A Potent Medicinal Plant: Polygala Tenuifolia

  • Polygala Tenuifolia, also described as Yuan Zhi, is a conventional botanic plant found in Korea and China. It's most well- known promise is to improve cognition and guard against mental disorders, cure sputum, anxiety, and sleeplessness, and keep the central nervous system health. The pharmacological aspects of Polygala Tenuifolia's genesis and component compounds reveal the neuroprotective potential in connection to Alzheimer's disease. It contains three herbs: Bokshin, Sukchangpo, and Wongi. P. Tenuifolia's primary ingredients are Xanthone glycosides, Triterpenoid saponins, and Oligosaccharides. Polygalasaponins and Etrahydrocolumbamine are the major components, and they've been widely used for more than a century to relieve mood and psychological illnesses, particularly in North Asian countries such as Korea, China, Japan, and Taiwan. P. Tenuifolia extract eliminates allergic illnesses such as eczema and contact dermatitis by modulating Protein kinase-A and Mitogen-protein kinase-38. In vitro and in vivo studies linking P. tenuifolia root ingredients to a variety of pharmacological effects pertinent to AD show that this species' isolates may function through polyvalency. In great health, people can take up to 250-300 mg per day. It was given in peer-reviewed studies at dosages of 100-150 mg many times each day. There is minimal evidence that it improves verbal memory in experimental animals.

  • Five compounds were isolated from the roots of Polygala tenuifolia (Polygalaceae). On the basis of spectroscopic evidences, the structures of these compounds were characterized as ${\alpha}-D-(6-O-sinapoyl)-glucopyranosyl(1{\rightarrow}2')-{\beta}-D-(3'-O-sinapoyl)-fructofuranoside$ (P3), ${\alpha}$-D-{6-O-(p-methoxybenzoyl)}-glucopyranosyl-$(1{\rightarrow}2')$-${\beta}$-D-{3'-O-(3',4',5'-trimethoxycinnamoyl)}-fructofuranoside(P4), ${\alpha}$-D-{6-O-(p-hydroxybenzoyl)}-glucopyranosyl-$(1{\rightarrow}2')$-${\beta}$-D-{3'-O-(3',4',5'-trimethoxycinnamoyl)}-fructofuranoside(P5), ${\alpha}-D-glucopyranosyl-(1{\rightarrow}2')-{\beta}-D-(1'-O-sinapoyl)-fructofuranoside$(P6), $1,5-anhydro-D-glucitol$(P7) respectively. ${\alpha}$-D-{6-O-(p-Methoxybenzoyl)}-glucopyranosyl-$(1{\rightarrow}2')$-${\beta}$-D-{3'-O-(3',4',5'-trimethoxycinnamoyl)}-fructofuranoside(P4) and ${\alpha}-D-glucopyranosyl-(1{\rightarrow}2')-{\beta}-D-(1'-O-sinapoyl)-fructofuranoside$(P6) were isolated for the first time from the genus of Polygala. 1,5-Anhydro-D-glucitol(P7) was isolated without hydrolysis for the first time from the root of Polygala tenuifolia.

Protective effects of Polygala tenuifolia on ischemia-induced 4 vessel occlusion in rats

  • The root of Polygala tenuifolia Willd (PT) is known to have neuroprotective effects and as an antidementic herb in Chinese and Japanese traditional medicine. We examined potential neuroprotective effects of PT using the 4-vessel occlusion model in rats. In this study, the efficacy of PT for the prevention of neuronal damage and for the reduction of memory impairment was investigated. The results indicate that PT confers significant neuroprotection especially for ischemic hippocampal neurons.

Anxiolytic-like Effects of Polygala tenuifolia Willdenow Using the Elevated Plus Maze and Hole-board Apparatus in Mice

  • The purpose of this study was to characterize the putative anxiolytic-like effects of the aqueous extract of the root of Polygala tenuifolia ( AEPT) using an elevated plus maze (EPM) and hole-board apparatus in mice. The AFPT was orally administered at 50, 100, 200 or 400 mg/kg to ICR mice, 1 h before the behavioral evaluation in the EPM respectively. Control mice were treated with an equal volume of saline, and positive control mice with buspirone (2 mg/kg). Single treatments of the AEPT significantly increased the percentage of time spent and arm entries into the open arms of the EPM vedrsus saline controls (P<0.05). Moreover, there were no changes in the locomotor activity and myorelaxant effects in any group compared with the saline controls. In the hole-board test,single treatments of the AEPT (200 and 400 mg/kg) significantly increased the number of headdips versus saline controls (P<0.05). In addition, the anxiolytic-like effects of the AEPT were blocked by WAY 100635(0.3mg/kg, I.p), a5-$HT_{1A}$ receptor antagonist not by flumazenil, a $GABA_{A}$ antagonist. These results indicate that P. tenuifolia is an effective anxiolytic agent, andsuggest that the anxiolytic-like effects of P. tenuifolia is mediated via the serotonergic nervous system.

  • We investigated whether an aqueous extract of Polygala tenuifolia root (PTAE) inhibits secretion of inflammatory cytokines from primary cultures of mouse astrocytes. PTAE dose-dependently inhibited the Tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ secretion by astrocytes stimulated with substance P (SP) and lipopolysaccharide (LPS). Interleukin-1 (IL-1) has been shown to elevate $TNF-{\alpha}$ secretion from LPS-stimulated astrocytes while having no effect on astrocytes in the absence of LPS. We therefore also investigated whether IL-1 mediated inhibition of $TNF-{\alpha}$ secretion from primary astrocytes by PTAE. Treatment of PTAE to astrocytes stimulated with both LPS and SP decreased IL-1 secretion to the level observed with LPS alone. Moreover, incubation of astrocytes with IL-1 antibody abolished the synergistic cooperative effect of LPS and SP. Reverse transcriptase-polymerase chain reaction analysis demonstrated the significantly reduced level of the $TNF-{\alpha}$ mRNA was expressed in astrocytes treated with PTAE. These results suggest that PTAE has an antiinflammatory activity on the central nervous system curing some pathological disease states.

원지(遠志) Saponin의 이뇨효과(利尿效果) 및 중추억제작용(中樞抑制作用)에 관한 연구(硏究) (Effects of the Saponin of Polygalae Radix on the Renal Function and CNS-Depression)

  • Polygalae Radix was used as diuretics, analgesics and expertorants in oriental medicine. The root of Polygala tenuifolia Willd. (Polygalaceae) is comprised saponin (Onjisaponin A,B,C,D,E,F and G) polygalitol, onsitin and sugars. The pharmacological action of crude Polygala-saponin (PS) obtained from the roots are studied. The following results were obtained; 1) The median lethal dose $(LD_{50})$ of PS in mice is presented 71.1mg/kg s.c. and 694. 5mg/kg p.o.. 2) PS demonstrated diuretic action of relatively long acting duration in mice. 3) The diuretic mechanism of PS was found due to inhibitory effect of renal tubular reabsorption of electrolytes and glomerular vascular dilatation. 4) The group, administered simultaneously PS and cefadroxil monobydrate was significantly increased with PS alone group on diuretic action. Synergistic effect cefadroxil monohydrate on the diuretic action of PS seems due to competitive inhibition of plasma protein binding with PS. 5) PS demonstrated analgesic action by the acetic acid stimulating method and Randall-Selitto test in mice. 6) PS presented antipyretic action against febrile treated with the typhoid vaccine. 7) PS was significantly prolonged against the hypnotic duration of pentobarbital in mice. 8) Onset time convulsion and death induced by picrotoxin and strychnine in mice were not delayed. According to the above results, the PS was identified as a pharmacological active component obtained from roots of Polygala tenuifolia Willd.

알루미늄호일 이용 배추좀나방(Plutella xylostella L.) 살란활성 검정법 개발 및 살란활성 식물추출물의 선발 (Development of a Bioassay Method Using Aluminium Foil Sheet for Screening Ovicidal Activity Against Diamondback Moth Egg, Plutella xylostella L. and Selection of Plant Extracts with High Ovicidal Activity)

  • 배추좀나방(Plutella xylostella L.)은 세계적으로 배추과 작물의 가장 문제 해충이다. 지금까지 배추좀나방 방제는 유충을 대상으로 이루어졌으나, 더욱 효과적인 방제를 위해서 알의 부화를 억제하거나 살란활성을 가진 새로운 살충제 개발이 요구된다. 이에 알루미늄 호일을 활용하여 배추좀나방의 살란활성을 효율적으로 생물검정하는 방법을 개발하였다. 알루미늄호일($4{\times}12$ cm, 산란호일)에 배추잎 즙액을 도포하여 아크릴원통에 넣고 갓 우화한 성충 300마리를 24시간 접종한 결과 알을 충분하게 확보할 수 있었으며, 이는 4일간 연속으로 채란 가능하였다. 살란활성 검정은 난괴 크기별로 산란호일을 6~12조각으로 나눈 농도별로 준비된 메탄올 용액에 2~10초간 핀셋으로 침지한 후 음건(15~30분)하여 소형사육통에 넣어 부화율을 조사하였다. 산란호일은 50% 메탄올에 10초간 침지하여도 활성검정에 어떠한 영향도 미치지 않았다. 하지만 소형 사육통의 뚜껑을 개폐하면 활성물질의 성질에 따라 영향을 받았다 이러한 방법으로 식물체 50종의 메탄올 추출물에 대한 배추좀나방 살란활성을 검정한 결과, 고본(Angelica tenuissima) 뿌리, 원지(Polygala tenuifolia) 뿌리, 지골피(Lycium chinense) 뿌리, 천궁(Cnidium officinale) 뿌리 등은 90% 이상의 높은 살란활성을 나타내었다.

Protective Effects of Cinnamic Acid Derivatives on Gastric Lesion

  • P-methoxycinnamic acid and 3,4,5-trimethoxycinnamic acid are the compounds found in Polygalae Radix, the root of Polygala tenuifolia Willdenow, and have been reported to have hepatoprotective and anti-neurodegenerative effects. On the other hand, there are no reports of their effects on gastric lesions. This study examined the inhibitory effects of cinnamic acids, including p-methoxycinnamic acid, 3,4,5-trimethoxycinnamic acid, and 8 compounds (cinnamic acid, 2-(trifluoromethyl) cinnamic acid, 3-(trifluoromethyl) cinnamic acid, trans-4-(trifluoromethyl) cinnamic acid, 4-(dimethylamino) cinnamic acid, 3,4-(methylenedioxy) cinnamic acid and 3,4-dihydroxycinnamic acid), which were selected based on their presence in medicinal herbs and molecular weight, against gastric lesions. Animal models were used to confirm the protective effects on acute gastritis caused by the administration of HCl/EtOH. Gastric acid inhibition was examined by an acid-neutralizing test and the proton pump ($H^+/K^+$-ATPase) inhibiting activity. In addition, antioxidant tests were performed and the gastric emptying rate was determined. The results showed that cinnamic acid, p-methoxycinnamic acid, and 3,4,5-trimethoxycinnamic acid had an inhibitory effect on gastric lesions.

원지(遠志)가 만성적 뇌혈류저하 흰쥐의 β-Amyloid 축적과 기억장애에 미치는 영향 (Effects of Polygalae Radix on β-Amyloid Accumulation and Memory Impairment Induced by Chronic Cerebral Hypoperfusion in Rats)

  • Objectives : This study was investigated the effects of the root of Polygala tenuifolia (POL) on learning and memory impairment induced by chronic cerebral hypoperfusion in rats. Methods : Chronic cerebral hypoperfusion was produced by permanent bilateral common carotid artery occlusion (pBCAO). POL was administered orally once a day (130 mg/kg of water-extract) for 28 days starting at 4 weeks after the pBCAO. The acquisition of learning and the retention of memory were tested on 9th week after the pBCAO using the Morris water maze. In addition, effects of POL on $A{\beta}$ generation and expressions of APP and BACE1 were observed in the hippocampus of rats. Results : POL significantly prolonged the swimming time spent in target quadrant and significantly reduced the swimming time spent in the quadrant far from the target. POL significantly increased the percentage of swim in the targer quadrant in the retention test, while POL was not effective on the escape latencies in the acquisition training trials. POL significantly reduced the levels of $A{\beta}_{(1-40)}$ and $A{\beta}_{(1-42)}$ in the cerebral cortex and the level of $A{\beta}_{(1-42)}$ in the hippocampus produced by chronic cerebral hypoperfusion. POL also significantly attenuated the up-regulation of APP and BACE1 expression in the hippocampus produced by chronic cerebral hypoperfusion. Conclusions : The results show that POL alleviated memory deficit and up-regulation of $A{\beta}$ and BACE1 expressions in the hippocampus. This result suggests that POL may exert ameliorating effect on memory deficit through inhibition of ${\beta}$-secretase activity and $A{\beta}$ generation.
