV). Solubility of myofibrillar proteins, bacterial counts, and thiobarbituric acid reactive substance (TBARS) values were determined for raw meat samples. Bologna sausage was manufactured using the irradiated lean pork, and total bacterial counts, TBARS values, and quality properties (color differences, cooking yield, texture, and palatability) were determined. Irradiation increased the solubility of myofibrillar proteins in a dose-dependent manner (p<0.05). Bacterial contamination of the raw meat was reduced as the absorbed dose increased, and the reduction was the same for both radiation types. Differences were observed only between irradiated and non-irradiated samples (p<0.05). X-ray irradiation may serve as an alternative to gamma irradiation and electron beam irradiation.

인천지역 도축장에서 생산된 돼지고기의 미생물 오염도 조사 (Survey on the contamination of microorganisms in pork from slaughterhouse in Incheon area)

  • For measuring general bacterial count and Escherichia coli count, the standard plate count was used to conduct a test on 113 cases of pork carcass surfaces from slaughterhouses in Incheon area from January to February 2003. And Salmonella spp, Staphylococcus aureus and Listeria monocytogenes was used to conduct a test on 68 cases of pork carcass surfaces and 76 cases of feces, that is to say, a total of 144 cases. The results were obtained as follows: In the case of general bacterial count, 29 cases(25.7%) were in the range of 100∼999 and 62 cases(54.9%) were in the range of 1,000∼9,999 and 22 cases(19.4%) were in the range of 10,000∼99,999. Meanwhile as regards E coli count, 22 cases(19.4%) were in the range of 1∼9 and 69 cases(61.2%) were in the range of 10∼99 and 22 cases(19.4%) were in the range of 100∼999. On 68 cases of pork carcass surfaces, 7 strains(10.2%) of Salmonella spp, and 12 strains(17.6%) of S aureus were detected and 27 strains(39.7%) of L monocytogenes, respectively. As for the detected Salmonella spp, 6 strains of the B group, 3 strains of S enterica subsp salame and 2 strains of S typhimurium were detected, respectively. On 76 cases of feces, 14 strains(18.4%) of Salmonella spp, and 15 strains(19.7%) of L monocytogenes and 14 strains(18.4%) of S aureus were detected respectively. As for the detected Salmonella spp, 6 strains of the B group, 4 strains of S derby and 8 strains of the C group, 5 strains of S rissen were detected, respectively. All of 42 strains of L monocytogenes were type 1. As a result of conducting a toxin test on the detected S aureus, all of 26 strains were found to be non-toxin.
