Global fates of polychlorinated biphenyl(PCB) were investigated with a fugacity based multimedia transport and fate model, Globe-POP(persistent organic pollutant). The accumulation of PCB was directly affected by the emission patterns of PCB into the atmosphere and surface areas of environmental compartments. Partition coefficients and reaction rates also influenced on the accumulation patterns of PCB. The emission patterns of PCB in 10 climate zones were consistent for the past 70 years, while the contribution of PCB in high-latitude zones to the globe has increased by cold condensation. Considering the amounts of emission and accumulation of PCB, the North temperature zone is regarded as an important source and sink of PCB. Meanwhile, in spite of no significant sources, POPs accumulate in Antarctic environments mainly due to extremely low temperature. Finally we suggested that a global water balance accounting for snow/ice should be incorporated into multimedia environmental models for high-latitude zones and polar regions with the seasonal snow pack and/or permanent ice caps. The modified model will be useful to evaluate the influence of climate change on the fate of POPs.
This study presents spatial characteristics of cloud using satellite image in the extreme heavy snowfall of the Yeongdong region. 3 extreme heavy snowfall events in the Yeongdong region during the recent 12 years (2001 ~ 2012) are selected for which the fresh snow cover exceed 50 cm/day. Spatial characteristics (minimum brightness temperature; Tmin, cloud size, center of cloud-cell) of cloud are analyzed by tracking main cloud-cell related with these events. These characteristics are compared with radar precipitation in the Yeongdong region to investigate relationship between cloud and precipitation. The results are summarized as follows, selected extreme heavy snowfall events are associated with the isolated, well-developed, and small-scale convective cloud which is developing over the Yeongdong region or moving from over East Korea Bay to the Yeongdong region. During the period of main precipitation, cloud-cell Tmin is low ($-40{\sim}-50^{\circ}C$) and cloud area is small (17,000 ~ 40,000 $km^2$). Precipitation area (${\geq}$ 0.5 mm/hr) from radar also shows small and isolated shape (4,000 ~ 8,000 $km^2$). The locations of the cloud and precipitation are similar, but in there centers are located closely to the coast of the Yeongdong region. In all events the extreme heavy snowfall occur in the period a developed cloud-cell was moving into the coastal waters of the Yeongdong. However, it was found that developing stage of cloud and precipitation are not well matched each other in one of 3 events. Water vapor image shows that cloud-cell is developed on the northern edge of the dry(dark) region. Therefore, at the result analyzed from cloud and precipitation, selected extreme heavy snowfall events are associated with small-scale secondary cyclone or vortex, not explosive polar low. Detection and tracking small-scale cloud-cell in the real-time forecasting of the Yeongdong extreme heavy snowfall is important.
A study on the biological and chemical characteristics in the middle last Sea of Korea was carried out at 31 stations in October $11\~18$, 1995 on board the R/V Tam-Yang. The chlorophyll a concentration, new and regenerated production, and the vertical diffusion of nitrate from the thermocline structure were investigated. From the vertical distribution of chlorophyll a, subsurface maxima were observed near the thermorline at most stations including the frontal zone, except at the southern stations where the maximum chloropyll a concentration occurred at the surface, The nanophytoplankton was the most dominant fraction comprising $83.5\%$ of total phytoplankton cell numbers, but netphytoplankton were common at the southern stations where the dominant species were Rhizosolenia sp. Nitrogenous new production and regenerated productions were measured using the stable isotope $^{15}N$ nitrate and ammonia uptake method. The vertically integrated nitrogen production varied between 8.470 and $72.945\;mg\;N\;m^{-2}\;d^{-1}$. The f-ratio, which is the traction of new production from primary production, waried between 0.03 and 0.72, indicating that $3\%$ to $72\%$ of primary production was supported by the input of nutrients from below the euphotic zone and the rest are supported by ammonia recycled within the euphotic layer. This range of f-ratio encompasses from extremely oligotrophic to eutrophic area characteristics. The differences in productivity and f-ratio among stations were related to frontal structure and the bottom topography. The values were high near the frontal zone and low outside of it, and the station near Ulleng Island showed the highest f-ratio. Vertical diffusion coefficients were calculated from both the water column stability (Kz-1) of King and Devol's equation (1979) and new nitrogen requirement (Kz-2). The values of Kz-2 ($0.11\~0.55\;cm^2/s$) were relatively low compared to the values reported previously.
In Summer 2008, a multidisciplinary survey was conducted onboard R/V Haeyang 2000 to understand plankton response to the three distinct physico-chemical settings that developed in the Ulleung Basin of the East Sea. Baseline settings of hydrographic conditions included the presence of the thin (<20 m) Tsushima Surface Water (TSW) on top of the Tsushima Middle Water (TMW). It extends from the Korea Strait to $37^{\circ}N$ along the $130^{\circ}E$ and then turns offshore and encompasses the relatively saline (T>$26^{\circ}C$, S>33.7) Ulleung Warm Eddy surface water centered at $36.5^{\circ}N$ and $131^{\circ}E$. A relatively colder and saline water mass appeared off the southeastern coast of Korea. It was accompanied by higher nutrient and chlorophyll-a concentrations, suggesting a coastal upwelling. Most of the offshore surface waters support low phytoplankton biomass (0.3 mg chl-a $m^{-3}$). A much denser phytoplankton biomass (1-2.3 mg $m^{-3}$) accumulated at the subsurface layer between 20-50 m depth. The subsurface chlorophyll-a maximum (SCM) layer was closely related to the nutricline, suggesting an active growth of phytoplankton at depth. The SCM developed at shallow depth (20-30 m) near the coast and deepened offshore (50-60 m). A fucoxanthin/zeaxanthin ratio was high in coastal waters while it was low in offshore waters, which indicated that diatoms dominate coastal waters while cyanobacteria dominate offshore waters. The community structure and biomass of phytoplanktonare closely related to nitrogen availability. Zooplankton biomass was higher in the coastal region than in the offshore region while species richness showed an opposite trend. Zooplankton community structure retained a coastal/offshore contrast. These suggest that summer hydrography is a stable structure, lasting long enough to allow a hydrography-specific plankton community to evolve.
Recently, Psychrobacter sp. ArcL13 strain showing the extracellular lipase activity was isolated from the Chuckchi Sea of the Arctic Ocean. However, due to the low expression levels of the enzyme in the natural strain, the production of recombinant lipase is crucial for various applications. Identification of the gene for the enzyme is prerequisite for the production of the recombinant protein. Therefore, in the present study, a novel lipase gene (ArcL13-Lip) was isolated from Psychrobacter sp. ArcL13 strain by gene prospecting using PCR, and its complete nucleotide sequence was determined. Sequence analysis showed that ArcL13-Lip has high amino acid sequence similarity to lipases from bacteria of some Psychrobacter genus (84-90%) despite low nucleotide sequence similarity. The lipase gene was cloned into the bacterial expression plasmid and expressed in E. coli. SDS-PAGE analysis of the cells showed that ArcL13-Lip was expressed as inclusion bodies with a molecular mass of about 35 kDa. Refolding was achieved by diluting the unfolded protein into refolding buffers containing various additives, and the highest refolding efficiency was seen in the glucose-containing buffer. Refolded ArcL13-Lip showed high hydrolytic activity toward p-nitrophenyl caprylate and p-nitrophenyl decanoate among different p-nitrophenyl esters. Recombinant ArcL13-Lip displayed maximal activity at $40^{\circ}C$ and pH 8.0 with p-nitrophenyl caprylate as a substrate. Activity assays performed at various temperatures showed that ArcL13-Lip is a cold-active lipase with about 40% and 73% of enzymatic activity at $10^{\circ}C$ and $20^{\circ}C$, respectively, compared to its maximal activity at $40^{\circ}C$.
The studies were carried out to disclose the physical and chemical properties of sericin fraction obtained from silk cocoon shells and its characteristics of swelling and solubility. The following results were obtained. 1. The physical and chemical properties of sericin fraction. 1) In contrast to the easy water soluble sericin, the hard soluble sericin contains fewer amino acids include of polar side radical while the hard soluble amino acid sach as alanine and leucine were detected. 2) The easy soluble amino acids were found mainly on the outer part of the fibroin, but the hard soluble amino acids were located in the near parts to the fibroin. 3) The swelling and solubility of the sericin could be hardly assayed by the analysis of the amino acid composition, and could be considered to tee closely related to the compound of the sericin crystal and secondary structure. 4) The X-ray patterns of the cocoon filament were ring shape, but they disappeared by the degumming treatment. 5) The sericin of tussah silkworm (A. pernyi), showed stronger circular patterns in the meridian than the regular silkworm (Bombyx mori). 6) There was no pattern difference between Fraction A and B. 7) X-ray diffraction patterns of the Sericin 1, ll and 111 were similar except interference of 8.85A (side chain spacing). 8) The amino acids above 150 in molecular weight such as Cys. Tyr. Phe. His. and Arg. were not found quantitatively by the 60 minutes-hydrolysis (6N-HCI). 9) The X-ray Pattern of 4.6A had a tendency to disappear with hot-water, ether, and alcohol treatment. 10) The partial hydrolysis of sericin showed a cirucular interference (2A) on the meridian. 11) The sericin pellet after hydrolysis was considered to be peptides composed with specific amino acids. 12) The decomposing temperature of Sericin 111 was higher than that of Sericin I and II. 13) Thermogram of the inner portioned sericin of the cocoon shell had double endothermic peaks at 165$^{\circ}C$, and 245$^{\circ}C$, and its decomposing temperature was higher than that of other portioned sericin. 14) The infrared spectroscopic properties among sericin I, II, III and sericin extracted from each layer portion of the cocoon shell were similar. II. The characteristics of seriein swelling and solubility related with silk processing. 1) Fifteen minutes was required to dehydrate the free moisture of cocoon shells with centrifugal force controlled at 13${\times}$10$^4$ dyne/g at 3,000 R.P.M. B) It took 30 minutes for the sericin to show positive reaction with the Folin-Ciocaltue reagent at room temperature. 3) The measurable wave length of the visible radiation was 500-750m${\mu}$, and the highest absorbance was observed at the wave length of 650m${\mu}$. 4) The colorimetric analysis should be conducted at 650mu for low concentration (10$\mu\textrm{g}$/$m\ell$), and at 500m${\mu}$ for the higher concentration to obtain an exact analysis. 5) The absorbing curves of sericin and egg albumin at different wave lengths were similar, but the absorbance of the former was slightly higher than that of the latter. 6) The quantity of the sericin measured by the colorimetric analysis, turned out to be less than by the Kjeldahl method. 7) Both temperature and duration in the cocoon cooking process has much effect on the swelling and solubility of the cocoon shells, but the temperature was more influential than the duration of the treatment. 8) The factorial relation between the temperature and the duration of treatment of the cocoon cooking to check for siricin swelling and solubility showed that the treatment duration should be gradually increased to reach optimum swelling and solubility of sericin with low temperature(70$^{\circ}C$) . High temperature, however, showed more sharp increase. 9) The more increased temperature in the drying of fresh cocoons, the less the sericin swelling and solubility were obtained. 10) In a specific cooking duration, the heavier the cocoon shell is, the less the swelling and solubility were obtained. 11) It was considered that there are differences in swelling or solubility between the filaments of each cocoon layer. 12) Sericin swelling or solubility in the cocoon filament was decreased by the wax extraction.. 13) The ionic surface active agent accelerated the swelling and solubility of the sericin at the range of pH 6-7. 14) In the same conditions as above, the cation agent was absorbed into the sericin. 15) In case of the increase of Ca ang Mg in the reeling water, its pH value drifted toward the acidity. 16) A buffering action was observed between the sericin and the water hardness constituents in the reeling water. 17) The effect of calcium on the swelling and solubility of the sericin was more moderate than that of magnecium. 18) The solute of the water hardness constituents increased the electric conductivity in the reeling water.
To understand the scales of the spatial distribution and temporal duration of the subsurface chlorophyll-a maximum (SCM) observed in the Ulleung Basin of the East Sea, we analyzed physical and chemical data collected during the East Asian Seas Time-series-I (EAST-I) program. The SCM layer occurred at several observation lines from the Korea Strait to $37.9^{\circ}N$ in the Ulleung Basin during August of 2008 and 2011. At each observation line, the SCM layer extended from the coast to about 200 km off the coast. The SCM layer was observed between 30 and 40 m depth in the Ulleung Basin as well as in the northwestern Japan Basin along $132.3^{\circ}E$ from $38^{\circ}N$ to $42.3^{\circ}N$ during July 2009, and was observed around 50 m depth in the northeastern Japan Basin ($135-140^{\circ}E$ and $40-45^{\circ}N$) during July 2010. From these observed features, we hypothesize that the SCM layer observed in the Ulleung Basin may exist in most of the East Sea and may last for at least half-year (from the early May to late October). The nutrient supply mechanism for prolonged the SCM layer in the East Sea was not known, but it may be closely related to the horizontal advection of the nutrient rich and low oxygen waters observed in the Korea Strait between a 50 m depth to near the bottom. The prolonged development of the SCM layer in the Ulleung Basin may result in high primary production and would also be responsible for the high organic carbon content observed in the surface sediment of the region.
The paleoceanography since 14 ka was reconstructed based on the planktic foraminiferal assemblages of core sediments from the outer shelf of the Korea Strait. Planktic foraminifera in the core sediments can be divided into four assemblages: A, B, C, and D. Assemblage A consists mainly of Globigerinoides ruber group and Globigerinoides conglobatus with low abundance (less than 10%), indicating the tropical-subtropical water mass. Assemblage B is composed of Pulleniatina obliquiloculata and Neogloboquadrina dutertrei, the indicator of Kuroshio Current, and shows the aspect of the inflow of the Tsushima Current into the Korea Strait. Assemblage C yields polar-subpolar species, mainly Neogloboquadrina incompta and N. pachyderma. It decreases upward of the core. Assemblage D contains coastal water species such as Globigerina bulloides and G. quinqueloba. It is abundant in the lower to middle region of the core. From the analysis of distributions of each assemblage and the result of age datings in the core, it is suggested that the Korea Strait played a role of channelling the East China Sea and the East Sea after the LGM (ca. 14 ka). During this time, the coastal water, affected by fresh waters originated from the river systems of China and/ or the Korean Peninsula, flourished around the Korea Strait and theses coastal water might entered to the East Sea. Around 8.5 ka, the effect of the Tsushima Current started to strengthen in this region, and the present current system seems to be formed at about $7{\sim}6ka$.
This study was to evaluate the in vitro survival of bovine enucleated MII (eMII) oocytes according to minimum volume cooling (MVC) freezing method and activation timing, and their in vitro development after somatic cell nuclear transfer (SONT). in vitro matured bovine oocytes for 20 h were stained with 5 $\mu\textrm{g}$/$m\ell$ Hoechst, and their 1st polar body and MII plate were removed by enucleation micropipette under UV filter. Also, eMII oocytes were subjected to activation after (group II) and before (group III) vitrification in 5 ${\mu}{\textrm}{m}$ ionomycin added CRlaa medium for 5 min. For vitrification, eMll oocytes were pretreated with EG10 for 5 min, exposed to EG30 for 30 sec and then directly plunged into L$N_2$. Thawing was taken by 4-step procedures at 37$^{\circ}C$. Survived eMII oocytes were subjected to SONT with cultured adult bovine ear cells. Reconstructed oocytes were cultured in 10 $\mu\textrm{g}$/$m\ell$ of cycloheximide and 2.5 $\mu\textrm{g}$/$m\ell$ of cytochalasin D added CRlaa medium for 1 h, and then in 10 $\mu\textrm{g}$/$m\ell$ of cycloheximide added CRlaa medium for 4 h. Subsequently, the reconstructed oocytes were incubated for 2 days and cleaved embryos were further cultured on cumulus-cell monolayer drop in CRlaa medium for 6 days. Survival rates of bovine vitrified-thawed eMII oocytes in group II (activation after vitrification and thawing) and III (activation before vitrification) were 81.0% and 84.9%, respectively. Fusion rates of cytoplasts and oocytes in group II and III were 69.0% and 70.0%, respectively, and their results were not different with non-frozen NT group (control, 75.2%). Although their cleaved rates (53.4% and 58.4%) were not different, cytoplasmic fragment rate in group II (32.8%) was significantly higher than that in group III (15.6%)(P<0.05). Also, subsequent development rate into >morula in group II (8.6%) was low than that in group III(15.6%). However, in vitro development rate in group III was not different with that in control (24.8%). This result suggested that MVC method was appropriate freezing method for the bovine eMII oocytes and vitrified eMII oocytes after pre-activation could support in vitro embryonic development after SONT as equally well as fresh oocytes.

This study was conducted to examine the effects of demecolcine-assisted enucleation and recipient cell cycle stage on the development of bovine somatic cell nuclear transfer (NT) embryos. In vitro cultured oocytes for $16\~20$ h were classified by first polar body (1st PB) extrusion and cell cycle stage (MI and MII) and treated $0.4\;{\mu}L/mL$ demecolcine for 40 min before enucleation. Enucleated oocytes were fused electrically with bovine ear skin cells, activated by Ca-ionophore+DMAP, and cultured in vitro. Most of eggs ($86.2\%$) treated with demecolcine protruded a chromosome mass and enucleated efficiently ($98.8\%$, (P<0.05). Demecolcine did not have a deteriorative effect on the development of NT embryos. Developmental rate of NT embryos reconstituted with oocytes extruded 1st PB significantly higher than that of NT embryos produced by oocytes without 1st PB ($18.2\%\;vs.\;4.6\%\cdot$, P<0.05). Cleavage and blastocyst formation rate of embryos reconstituted with MI oocytes ($69.4\%\;and\;5.9\%$, respectively) were significantly lower than those of embryos reconstituted with MII oocytes ($96.7\%\;and\;23.9\%$, respectively, P<0.05). From the present result, it is suggested that domecolcine is useful for the enucleation of recipient oocytes in bovine NT procedures, and MII oocytes rather than MI oocytes are more appropriate for recipient cytoplasm. Although, the potential to develop into blastocysts of NT embryos produced by 1st PB-nonextruded and MI oocytes was very low, these oorytes could be used for NT.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
