Induction of Somatic Hybrid by Protoplast Fusion between Populus koreana × P. nigra var. italica and P. euramericana cv. Guardi (수원포플러와 구아디 포플러 원형질체(原形質體) 융합(融合)에 의한 체세포잡종체(體細胞雜種體) 유도(誘導))

  • Protoplasts isolated from leaf mesophyll tissues of Populus koreana ${\times}$ P. nigra var. italica were fused with those of P. euramericana cv. Guardi. Well expended healthy leaves of 5 to 7 week-old-plantlet grown in vitro were used as source materials. Leaves from P. koreana ${\times}$ P. nigra var. italica and P. euramericana cv. Guardi were digested in enzyme solution I (2.0% Cellulase, 1.2% Hemicellulase, 0.4% Macrozyme, 2.0% Driselase, 0.05% Pectolyase ; w/v) and enzyme solution II (1.0% Cellulase, 1.2% Hemicellulase, 0.4% Macrozyme, 2.0% Driselase, 0.05% Pectolyase ; w/v), respectively, The highest frequency of fusion among the protoplasts originated from the two source materials was approximately 21% using 40% PEG or 15% dextran. In addition, fusion frequency was enhanced by incorporating 30mM of $Ca^{2+}$ in eluting solution at pH 10.5. Dividing cells and/or mint-calli were obtained by culturing the fusion products in a liquid 8p-KM medium supplemented with 0.6M sucrose, $0.45{\mu}M$ 2, 4-D, and $0.5{\mu}M$ BA. Shoots were regenerated from the fusion product-derived calli after culture on MS medium containing $5.0{\mu}M$ zeatin. To verify the putative hybrid or cybrid, SDS-PAGE was carried out. From the 24 regenerants, just two plants showed intermediate protein band patterns compared with those of the original source plants.

Pretreahent and Saccharification of Poplar by Solvent Extraction (유기용매 추출법에 의한 포플라의 전처리 및 당화)

  • The effect of the pretreatment by solvent extraction on the saccharification of poplar (Populus euramencana) was studied. The solvent system was Phenol/H2O(Uncatalyzed)and Phenol/H2O/HCl catalyst system. When the poplar was pretreated by uncatalyzed system, the best result of the saccharification was total of 43.9 g/1 reducing sugar produced and 83.5% of carbohydrated conversion was obtained at 19$0^{\circ}C$, 60 minutes. Total wood yield and the lignin removal were 46.3% and 98.2% respectively. The use of acid catalyst was unsuitable to increase he efficiency of saccharification.

A Study on the Awareness Survey of Government Officials for Improvement of Urban Roadside Tree Management (도시 가로수의 관리개선을 위한 공무원 의식조사에 관한 연구)

  • This study aimed to provide basic data for future management of roadside trees by analyzing problems associated with plantation and management of roadside trees through categorization of functions and organizational structure, plantation and management, and management regulations and generating improvement opportunities based on the result of a survey on the awareness of government officials who are directly in charge of managing roadside trees in 31 cities & counties within Kyonggi province. The summary of key results of this study is as follows. First, "roadside tree-related function" is still not considered to be independent. About a half of government officials in charge did not majored in this field and they spent only about "2.09hours", in average, in the roadside tree-related function out of 8 working hours per day. Second, regarding problems and improvement opportunities in roadside tree-related function, lack of time was considered to be the biggest problem in overall management of roadside trees. As for improvement opportunities, "increase of dedicated manpower" and "system development" to facilitate efficient processing were suggested. Third, an annual budget for roadside tree-related affairs was merely 92 million won per city/county. A registry for management of roadside trees was kept manually. As for the roles of roadside trees, improvement of landscape and securing of green area itself were valued highly. Fourth, Ginkgo biloba, Zelkova serrata, and Prunus yedoensis were suggested to be the most appropriate species for roadside tree and Platanus occidentalis, Populus albaglandulosa, Populus euramericana, and Robinia pseudo-acacia were mentioned to be the least appropriate species.

The Cd and Pb Accumulation in Various Tissues of Rooted Cuttings of Four Populus Species Inoculated with Ectomycorrhizal Fungi, Pisolithus tinctorius (모래밭버섯 균근균(菌根菌)으로 접종(接種)한 포플러 4개(個)개 수종(樹種) 삽목묘(揷木苗)의 체내(體內) 부위별(部位別) Cd과 Pb 축적(蓄積) 특성(特性))

  • The objectives of this study were to understand the characteristics of Cd and Pb accumulation in various tissues of poplar species and the effects of ectomycorrhizal inoculation on the accumulation of above two heavy metals in the tissues. The mycelial inoculum of ectomycorrizal fungus, Pisolithus tinctorius was produced in peatmoss and vermiculite mixtures, and inoculated into potted soil with fresh cuttings of four species of Populus, P. alba ${\times}$ glandulosa (Pag). P. koreana ${\times}$ nigra var. italica (Pkn), P. nigra ${\times}$ maximowiczii(Pnm), and P. euramericana(Pe). The potted soils were added with 0, 30, and 80 ppm Cd, and 0, 50, and 300 ppm Pb. The cuttings were grown outdoors for about five months until the plants were harvested for measurement of growth, mycorrhizal infection, and metal contents in leaves, stems, and roots. The total dry weight of Pe treated with Cd and Pb was increased by mycorrhizal inoculation, while that of three other species was not affected by the inoculation. Cd was accumulated in the highest concentration in Pag and its concentration was increased by four times by mycorrhizal inoculation. The Pag accumulated Cd in the highest concentration in the leaves, while three other species accumulated Cd most in the roots. Pb was accumulated in the highest concentration in the roots of all the four species, while Pkn accumulated Pb in the leaves as much as in the roots. Without mycorrhizal inoculation Pe accumulated Pb most among the four species, while Pkn with mycorrhizal inoculation accumulated Pb two times more than in the same species without mycorrhizal inoculation. It was concluded that Pag was the most effective species among the four poplar species in Cd absorption from contaminated soil, and that Pe instead effectively absorbed Pb. Mycorrhizal inoculation increased the Cd accumulation in the tissues by four times in Pag and also increased Pb accumulation by two times in Pkn, with leaves being the major sites of metal accumulation. It may be possible to use these two poplar species in remedying the metal in the soil through the raking and removing the litter out of the contaminate site.

  • Italian poplar (Populus euramericana) rotary veneers were press-dried and air-dried to study drying curves, thickness shrinkage and width shrinkage of them under several drying conditions such as drying temperatures and veneer thicknesses. The results of the study are summarized as follows; 1. In press-drying, drying times from green condition to about final moisture content of 10 percent with veneer thickness of 0.6mm by platen temperatures required about 10 minutes at platen temperature of $100^{\circ}C$, 3 minutes at $110^{\circ}C$, 1 minute at $120^{\circ}C$ and $130^{\circ}C$, respectively. 2. In press, drying, drying times from green condition to about final moisture content of 10 percent by veneer thicknesses required 2 minutes at veneer thickness of 0.6mm, 4 minutes at 1.2mm,6 minutes at 1.8mm and 9 minutes at 3.6mm, respectively. 3. In air-drying, drying times from green condition to air-dried moisture content by veneer thicknesses required 15 hours at veneer thickness of 0.6mm, 18 hours at 1.2mm and 23 hours at 2.4mm, respectively. 4. Thickness shrinkage of press-drying was remarkably greater than that of air-drying, but width shrinkage of press-drying was rather smaller. 5. Difference of thickness shrinkage among platen temperatures was insignificant, and width shrinkage at platen temperature of $130^{\circ}C$ was the least.

Comparative Analysis of Gross Calorific Value by Determination Method of Lignocellulosic Biomass Using a Bomb Calorimeter

  • This study was conducted to compare and analyze gross calorific values from measurement methods of lignocellulosic biomass and calculation data from calorific value prediction models based on the elemental content. The deviation of Liriodendron tulipifera (LT) and Populus euramericana (PE) was shown 7.7 cal/g and 7.4 cal/g respectively in palletization method, which are within repeatability limit 28.8 cal/g of ISO FDIS 18125. In the case of Thailand charcoal (TC), nontreatment method and palletization method was satisfied with repeatability limit as 22.8 cal/g and 8.8 cal/g respectively. Seowon charcoal (SC) was shown deviation of 11.4 cal/g in nontreatment method, because the density and chemical affinity of sample increases as the carbon content increases from heat treatment at high temperature in the case of TC and SC. In addition, after applying the elemental content of each of these samples to the calorific value prediction models, the study found that Model Equation (3) was relatively consistent with measured calorific values of all these lignocellulosic biomass. Thus, study about the correlation between the density and size of particle should be conducted in order to select the measurement method for a wide range of solid biofuels in the future.

Enzymatic hydrolysis and micro-structure of ozone treated wood meal (오존 처리에 의한 목재 세포벽의 미세구조변화와 효소가수분해)

  • Pine (Pinus densiflora) and aspen (Populus euramericana) wood meals were treated with ozone at various time schedule in acidic condition. The lignin contents and surface area of the ozone treated wood meals were determined and the enzymatic hydrolysis rate of ozonated wood meals was evaluated. The feasibility of enzymatic hydrolysis of the ozone treated wood meal was obviously influenced with the degree of delignification. After ozone treatment of wood meal for 10min, total pore volume were slightly increased in the surface of wood meal. When wood meals were treated with ozone longer than 10min, few change in the pore volume was observed. However, the area of over $50{\AA}$ of pore size is increased with ozonation time. As a conclusion, the rate of enzymatic hydrolysis of wood is more effective with the pore size distribution than the total pore volume.

Dimensional Stability of Plastic Processing Wood Material - Compression Wood and Bentwood -

  • This study was carried out to assess the dimensional stability of wood material treated by plastic processing for bentwood and compression wood. The evaluation method was different between two wood materials, but the treatments for them were very similar to each other. One of the main methods is heat treatment with sufficient water vapor. In bentwood, the used species were painted maple (Acer mono), bitter wood (Picrasma quassioides) and birch (Betula schmidtii). Steaming was the worst treatment method for dimensional stabilization of bentwood. The best results could be attained with PEG treatment for dimensional stabilization of bentwood. Dimensional stability of bitter wood was found to be conspicuous. However the steaming treatment at lower temperatures, i.e., about $130^{\circ}C$ was not suitable for dimensional stability of bentwood. In compression wood, the used specimen was Italian poplar wood (Populus euramericana). Two heat compressive pressing conditions, an open-press system and an air-tighten closed-press system, were used. The recovery rate was measured after boiling and/or absorbing in water to estimate the dimensional stability of heat compressed wood. The best dimensional stability of compressed wood in the air-tighten closed-press system was found to be better at $200^{\circ}C$ than $180^{\circ}C$. The best compression rate for dimensional stability was 73 percent.

Comparison of Dynamic Sorption and Hygroexpansion of Wood by Different Cyclic Hygrothermal Changing Effects

  • To investigate the dynamic sorptive and hygroexpansive behaviors of wood by different cyclic hygrothermal changing effects, poplar (populus euramericana Cv.) specimens, were exposed to dynamic sorption processes where relative humidity (RH) and temperature changed simultaneously in sinusoidal waves at 75-45% and $5-35^{\circ}C$ (condition A) and where RH changed sinusoidally at 75-45% but temperature was controlled at $20^{\circ}C$ (condition B), both for three cyclic periods of 1, 6, and 24 h. Moisture and dimensional changes measured during the cycling gave the following results: Moisture and transverse dimensional changes were generally sinusoidal. Moisture and dimensional amplitude increased with increasing cyclic period but all were lower for thicker specimens. The amplitude ratio of condition A to condition B ranged from 1.0 to 1.6 with the maximum value of 1.57 occurring at the shortest cyclic period, not as much as expected. T/R increased as cyclic period increased or specimen thickness decreased. T/R from condition B was weaker than that from condition A. Sorption and swelling hysteresis existed in both conditions. Sorption hysteresis was negatively related to cyclic period but in positive correlation with specimen thickness. Sorption hysteresis was found more obvious in condition B, while moisture sorption coefficient and humidity expansion coefficient showed the opposite results.

Heating Compression of Italian Poplar (Populus euramericana) Wood - Dimensional Stability Against Moisture -

  • This study was carried out to estimate the property of dimensional stability of heat compression of italian poplar wood with low density. Firstly, two levels of pressure conditions were applied using the closed and open-press system. The thermal treatment temperatures were 180℃ and 200℃, respectively. Water absorption tests were conducted in water bath at 25℃ and 100℃ for 35 hours and 1 hour, respectively. The compression rates of wood were 47 percent, 60 percent, and 73 percent, respectively. From these tests, it was found that the dimensional stability of the closed-press system was superior to that of the open-press system. Furthermore, the dimensional stability of compressed wood in the closed-press system was better at 200℃ than 180℃. In compression rate, dimensional stability of 73 percent compression rate was the best result. Considering these results, the best conditions for the dimensional stability of compressed wood were those of the closed-compressing system at high temperatures above 200℃ and larger compression rate. Therefore, it was concluded that the dimensional stability of wood is improved at higher temperature and larger deformation.
