Ecological Characteristics of Tachanovsky's Gudgeon, Ladislabia taczanowskii in Songcheon Stream, Korea (송천에 서식하는 새미(Ladislabia taczanowskii)의 생태 특성)

  • This study surveyed the ecological characteristics of Ladislabia taczanowskii at Songcheon Stream from March to November 2019. The species inhabited upstream, where the riverbed structure was mostly covered with boulders and cobbles. The water depth was 31-148 cm, and the stream velocity was fast at 0.94±0.23 (0.51-1.39) m/sec. The gender ratio of females to males was 1 : 0.89. The age according to the total length frequency distribution indicated that the group with 38-70 mm below in total length was one year old, the group with 70-100 mm was two years old, the group with 100-120 mm was three years old, and the group over 120-128 mm was over four years old. The total length of sexually mature fishes was 70 mm for females and 75 mm or more for males. The spawning season was from May to August, and the water temperature was 15.5-20.1℃ during the period. The prosperous spawning season was from June to July, and the water temperature was 15.8-17.2℃ during the period. The spawning ground was where the riffle began in the pool, and its bottom was formed of sand and gravel. The width was about 150 cm, and the water depth was 20 to 50 cm. The average number of eggs in the ovaries was 821 (401-1,314) per matured female, and the matured eggs were yellowish and spherical with a mean diameter of 1.62±0.02 (1.43-2.01) mm. The live foods of L. taczanowskii Cyanophyta, Chlorophyta, Bacillariophyta, Ephemeroptera, and Diptera. The feeding habit of L. taczanowskii is omnivorous, but more than 90% of the stomach content was attached algae.

Triclosan Resistant Bacteria from Sewage Water: Culture Based Diversity Assessments and Co-Resistance Profiling to Other Antibiotics

  • Triclosan (TCS) is an antimicrobial agent used in various human personal care products against both gram-positive and gram-negative bacteria. The purpose of this study was to evaluate the presence of TCS-resistant bacteria in sewage water in Peshawar, Khyber Pakhtunkhwa (KPK), Pakistan, for the first time. TCS-supplemented Luria Bertani (LB) agar was used to isolate TCS-tolerant bacteria. A total of 17 TCS-resistant isolates were randomly selected from a large pool of bacteria that showed growth on TCS-supplemented LB agar. Based on gram staining and physiochemical characteristics, the isolated strains were identified as Salmonella typhi (n = 6), Escherichia coli (n = 4), Citrobacter freundii (n = 4), Proteus mirabilis (n = 1), Enterobacter cloacae (n = 1), and Pseudomonas aeruginosa (n = 1). The Triclosan mean minimum inhibitory concentrations (MICs) for the isolates of Salmonella typhi, Escherichia coli, Citrobacter freundii, Proteus mirabilis, Enterobacter cloacae, and Pseudomonas aeruginosa were 23.66 ㎍ ml-1, 18.75 ㎍ ml-1, 42 ㎍ ml-1, 32 ㎍ ml-1, 64 ㎍ ml-1, and 128 ㎍ ml-1, respectively. The antibiogram revealed that all isolates were resistant to penicillin G (100%) and linezolid (100%), followed by ampicillin (94%), tetracycline (76%), tazobactam (76%), sulbactam/cefoperazone (64%), polymyxin PB (58%), amikacin (29.41%), aztreonam (29.41%), imipenem (5%), and gentamicin (5%). This is the first known study regarding the isolation of TCS-tolerant bacteria from sewage water in Peshawar, KPK, Pakistan. It was concluded that all the TCS-resistant isolates were multidrug resistant (MDR) gram-negative rod-shaped bacteria, mostly belonging to the Enterobacteriaceae family.

Warm Season Hydro-Meteorological Variability in South Korea Due to SSTA Pattern Changes in the Tropical Pacific Ocean Region (열대 태평양 SSTA 패턴 변화에 따른 우리나라 여름철 수문 변동 분석)

  • In this study, we analyzed the effects of regional hydrologic variability during warm season (June-September) in South Korea due to ENSO (El $Ni{\tilde{n}}o$-Southern Oscillation) pattern changes over the Tropical Pacific Ocean (TPO). We performed composite analysis (CA) and statistical significance test by Student's t-test using observed hydrologic data (such as, precipitation and streamflow) in the 113 sub-watershed areas over the 5-Major River basin, in South Korea. As a result of this study, during the warm-pool (WP) El $Ni{\tilde{n}}o$ year shows a significant increasing tendency than normal years. Particularly, during the cold-tongue (CT) El $Ni{\tilde{n}}o$ decaying years clearly decreasing tendency compared to the normal years was appeared. In addition, the La $Ni{\tilde{n}}a$ years tended to show a slightly increasing tendency and maintain the average year state. In addition, from the result of scatter plot of the percentage anomaly of hydrologic variables during warm season, it is possible to identify the linear increasing tendency. Also the center of the scatter plot shows during the WP El $Ni{\tilde{n}}o$ year (+17.93%, +26.99%), the CT El $Ni{\tilde{n}}a$ year (-8.20%, -15.73%), and the La $Ni{\tilde{n}}a$ year (+8.89%, +15.85%), respectively. This result shows a methodology of the tele-connection based long-range water resources prediction for reducing climate forecasting uncertainty, when occurs the abnormal SSTA (such as, El $Ni{\tilde{n}}o$ and La $Ni{\tilde{n}}a$) phenomenon in the TPO region. Furthermore, it can be a useful data for water managers and end-users to support long-range water-related policy making.

Latitudinal Variation of Nutritional Condition and Diet for Copepod Species, Euchaeta sp. and Pleuromamma spp., from the Northwest Pacific Ocean Using Lipid Biomarkers (지방 바이오마커를 활용한 북서태평양에서 요각류(Euchaeta sp. and Pleuromamma spp.)의 서식 위도별 영양상태 및 먹이원 연구)

  • In order to ascertain latitudinal variation of lipid contents and compositions in copepods, we collected warm water copepod species (Euchaeta sp. and Pleuromamma spp.) from four different regions from low (sub-tropical) to mid (temperate) latitudes in the Northwest Pacific Ocean. Total lipid contents of Pleuromamma spp. were about 11 $ug{\cdot}ind^{-1}$ with little latitudinal variation, whereas Euchaeta sp. showed slightly higher lipid content (20 $ug{\cdot}ind^{-1}$) than Pleuromamma spp. with latitudinal gradient (low at subtropic and high at temperate). Wax esters, known as the major storage lipid classes, were found to be the dominant lipid classes (accounting for more than 35% of total lipids) in Euchaeta sp., whereas in Pleuromamma spp., phospholipids, known as cellular membrane components, were the dominant lipid classes. However, the exception was specimens from warm pool region exhibiting dominance in storage of lipids as a form of triacylglycerols. Among fatty acids, polyunsaturated fatty acids (PUFA), especially docosahexaenoic acid (DHA : 22:6(n-3)) (about 35% of total fatty acids), were most abundant in Euchaeta sp., while saturated fatty acids (SAFA), specially hexadecanoic acid (16:0) (about 30% of total fatty acids), were most abundant in Pleuromamma spp.. Among the neutral fraction of lipids, phytol, originated from the side chain of chlorophyll, was found in all samples which generally indicate active copepods feeding on algae. While only trace amounts of short-chain fatty alcohols were found in Pleuromamma spp., significant amounts of fatty alcohols were found in Euchaeta sp.. Particularly, significant amounts of long chain monounsaturated fatty alcohols (20:1 and 22:1), generally found in cold water species, were found in Euchaeta sp. from low latitudes. The latitudinal variation of trophic lipid markers in these copepods could be significantly related with in-situ food availability and species-specific diet preference. The result of this study suggests that the lipid contents and compositions in copepods may not only indicate their nutritional condition and feeding ecology but also provide insight into species-specific living strategies under different environmental conditions (i.e. water temperature, food availability).

Experimental Studios on the Mechanism of Reproductive Cycle in the Longchin Goby Chasmichthys dolichognathus (HILGENDORF) (점망둑, Chasmichthys dolichognathus의 생식기구에 관한 실험적 연구)

  • The mechanism of reproductive cycle of longchin goby Chasmichthys dolichognathus was investigated for the annual variations of gonadosomatic index(GSI), histological changes of gonad, reproductive cycle, and initiating and terminating factors of the breeding season. The experimental fish were captured in the tide pool of Tongbaeksom, Pusan, Korea from February 1983 to September 1984. Experimental fish for the study of initiating and terminating factors of the breeding season were maintained in filtered recirculating aquariums and exposed to various temperature and photoperiod regimes. The ovary is a pair of sac-shaped organ, and the testicular structure is tubular and it is connected to the seminal vesicle which is located at the posterior end of the testis. GSI began to increase from February when the water temperature began to increase and reached peak in April. It began to decrease from August, the top water temperature season, and maintained relatively low values until January. The annual reproductive cycle includes successive six developmental stages : early growing from December to February, late growing from January to March, early mature from February to May, late mature and ripe from March to August, spawning from April to July, degeneration and resting stage from May to December. An experimental study based on the reproductive cycle of this species indicated that rising temperature under long photoperiod during spring stimulated gonadal maturation, whereas terminating factor of the breeding season with recrudescence of the gonads was demonstrated as high water temperature during summer regardless of photoperiod.

Effects of Bacterial Inoculants and Organic Acids on Silage Quality : Meta-analysis (미생물제제 및 유기산제제의 처리가 사일리지 품질에 미치는 영향 : 메타분석)

  • This study was conducted to estimate and compare the effects of bacterial inoculants and organic acids on silage quality. Silage pH, lactate, acetate, lactate:acetate ratio, propionate, butyrate, water-soluble carbohydrate, crude protein, ammonia-N, neutral detergent fiber and acid detergent fiber (ADF) were used as parameters for quality analysis and a meta-analysis technique was employed to determine the effect size. As a data pool for analysis, we examined 14 research papers. Bacterial inoculants were found to elevate pH, lactate, acetate, lactate:acetate ratio, propionate and ADF contents compared to the controls (p<0.01). In contrast bacterial inoclulants decreased butyrate, water-soluble carbohydrate, crude protein and ammonia-N contents (p<0.01). In the organic acid treatments, all parameters except ADF showed higher contents than the control (p<0.01). In the comparison of effect sizes between the two treatments, significant differences were detected in butyrate, water-soluble carbohydrate, crude protein and ammonia-N (p<0.05). It may be concluded that bacterial inoculants could improve silage quality in terms of the aforementioned four parameters compared with organic acid treatments.

Chemical Effects on Head Loss across Containment Sump Strainer under Post-LOCA Environment (LOCA이후 환경에서 원자로건물집수조 여과기의 수두손실에 대한 화학적 영향)

  • A test apparatus has been fabricated to simulate chemical effect on head loss through a strainer in a pressurized water reactor (PWR) containment water pool after a loss of coolant accident (LOCA). Tests were conducted under condition of same ratio of strainer surface area to water volume between the test appratus and the containment sump. A series of tests have been performed to investigate the effects of spray, existence of calcium-silicate with tri-sodium phosphate (TSP), and composition of materials. The results showed that head loss across the chemical bed with even a small amount of calcium-silicate insulation instantaneously increased as soon as TSP was added to the test solution. Also, the head loss across the test screen is strongly affected by spray duration and is increased rapidly at the early stage, because of high dissolution and precipitation of aluminum and zinc. After passivation of aluminum and zinc by corrosion, the head loss increase is much slowed down and is mainly induced by materials such as calcium, silicon, and magnesium leached from NUKONTM and concrete. Furthermore, it is newly found that the spay buffer agent, tri-sodium phosphate, to form protective coating on the aluminum surface and reduce aluminum leaching is not effective for a large amount of aluminum and a long spray.

Experiment and Analysis of Backscattering Signals According to Presence or Absence of Chloroform (클로로폼 침적 유무에 따른 후방산란신호 측정 실험 및 분석)

  • Because it is difficult to apply direct and optical detection techniques to sunken hazardous and noxious substances (HNS), effective acoustic detection techniques are required to detect sunken HNS in water. In this study, the possibility of acoustic detection of sunken HNS was investigated through backscattering signal measurement experiments using chloroform, a sunken HNS. After establishing a pool in an acrylic tank, backscattering signals were measured according to the presences or absence of chloroform by varying the grazing angle from 90° to 50° in 0.5° intervals using a pan&tilt system. A directional transducer transmitted and received sinusoidal signals with a frequency of 200 kHz and a pulse length of 25 ㎲ in a monostatic state. When chloroform was deposited, the received level of the backscattering signal at the interface between water and chloroform became low at a grazing angle of approximately 80° or smaller. Based on the backscattering signal results obtained at the interface between water and chloroform, the possibility of acoustic detection of sunken HNS was demonstrated.

Evaluation of Flooding Characteristics of Dam Reservoir using Cumulative Flooding Area Curve (누가침수면적곡선을 이용한 댐 저수구역의 침수특성 평가)

  • Floodplain in a reservoir is defined as the area naturally formed between the design flood level and the normal pool level. Located around the dam reservoir, floodplain has been damaged in many different ways including cultivation. As it is impossible to restore all the damaged floodplain at once, it is necessary to determine their order of priority. This process considers various factors, among which the flooding frequency is an important hydrologic characteristic, Different from the floodplains in a river, all the floodplains around the given dam reservoir have the same flooding frequency. To overcome this problem, this study proposes to use the cumulative flooding area curve, which represents the cumulative flooding area corresponding to the reservoir water level. Especially, this study evaluates the flooding frequency of those water levels corresponding to the cumulative flooding area of 30%, 50% and 70%. As application examples, this study considers the five restoration candidates each selected in the Andong Dam, Imha Dam, Youngju Dam and Nam river Dam of the Nakdong River Basin. As a result, the cumulative flooding area curve was found to well represent the overall shape of the floodplain (i.e., steep-to-mild slope or mild-to-steep slope). Also, the flooding frequency of those water levels corresponding to the cumulative flooding area of 30%, 50% and 70% was found to be so effective to quantify the hydrologic characteristics of a floodplain.

Pigment production in Monascus anka (Monascus anka의 균주선발 및 색소생성 조건)

  • Monascus anka strains with higher pigment production were developed using UV mutation and natural selection. To obtain organic solvent soluble pigments from Monascus anka, the following culture conditions were compared : standing and shaking culture with Nishikawa's medium, and shaking culture with Lin's medium. Shaking culture in Lin's medium exhibited decrease in solvent-soluble intracellular pigments after initial increase. The decrease was accompanied by the increase in water-soluble extracellular pigments. Monascus anka preferred sucrose and ethanol among 7 carbon sources tested. Treatment of sterol biosynthesis inhibitors, $({\pm})$-miconazole and chlorocholine chloride(CCC) , directed carbon pool to the biosynthetic pathway leading to the pigments with CCC's more pronounced effect. Two dimensional TLC revealed at least 7 yellow pigments suggesting existence of hereto unreported pigment. One of the most abundant yellow pigments was isolated and found to be ankaflavin by NMR and MS analysis.
