Anti-lipid Peroxidative Effect of Extracts and its Fractions Polygonatum odoratum (둥굴레 추출물 및 분획의 과산화지질 생성 저해효과)

  • The rhizoma of Polygonatum odoratum (Liliaceae) promotes the production of body fluid and relives dryness symptoms and has hypoglycemic effect. In order to evaluate the prevention of lipid peroxidative efficacy of P. odoratum, its extracts (Et$_2$O and MeOH ex.) and its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH and 100% MeOH fr.) were measured by TBARS assay on rat liver S9 and human erythrocyte ghost membrane. The order of anti-lipid peroxidative effect was as follows: Et$_2$O ex. >40% MeOH fr. >60% MeOH fr.>100% MeOH fr. Based on these results, we conclude that membrane lipid peroxidation is inhibited in vitro by addition of P. odoratum ether extract and its gradient MeOH fractions excepts $H_2O$ fr., and which have significant hepatoprotective activity in $CCl_4$-treated rats.

  • The rhizoma of Polygonatum odoratum (Liliaceae) has been used as the treatment of body fluid deficiency, dryness symptoms and hyper glycemia. To study effects on acute renal failure of P. odorati rhizoma, urinary volume, urinary electrolytes and serum factors associated with renal dysfunction were measured in $HgCl_2-induced$ acute renal failure rats. It was revealed that its ether ex. had significant diuretic effects, regulated hypoelectrolytes$(Na^+,\;K^+,\;Cl^-)$ and inhibited the increase of creatinine, BUN in acute renal failure rats. From the ether ex. which was more effective than MeOH ex., three lipophilic compounds were isolated and elucidated hydrocarbon(l), ${\beta}-sitosteryl$ stearate(2), ${\beta}-sitosterol$(3). The ${\beta}-sitosteryl$ stearate(2) were isolated from Polygonatum sp. at first.

Effect of Steaming Prior to Roasting of Polygonatum odoratum Roots on Its Water Solubles and Browning (볶음전 증자처리가 둥굴레 근경의 수용성 성분 및 갈색화에 미치는 영향)

  • This work was designed to determine the effect of steaming-Pretreatment on Physicochemical and functional properties of roasted Polygonatum odoratum roots. Steaming treatments led to some reduction in the contents of water solubles, such as total solid, reducing sugar and free-amino nitrogen of the unroasted samples. Moreover, roasting processing caused decrease in reducing sugar and free-amino acid, with increase in total soluble solid and browning color of the samples, which showed the dependence of steaming treatment as well as roasting temperature. Electron-donating ability and nitrite-scavenging ratio of the samples were also highly developed along with roasting processing at 17$0^{\circ}C$ within 35min. Considering the physicochemical and some functional parameters of water extracts of roasted samples, roasting at above 14$0^{\circ}C$ for over 55min or at 17$0^{\circ}C$ for around 35min was recommendable for the higher quality of Polygonatum odoritum tea, which conditions were similarly adapted for both steamed and unsteamed samples.

Optimization of Preparation Conditions of Houttuynia cordata Beverage Containing Saururus chinensis and Polygonatum odoratum by Sensory Evaluation and Response Surface Methodology (관능 평가 및 반응 표면 분석에 의한 삼백초와 둥굴레 가미 어성초 음료의 제조 조건 최적화)

  • The optimization of the addition ratios of Houttuynia cordata, Saururus chinensis, and Polygonatum odoratum var. pluriflorum hot water extract to Houttuynia cordata beverage was evaluated by applying response surface methodology. The quality characteristics of a selected best product were investigated. Sweet, savory, and fishy taste, and overall acceptability were optimized by response surface methodology using a central composite design. The concentration of effect of Houttuynia cordata, Saururus chinensis, and Polygonatum odoratum var. pluriflorum hot water extract was modeled using an equation. Hot water extracts of Houttuynia cordata ($1.75{\sim}1.89%$), Saururus chinensis ($1.03{\sim}1.04%$), and Polygonatum odoratum var. pluriflorum ($2.17{\sim}2.31%$) represented the optimal concentration conditions to obtain the best taste. The physical and chemical characteristics of the beverage manufactured at optimized conditions were pH 4.18, soluble solids 0.41 $^{\circ}Brix$, total acidity 0.21%, color value ($L^*$ value 40.08, $a^*$ value 4.53, and $b^*$ value 10.69), total polyphenol 54.63 mg/L, and electron donating ability 26.98%.

Attenuation of insulin resistance using steamed Polygonatum odoratum var pluriflorum extract in rat skeletal muscle cells L6 myoblast (렛 근육세포 L6에서 둥굴레 추출물의 인슐린저항성 개선)

  • Objectives : The purpose of this study was to investigate inhibitory effects of steamed Polygonatum odoratum extract (POE) on insulin resistance in rat skeletal muscle cells, L6 cells.Methods : Polygonatum odoratum (P. odoratum) extract was extracted with ethyl acetate. Activity of α-glucosidase in POE was measured for blood glucose regulation. MTT assay was examined for cell toxicity. Western blot analysis for measurement of adiponectine, peroxisome proliferator-activated receptorγ (PPARγ), insulin receptor substrate (IRS), glucose transporter 4 (Glut-4) and phosphorylation of serine/threonine-specific protein kinase (Akt) expressions were performed. Akt signaling pathway were analyzed with LY294002, which is a specific PI3K/Akt inhibitor.Results : The results revealed that POE inhibited α-glucosidase activity. Treatment of POE in L6 cells inhibited the differentiation of L6 cells compared to those of vehicl control. Additionally, protein expressions of adiponectine, PPARγ, IRS and Glut-4 were significantly regulated compared to those of vehicle control (p < 0.05), respectively. Futhermore, phosphorylation of Akt was increased in L6 cells treated with POE compared to that of vehicle control (p < 0.05). pAkt expression was significantly accentuated with Akt inhibitor (LY294002).Conclusions : These results suggest that POE may have potential as a natural agent for prevention/improvement of diabetes, especially, regulation of blood glucose. Therefore, further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

Phylogeny of the Polygonatum odoratum Complex Inferred from Multiple cpDNA and Nuclear RNApol2_i23 Sequence Data (Ruscaceae) (엽록체 DNA 및 핵 DNA RNApol2_i23에 근거한 둥굴레복합체 (Ruscaceae)의 계통 연구)

  • The sequence data of the plastid DNA (trnL-F IGS, trnL intron, and trnH-psbA) and nuclear DNA (RNApol2_i23) markers were utilized to study phylogenetic relationships among the taxa in the Polygonatum odoratum complex (Ruscaceae). European P. odoratum individuals form a clade with a high bootstrap value, which is a sister to the clade of Korean P. odoratum var. odoratum, P. odoratum var. pluriflorum and P. robustum. The formation of the clade with P. odoratum var. odoratum, P. robustum, and one accession of P. odoratum var. pluriflorum indicates geological speciation in isolated populations in the islands following dispersal events from the mainlands. All data sets form two major clades, which are congruent with the subgroups divided by the basic chromosome numbers (x = 9 and x = 10). Although it is not easy to test the hypothesis of the decrease in the basic chromosome number due to scatter taxon sampling in this study, the molecular data strongly suggested that aneuploidy plays an important role in lineage diversification in the genus Polygonatum. The cytological data was not strongly supported by the cpDNA sequences. Further investigations of the cytological, morphological, and geographical characteristics with comprehensive sampling are desired to understand the evolution and lineage diversification in the genus.

  • The phylogenetic relationship of Polygonatum species were examined by RAPD analysis. Polygonatum Rhizoma is called 'Whang-jung', and used Polygonatum sibiricum Redoute in defined as a source plant in Korean Pharmacopoeia. Polygonati Odorati Rhizoma is called 'Wui-yu', and P. odoratum Druce var. pluriflorum Ohiwi and related species are defined is source plants in Korean Herbal Pharmacopoeia. In UPGMA analysis, Polygonum was divided into two major groups. Polygonatum sibiricum and P. stenophyllum was placed in a cluster. On the other hand, P. falcatum A. Gray was included in the other cluster comprising. P. odoratum and related species, which are used as source plants for Polygonati Odorati Rhizome.

Hypoglycemic Effect of Polygonatum Odoratum var. Pluriflorum Ohwi Extract in Streptozotocin-Induced Diabetic Rats (둥굴레(Polygonatum Odoratum var. Pluriflorum Ohwi)추출물의 당뇨 유발 흰쥐에 대한 혈당강화 효과)

  • The hypoglycemic effect of Polygonatum odoratum var. Pluriflorum Ohwi was investigated after extracted with methanol. The methanol extract was fractionated into 4 layers ; hexane, chloroform, butanol and aqueous. Fifty male Sprague-Dawley rats(200-300g) were induced diabetes mellitus by the streptozotocin injection(45mg/kg B.W) into the tail vein and were divided into 5 groups ; diabetic control and 4 experimental groups. All groups of the rats were fed on a AIN-76 diet, and the 4 experimental groups were orally administered with each fraction(500mg/kg B.W) for 12 days and the diabetic control group was orally administered CMC. The body weight gain was monitered and the blood levels of glucose and cholesterol were measured. Levels of protein, triglyceride, and free fatty acid in plasma were analysed. The contents of protein, triglyceride and glycogen in liver and of protein and glycogen in muscle were also determined. The body weight gain was shown significantly higher in the H2O fraction group than that in the diabetic control group and the heart weight was greater in the CHCl3 group. The extents of blood glucose decrement were greater in the BuOH and H2O groups than that in the control group. The urinary glucose excretion was shown relatively small amount in the BuOH and H2O groups. The plasma cholesterol and protein levels were not influenced by these four fractions in diabetic rats. The liver glycogen level was significantly higher in the BuOH group. The results suggest that the orally administered BuOH and H2O fraction of Polygonatum odoratum var. Pluriflorum Ohwi exhibited hypoglycemic effect in streptozotocin induced diabetic rats.

Sclerotium rolfsii Causing Collar Rot on Doonggulle (Polygonatum odoratum var. pluriflorum) (둥굴레에 흰비단병을 일으키는 Sclerotium rolfsii에 관하여)

  • A destructive collar rot of Doonggulle (Polygonatum odoratum var. pluriflorum) occurred in an experimental farm at Hamyang Medicinal Plant Experiment Station, Kyonguam ARES in 1999. The disease also observed in farmer's fileds in Hamyang and the infection rate reached up to 12.3%. Upper parts of the infected plants were mostly blighted and white mycelial mats were formed on the lesions or soil lines near the stem. The fungus readily formed sclerotia on PDA and then shapes were globoid or irregular, and sized $0.9{\sim}6.8{\times}0.6{\sim}5.2\;mm$ (av. $2.7{\sim}2.3\;mm$). Although the fungus showed wide range of mycelial growth temperature, the optimum temperature was about $30^{\circ}C$. The typical sclerotium type and clamp connections of hyphae were found in the fungus grown in PDA. The causal fungus was identified as Sclerotium rolfsii on the basis of mycological characteristics and pathogenicity that on the host plants. This is the first report on the collar rot of Polygonatum odoratum caused by Sclerotium rolfsii in Korea.

Optimization of Roasting Conditions for High-Quality Polygonatum odoratum Tea (둥굴레차의 고품질화를 위한 볶음조건의 최적화)

  • Response surface methodology was applied to determine the optimum roasting conditions (roasting temperature and time) for the high-quality Polygonatum odoratum tea which has been roasting with a traditional means. As quality criteria of Polygonatum odoratum tea, water-soluble solids, browning color, total phenolic compounds and electron-donating ability were proportionally increased with increased temperature and time of roasting conditions up to around $145^{\circ}C$ and 55 min, respectively, while they were decreased under the extended-roasting conditions. The optimum roasting temperature and time based on the organoleptic overall acceptability were $146^{\circ}C$ and 52 min, respectively. On the basis of superimposed contour maps for the tea characteristics, the optimum range of roasting conditions were $135{\sim}140^{\circ}C$ and $58{\sim}64$ min. Predicted values at the optimum conditions $(137^{\circ}C,\;60\;min)$ were in good agreement with experimental values.
