Effects of vegetation structure and human impact on understory honey plant richness: implications for pollinator visitation

  • Background: Though the biomass of floral vegetation in understory plant communities in a forested ecosystem only accounts for less than 1% of the total biomass of a forest, they contain most of the floral resources of a forest. The diversity of understory honey plants determines visitation rate of pollinators such as honey bee (Apis mellifera) as they provide rich food resources. Since the flower visitation and foraging activity of pollinators lead to the provision of pollination service, it also means the enhancement of plant-pollinator relationship. Therefore, an appropriate management scheme for understory vegetation is essential in order to conserve pollinator population that is decreasing due to habitat destruction and disease infection. This research examined the diversity of understory honey plant and studied how it is related to environmental variables such as (1) canopy density, (2) horizontal heterogeneity of canopy surface height, (3) slope gradient, and (4) distance from roads. Vegetation survey data of 39 plots of mixed forests in Chuncheon, Korea, were used, and possible management practices for understory vegetation were suggested. Results: This study found that 113 species among 141 species of honey plant of the forests were classified as understory vegetation. Also, the understory honey plant diversity is significantly positively correlated with distance from the nearest road and horizontal heterogeneity of canopy surface height and negatively correlated with canopy density. Conclusions: The diversity of understory honey plant vegetation is correlated to vegetation structure and human impact. In order to enhance the diversity of understory honey plant, management of density and height of canopy is necessary. This study suggests that improved diversity of canopy cover through thinning of overstory vegetation can increase the diversity of understory honey plant species.

Effect of Insect Pollinators for Chinese jujube (Zizyphus jujuba var. inermis (Bunge) Rehder) in Rain shelter house (대추 비가림하우스 내 화분매개곤충의 수분 특성 및 효과)

  • The flowering season of jujube (Zizyphus jujuba var. inermis (Bunge) Rehder) is overlaps with the rainy season and the abnormal weather conditions in Korea, thereby affecting pollination, fertilization, and fruit setting. We studied the pollinating activities of Apis mellifera L. and Bombus terrestris L. in rain shelter houses and their effects on fruit setting, with the ultimate aim of stabilizing fruit setting in Z. jujuba. A. mellifera and B. terrestirs were used for pollination in jujube orchard in the rain shelter house for approximately 55 days from June 1, 2016, to July 25, 2016. The peak time of the rate of outgoing and incoming A. mellifera was recorded in the afternoon. However, the diurnal activity of B. terrestris was constant between 09:30 and 17:30 h. The rate of jujube fruit set on current shoots by A. mellifera and B. terrestirs was 10.2 and 8.9%, whereas that in plots with no pollinators was 5.5%. Therefore, using pollinator in the rain shelter house in jujube orchard is effective in promoting jujube fruit setting.

  • Integrated pest management system in Korean apple has significantly advanced for last few decades. However harmful effects of pesticides threats the ecosystem services of natural enemies and pollinators. Apple require cross-pollination and Insect pollination with diversity and abundance is one of the keys to profitable apple production in quantity and quality as well. Thus crop protection tools are to be administered in harmony to meet the pest suppression and protection of beneficial organisms such as natural enemies and pollinators. Adding onto the established IPM system, integrated pollinator-pest management (IPPM) concept is proposed as the future direction of apple IPM. For this, ecological enginnering of creating habitats for pollinators, landscape management and agroecosytem diversification as well as selective soft pesticide uses on time guided by pest monitoring and phenologyand targeted delivery are further proposed. Recent shift of agroecosystem from climate change and new pest outbreaks require new paradigm of pest management for sustainable agricultural production.

Diversity and Interaction of Pollination Network from Agricultural Ecosystems during Summer (경북과 강원지역 농업생태계에서 여름철 화분매개네트워크 다양성과 상호작용)

  • Pollination is an important ecosystem service involved in plant breeding and reproduction. This study analyzed the pollination network, which is the interaction between flowering plants and flower-visiting insects in the agricultural landscape. Flower-visiting insects from blossoms of flowering crops and surrounding plants were quantitatively surveyed during summer time. The pollinator species and abundance on each flowering plant were analyzed. A total of 2,381 interactions were indentified with 154 pollinators on 30 species of plants. Species richness of the pollinators was highest in Coleoptera (34%) followed by Hymenoptera (28%), Diptera (28%) and Lepidoptera (10%). Apis mellifera dominated (50%) followed by Calliphora vomitoria (5.3%) and Xylocopa appendiculata among pollinators, and remaining wild pollinators provided complex interaction. Among plants, Platycodon grandiflorum, Perilla frutescen and Fagopyrum esculentum harbored most pollinators and showed highest interaction frequencies. In the modular analysis, Apis mellifera was located as a hub-species which connect the interaction of others, implying most important role in the network. This results provide the basic information on the pollinator species associated with each crop and pollinator habitat in which plant provide the nectar, pollen and habitat resources for wild pollinators.

Ex situ acclimatization of asymbiotically germinated seedlings of endangered lady's slipper orchid(Cypripedium macranthos Sw.) (멸종위기종 복주머니란 발아특성 및 실생묘 현지외 적응성 검토)

  • We use two different methods for laboratory propagation from seed of lady's slipper orchid(Cypripedium macranthos Sw.); immature seed which also called green capsule or fully mature seed about 120~130 days from pollination. In green capsule culture, the seed pods should be collected within precisely right time. The right time of seed collection could be diverse under the wether conditions or nutritional factors of the plants. In fully matured seed culture, the more complicated procedures are needed to break the dormancy of the seed; thermal or chemical treatment. The seedlings in this study were easily germinated from immature seeds in Harvais medium; 53 days after pollination(DAP) in Cypripedium pubescens, DAP 65 in C. parviflorum and C. macranthos. The germinated seedlings were transplanted to hormone free media immediately to avoid abnormal growth of seedlings. When the seedlings have roots with a minimum length of around 2-3cm and have visible dormant buds, the seedlings were removed from the flask and stored in refrigerator for vernalization. To examine the correlation of seedlings and maternal plants, the 125 seedlings of C. macranthos were transplanted in the soil bed at a distance of 20-100 cm from mother plants on April 20. The survival rate of seedlings were 92% in 20 cm distance from the ripe plants, and 56 % in 100 cm distance. The seedlings which were transplanted near mother plants showed vigorous growth in plant height, leaf width, and especially dormant buds. Considering the existence of mycorrhiza which is a symbiotic association between a fungus and the roots of a orchid vascular, the various fungus from mother plants could affect the growth of the seedlings. These results indicate the possibility of high and stable production and practical industrialization of endangered lady's slipper orchids.

  • 용과(Hylocereus undatus)는 멕시코 및 과태말라가 원산지이며 아열대 및 열대지역에서 재배되고 있으며 아시아는 베트남 타이완 중국 일본 등 국가에서 재배하고 있다. 가지에 열매가 있는 모습이 용이 여의주를 물고 있는 형과 닮았다하여 붙여진 이름이다. 지구온난화로 인한 기온상승으로 전남지역 재배에 적합한 대체 열대 및 아열대과수 개발이 필요하다. 용과는 열대과수 중 비교적 낮은 온도($10^{\circ}C$이상) 조건에서 재배 가능하여 남부지역에서 시설내 가온재배하고 있다. 국내 시설재배 중 문제점은 용과는 밤에 꽃을 피기 때문에 밤에 활동하는 매개곤충이 없어서 인공수분으로 수분문제를 해결해야한다. 인공수분의 한계시간과 수분할 경우 과실비대에 미치는 영향을 알아보고자 인공수분 시간을 개화 1시간 이내, 4~5시, 11~12시 및 무 처리(방임)로 처리하였다. 개화시기는 1차 7월 13일, 2차 8월 8일, 3차 8월 26일로 3회에 걸쳐 개화가 이루어졌으며 이시기의 화분량는 4~5시가 0.68g, 11~12시 0.47g로 1시간 이내 0.16g보다 많았고 과실비대에서 1과중은 개화11~12시가 421g, 4~5시 400g, 1시간 이내가 377g으로 처리 간에 차이는 없었지만 무처리는 185g으로 처리구와 192~236g의 유의차가 있었다. 용과의 인공수분한계시간은 다음날 아침 7시(개화 11~12시)까지 인공수분 할 경우 정상적인 착과가 이루어졌으며 과실비대를 위해서 인공수분은 필수적으로 실시해야 한다.

  • About 50,000 hives of Bombus terrestris are used for pollination of tomato and other economical crops (fruit vegetables and fruit trees) in Korea. Therefore this study was conducted to find out the effective use of B. terrestris and its economic effect in the major beefsteak-tomato cultivation areas from February to August in 2008. The pollinating activities, rate of bite-mark and lifespan of B. terrestris's hive according to all beefsteak-tomato varieties (Super Momotaro, Zeus42, Mascara and Super Sunroad, 500 $m^2$, $9,600{\sim}12,000$plants) were apparent by regular supplement of pollen ($5{\sim}6g$/3 days) to the hive of B. terrestris. Especially, Super Momotaro and Super Sunroad variety were most distinctive in the effects by regular pollen supplement.

Regional Adaptation of the Genus Cypripedium in Korea and Micropropagation with Seed

  • Cypripedium is an orchid genus of over 50 species widely distributed in the northern temperate zone, with a few taxa extending central America and the Himalayas and the southwesternmost tip of China in Asia. Of the known species approximately two thirds are found in China. In Korea, 4 species were reported to find with rare popularity ; C. macranthos, C. guttatum, C. japonicum, and C. calceolus. The seed has a firm brown fusiform testa with an opening at one end where the micropyle of the ovule was situated. Cypripedium has small, fusiform, wind-dispersed seeds that weigh about $1{\sim}2{\mu}g$ and range from 0.1 to 2 mm long and 0.07 to 0.4 mm across. The germination of Cypripedium was previously thought to be hard to cultivate in one's garden and impossible to propagate artificially, nowadays lab-propagated seedlings are readily available in EU or United States. We had already reported the successful micropropagation of lady's slipper orchids with green pod sowing methods. The collected Cypripedium species and hybrids were planted in Korea National Arboretum (KNA) on Oct. 2018. The 11 species including C. candidum and the 31 hybrids such as C. Ingrid were introduced from Germany with CITES certification. The lady's slipper orchids bloomed on May to June, and the flowers were pollinated in the same species or hybrids to get seeds for in vitro germination on 2~3 days after fully flowering. The green pods were collected after 49~70 days after pollination. The 6 species of Cypripedium seeds were easily germinated on the POM within 70 days after sowing, and the 12 hybrids PLBs were observed with small roots. We had established the scheme of lady's slipper orchids germination in 3 months with green pods, and get the possibility of seedling in 10 months of sowing in vitro.

Analysis of factors on the asymbiotic germination of white lady's slipper orchid(Cypripedium macranthos Sw. albiflorum) (멸종위기 흰복주머니란 종자발아에 미치는 요인 분석)

  • Cypripediums, popularly called lady's slippers or moccasin flowers, are the showiest and most sought after hardy terrestrial orchids, collected and grown by orchid and alpine plant enthusiasts alike. In Korea, 4 species of cypripedium are reported as Cypripedium japonicum, C. macranthos, C. guttatum, and C. calceolus. We had already reported the feasibilities of C. macranthos and C. guttatum with in vitro germination methods from immature seeds. The seeds of white lady's slipper orchid (Cypripedium macranthos Sw. alba) were collected 65 days after pollination in 2018. The green pods were sterilized with flame and sowed immediately on the POM(Phytomax orchid maintenance media(R), Sigma) supplemented with BAP 0, 0.5, 1.0 mg/L and NAA 0, 1, 2mg/L. The germination of seed was observed 90 days after sowing, and the plantlets were subcultured to the same media according to the size of the protocorm with 1~2, 2~3, 3~4, 5~6, 7~8mm. The time of the subculture to the new media seems to be critical factors of forming rhizoids which is the hairy root of the cypripediums. As a results, the protorms of the white lady's slipper orchid was successfully germinated in the POM media supplemented BAP 0.5 and NAA 1.0 mg/L. The roots and rhizoids were formed in 5~6mm protocorms subculture over 95% survival ratio. We also tried to subculture to liquid medium without activated charcoal, however the browning or malformation of the roots was observed in the root. The formation of shoots from the protocorm was effectively enhanced in the POM media with non-additives of plant growth regulators. These results indicate the possibility of high and stable production and practical industrialization of endangered white lady's slipper orchids.

Use of Microsatellite Markers to Identify Commercial Melon Cultivars and for Hybrid Seed Purity Testing (Microsatellite Marker를 이용한 멜론 시판품종의 품종식별과 F1 순도검정)

  • Microsatellite markers were used to identify 58 major commercial melon cultivars, and to assess hybrid seed purity of a melon breeding line known as '10H08'. A set of 412 microsatellite primer pairs were utilized for fingerprinting of the melon cultivars. Twenty-nine markers showed hyper-variability and could discriminate all cultivars on the basis of marker genotypes, representing the genetic variation within varietal groups. Cluster analysis based on Jaccard's distance coefficients using the UPGMA algorithm categorized 2 major groups, which were in accordance to morphological traits. The DNA bulks of female and male parents of breeding line '10H08' were tested with 29 primer pairs based on microsatellites to investigate purity testing of $F_1$ hybrid seeds, and 5 primer pairs exhibited polymorphism. One microsatellite primer pair (CMGAN12) produced unambiguous polymorphic bands among the parents. Among 192 seeds tested with CMGAN12, progeny possibly generated by self-pollination of the female parent were clearly distinguished from the hybrid progeny. These markers will be useful for fingerprinting melon cultivars and can help private seed companies to improve melon seed purity.
