Effects of Three Strains of Bacillus subtilis Supplemented to Diets on Egg Quality, Intestinal Microflora and Tibia in The Late Stage of Laying Hens (산란계 사료 내 3종류의 Bacillus subtilis의 첨가가 산란후기 계란 품질과 장내 미생물 및 경골에 미치는 영향)

  • The objective of this study was to investigate the effects of three strains of Bacillus subtilis (B. subtilis) supplemented to diets on egg production, egg quality, egg yolk cholesterol levels, the profile of cecal microflora, and tibia characteristics in laying hens. One hundred sixty 76-week-old Hy-Line Brown layers were randomly divided into 4 groups with 4 replicates per group (10 birds per replicate). Birds in the control group were fed a corn-soybean meal based diet. The remaining three treated groups were fed the control diet containing either 0.05% B. subtilis Ch3 (T1), 0.05% B. subtilis Ch3 + B. subtilis W1 (T2) or 0.05% B. subtilis commercial product (T3) for 6 weeks, respectively. There were no differences in feed intake, egg weight, egg production and egg mass among the groups. The dietary supplementation of B. subtilis improved eggshell strength and Haugh units compared to those of control (P<0.05). The activities of GOT and GPT in serum were not also affected by the dietary treatments. The population of total microbes and lactic acid bacteria in cecum were significantly increased by the dietary B. subtilis (P<0.05), but not the coliforms. The cholesterol concentration in egg yolk and serum in the treated groups were significantly decreased compared to those of control (P<0.05). Also, The levels of phospholipids in serum were significantly decreased compared to those of control (P<0.05). The supplementation of three strains of B. subtilis to diets significantly increased the contents of tibia ash compared to that of control (P<0.05). Thus, this study showed significant improvements in egg quality, such as eggshell strength and Haugh unit, by dietary B. subtilis strains. The B. subtilis strains added to the diets modulated the profiles of cecal microflora, reflecting beneficial effects in laying hens.

Investigation of Coat Color Candidate Genes in Korean Cattle(Hanwoo) (한우에서 모색관련 유전자 변이에 관한 연구)

  • Most cattle breeds have a coat color pattern that is characteristic for the breed. Korean cattle(Hanwoo) has a coat color ranging from yellowish brown to dark brown including a red coat color. Variation in the Hanwoo coat color is likely to be the effects of modified genes segregating within the Hanwoo breed. MC1R encoded by the Extension(E) locus was almost fixed with recessive red e allele in the Hanwoo, but other gene(s) might be affecting the variation of the Hanwoo coat color into yellowish to red brown. We have analyzed a segregation of coat color in the F2 families generated from two Hanwoo bulls(yellowish brown) mated to six F1 dams(black) derived from Hanwoo and Holstein crosses. Segregation of coat color in the offspring found a ratio of 1(yellowish brown) : 1(black) and this ratio indicates that a single gene may play a major role for the Hanwoo coat color. We further investigated SNPs in MC1R, ASIP and TYRP1 loci to determine genetic cause of the Hanwoo coat color. Several polymorphisms within ASIP intron 2 and TYRP1 exons were found but not conserved within the Hanwoo population. However, the segregation of the MC1R e allele was completely associated with the Hanwoo coat color. Based on this information, it is clear that the MC1R e allele is mainly responsible for the yellowish red Hanwoo coat color. Further study is warrant to identify possible genetic interaction between MC1R e allele and other coat color related gene(s) for the variation of Hanwoo coat color from yellowish brown to dark brown. Analysis of Dietary Fiber Intake in the Korean Adult Population Using 2001 Korean National Health and Nutrition Survey Data and Newly Established Dietary Fiber Database (식이섬유 D/B 구축과 2001 국민건강 영양조사 식이섬유 섭취량 재평가 -20세 이상 성인을 대상으로-)

  • The present study estimates intake levels of dietary fiber (DF) in Korean adults aged 20 and over, using a newly established dietary fiber database for 3,149 food items, as well as 24-hour recall method data from the 2001 Korean National Health and Nutrition Survey. Dietary fiber intake and food groups are analyzed by gender, age, and region. The average dietary fiber intake, per capita of Korea was estimated to be 12.24g/1,000kcal or 23.58g/day. Calorie-based dietary fiber intake for 20-49 year-old-Korean males, 20-29 year-old females, and the adults who resided in metropolitan areas was under the Adequate Intake for DF, 12 g/1,000 kcal. Further, the dietary fiber intake after adjusting energy intake in people over 75 year-old was estimated to be 75% of AI. Vegetables, cereals and fruits were three major sources of DF for Korean, making up approximately 75% of DF. Regarding the subjects of this study, major sources of dietary fiber were Kimchi and well-polished rice, which supplied 13.98% and 9.16% of total dietary fiber intake, respectively. The result of this study could contribute to the establishment of DRIs for dietary fiber, after adjusting energy intake for Korean aged 75 years and over. The beneficial health effects of DF and the necessity of nutritional education in this area should be continuously emphasized concerning 20-29 year-old people and metropolitan adults.

Estimated flavonoid intakes according to socioeconomic status of Korean adults based on the Korea National Health and Nutrition Examination Survey 2007~2012 (우리나라 성인의 사회경제적 수준에 따른 플라보노이드 섭취현황 : 2007~2012년 국민건강영양조사 자료를 이용하여)

  • Purpose: The purpose of this study was to estimate the dietary flavonoid intakes of Korean adults according to socioeconomic status. Methods: Using data from the 2007~2012 Korea National Health and Nutrition Examination Survey, a total of 31,112 subjects aged over 19 years were included in this study. We estimated individuals' daily intakes of total flavonoids and seven flavonoid subclasses, including flavonols, flavones, flavanones, flavan-3-ols, anthocyanins, proanthocyanidin, and isoflavones,by linking food consumption data with the flavonoids database for commonly consumed Korean foods. We compared intakes of flavonoids according to the levels of household income and education. Results: Average dietary flavonoid intakes of the study subjects were 321.8 mg/d in men and 308.3 mg/d in women. Daily flavonoid intakes were positively associated with household income level (p < 0.0001) and education level (p < 0.0001). The subjects in the highest household income and highest education level group (OR 0.37, 95% CI 0.30~0.45, p < 0.0001 in men, OR 0.50, 95% CI 0.41~0.60, p < 0.0001 in women) had a lower likelihood of having low total flavonoid intake (less than 25 percentile) compared to the lowest household income and lowest education level group. The food group that contributed to total flavonoid intake with the biggest difference between the lowest and highest groups for both household income level and education level was beverages. Conclusion: This study shows that socioeconomic status was positively associated with flavonoid intake in a representative Korean population. Further research is needed to analyze the association of flavonoid intake with health outcomes according to socioeconomic status such as household income and education level.

Studies on the establishment and characteristics of habitat use of muskrat(Ondrtra zibethicus) in South Korea (사향쥐(Ondrtra cuniculus)의 정착 및 서식지 이용에 관한 연구)

  • This study was conducted to determine whether invasive muskrats settle in the natural ecosystem of Korea. The study also aimed to provide information on the selection of target areas, basic behavioral characteristics, and habitat use-related data, all of which are essential for effective management strategy establishment and site management. To this end, a trend in international muskrat management was identified to examine the management direction according to the settlement stage. We also secured the location of 72 domestic farmers and classified five regions in Gyeonggi-do, Chungcheongnam-do, Gyeongsangbuk-do, and Sejong Special Self-governing City into regions with high potential for natural leakage to raise the need for a quick survey. We determined that muskrats successfully settled in the natural ecosystem of Korea, based on the fact that we consistently identified a muskrat habitat at the same point for three years, from 2016 to 2018, the age of the captured population was estimated to be 1.2 years, and there had been no additional make-up factors near the habitat since 2012. The muskrats that settled in Korea showed a small home range of 0.0027 ㎢ (MCP 95%) and were estimated to show regular mobility before Spring and Autumn. Also, the areas where the muskrat habitat was observed had a high level of habitat environment disturbance and their ecological and food intake characteristics reflected that disturbance. Muskrat traces were high in hydrophyte vegetation, which can be used as a hideout in open waters and where the growth of both floating leaved plants and floating plants are prevalent. Since international invasive muskrats have already settled in the domestic ecosystem and there is concern about their proliferation in the future, the government must consider a preemptive response and scientific management.

A Study on Development of Guideline on Writing Technical Document for Electrical Medical Devices: Dental X-ray Equipment (치과용엑스선장치의 기술문서 작성을 위한 가이드라인 개발 연구)

  • Due to recent population aging, the number of check-up for senior citizens has increased steadily. According to this trend, the market size of dental X-ray equipment and the number of approval and review for these devices have simultaneously increased. The technical document of medical device is required for approval and review for medical device, and medical device companies needs to have work comprehension and expertise, as the document needs to include the overall contents such as performances, test criteria, etc.. Yet, since most of domestic manufacturers or importers of medical devices are small businesses, it is difficult for them to recruit professional manpower for approval of medical devices, and submission of inaccurate technical documents has increased. These problems lead to delay of the approval process and to difficulties in quick entering into the market. Especially, the Ministry of Food and Drug safety (MFDS) standards of a dental extra-oral X-ray equipment, a dental intra-oral X-ray equipment, an arm-type computed tomography, and a portable X-ray system have been recently enacted or not. this guideline of dental X-ray equipment adjusting revised standards was developed to help relative companies and reviewers. For this study, first, the methods to write technical document have been reviewed with revised international and domestic regulations and system. Second, the domestic and foreign market status of each item has been surveyed and analyzed. Third, the contents of technical documents already approved by MFDS have been analyzed to select the correct example, test items, criteria, and methods. Finally, the guideline has been developed based on international and domestic regulation, through close review of a consultative body composed of academic, industrial, research institute and government experts.

Effect of Rice Bran and Barley Bran Application on Growth and Yield of Chinese Chive (Allium tuberosum Rottler) and Taro (Colocasia esculenta) and Weed Control (쌀겨, 보릿겨 처리가 부추와 토란의 생육과 수량 및 잡초방제에 미치는 영향)

  • This study was carried out to examine the effect of rice bran, barley bran, burned rice bran, and burned barley bran on the growth and yield of Chinese chive (Allium tuberosum Rottler), taro (Colocasia esculenta), and weed control. When the above 4 brans were examined 13, 27, 41 and 57 days respectively after application, the plant height of Chinese chive applied with burned barley bran was significantly higher than non-treated control, whereas the other brans did not have any distinct effect on the plant height or population number of Chinese chive. However, when examined 57 days after the application of the above 4 brans, all the plants applied with brans showed more than twice the improvement in shoot fresh weight compared with non-treated control. A chemical analysis of soil 57 days after the application of the above 4 brans showed that the soils were richer in available phosphate and organic matter. Shoot fresh weight of Chinese chive at 2 weeks after cutting was significantly higher in barely bran treated plot than in non-treated plot. In the case of taro, only taro plots transplanted when 10 cm tall and applied with barley bran showed an improvement in growth increment of both the underground and above parts. However, when sowed seeds after the application of the 4 brans, the yield of taro was reduced by the brans. Thus this research indicates that the effect of brans is differ based on the amount of bran application as well as crops. The effect of weed control on Echinochloa crus-galli, Digitaria clliaris, Chenopodium album, and Solanum nigrum as affected by brans was very low in pot conditions. Weed efficacy of the brans was also very low in field conditions. Growth of Chinese cabbage and garland chrysanthemum was inhibited 63% and 37% by rice bran at $4,000kg\;ha^{-1}$, respectively, but other crops such as maize, squash, cucumber, and Chinese chive were inhibited by 0-20%. These results were similar to that of barley bran except for Chinese cabbage.

Evaluation of Feed Values for Imported Hay Using Near Infrared Reflectance Spectroscopy (근적외선분광법을 이용한 수입 건초의 사료가치 평가)

  • Near infrared reflectance spectroscopy (NIRS) has become increasingly used as a rapid and accurate method of evaluating some chemical compositions in forages. The objective of this study was to evaluate the potential of NIRS, applied to imported forage, to estimate the moisture and chemical parameters for imported hays. A population of 392 imported hay representing a wide range in chemical parameters was used in this study. Samples of forage were scanned at 1 nm intervals over the wavelength range 680-2500nm and the optical data was recorded as log 1/Reflectance(log 1/R), which scanned in intact fresh condition. The spectral data were regressed against a range of chemical parameters using partial least squares(PLS) multivariate analysis in conjunction with spectral math treatments to reduced the effect of extraneous noise. The optimum calibrations were selected based on the highest coefficients of determination in cross validation(R2) and the lowest standard error of cross-validation(SECV). The results of this study showed that NIRS predicted the chemical parameters with very high degree of accuracy. The R2 and SECV for imported hay calibration were 0.92(SECV 0.61%) for moisture, 0.98(SECV 0.65%) for acid detergent fiber, 0.97(SECV 0.40%) for neutral detergent fiber, 0.99(SECV 0.06%) for crude protein and 0.97(SECV 3.04%) for relative feed value on a dry matter(%), respectively. Results of this experiment showed the possibility of NIRS method to predict the moisture and chemical composition of imported hay in Korea for routine analysis method to evaluate the feed value.

Studies on the Improvement of the Fishing Efficiency of Purse Seine in the Sea Area of Cheju Island -The Changes of Seine Volume and Tension in the Purseline During Pursing- (제주도 주변해역 선망의 어획성능 향상에 관한 연구 -짐줄 체결 중 선망의 용적과 짐줄의 장력 변화 -)

  • A simple experimental method was used in an attempt to realize the elevation of the fishing ability of purse seine in the sea area of Cheju Island, the changes of seine volume and tension in the purseline during pursing. Experiments carried out on the six types simplified reduced model seines which were made of knotless nettings. The nettings were woven in different leg length 4.3, 5.0, 5.5, 6.0, 6.6 and 7.7mm of polyester 28 tex two threads two-ply twine, and each of the seines were named I, II, III, IV, V and Ⅵ seine. Dimension of seine models were 450cm for corkline and 85cmfor seine depth, each seines rigged up 160g of float for a floatline and 50g (underwater weight) of lead for a leadline. These model purse seines were made of the scale of 1/200 of its full scale, a 120 ton in the near sea of Cheju Island. Designing and testing for the model purse seines were based on the Tauti's law. Experiments were measured in the observation channel of a flume tank at the static conditions set up shooting and pursing equipments. Motion of purse seine during purse line was recorded by the two sets video camera for VTR which were placed in top and front of the model seine. The reading coordinate of seine volume carried out by the video digitization system, disk data for the purseline tension. An analysis were performed on the changes seine volume and tension in the purseline during pursing. The results obtained were as follows: 1. The seine volume during pursing was largest for Ⅵ seine with smallest d/l followed by V, IV, III, II and I seines, and tension in the purseline was small. 2. Seine volume during pursing can be expressed by the following equation; CVt=l-EXP[{2.79 (d/l)+0.35}t-33.37 (d/l) + 0.57] Where CVt is volume ratio, d is twine diameter, l is leg length and t is pursing time (sec). 3. Tension in the purse line during pursing can be expressed by the following equation; T= 1- EXP {0.57t + 13.36 (d/l)+2.97} Where T is tension (kg) in the purseline during pursing.

Evaluation of Antibodies Against Haemophilus influenzae Type b in Korean Adults (우리나라 성인에서 Haemophilus influenzae type b에 대한 항체 평가)

  • Purpose: After the introduction of Haemophilus influenzae type b (Hib) vaccine in 1995 in Korea, it was included in the national immunization program in 2013. In the post-Hib vaccine era, some studies in other countries reported that invasive Hib disease affects adults, especially the elderly and immunocompromised persons, more often than it affects children. To evaluate disease susceptibility, quantitative and qualitative analysis of anti-polyribosylribitol phosphate (PRP) antibodies were carried out in Korean adults aged 20 to 85 years. Methods: Sera were collected from 39 healthy adults (20 to 50 years of age) and from 30 elderly adults (75 to 85 years of age) who did not have immune-compromising conditions. The concentration of anti-PRP immunoglobulin G (IgG) and serum bactericidal indices (SBIs) were measured by enzyme-linked immunosorbent assay and serum bactericidal assay. Results: Geometric mean concentrations of anti-PRP IgG and geometric mean SBIs were $0.88{\mu}g/mL$ (95% confidence interval [CI], 0.17 to 3.85) and 354 (95% CI, 50 to 2,499) in young adults and $1.67{\mu}g/mL$ (95% CI, 0.53 to 5.24) and 449 (95% CI, 146 to 1,376) in elderly adults, respectively. When the threshold of seropositivity for anti-PRP IgG was applied as 0.15 or $1.0{\mu}g/mL$, which is the protective antibody level in children, seropositive rates were 87.2% or 53.8% in young adults and 100% or 60% in elderly adults. The seropositivity rates of the SBI ($SBI{\geq}4$) were 82.1% and 100% in the groups, respectively. Conclusions: Most subjects in the adult and elderly adult groups display immunity to Hib based on quantitative and qualitative antibody levels, but not all. Because high immunization and low Hib circulation rates may reduce the natural Hib immunity in the population, monitoring Hib immunity as well as disease are needed continuously.
