  • The phylogenetic relationship of Polygonatum species were examined by RAPD analysis. Polygonatum Rhizoma is called 'Whang-jung', and used Polygonatum sibiricum Redoute in defined as a source plant in Korean Pharmacopoeia. Polygonati Odorati Rhizoma is called 'Wui-yu', and P. odoratum Druce var. pluriflorum Ohiwi and related species are defined is source plants in Korean Herbal Pharmacopoeia. In UPGMA analysis, Polygonum was divided into two major groups. Polygonatum sibiricum and P. stenophyllum was placed in a cluster. On the other hand, P. falcatum A. Gray was included in the other cluster comprising. P. odoratum and related species, which are used as source plants for Polygonati Odorati Rhizome.

  • This work was designed to determine the optimum roasting conditions with application of a pressure roaster for improving the roasting process of the Polygonatum sibiricum tea which has been roasting with a traditional method. Soluble solid content of the Polygonatum cibiricum tea was 63.16% in the tea roasted at 0.85kgf/$\textrm{cm}^2$(roasting pressure) and 13.9min(roasting time) which were maximum point by the ridge analysis. The optimum conditions predicted for each corresponding sensory properties of the Polygonatum cibirium tea were 0.87kgf/$\textrm{cm}^2$ and 9.2min in color, 0.89kgf/$\textrm{cm}^2$ and 9.7min in sweet 0.72kgf/$\textrm{cm}^2$ and 5.4min in burnt flavor, 0.89kgf/$\textrm{cm}^2$ and 9.9min in appetizing flavor, 0.87kgf/$\textrm{cm}^2$ and 9.3min in aftertaste, 0.89kgf/$\textrm{cm}^2$ and 10.0min in aroma, and 0.88kgf/$\textrm{cm}^2$ and 9.6min in overall acceptability of the tea. The optimum conditions based on the soluble solid content and sensory properties of the Polygonatum cibiricum tea, were 0.83~0.88kgf/$\textrm{cm}^2$ and 9.7~10.4min.

  • Response surface methodology(RSM) was applied in roasting processes of Polygonatum odoratum roots in order to monitor Hunter color parameters and to optimize organoleptic properties of the tea. In roasting processing based on the central composite design with variations in roasting temperature (110~19$0^{\circ}C$) and roasting time(15~75min), coefficients of determinations($R^{2}$) of the models were above 0.9827(p<0.01) in Hunter color parameters and above 0.7748 in organoleptic properties. Hunter color L and $\Delta$E values of the powdered samples decreased with the roasting processes, whereas Hunter color a and b values increased up to around 59 min at 136$^{\circ}C$ and thereafter showed a decreased tendency. Organoleptic qualities on color, taste and aroma of Polygonatum odoratum tea were improved along with roasting processes, thereby estimating the optimum roasting conditions by RSM for the maximized qualities of organoleptic properties as a range from 127$^{\circ}C$ to 135$^{\circ}C$ and from 60min to 66 min. Hunter color a value(redness) was found to generate a similar response surface to that of organoleptic properties, showing the possibility of quality index in roasting processes of Polygonatum odoratum roots.

  • Micropropagation and adventitious root production via the culture of Polygonatum odoratum were performed. Stem segments of seedlings of Polygonatum odoratum were the most efficient explants for adventitious shoot formation compared to leaf and root segments. Exogenous cytokinin treatment was required for adventitious shoot formation. Among the cytokinin (BA, Kinetin and Zeatin) tested, BA was most effective for shoot formation from stem segments. Auxin (NAA or IBA) in combination with cytokinin significantly enhanced adventitious shoot formation. Twenty five percent of explants produced adventitious shoots on medium with 2.0 mg/L BAP alone, while 83% of explants produced adventitious shoots on medium with the combination of 2.0 mg/L BAP and 0.1 mg/L IBA. Rooting of adventitious shoots was achieved after transferring to 112 MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L zeatin. When stem segments were cultured on MS medium with various kinds of auxin (IBA, NAA and 2,4-D), adventitious roots were formed from callus. frequency of adventitious root formation was highest in 2,4-D than NAA and IBA. When roots were in clusters together with parental stem segments, growth of roots actively occurred in hormone-free MS liquid medium. The above results represent that possible application for the mass production of roots and plantlets through in vitro culture system of Polygonatum odoratum.

  • Biologically active compounds in Polygonatum sibiricum were extracted using organic solvents as hexane, CHC1$_3$, n-butanol corresponding each component. Compound II was purified from hexane layer and the chemical structure of compound II was characterized using IH-nmr, 13C-nmr, DEPT135, COSY, HMQC, HMBC spectrum and MS-spectrum. Consequently, the chemical structure of compound II was determined as 2-Hydroxy-3-(9,12-(9E,12E)-Octadecadienoyloxy) propanoic acid.

  • The rhizoma of Polygonatum odoratum (Liliaceae) has been used as the treatment of body fluid deficiency, dryness symptoms and hyper glycemia. To study effects on acute renal failure of P. odorati rhizoma, urinary volume, urinary electrolytes and serum factors associated with renal dysfunction were measured in $HgCl_2-induced$ acute renal failure rats. It was revealed that its ether ex. had significant diuretic effects, regulated hypoelectrolytes$(Na^+,\;K^+,\;Cl^-)$ and inhibited the increase of creatinine, BUN in acute renal failure rats. From the ether ex. which was more effective than MeOH ex., three lipophilic compounds were isolated and elucidated hydrocarbon(l), ${\beta}-sitosteryl$ stearate(2), ${\beta}-sitosterol$(3). The ${\beta}-sitosteryl$ stearate(2) were isolated from Polygonatum sp. at first.

  • Polygonatum taxa of Korea were classified as two sections, three series and 14 species and 2 varieties and reviewed taxonomically. Among the examined taxa, endemics in Korea were recorded as two species; P. grandicaule Y. S. Kim, B. U. Oh & C. G. Jang and P. infundiflorum Y. S. Kim, B. U. Oh & C. G. Jang. The habitat of Korean unrecorded species, P. acuminatifolium Kom. and P. odoratum var. odoratum were confirmed in this study. Keys to the sections, series and species including description of each taxon were provided here.

  • This work was designed to determine the effect of steaming-Pretreatment on Physicochemical and functional properties of roasted Polygonatum odoratum roots. Steaming treatments led to some reduction in the contents of water solubles, such as total solid, reducing sugar and free-amino nitrogen of the unroasted samples. Moreover, roasting processing caused decrease in reducing sugar and free-amino acid, with increase in total soluble solid and browning color of the samples, which showed the dependence of steaming treatment as well as roasting temperature. Electron-donating ability and nitrite-scavenging ratio of the samples were also highly developed along with roasting processing at 17$0^{\circ}C$ within 35min. Considering the physicochemical and some functional parameters of water extracts of roasted samples, roasting at above 14$0^{\circ}C$ for over 55min or at 17$0^{\circ}C$ for around 35min was recommendable for the higher quality of Polygonatum odoritum tea, which conditions were similarly adapted for both steamed and unsteamed samples.
