Studies on the Fertilization of Pulsatilla koreana (할미꽃(Pulsatilla koreana)의 수정현상(受精現象)에 관(關)한 연구(硏究))

  • This experiment was carried out to investigate the fertilization and the size of mature female and male gametophytic parts of Pulsatilla koreana after artificial pollination. The size of pollen is $26.5{\mu}m$ at the time of anther dehiscence, that is, about $3{\sim}4$ days after pollination. Synergid nucleus, egg nucleus, and polar nucleus are 10.0, 15.0, and $32.5{\mu}m$ respectively at the time of completing egg apparatus formation, that is, about 2 days after pollination. Poller tubes germinate on stigma about 10 hours, passing lower part of style about 30 hours, penetrating into micropyle about 35 hours after pollination. Sperm nucleus penetrates into polar nucleus about 40 hours and egg cell about 48 hours after pollination. But, there seems to be different among the individuals. Multinuclei and multinucleoli are formed in egg cell, synergid, and polar nucleus about the time of fertilization. Proembryo is formed about 4 days, being changed to large globular form about $6{\sim}8$ days after pollination. Endosperm nuclei divide into free nuclei after fertilization and change to cotyledon in gymnosperm. There seems to be same phenomena in Pulsatilla koreana.

Environment of Fluvial Sedimentary Deposits and Palynological Occurrence in the Geochang Area (거창 지역 하성퇴적층 형성환경과 화분산상 연구)

  • Jeonjangri site of Geochang area is located in the Geochang Basin, and lies on the river terrace of upstream part of Hwang River. Fluvial deposits are well distributed at the northern and southern walls of trench 2(district 2) in the Jeonjangri archeological site. This study aims to interpret the occurrences of fluvial sedimentary deposits on the basis of grain size analysis and palynological analysis in the representative sections of Jeongjangri site. The sedimentary profile shows that the upper units are typified by paleosols with soil wedge formed at about $25,000{\sim}30,000yr$ B.P, and the lower units are characterized by reddish brown muddy sands, organic muds and sand/gravel downwards in the profile. Particularly palynological study on the organic muds of southern wall section showed a result that lower unit is dominant with grass vegetation, and upper unit with Alnus-Quercus-Pinus vegetation. The former is interpreted to be formed at $60,000{\sim}50,000yr$ B.P (stadial), while the latter at $80,000{\sim}70,000yr$ B.P. In general broad-leaved/coniferous mixed forests are mostly dominant in Jeongjangri site and the climate was presumed to be cool temperate at that time.

The Comparison of Volatile Organic Compounds (VOCs) Analysis and Tradescantia Micronucleus (Trad-MCN) Bioassay for Evaluation of Hazardous Materials in Chemical Workplace Field (화학공장 실내 작업장에서의 유해물질 평가를 위한 VOC 분석법과 자주달개비 미세핵 분석법의 비교)

  • This research examined the presence of hazardous materials in chemical workplace field using an integrated chemical/biological monitoring. Chemical workplace field air for volatile organic compounds (VOCs) analysis was collected using a collection tube packed with Tena.x TA adsorbent 400 mg. Workplace field air samples were analyzed by gas chromatography/mass spectrometry (GC/MS). Simultaneously, Tradescantia BNL 4430 clone was exposed in situ to monitor hazardous materials in chemical workplace field. GC/MS analysis showed the presence of various VOCs such as trichloroethylene, toluene, ethylbenzene, (m,p,o)-xylenes, styrene, 1,3,5-trimethylbenzene, and 1,2,4-trimethylbenzene. The results showed that in situ monitoring of VOCs with the Tradescantia-micronucleus (Trad-MCN) assay gave positive results in chemical workplace field and negative response at outdoor air. In conclusion, inhalation of these field air by workers may affect chronic demage to their health by inducing micronuclei formation in Tradescantia pollen mother cells. The combination of chemical/biological monitoring is very effective to evaluate hazardous materials in workplace field and can be alternatively used for screening hazardous materials.

Formation of Genetic Tumor and Characteristics of Teratoma Shoot from Tobacco Interspecific Reciprocal Hybrids (연초종간 상호교잡에 의한 Genetic Tumor의 유도 및 Teratoma Shoot의 특성)

  • Reciprocal interspecific hybrids between N. glauca(2n=24) and N. langsdorffii(2n=18) were obtained by intercrossing. One hundred percent of F$_1$ seeds was produced from intercrossing of N. glauca $\times$ N. langsdorffii, whereas the frequency of F$_1$ hybrid seed formation from N. langsdorffii $\times$ N. glauca was very low. However, all the hybrid seeds were germinated well and then grown to normal plantlets. All the plants of F$_1$ hybrids have chromosome number of interspecific hybrids (2n=21). From observation of morphological characteristic, the structure of petrol, leaf, flower, and the morphology of pollen have characteristics of F1 hybrid. Spontaneous tumors (genetic tumor) were formed from each F$_1$ hybrid; the genetic tumor arose at the reproductive phase when the maternal type of F$_1$ hybrid came from N. glauca, while the genetic tumor arose only after reproductive phase when the maternal type of F$_1$ hybrid came from N. langsdorffii. The genetic tumor actively proliferated on hormone-free medium and produced numerous teratoma shoots. In addition, normal leaf or stem explants of F$_1$ hybrid produced calli on hormone-free medium after 15 days of culture, the calli produced new numerous teratoma shoots after 30 days. The frequency of teratoma shoot formation from rnterspecific hybrid was higher in the N. glauca $\times$ N. langsdorffii than in the N. langsdorffii $\times$ N. glauca. Root development from the teratoma shoots was hardly obtained. Teratoma shoots without roots in vitro can form genetic tumor at the vegetative growth phase after tissue culture.

  • Anther is the major organ of flower in responsible to reproduction and outward appearance. From anther-specific cDNA library of Lilium Oriental Hybrid 'Acapulco', 2000 expressed sequence tags were selected randomly. Differential slot blot analysis with cDNA probes from the anther and leaf was used to get anther-expressed clone and 570 non-redundant ESTs were obtained and sequenced. Compared to the GenBank database using BLASTX algorithm, 191 clones showed significant similarity but others (66.5%) did not measured to known sequence. Functional categories according to gene ontology (GO) annotation included sequence representing a significant portion of protein in cell and cell part respectively. A transcriptional analysis at 7 different organs and developmental stage was performed using northern blot with thirty ESTs as putative anther specific gene. This report suggest that selection of anther expressed clone using differential slot blot was considered as very effective tool and our current study can provide fundamental information on the lily anther including pollen furthermore.

Changes of Lipids in Flint Corn by a Growth Process I. A Study on the Non-Polar Lipid (경립종 옥수수의 성장과정에 따른 지질변화 I. 비극성 지질에 대한 연구)

  • In order to investigate the changes of lipid ingredient and fatty acid content in flint corn by a growth process, total lipids from flint corn(Zea may l.) were extracted, purified and fractionated into three lipid classes (neutral lipid ; NL, glycolipid ; GL, phospolipid ; PL) by silicic acid column chromatography (SACC). Non-polar lipid ingredients and fatty acid contents were determined by thin layer chromatography (TLC) and gas chromatography (GC). And non-polar lipid ingredients were quantitatived by TLC scanner. The crude oil among proximare composition increased 2.74% to 4.43% and NL and PL were fractionated by SACC increased 48.5% to 86.02%, 1.01% to 2.73%, respectively. But GL decreased 50.4% to 11.25%. Neutral lipid ingredients identified monoglyceride, were 1.1~3.7%, 11.9~16.1%, 2.3~4.2%, 16.7~21.5% and 58.5~63.6%, respectively. The major fatty acids of NL were linoleic (48.9~61.0%), oleic(22.3~34.2%), palmitic (10.3~11.7%) acids and stearic, linolenic acids were also detected as minor components. The ratio of total unsaturated fatty acid to total saturated fatty acid presented a higher percentage of 5.87~6.34. The physico-chemical characteristics of flint corn lipids were presented as specific gravity presented 0.920, refractive index 1.468, saponification value 191.2, unsaponificable matter 0.97%, acid value 0.58 and iodine value 124.6, respectively.

Effects of Plant Growth Regulators on Haploid Production of Wheat (밀 반수체 생산에서 식물생장조절제의 영향)

  • The introduction of doubled haploid (DH) approach into breeding programs has reduced the times and population sizes required for the production of pure lines. We carried out the experiment for development on effective method of producing haploid in wheat. Emasculated spikelets of wheat were pollinated with maize pollen and cultured in the solution containing 40 g/L sucrose and 2,4-D, NAA, 2,4,5-T and dicamba 24 h after pollination, and then incubated until embryo rescue. Fourteen to sixteen days after pollination, the embryos are excised and cultured in half-strength MS basal medium supplemented with 20 g/L sucrose and 1 mg/L NAA. The type of plant growth regulators was found to be most significant in production of haploid plants. The application of synthetic auxins to pollinated florets, stimulates haploid embryo development to a stage where the embryos can be rescued onto nutrient media. The percentage of seeded florets was significantly affected by 100 mg/L 2,4-D, 150 mg/L 2,4,5-T and 50 mg/L dicamba. The percentage of embryos formed was significantly increased by treatment with 2,4-D and 2,4,5-T at 100 mg/L, and dicamba at 50 mg/L, but the treatments with 150 mg/L 2,4-D inhibited embryo development and plant regeneration. The optimum application time of plant growth regulators was 24 hrs after pollination.

'Glory Pink' a FA Intersectional Lily with Unspotted Pink Petals (연한 분홍색 FAA 아속간 교잡나리 'Glory Pink' 육성)

  • 'Glory Pink' an interspecific lily cultivar was released in 2007 at National Horticultural Research Institute (NHRI), Rural Development Administration (RDA), Suwon, Korea. The cross and ovule culture was made between FA interspecific hybrid 'FA97-30' (L. formolongi 'Raizan' ${\times}$ Asiatic lily 'A61'), a red colored cultivar, and Asiatic lily 'Sanzio', light red colored cultivar in 2001. Multiplication and bulbing, and characteristic tests were conducted from 2005 to 2006. Flowering time is around the first of July and the plant height is 131.7 cm. Flowers are upward-facing and red-pink (RHS 63C), with size of 11.3 cm. Petal length and width is 9.1 cm and 4.1 cm, respectively. Leaves are 13.8 cm long, 1.4 cm wide. The throat is dark green, and the stigma is light yellow and pollen is light brown. The weight and size of bulb is 51.1 g and 13.2 cm, respectively. For long-term storage, bulbs can be stored under -1 to -2 for year-round forcing.

A FA Intersectional Hybrid Lily 'Cherry Pink' with Deep Pink Petals (진분홍색 FA 아속간 잡종나리 '체리핑크' 육성)

  • A FA intersectional hybrid lily cultivar 'Cherry Pink' was developed in 2006 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) Korea. The cross and immature embryo rescue was conducted between female parents Lilium FA hybrid 'Supia' and male parents L. Asiatic hybrid 'A94-9' by cut style pollination method (CSM) at Suwon in 1999. The first selection was done and was tentatively named as 'FA01-17' in 2001. After in vitro multiplication, bulbing production of 'FA01-17' line, growth and flowering characteristic tests were conducted from 2002 to 2005. The evaluation of characteristics and consumer preferences were surveyed at a lily flower show of NIHHS at Taean in 2006. 'Cherry Pink' flowered in the middle of June and grew more than 120 cm stem in length. Flowers bloomed facing upward, a little spotted in petals and deep pink (RHS, RP27D). The pollen of 'Cherry Pink' was sterile. Year-round flowering can be done by storing the bulb under $-1.5^{\circ}C$ conditions. It was needed to control Botrytis disease in wet season.

Hybridity Verification of Progenies Obtained from Ovule Culture by Using RAPD Markers in Reciprocal Crosses of Alstroemeria (알스트로메리아 배주배양을 통하여 획득한 정역교배 자손의 혼종성 분석)

  • In this study, we performed ovule culture after reciprocal crosses of two Alstroemeria accessions and investigated genetic contribution of parents by using RAPD markers. The best method was half-ovule culture on MS medium supplemented with $60g{\cdot}L^{-1}$ sucrose and $2.2g{\cdot}L^{-1}$ gelrite at 14 days after pollination. Embryos began to germinate after 6 weeks of culture. The complete plantlets were formed after 4 months of culture. In eight progenies and two parental cultivars, 59 polymorphic bands were obtained out of 89 total bands by RAPD analysis using 7 primers. Eight $F_1$ progenies from the crosses between two accessions using reciprocal crosses showed 1:1 contribution of maternal and paternal parents. It is confirmed that $F_1$ progenies were obtained from parental accessions by using RAPD markers. We conclude this cross combination showed pre-fertilization barriers with incompatibility between stigma or style, and pollen because progeny number was different in each cross combination. Thereby, it warrants overcoming pre-fertilization barrier together with post-fertilization barrier in order to broaden the heterozygosity within progeny populations in Alstroemeria breeding program.
