Analysis of Genes Activated by Salt and ER Stress in bZIP17 and bZIP28 Gene Transgenic Potato Plants

  • Potato (Solanum tubersosum L.) is susceptible to various environmental stresses such as salt, high temperature, and drought. Especially, potato tuber growth is greatly affected by drought that causes not only yield reduction but also loss of tuber quality. Since unpredictable global weather changes cause more severe and frequent water limiting conditions, improvement of potato drought tolerance can minimize such adverse effects under drought and can impact on sustainable potato production. Genetic engineering can be utilized to improve potato drought tolerance, but such approaches using endogenous potato genes have rarely been applied. We were obtained AtbZIP28 gene transgenic potato plants. It is identified transcript levels at various stress conditions, polyethylene glycol (PEG), NaCl, (ABA). Also, For identification to regulate ER stress response genes in AtbZIP28 gene transgenic potato plant, we screened seven potato genes from RNA-seq analysis under TM treatment. Five and two genes were up- and down-regulated by TM, respectively. Their expression patterns were re-examined at stress agents known to elicit TM, DTT, DMSO and salt stress.

Effect of Surface Modifying Agents Towards Enhancing Performance of Waste Gypsum Based PBAT Composite

  • Stearic acid (SA), polyethylene glycol (PEG), and malic acid (MA) have been used to modify the surface of waste gypsum to develop corresponding poly (butylene adipate-co-terephthalate) (PBAT) composites. According to the mechanical properties, MA-treated gypsum (MA-gypsum) showed the best performance, whereas SA-gypsum showed the worst performance. In contrast to SA and PEG (having -COOH and -OH as polar functional groups, respectively), the presence of both -OH and -COOH in MA is responsible for the superior surface treatment of gypsum and its better dispersion in the polymer matrix (as revealed by FE-SEM analyses). The presence of long aliphatic chain in SA is supposed to inhibit the dispersion of SA-gypsum. Further, the performance of MA-gypsum/PBAT was enhanced by adding polylactic acid (PLA). The maximum optimized contents of MA-gypsum and PLA are 20 and 7.5 wt% for developing a high-performance PBAT composite.

Desalination performance of Al2O3 positively charged nanofiltration composite membrane

  • Al2O3 positively charged nanofiltration composite membrane was successfully prepared with aluminate coupling agent (ACA) as modifier, sodium bisulfite (NaHSO3) and potassium persulfate (K2S2O8) as initiator and methacryloyloxyethyl trimethylammonium chloride (DMC) as crosslinking monomer. The surface of the membrane before grafting and after polymerization were characterized by SEM and FT-IR. Three factor and three-level orthogonal experiments were designed to explore the optimal conditions for membrane preparation, and the optimal group was successfully prepared. The filtration experiments of different salt solutions were carried out, and the retention molecular weight was determined by polyethylene glycol (PEG). The results showed that the polymerization temperature had the greatest effect on the rejection rate, followed by the reaction time, and the concentration of DMC had the least effect on the rejection rate. The rejection rates of CaCl2, MgSO4, NaCl and Na2SO4 in the optimal group were 83.8%, 81.3%, 28.1% and 23.6% (average value), respectively. The molecule weight cut-off of 90% (MWCO) of the optimal group was about 460, which belongs to nanofiltration membrane.

Efficacy and safety of electrolytes-free polyethylene glycol (PEG) 4000 for disimpaction in children with chronic functional constipation (소아 만성 기능성변비 치료에서 전해질이 함유되지 않은 polyethylene glycol (PEG) 4000의 분변박힘 제거효과 및 안전성)

  • Purpose : This study was performed evaluate optimal dose for disimpaction, efficacy and safety of PEG 4000 in children with chronic functional constipation. Methods : Eighty six children with chronic functional constipation were enrolled in this prospective study at Konkuk university hospital March, 2003 through August, 2006. Success in disimpaction with PEG 4000 was defined as meeting at least two out of three criteria; resolution of chief complaint, getting easiness of defecation with respect to frequency of bowel movement and hardness of stool based on defecation diary, and decrease in fecal impaction on simple abdominal X-ray test. Adverse effects of PEG 4000 were monitored clinically and biochemically. Results : Eighty three out of 86 children completed the study, and success rate of disimpaction was 99% (82/83). The mean dose of PEG 4000 for disimpaction was $0.93{\pm}0.28g/kg/day$ (0.4-2.0 g/kg/day, Max.: 30 g/day). Frequency of bowel movement increased ($5.02{\pm}2.71/wk$ vs. $11.25{\pm}5.43/wk$) in most children (79/83). Fecal impaction on simple abdominal X-ray test improved with statistical significance in 25 children (P=0.0007). Because of adverse effect of PEG 4000, 3 children did not complete the study; urticaria, severe diarrhea, diarrhea and abdominal pain. One 6-year-old girl who completed the study complained tingling sensation in the hand and foot without laboratory abnormality (4/86, 4.7%). Laboratory test revealed hyperosmolality without clinical symptom in 1 child, and eosinophilia in 6 children. Conclusion : The average safe and effective dose of PEG 4000 for disimpaction was $0.93{\pm}0.28g/kg/day$ (0.4-2.0 g/kg/day, Max : 30 g/day) in children with chronic functional constipation.

Polyethersulfone (PES) ultrafiltration (UF) membranes loaded with silver nitrate for bacteria removal

  • PES UF membranes containing silver were prepared to impart antibacterial properties for waste water treatment. Asymmetric membranes for antibacterial application were prepared from polyethersulfone (PES) and silver nitrate ($AgNO_3$) (PES/$AgNO_3$=15/2 by weight) solution in N-Methyl-2-pyrrolidone (NMP) via simple wet phase inversion technique. These membranes were characterized by polyvinylpyrrolidone (PVP) and polyethylene glycol (PEG) of different molecular weights (1000 ppm in water) at room temperature and on operating pressure of 5 bars. It was observed that the water flux of PES-$AgNO_3$ membrane is slightly lower than virgin PES but still increased linearly with the increment of pressure applied. The morphology of the resulting membranes was examined using Field-Emission Scanning Electron Microscope (FESEM) coupled with Energy Dispersive Spectroscopy (EDS). Elemental analysis using EDS proved that silver is successfully loaded on the membrane surfaces. Due to the success of loading silver on membrane surfaces, antibacterial activities were evaluated via agar diffusion method against Escherichia coli (E.coli) and Staphylococcus aureus (S.aureus) culture. By incorporating 2 wt% of silver nitrate, PES-$AgNO_3$ showed significant inhibition ring on both E.coli and S.aureus. Filtration of E.coli solution (OD 0.31) showed satisfactory rejection data with ~100% inhibition growth after 24 hours incubation at $37^{\circ}C$. Resultant membranes also exhibit better tensile strength (compared to virgin PES) up to 71% may be due to the suggested interactions. The residual silver during fabrication was measured using ICP-MS and result showed that the residual silver content of PES-$AgNO_3$ membrane was only ~1% of the original silver added in the polymer solution. These studies have shown that PES-$AgNO_3$ UF membranes are potential in improving the filtration in water treatment.

Germination and Biochemical Changes in Accelerated Aged and Osmoprimed Pinus thunbergii Seeds

  • The aim of this study was to investigate relationship among seed viability and enzymes activities involved in scavenging reactive oxygen species (ROS), especially, superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT). In other respects, osmopriming has been demonstrated to reinvigorate aged seeds. Various viabilities of seeds that were ranged from 80 to 100% of germination rate could be produced using osmopriming and accelerated aging treatments. Priming treatment of Pinus thunbergii seeds for 3 days at $15^{\circ}C$ with a polyethylene glycol solution at -1.2 MPa improved their subsequent germination at $25^{\circ}C$. Accelerated aging (3, 6, 9, and 12 days at $41^{\circ}C$ and 100% relative humidity) decreased seed germination percentage depending on aging treatment duration. Electrolyte conductivities of seeds were measured as assay of membrane integrity. The conductivity from electrolyte leakage of P. thunbergii seed was also correlated with seed germinability. Conductivity for control seeds that had 95% of germination percentage was 3.48 ${\mu}S\;g^{-1}$, but jumped as doubled (7.98 ${\mu}S\;g^{-1}$) in 12-day-aged seed that had 80% of germination percentage. Our results demonstrate that aging of P. thunbergii seeds is associated with changes in the electrolyte leakage, lipid peroxidation, and antioxidant defense system. Priming of aged seeds progressively restored the initial germinative ability and resulted in a marked decrease in the levels of MDA and conductivity of seed leachate. These effects of priming were also well recovered of GR and CAT activities in aged seed. The improved seed quality by priming treatment appears at least partly attributable to reduced lipid peroxidation, resulting from enhanced antioxidative enzyme activities that are suggesting the antioxidant defense systems play a key role in seed vigor.

Photosynthetic Responses to Dehydration in Green Pepper(Capsicum annuum L.)Leaves

  • Photosynthetic responses to dehydration were examined by the simulataneous measurement of O2 evolution and chlorophyll (Chl) fluorescence in green pepper leaves. Dehydration was induced by immersing the plant roots directly in the Hoagland solution containing varying concentration (2-30%) of polyethylene glycol(PEG-6000) . Water potential of the leaf was decreased time-and concentation -dependently by PEG-treatment. The decrease in water potential of leaf was correlated with the decrease in both the maximal photosynthesis (Pmax) and quantum yield of O2 evolution, but Pmax dropped more rapidly than quantum yield at all water deficit conditions tested. However, Chl fluorescence parameters were not affected much. Dehydration did not change the initial fluorescence (Fo) and maximum photochemical efficiency(Fv/Fm) of photosystem(PS) II. Both the photochemical quenching (qP) and non-photochemical quenching(NPQ) were not changed by dehydration under low PFR(50 $\mu$mols m-2s-1 ). In contrast, under high PFR(270$\mu$mols m-2s-1)qP was slightly decreased while NPQ was greatly increased. The fast induction kinetics of Chl fluroecence showed no change in Chl fluorescence pattern by dehydration at high PFR (640 $\mu$mols m-2s-1 ), but exhibited a significant drop in peak level(Fp)at low PRFR (70$\mu$mols m-2s-1 ). PS I oxidation and reduction kinetics revealed normal reduction but delayed oxidation to P-700+, suggesting no lesionin electron flow from PSII to PSI , but impaired electron transport to NADP+,These results suggest that water stress caused by PEG-treatment results in the reduction of photosynthesis, promarily due to the reducted electron trasport from PSI to NADP+ or hampered subsequent steps involving Calvin Cycle.

Effects of Pre-sowing Treatments of Tobacco Seed on Germination and Seedling Uniformity (담배종자의 파종 전처리가 발아 및 묘의 균일성에 미치는 영향)

  • Pre-sowing treatments of tobacco seeds (cv. NC82) were evaluated as a means of improving the speed of emergence, early seedling growth and uniformity of the seedlings. Seeds were treated through the use of polyethylene glycol 6000(PEG), solid matrix priming(SMP), chilling and dark-preincubati(DPI). Under the laboratory condition of 15$^{\circ}C$, all the treated seeds germinated more rapid and the time to 50% germination(T50) reduced than control. Among the treatments, SMP was the best to reduce the T50 for germination. All the treatments did not affect the final germination percentage, while the chilling showed of little benefit for germination and T50. Under the greenhouse condition, seedling from SMP emerged earlier and more uniformity and also greater mean fresh and dry weight showed than other treatments. The advantage of T50 from SMP was affected more in temperature than in water content of Agro-Lig during the treatment.

Endophytic Bacteria Improve Root Traits, Biomass and Yield of Helianthus tuberosus L. under Normal and Deficit Water Conditions

  • Drought is more concerned to be a huge problem for agriculture as it affects plant growth and yield. Endophytic bacteria act as plant growth promoting bacteria that have roles for improving plant growth under stress conditions. The properties of four strains of endophytic bacteria were determined under water deficit medium with 20% polyethylene glycol. Bacillus aquimaris strain 3.13 showed high 1-aminocyclopropane-1-carboxylate (ACC) deaminase production; Micrococcus luteus strain 4.43 produced indole acetic acid (IAA). Exopolysaccharide production was high in Bacillus methylotrophicus strain 5.18 while Bacillus sp. strain 5.2 did not show major properties for drought response. Inoculation of endophytic bacteria into plants, strain 3.13 and 4.43 increased height, shoot and root weight, root length, root diameter, root volume, root area and root surface of Jerusalem artichoke grown under water limitation, clearly shown in water supply at 1/3 of available water. These increases were caused by bacteria ACC deaminase and IAA production; moreover, strain 4.43 boosted leaf area and chlorophyll levels, leading to increased photosynthesis under drought at 60 days of planting. The harvest index was high in the treatment with strain 4.43 and 3.13 under 1/3 of available water, promoting tuber numbers and tuber weight. Inulin content was unchanged in the control between well-watered and drought conditions. In comparison, inulin levels were higher in the endophytic bacteria treatment under both conditions, although yields dipped under drought. Thus, the endophytic bacteria promoted in plant growth and yield under drought; they had outstanding function in the enhancement of inulin content under well-watered condition.

Water desalination by membrane distillation using PVDF-HFP hollow fiber membranes

  • Poly(vinylidene fluoride-co-hexafluoropropylene), PVDF-HFP, hollow fiber membranes were prepared by the dry/wet spinning technique using different polyethylene glycol (PEG) concentrations as non-solvent additive in the dope solution. Two different PEG concentrations (3 and 5 wt.%). The morphology and structural characteristics of the hollow fiber membranes were studied by means of optical microscopy, scanning electron microscopy, atomic force microscopy (AFM) and void volume fraction. The experimental permeate flux and the salt (NaCl) rejection factor were determined using direct contact membrane distillation (DCMD) process. An increase of the PEG content in the spinning solution resulted in a faster coagulation of the PVDF-HFP copolymer and a transition of the cross-section internal layer structure from a sponge-type structure to a finger-type structure. Pore size, nodule size and roughness parameters of both the internal and external hollow fiber surfaces were determined by AFM. It was observed that both the pore size and roughness of the internal surface of the hollow fibers enhanced with increasing the PEG concentration, whereas no change was observed at the outer surface. The void volume fraction increased with the increase of the PEG content in the spinning solution resulting in a higher DCMD flux and a smaller salt rejection factor.
