The Introduction of Polycrylamide Gel into the Solid Culture of Streptomyces spp

  • It is proposed the polyacrylamide gel, instead of agar, could be used for the solid cultures of microorganisms including Streptomyces strains. Polymerization and gellation of 5% acrylamide solution were done by autoclaving for 5 min at 121.deg.C and no hindered by the addition of nutrient-rich media. In particular, pH buffer solution suitable for corresponding microorganisms must be used in the preparation of culture media. Comparing with agar, it was discussed that polycrylamide gel had many advantages such as gellation within the wide range of strong acid Carbon and Nitrogen sources, requirement tests of growth factors and minerals, sterization at high temperature, diffusion assays of products depending on the pore size of gel, and stability and standarization of microbial cultures.

Saccharomyces 종의 등전점 전기영동에 의한 단백질 분획상 차이에 관한 연구 (Studies on the Differentiation of Protein Patterns from Saccharomyces species by Isoelectric Focusing in Polyacrylamide Gels)

  • The whole proteins from 10 different Saccharomyces species were separated by isoelectric focusing, which was carried out in pH gradient polyacrylamide gels with the carrier ampholytes of various pH ranges. About 25 protein bands were found in the gel using pH 3.0-10.0 carrier ampholytes. In gel using pH 4.0-7.0 carrier ampholytes, the protein band of pI 6.3 was found in Sacch. cerevisiae NCYC 478, ATCC 26787, Sacch. rosei and Sacch. uvarum, but it was absent in Sacch. cerevisiae ATCC 24903, ATCC 42949, ATCC 36029, Sacch. steineri var hara, Sacch. bayanus, and Sacch. diastaticus.

포도주의 신맛 조절을 위한 Leuconostoc oenos 세포의 고정화 (Immobilization of Leuconostoc oenos Cells for Wine Deacidification)

  • Leuconostoc oenos ML-34균의 세포를 polyacrylamide gel 속에 고정시키고, 이것을 이용하여 포도즙과 포도주의 신맛을 감소시켜 보았다. 세포가 가졌던 malo-lactic 발효능은 고정화 시킴으로서 감소되지는 않았다. 그러나 고정화 세포에 의한 사과산 분해의 속도는 느려졌는데, 이것은 기질이 gel층을 통과하는데 시간이 걸리기 때문이었다. 고정화 세포로서 포도즙 속의 사과산의 양을 적절한 수준까지 감소시킴으로서 포도주의 신맛을 조절할 수 있을 것 같다.

  • 1986년 12월부터 87년 11월까지 채집된 애응애과 응애를 분류학적으로 정리한 바 Brevipalpus속의 B.californicus와 B.ovovatus 2종과 다음의 한국4미기록종을 얻었다: Aegyptobia nothus Pritchard and Baker,Pentomerismus taxi Hailer, P. oregonensis McGregor,Brevipalpus lewisi McGregor. 따라서, 한국산 애응애과 응애는 Tenuipalpus속의 T.zhizhilashiliae(감나무애응애)를 포함하여 4속 7종이 보고되는 셈이다. 또한 P. oregonensis,B.californicus,B.ovovatus 및 B.lewisi에 대하여 polyacrylamide slab gel electrophoresis로 esterase와 alkaline phosphatase pattern을 조사한 바 esterase는 band수와 이동도에서 종과 속간에 뚜렷한 차이를 보였다.

  • Seeds or beans of 55 plants belonging to 31 families were screened by using several different types of red blood cells to find new lectins. In this paper, white kidney been (phaseolus vulgaris C.) was chosen to study biochemical properties of hemagglutinating proteins(lectins). An anion exchanger, DEAE Sephadex A-50, and polyacrylamide disc gel electrophoresis were main techniques used. From three main fractions eluted by stepwise NaCl gradient in 25mM Tris-HCI buffer on DEAE Sephadex column, principal lectin was identified.

  • 사람의 alpha-fetoprotein(AFP)에 대한 모노클론 항체의 생산 및 분석을 위하여, 태아조직을 재료로 추출법, DEAE-cellulose 및 concanavalin A-Sepharose, Cibacron blue F3GA-agarose, immunoadsorbent column등의 흡착크로마토그라피법에 의해 AFP를 분리하였다. 총 534g의 태아조직에서 AFP의 量은 8.76 mg으로서 순수분리되었음을 확인하였다.

  • Bacillus stearothermophilus의 mutant가 생산하는 cyclodextrin glucanotransferase(CGTase)를 affinity chromatography법을 이용하여 정제하였다. 이 CGTase의 회수율은 95%이었고 specific activity는 26.2U/mg protein에서 485.5U/mg protein으로 증가하였다. 정제된 CGTase는 SDS-polyacrylamide gel 전기영동 결과 단일 band로 나타났다. CGTase는 affinity chromatography를 이용하여 one-step으로 정제할 수 있었다.

Bacillus sp. CW-1121이 생성하는 Alkaline Proteas의 생산 및 정제 (Production and Purification of Alkaline Protease from Bacillus sp. CW-1121)

  • Alkaline protease producing bacteria were isolated from soil and identified as Bacillus sp. CW-1121. It was found that the production of alkaline protease reached to maximum in 5 day of fermentation at 4$0^{\circ}C$. The enzyme was purified by ammonium sulfate precipitation, gel filtration on Sephadex G-150 and DEAE-cellulose ion-exchange chromatography. The homogeneity of the purified enzyme was verified by polyacrylamide gel electrophoresis. The enzyme was purified 5.72 fold and yield of the enzyme purification was 16.71%. When the purified enzyme was applied to sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the molecular weight was estimated to be 55, 000.

인삼단백질분획에 대한 SDS-폴리 아크릴아미드 전기영동 (SDS-polyacrylamide Gel Electrophoresis on Partially Purified Ginseng Proteins)

  • Ginseng proteins have been extracted and partially purified by the methods of ammonium sulfate fractionation, heat inactivation and Sephadex G-75 column chromatography. The final three fractions obtained (GI, GII and GIII) were subjected to paper chromatography and SDS-polyacrylamide gel electrophoresis. Molecular weights of polypeptide chains from each fraction were est-mated by the standard line obtained from the plot of electrophoretic mobilities against the logarithm of molecular weights of standard proteins. Results are as follows: 1) GI showed three protein spots and four polypeptides of which M.W. were 63,000, 60,000, 56,000, 51,000 and 34,200. 2) GII showed four protein spots and five polypeptides with their M.W. of 64,600, 56,000, 45,400, 35,800, end 25,200, 3) GIII showed three protein spots and four polypeptides with their M.W. of 66,000, 63,000, 56,000 and 22,000.

  • 酸性 mucopolysaccharide는 鯨胎兒의 nucleus pulposus에서 分離하였다. Gel electophoresis에 있어서 酸性 mucopolysaccharide의 分離는 $0.03{\%}$의 hexamine cobaltic chloride 를 含有하는 0.05M 醋酸소-다 緩衝溶液(pH4.8)中에서 가장 좋았다. Spacer gel 上의 mucopolysaccharide 溶液을 $40{\%}$ sucrose 溶液으로 덮었을 때 mucopolysaccharide의 移動度에 變化가 나타나는 것을 觀察하였으며 이 효과는 醋酸소-다 緩衝溶液에 hexamine cobaltic chloride 을 加하면 消失하는 것을 觀察하였다.
