침자 술기 실습 모델에서 침감의 정량적 특성에 관한 연구 (A Study on the Quantitative Characteristics of Needle Force on the Acupuncture Practical Model)

  • Objectives : In this study, we quantitatively evaluated the needle forces using needle insertion-measurement system and compared the needle sensation of each acupuncture practical model. Methods : After inserting acupuncture with a sensor to six models, a lifting-thrusting motion was implemented using the needle insertion-measurement system. The needle force was measured repeatedly, and the measurement was analyzed based on the modified Karnopp friction model for a comparison of friction coefficients. After the insertion, practitioners did lifting-thrusting manipulations. They quantified the similarity of needle sensation with VAS (Visual Analogue Scale). Results : When friction force and coefficients of friction in five different models were compared with a porcine shank model, all five models were significantly different from a porcine shank model, cotton and apple showing the closest frictional values to that of a porcine shank model. In the Cp and Cn values of cotton and in the Cp values of IM injection pad, there was no statistically significant difference. The similarity of the needle sensation between the porcine shank and five models was the highest in the apple, and overlapping papers was the lowest. Conclusions : This study quantitatively compared the physical forces in the practical model when implementing lifting-thrusting manipulations, using a needle insertion-measurement system. We suggest that a reproducible exercise model that reflects the characteristics of various human tissues, such as viscoelasticity or strength, needs to be further developed. This will contribute to establishing standardized acupuncture practice training.

Ex Vivo Permeability Characteristics of Porcine Buccal Mucosa to Drugs with Various Polarity

  • The aim of this study was to analyze characteristics of the barrier function of excised porcine buccal mucosa to the test compounds, estradiol, propranolol HCI, melatonin, and mannitol with a wide range of partition coefficient values. The permeability of melatonin was measured through frozen, stored, and fresh porcine buccal mucosa to examine the impact of storage conditions on the permeability of porcine buccal mucosa. The results demonstrated that the ex vivo permeability of the porcine buccal mucosa was greater for more lipophilic solutes, which was consistent with a series of molecules transported by passive transepithelial diffusion. The melatonin permeation profiles through frozen, stored, and fresh mucosa illustrated that damage was incurred by the freezing process of the mucosal tissue, leading to loss of the barrier function and thereby an increased permeation coefficient. It can be observed that the influence of compound lipophilicity on the association of the compounds with buccal mucosa was clear. The relationship between permeation coefficient and Log P values for the four compounds investigated demonstrated a proportional relationship, further confirming the importance of the lipophilicity of a compound to permeate the buccal mucosa. These results showed that the ex vivo porcine buccal mucosa model is a suitable tool to screen oral mucosal permeability.

Freeze-dried bovine amniotic membrane as a cell delivery scaffold in a porcine model of radiation-induced chronic wounds

  • Background Locoregional stem cell delivery is very important for increasing the efficiency of cell therapy. Amnisite BA (Amnisite) is a freeze-dried amniotic membrane harvested from bovine placenta. The objective of this study was to investigate the retention of cells of the stromal vascular fraction (SVF) on Amnisite and to determine the effects of cell-loaded Amnisite in a porcine radiation-induced chronic wound model. Methods Initially, experiments were conducted to find the most suitable hydration and incubation conditions for the attachment of SVF cells extracted from pig fat to Amnisite. Before seeding, SVFs were labeled with PKH67. The SVF cell-loaded Amnisite (group S), Amnisite only (group A), and polyurethane foam (group C) were applied to treat radiation-induced chronic wounds in a porcine model. Biopsy was performed at 10, 14, and 21 days post-operation for histological analysis. Results Retaining the SVF on Amnisite required 30 minutes for hydration and 1 hour for incubation. A PKH67 fluorescence study showed that Amnisite successfully delivered the SVF to the wounds. In histological analysis, group S showed increased re-epithelialization and revascularization with decreased inflammation at 10 days post-operation. Conclusions SVFs had acceptable adherence on hydrated Amnisite, with successful cell delivery to a radiation-induced chronic wound model.

Porcine growth hormone induces the nuclear localization of porcine growth hormone receptor in vivo

  • Objective: Recent studies have challenged the traditional paradigm that growth hormone receptor (GHR) displays physiological functions only in the cell membrane. It has been demonstrated that GHR localizes to the cell nucleus and still exhibits important physiological roles. The phenomenon of nuclear localization of growth hormone (GH)-induced GHR has previously been described in vitro. However, until recently, whether GH could induce nuclear localization of GHR in vivo was unclear. Methods: In the present study, we used pig as an animal model, and porcine growth hormone (pGH) or saline was injected into the inferior vena cava. We subsequently observed the localization of porcine growth hormone receptor (pGHR) using multiple techniques, including, immunoprecipitation and Western-blotting, indirect immunofluorescence assay and electronmicroscopy. Results: The results showed that pGH could induce nuclear localization of pGHR. Taken together, the results of the present study provided the first demonstration that pGHR was translocated to cell nuclei under pGH stimulation in vivo. Conclusion: Nuclear localization of pGHR induced by the in vivo pGH treatment suggests new functions and/or novel roles of nuclear pGHR, which deserve further study.

The Effect of Storage Conditions on the Permeability of Porcine Buccal Mucosa

  • The impact of storage conditions on the permeability of porcine buccal mucosa to [$^3H$]water and [$^{14}C$]mannitol was assessed. The fresh pocine buccal tissue (fresh tissue) was obtained by utilizing pig heads within 24 hours of slaughter. The stored and frozen porcine buccal tissues (stored tissue and frozen tissue) were obtained after the storage of the tissue intact in the pig heads at $4^{\circ}C$ or -$20^{\circ}C$, respectively, for 24 h. The results demonstrated that the barrier properties of the porcine buccal mucosa were maintained with regard to [$^3H$]water permeability when stored at $4^{\circ}C$ for 24 h. However, freezing the tissue resulted in tissue damage illustrated by a significant increase in [$^3$H]water permeability. [$^{14}C$]Mannitol does not appear to be a suitable model solute to assess the ex vivo permeability of porcine buccal mucosa due to its extremely low permeability.

이종이식재 개발을 위한 감마선 조사 돼지 인대의 미생물학적 안전성 및 인장강도 (Microbiological Safety and Tensile Strength of Gamma Irradiated Porcine Tendon for the Development of Xenografts)

  • This study was to determine the microbiological safety and tensile strength of gammairradiated porcine tendon for the development of safe xenografts. Escherichia coli and Bacillus subtilis were used as model pathogens and inoculated as $10^6{\sim}10^7log$ colonies forming unit $(CFU)g^{-1}$. As model virus from porcine, porcine parvovirus (PPV), bovine viral diarrhoea virus (BVDV) and poliovirus were inoculated as $10^5{\sim}10^6$ tissue culture infectious dose $(TCID)_{50}g^{-1}$ into porcine skin. The $D_{10}$ value of E. coli and B. subtilis was measured as $0.32{\pm}0.082kGy$ and $4.0{\pm}0.312kGy$, respectively. Additionally, the $D_{10}$ values of PPV, BVDV and poliovirus were also shown as $1.75{\pm}0.131kGy$, $3.70{\pm}0.212kGy$ and $6.26{\pm}0.332kGy$, respectively. Gamma irradiation decreased the tensile strength of porcine tendon. Results indicate that microbiological safety of porcine tendon can be improved significantly by gamma irradiation. However, further studies are needed to improve the tensile strength of gamma-irradiated porcine tendon.

Gene expression profiling after ochratoxin A treatment in small intestinal epithelial cells from pigs

  • Ochratoxin A (OTA) is a well-known mycotoxin that causes disease through the ingestion of contaminated food or feed, for example, in the porcine industry. The intestinal epithelium acts as the first barrier against food contamination. We conducted a study on the exposure of the porcine intestinal epithelium to OTA. We used the intestinal porcine epithelial cell line IPEC-J2 as an in vitro model to evaluate the altered molecular mechanisms following OTA exposure. Gene expression profiling revealed that OTA upregulated 782 genes and downregulated 896, totalling 1678 differentially expressed genes. Furthermore, immunofluorescence, quantitative real-time polymerase chain reaction, and western blotting confirmed that OTA damages the tight junction protein ZO-1. Moreover, OTA activated the expression of inflammatory genes (IL-6, IL-8, IL-10, NF-kB, TLR4, and TNF-α). In summary, this study confirmed that OTA alters various molecular mechanisms and has several adverse effects on IPEC-J2 cells.

  • 돼지 웅성 페르몬인 5a-androst-16-en-3-one을 대체할 수 있는 활성 분자를 탐색하여 가축의 생산과 수요를 조절하기 위한 생물학적 자극 통제 수단으로 활용하고자 냄새 분자로서 2-cyclohexyloxytetrahydrofurane (A) 및 2-phenoxytetrahydrofurane (B) 유도체들의 구조 변화와 수용체인 porcine odorant binding protein (pOBP)에 대한 결합친화력 상수(p[Od.]/sub 50/) 사이의 정량적인 구조-활성관계에 관한 분자 HQSAR 모델XI을 유도하였다. 냄새 분자 중에서 cyclohexyl-치환체(A)가 phenyl-치환체(B)보다 높은 결합 친화력을 나타내었으며(A>B) 모델XI은 분자 조각크기(5∼8), 홀로그램 길이(97 bin)의 키랄성(chirality)조건에서 예측성(q²=0.916)과 상관성(r²=0.988)이 매우 양호하였다. 기여도(contribution map)로부터 냄새 분자의 결합 친화력 상수에 기여하는 부분은 2-oxyfuryl group의 C3 및 C5 원자인 반면에 cyclohexyl 고리상 tert-butyl group의 중심 탄소 원자와 furyl group의 C4 원자 부분은 기여하지 않았다.

Comparison of porcine and human acellular dermal matrix outcomes in wound healing: a deep dive into the evidence

  • Acellular dermal matrices (ADM) are a novel graft. The goal of this systematic review is to evaluate the evidence behind differences in human and porcine ADM, irrelevant of manufacturing method, and to determine if there is enough of an evidence base to change clinical practice. An extensive literature search was performed through MEDLINE and Embase with search terms defining a population, intervention and outcome. Title and abstract exclusion were performed with other exclusion criteria. In 191 articles were found after exclusion of duplicates, with only 29 remaining following exclusions. Ten studies were found to have level I and II evidence (I=3, II=8), of which two were histopathological, one was an animal model, one was a systematic review, and six were clinical. The remaining studies were reviewed and considered for discussion, but did not hold high enough standards for medical evidence. Strong clinical evidence already exists for the use of human ADM, but questions of access, cost, and ethics require consideration of a xenograft. Histopathologically, evidence suggests minimal long-term differences between human and porcine ADM, although there is a short acute immune response with porcine ADM. Clinically, there is limited difference in outcomes, with a small range in effect of different ADM preparations. Considering the effectiveness of ADM in wound healing, more high-level research with appropriate statistical analysis to facilitate a future meta-analysis is recommended to justify a transition from human to porcine ADM.

Use of a multinomial logistic regression model to evaluate risk factors for porcine circovirus type 2 infection on pig farms in the Republic of Korea

  • The current study identified risk factors associated with porcine circovirus type 2 (PCV2) infection on pig farms in the Republic of Korea using a multinomial logistic regression model to evaluate the PCV2 infection status of pigs at different growth stages. Compulsory disinfection of visitors (odds ratio [OR]: 0.019, 95% confidence interval [CI]: <0.001-0.378, p=0.0095), compulsory registration of visitors (OR: 0.002, 95% CI: <0.001-0.184, p=0.0070), regular blood testing (OR: 0.012, 95% CI: <0.001-0.157, p=0.0007), and running on-farm biosecurity learning programs for workers (OR: 0.156, 95% CI: 0.040-0.604, p=0.0072 and OR: 0.201, 95% CI: 0.055-0.737, p=0.0155, respectively) were identified as factors which could reduce the risk of PCV2 infection. However, visitation by a regular veterinarian (OR: 32.733, 95% CI: 3.768-284.327, p=0.0016) was associated with PCV2 infection.
