융합 환제의 PAHs 함량 및 위해성 노출 평가에 대한 연구 (Study on the Assessment of PAHs Content and Risk Exposure of Convergence Herbal Pills)

  • 본 연구는 서울에서 유통되는 식품용 융합 환제에 대하여 위해물질인 벤조피렌 등의 PAHs (polycyclic aromatic hydrocarbons) 오염실태를 조사하고 위해도를 평가하였다. 2010~2013년 서울약령시 한약재 판매업소 등에서 31종 93건의 환제를 수집한 후 시료를 추출, 여과, 농축하여 SPE (Sep-pak florisil)로 유출시킨 후 다시 농축하여 액체크로마토그래프로 분석하였다. 분석한 결과 총 8종 PAHs 평균 함량은 $10{\mu}g/kg$ 이하이었고, PAHs 위해성 평가 결과 초과발암위해도는 일상 생활에서의 노출에 의한 것보다 낮았고, 노출안전역(MOEs)은 대부분의 시료가 위해를 무시할 만한 수준으로 안전한 것으로 평가되었다. 향후 의약용 환제와 식품용 환제의 위해물질에 대한 비교 융합연구가 필요하다.

Chemopreventive Effects of Korean Red Ginseng (Panax ginseng Meyer) on Exposure to Polycyclic Aromatic Hydrocarbons

  • Polycyclic aromatic hydrocarbons (PAHs) are well known environmental carcinogens. PAH metabolites, especially BaP-7,8- dihydrodiol, 9,10 epoxide, initiate carcinogenesis via high specificity binding to DNA to form DNA adducts. The Korean red ginseng (KRG) from Panax ginseng has been suggested to protect against damages due to PAH exposure but the mechanism is unknown. Therefore, we investigated effects of KRG on PAH exposure using toxicokinetic methods and changes of PAH-induced oxidative damage during a 2 week-clinical trial (n=21 healthy young female, $23.71{\pm}2.43$ years). To analyze antioxidative effects of KRG, we measured changes in the levels of urinary malondialdehyde (MDA) before and after KRG treatment. We observed a significant positive association between levels of urinary MDA and 1-hydroxypyrene, a biomarker of PAH exposures (slope=1.47, p=0.03) and confirmed oxidative stress induced by PAH exposures. A reverse significant correlation between KRG treatment and level of urinary MDA was observed (p=0.03). In summary, results of our clinical trial study suggest that KRG plays a significant role in antioxidative as well as toxicokinetic pathways against PAHs exposure.

Quality Properties of Seasoned-Dried Pacific Saury Treated with Liquid Smoke -1. Volatile Flavor Compounds in Commercial Liquid Smokes-

  • In order to identify of volatile flavor compounds and polycyclic aromatic hydrocarbons (PAHs) in commercial liquid smokes, this study was conducted to analyze volatile flavor compounds by solvent extraction and/or Purge & Trap method/GC/MSD. A total of 156 volatile flavor compounds were detected in 6 commercial liquid smokes, and these compounds were composed mainly of 12 aldehydes, 60 ketones, 7 alcohols, 14 acids, 20 esters, 24 aromatic compounds, 7 furans and 12 miscellaneous compounds. Ketones $(806.6-7,573.9\mu g/mL)$ and aromatic compounds $(282.6-7,896.3 \mu g/mL)$ were more abundant than others. The PAHs known as carcinogen have not been detected in this study. The acids $(422.9-4,903.1\mu g/mL)$ was identified in relatively high concentration compared to other groups. Phenol and its derivatives among aromatic compounds were in relatively high concentration. Especially, the phenol and its derivatives including o-cresol, guaiacol, 4-ethylguaiacol and syringol were in higher concentration.

  • 나프탈렌(Naphthalene)과 같은 다환방향족탄화수소류(Polycyclic aromatic hydrocarbons, PAHs)는 수계에 유입되어 해양생물들에게 영향을 미치게 된다. 나프탈렌은 유류속의 PAHs 내에서 함량이 가장 높으며, 독성이 강하다. 본 연구에서는 대조구(0 ${\mu}g/L$), 1000, 1800, 3200, 5600, 10000 ${\mu}g/L$의 6개 나프탈렌 농도구를 설정하고 넙치 치어에 대한 나프탈렌의 24시간 반수치사농도(24h-$LC_{50}$)을 구했으며, 아가미, 간, 신장, 근육내의 나프탈렌 농도를 분석하였다. 넙치 치어의 24h-$LC_{50}$은 Large 그룹에서 2410.76 ${\mu}g/L$, Small 그룹에서 2230.67 ${\mu}g/L$을 나타내었다. 각 조직에서의 나프탈렌 농도는 간 및 신장에서 아가미 및 근육보다 높았다.

Proteomic Analysis of Polycyclic Aromatic Hydrocarbons (PAHs) Degradation and Detoxification in Sphingobium chungbukense DJ77

  • Polycyclic aromatic hydrocarbons (PAHs) are commonly present xenobiotics in natural and contaminated soils. We studied three (phenanthrene, naphthalene, and biphenyl) xenobiotics, catabolism, and associated proteins in Sphingobium chungbukense DJ77 by two-dimensional gel electrophoresis (2-DE) analysis. Comparative analysis of the growth-dependent 2-DE results revealed that the intensity of 10 protein spots changed identically upon exposure to the three xenobiotics. Among the upregulated proteins, five protein spots, which were putative dehydrogenase, dioxygenase, and hydrolase and involved in the catabolic pathway of xenobiotic degradation, were induced. Identification of these major multifunctional proteins allowed us to map the multiple catabolic pathway for phenanthrene, naphthalene, and biphenyl degradation. A part of the initial diverse catabolism was converged into the catechol degradation branch. Detection of intermediates from 2,3-dihydroxy-biphenyl degradation to pyruvate and acetyl-CoA production by LC/MS analysis showed that ring-cleavage products of PAHs entered the tricarboxylic acid cycle, and were mineralized in S. chungbukense DJ77. These results suggest that S. chungbukense DJ77 completely degrades a broad range of PAHs via a multiple catabolic pathway.

Determination and risk characterization of polycyclic aromatic hydrocarbons of tea by using the Margin of Exposure (MOE) approach

  • Tea is one of the most frequently consumed drinks due to its favourite taste and the health benefit. Tea is produced by several processes and drying is very important step to develop the flavour and destroys the enzymes in tea. However, during drying tea, polycyclic aromatic hydrocarbons some of which are carcinogen and genotoxin are naturally produced. The risk of PAHs by drinking tea was characterized by determining contents of 4 PAHs in tea. 4 PAHs including Benz(a)anthracene (BaA), Chrysene (CHR), Benzo(b)fluoranthene (BbF) and Benzo(a)pyrene (BaP) were investigated by GC-MS in total 468 tea products, which were contaminated up to $4.63ng\;g^{-1}$. Mate tea was the most highly contaminated by BaA, CHR, BbF and BaP and followed by Solomon's seal and Chrysanthemum. The Margin of Exposures calculated by the concentration of BaA, CHR, BbF and BaP and consumption amount of tea were higher than 10,000, and the risk of PAHs in tea were low concern to public health.

  • Polycyclic aromatic hydrocarbons(PAHs) are widely distributed in the environment and are often implicated as potential carcinogens. The chromatographic methods of detection and quantitative determination of PAHs in environmental samples are costly, time consuming, and do not account for all kinds of PAHs. This work describes a quantitative spectroscopic method for the analysis of mixtures of eight PAHs using multivariate calibration models for Fourier transform near infrared(FT-NIR) spectral data. The NIR spectra of mixtures of PAHs (anthracene, pyrene, 1,2-benzanthracene, perylene, chrysene, benzo(a)pyrene, 1-methylanthracene and benzo(ghi)perylene) were measured in the wavelength range from 1100 nm to 2500 nm. The spectral data were processed using a partial least squares regression. We have studied the spectral characteristics of NIR spectra of mixtures of PAHs. It was possible to determine each PAM used in this study at the environmental level(mg L-1) in the laboratory samples. Further development may lead to the rapid determination of more PAHs in typical environmental samples.

2002년 6월부터 11월까지 전주지역 대기 중 다환방향족 탄화수소의 특성 (Characteristics of Polycyclic Aromatic Hydrocarbons in Ambient Air in Jeonju between July and November in 2002)

  • Atmospheric concentrations of polycyclic aromatic hydrocarbons (PAHs) were measured at the Chonbuk National University located in Jeonju, four times between June and November 2002, each time for five days. Twenty-four compounds including five alkyl PAHs and byphenyl were analyzed. Average total concentration of 24 PAHs was 85 $\pm$ 15 ng/$m^3$ and about 94% of PAHs existed in the gas phase. On an average, naphthalene accounted for about 30% of the total PAHs concentration. The gas/particle partitioning was not much varied during the measurement period. High molecular weight PAHs with five and six rings were primarily associated with fine particles less than 1 $\mu$m. Lower molecular weight PAHs were evenly distributed in fine and coarse particles so that their distribution was similar to that of TSP.

Affecting Factors on the Variation of Atmospheric Concentration of Polycyclic Aromatic Hydrocarbons in Central London

  • In this study, a statistical investigation was carried out for the evaluation of any relationship between polycyclic aromatic hydrocarbons (PAHss) associated with ambient aerosols and other air quality parameters under varying meteorological conditions. Daily measurements for PAHs and air quality/meteorological parameters were selected from a data-base constructed by a comprehensive air monitoring in London during 1985-1987. Correlation coefficients were calculated to examine any significant relationship between the PAHs and other individual variables. Statistical analysis was further Performed for the air quality/meteorological data set using a principal component analysis to derive important factors inherent in the interactions among the variables. A total of six components were identified, representing vehicle emission, photochemical activity/volatilization, space heating, atmospheric humidity, atmospheric stability, and wet deposition. It was found from a stepwise multiple regression analysis that the vehicle emission component is overall the most important factor contributing to the variability of PAHs concentrations at the monitoring site. The photochemical activity/volatilzation component appeared to be also an important factor particularly for the lower molecular weight PAHs. In general, the space heating component was found to be next important factor, while the contributions of other three components to the variance of each PAHs did not appear to be as much important as the first three components in most cases. However, a consistency for these components in their negative correlations with PAHs data was found, indicating their roles in the depletion of PAHs concentrations in the urban atmosphere.

Distribution and Origin of Polycyclic Aromatic Hydrocarbons (PAHs) in Surface Sediments Inside Hallim Harbor of Jeju Island, Korea

  • The surface sediments inside Hallim Harbor, one of the major harbors of Jeju Island, were sampled three times (June, September and December, 2001) and analyzed for 16 polycyclic aromatic hydrocarbons (PAHs), recommended by US-EPA as priority pollutants, to assess their distribution levels and to suggest their possible origins. The concentrations of PAHs ranged from 19 to 496 ng/g on a dry weight basis with a mean value of 245 ng/g, and the levels were low to moderate in comparison with other areas in the world. Based on comparisons of individual and total PAH concentrations with effects-based and equilibrium partitioning-based on sediment quality guidelines, the potential for the biological effects were expected to be low. The sedimentary PAHs may be correlated with organic carbon and mud contents to some extent. From the examinations of the four PAH origin indices, such as LMW/HMW (low molecular weight 2-3 ring PAHs over high molecular weight 4-6 ring PAHs), phenanthrene/anthracene ratio, fluoranthene/pyrene ratio, chrysene/benzo〔a〕anthracene ratio, it can be concluded that the sediment PAH contaminations were ascribed to both pyrolytic and petrogenic origins.
