An Update on Occupation and Prostate Cancer

  • Background: Our aim was to identify gaps and limitations in the current literature and to make recommendations for future research required to address these. Materials and Methods: We reviewed occupational exposures and related factors associated with the risk of prostate cancer between 2000 and 2012. These included chemical, ergonomic, physical or environmental, and psychosocial factors which have been reported by epidemiological studies across a range of industries. Results: The results are inconsistent from study to study and generally this is due to the reliance upon the retrospectivity of case-control studies and prevalence (ecological) studies. Exposure assessment bias is a recurring limitation of many of the studies in this review. Conclusions: We consider there is insufficient evidence to implicate prostate cancer risk for ergonomic, physical, environmental or psychosocial factors, but there is sufficient evidence to implicate toxic metals, polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs). More research is required to identify specific pesticides that may be associated with risk of prostate cancer.

Relationships between Biodegradation and Sorption of Phenanthrene in Slurry Bioremediation

  • Bioremediation of hazardous hydrophobic organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), is a major environmental concern due to their toxic and carcinogenic properties. Due to their hydrophobicity, the hydrophobic organic compounds are mainly associated with the soil organic matter or nonaqueous-phase liquids. A major question concerns the relationships between biodegradation and sorption. This work develops and utilizes a non- steady state model for evaluating the interactions between sorption and biodegradation of phenanthrene, a 3-ring PAH compound, in soil-slurry systems. The model includes sorption/desorption of a target compound, its utilization by microorganisms as a primary substrate existing in the dissolved phase and/or the sorbed phase in biomass and soil, oxygen transfer, and oxygen utilization as an electron acceptor. Biodegradation tests with phenanthrene were conducted in liquid and soil-slurry systems. The soil-slurry tests were performed with very different mass transfer rate: fast mass transfer in a flask test at 150 rpm, and slow mass transfer in a roller-bottle test at 2 rpm. In the slurry tests, phenanthrene was degraded more rapidly than in liquid tests, but with a similar rate in both slurry systems. Modeling analyses with several hypotheses indicate that a model without biodegradation of compound sorbed to the soil was not able to account for the rapid degradation of phenanthrene, particularly in the roller bottle slurry test. Reduced mass-transfer resistance to bacteria attached to the soil is the most likely phenomenon accounting for rapid sorbed-phase biodegradation.

  • The fate and the behavior of polycyclic aromatic hydrocarbons (PAHs) and heavy metals in the environment are mainly controlled by their interactions with various components of soils and sediments. Due to their large surface area and abundance in many soils, smectites may greatly influence the fate and transport of the contaminants in the environment. In our experiment, PAH sorption by hexadecyltrimethylammonium (HDTMA)-modified smectite linearly increased in proportion to the amount of HDTMA added on the clay. However, trimethylammonium (TMA)-modified smectite did not show superiority in its sorption of PAH compared with the HDTMA-smectite or dodecyltrimethylammonium (DTMA)-smectite. Meanwhile, the smectites modified with the same cationic surfactants adsorbed Cd$^{2+}$ (heavy metal) significantly from water at low surfactant loading level, but the Cd$^{2+}$ adsorption linearly decreased as the loading of surfactant increased. The result shows that the sorption tendency of organoclays for organic or inorganic contaminants was significantly influenced by the amount and size of the surfactants added on the clay. It means that the stabilization and configuration of cationic surfactant formed on the clay interlayer according to the loading amount of each surfactant of different sizes may be an important factor in effectively sorbing environmental pollutants.nts.

담배연기응축물의 DNA 손상작용과 야채 및 과일추출물의 보호효과 (Antigenotoxicity of Vegetable or Fruit Extract against Cigarette Smoke Condensate)

  • Cigarette smoke condensate (CSC) is known to be carcinogenic compound. CSC contains many organic compounds such as polycyclic aromatic hydrocarbons (PAHs), and heterocyclic amine compounds (HCAs). Reactive oxygen species (ROS) are also generated and induce oxidative DNA damage during the metabolism of CSC. The rat microsome mediated and DNA repair enzyme treated comet assays together with conventional comet assay were performed to evaluate the mechanisms of CSC genotoxicity. The organic extract of CSC induced oxidative and microsome mediated DNA damage. Vitamin C as a model antioxidant reduced DNA damage in endonuclease III treated comet assay. One of flavonoid, galangin as a CYP1A1 inhibitor, reduced DNA damage in the presence of S-9 mixture. The ethanol extracts of the mixed vegetables (BV) or the mixed fruits (BF) showed potent inhibitory effects against CSC induced DNA damage with oxidative DNA lesions and in the prescence of S-9 mixture. These results indicate that BV and BF could prevent CSC-induced cellular DNA damage by inhibiting oxidative stress and suppressing cytochrome P450 in mammalian cells.

Effect of Chinese Yam on Benzo[a]pyrene Hydroxylase Activities in Rats Fed Dietary Benzo[a]pyrene

  • To investigate the effect of yam on the activity of benzo[a]pyrene hydroxylase(BPH), the key enzyme associated with polycyclic aromatic hydrocarbons(PAHs) metabolism, rats were fed a fiber free diet for 7 days, whereupon they were switched to experimental diets for another 7 days. Diets contained benzo[a] pyrene(BP, 400 mg/kg diet) and 25% or 50% yam powder (freezer dried and hot air dried ). Diets containing pectin and cellulose were compared with diets containing yam. BPH activities were assessed in the liver, lung, kidney, stomach, small intestine and large intestine of rats. BP induced BPH activities in various tissues ; 8 fold in liver, 28 in lung and stomach , and 32 in large intestine. The addition of yam significantly lowered BPH activity in liver, lung and stomach and hot air dried yam was nmor eeffectivie than freeze dried yams. These data suggested that yam containing diet may influence carcinogen metabolism in liver and extrahepatic target tissues by altering activities of BPH and may reduce exposure of these tissues to dietary carcinogens.

좀개구리밥(Lemna perpusilla Torr.)의 생육에 대한 PAHs와 암모늄의 영향 (Influences of PAHs and Ammonium on Growth of Duckweed (Lemna perpusilla Torr.))

  • 좀개구리밥(Lemna perpusilla J. Torrey)은 오염물질의 독성시험에 사용되어 온 대표적인 부유성 수생식물이다. 본 연구에서는 오염된 공장지대나 농경지에서 식물의 생육에 악영향을 미치는 독성물질인 PAHs(polycyclic aromatic hydrocarbons; 다환방향족탄화수소)와 암모늄($NH_4{^+}$)의 식물 독성 평가를 위해 좀개구리밥을 이용하였다. 독성물질인 PAHs와 암모늄이 들어있는 배양액에 좀개구리밥 엽상체를 배양하면서 반응을 조사하였다. 조사결과, 좀개구리밥의 생장은 PAHs의 종류와 농도에 따라 다르게 반응하였다. PAHs중 fluorene의 처리 때 좀개구밥의 생장이 가장 저해되었으며, fluoranthene과 pyrene은 생장 저해 효과가 적었다. fluoranthene과 pyrene의 경우 30ppm정도의 낮은 농도에서는 좀개구리밥의 생장 저해는 거의 일어나지 않았다. 식물색소 함량도 또한 처리한 독성물질의 종류에 따라 다르게 나타났다. Phenanthrene과 fluorene 처리 시 fluoranthene과 pyrene 처리 때보다 chlorophyll의 함량이 많이 감소되었다. 그러나, 항산화 화합물로 잘 알려진 carotenoids의 경우 fluoranthene과 pyrene을 30ppm 이하의 낮은 농도로 처리 시 오히려 함량이 약간 증가하는 것으로 나타났다. 이는 저농도의 fluoranthene 또는 pyrene 처리 시 이들 물질이 좀개구리밥의 황산화 작용을 자극하여 황산화 물질인 carotenoids를 많이 만들게 하는 작용을 하는 것으로 추측된다. 암모늄은 3mM의 낮은 농도에서도 좀개구리밥 엽상체의 생장에 강한 저해효과를 갖는 것으로 나타났다.

신평·장림 산단 인근 주민의 PAHs 및 VOCs 노출 (Exposure to PAHs and VOCs in Residents near the Shinpyeong·Jangrim Industrial Complex)

  • Objectives: This study aims to investigate the atmospheric concentration of polycyclic aromatic hydrocarbons (PAHs) and volatile organic compounds (VOCs) and the urinary concentration of biomarkers in residents near the Shinpyeong·Jangrim Industrial Complex to compare them with those of residents in a control area. Methods: Hazardous air pollutants (PAHs and VOCs) were measured in an exposure area (two sites) and a control area (one site). Urine samples were collected from residents near the industrial complex (184 persons) and residents in the control area (181 persons). Multiple linear regression analysis was used to identify which factors affected the concentration of PAHs and VOCs metabolites. Results: The average atmospheric concentration of PAHs in Shinpyeong-dong and Jangrim-dong was 0.45 and 0.59 ppb for pyrene, 0.15 and 0.16 ppb for benzo[a]pyrene, and 0.29 and 0.35 ppb for dibenz[a,h]anthracene. The average atmospheric concentration of VOCs was 1.10 and 0.99 ppb for benzene, 8.22 and 11.30 ppb for toluene, and 1.91 and 3.05 ppb for ethylbenzene, respectively. The concentrations of PAHs and VOCs in residents near the Shinpyeong·Jangrim Industrial Complex were higher than those of residents in the control area. Geometric means of urinary 2-hydroxyfluorene, 1-hydroxypyrene, methylhippuric acid, and mandelic acid concentrations were 0.45, 0.22, 391.51, and 201.36 ㎍/g creatinine, respectively. Those levels were all significantly higher than those in the control area (p<0.05). In addition, as a result of multiple regression analysis, even after adjusting for potential confounding factors such as gender and smoking, the concentration of metabolites in urine was high in residents near the Shinpyeong·Jangrim Industrial Complex. Conclusion: The results of this study show the possibility of human exposure to VOCs in residents near the Shinpyeong·Jangrim Industrial Complex. Therefore, continuous monitoring of the local community is required for the management of environmental pollutant emissions.

넙치 치어 Paralichthys olivaceus의 혈액학적 성상에 미치는 나프탈렌의 급성독성영향 (The Acute Toxicity of Naphthalene on Hematologic Properties in Juvenile Flounder Paralichthys olivaceus)

  • 넙치 치어에 대한 나프탈렌의 급성독성을 조사하기 위하여 대조구($0{\mu}g/L$), 1000, 1800, 3200, 5600, $10000{\mu}g/L$의 6개의 나프탈렌 농도구를 설정하여 24시간 동안 노출실험을 실시하고 혈액학적 성상을 분석하였다. 넙치 치어에 대한 나프탈렌의 24h-$LC_{50}$$3600{\mu}g/L$를 나타냈다. 넙치 치어의 핼액학적 반응에서 헤마토크리트값은 5600, $10000{\mu}g/L$의 농도구에서 대조구와 비교하여 유의하게 증가하였고, 글루코스는 $10000{\mu}g/L$의 농도 구에서 유의하게 증가 하였다(P<0.05). 삼투질 농도는 3200, 5600, $10000{\mu}g/L$의 농도구에서 유의하게 증가하였다. 반면, 이온분석 결과 [$Na^+$], [$K^+$]은 5600 및 $10000{\mu}g/L$ 나프탈렌 농도구에서 유의하게 증가하였으나 [$Cl^-$]는 큰 차이를 보이지 않았다.

Impact of AhR, CYP1A1 and GSTM1 Genetic Polymorphisms on TP53 R273G Mutations in Individuals Exposed to Polycyclic Aromatic Hydrocarbons

  • This study was to undertaken to investigate the impacts of AhR, CYP1A1, GSTM1 genetic polymorphisms on the R273G mutation in exon 8 of the tumor suppressor p53 gene (TP53) among polycyclic aromatic hydrocarbons (PAHs) exposed to coke-oven workers. One hundred thirteen workers exposed to PAH and 82 control workers were recruited. We genotyped for polymorphisms in the AhR, CYP1A1, GSTM1, and TP53 R273G mutation in blood by PCR methods, and determined the levels of 1-hydroxypyrene as PAH exposure marker in urine using the high pressure liquid chromatography assay. We found that the distribution of alcohol users and the urinary excretion of 1-OHP in the exposed workers were significantly higher than that of the control workers (p=0.004, p<0.001, respectively). Significant differences were observed in the p53 genotype distributions of smoking subjects (p=0.01, 95%CI: 1.23-6.01) and PAH exposure (p=0.008, 95%CI: 1.24-4.48), respectively. Further, significant differences were observed in the p53 exon 8 mutations for the genetic polymorphisms of Lys/Arg for AhR (p=0.02, 95%CI: 0.70-15.86), Val/Val for CYP1A1 (p=0.04, 95%CI: 0.98-19.09) and null for GSTM1 (p=0.02, 95%CI: 1.19-6.26), respectively. Our findings indicated that polymorphisms of PAH metabolic genes, such as AhR, CYP1A1, GSTM1 polymorphisms may interact with p53 genetic variants and may contribute to PAH related cancers.

NEAR- TO MID-INFRARED SLIT SPECTROSCOPIC OBSERVATIONS OF THE UNIDENTIFIED INFRARED BANDS IN THE LARGE MAGELLANIC CLOUD

  • We present the results of the near-infrared (NIR) to mid-infrared (MIR) slit spectroscopic observations of the diffuse emission toward nine positions in the nearby irregular galaxy Large Magellanic Cloud (LMC) with the Infrared Camera (IRC) on board AKARI. The unique characteristic of AKARI/IRC provides a great opportunity to analyze variations in the unidentified infrared (UIR) bands based on continuous spectra from 2.5 to $13.4{\mu}m$ of the same slit area. The observed variation of $I_{3.3}/I_{11.3}$ suggests destruction of small-sized UIR band carriers, polycyclic aromatic hydrocarbons (PAHs) in harsh environments. This result demonstrates that the UIR $3.3{\mu}m$ band provides us powerful information on the excitation conditions and/or the size distribution of PAHs, which is of importance for understanding the evolutionary process of hydrocarbon grains in the Universe. It also suggests a new diagnostic diagram of two band ratios, such as $I_{3.3}/I_{11.3}$ versus $I_{7.7}/I_{11.3}$, for the interstellar radiation conditions. We discuss on the applicability of the diagnostic diagram to other astronomical objects, comparing the LMC results with those observed in other galaxies such as NGC 6946, NGC 1313, and M51.
