Simple Electrochemical Immunosensor for the Detection of Hippuric Acid on the Screen-printed Carbon Electrode Modified Gold Nanoparticles

  • This paper describes an electrochemical immunosensor for simple, fast and quantitative detection of a urinary hippuric acid which is one of major biological indicator in toluene-exposed humans. The feature of this electrochemical system for immunoassay of hippuric acid is based on the direct conjugation of ferrocene to a hippuric acid. With the competition between the ferrocene-hippuric acid complex and hippuric acid for binding to the anti-hippuric acid monoclonal antibody coated onto gold nanoparticles, the electrical signals are turned out to be proportional to urinary hippuric acid in the range of 0.01-10 mg/mL, which is enough to be used for the point-of-care. The proposed electrochemical method could extend its applications to detect a wide range of different small molecules of antigens in the health care area.

일부지역 장애인구강진료센터 이용 만족도에 미치는 요인 (Factors affecting the satisfaction of persons with disabilities at community oral care centers)

  • Background: To identify the factors that affect the current status and satisfaction of people with disabilities at community oral care centers. Methods: A structured self-administered survey, including five questions on facility environment, five on usage procedure, four on medical skill, four on care cost, three on friendliness, and three on satisfaction, was administered to 218 residents of the G-disabled community care center. It comprised a Likert 5-point scale (strongly agree, 5 points; agree, 4 points; moderate, 3 points; disagree, 2 points; not at all, 1 point). The reliability of the measurement tool was 0.932 for Cronbach's α. Results: The evaluation of community oral care centers for the disabled showed that the environment was hygienic (4.42±0.73), reservation system was well maintained (4.18±0.95), and the dentist-in-charge was satisfied with the treatment (4.37±0.62). The participants agreed that the details were sufficiently explained (4.29±0.71). However, in terms of medical expenses, the score of "have fully heard the explanation of medical expenses and reductions" was 3.88±0.92. The factors affecting satisfaction were sex, final educational background in the facility environment, usage procedure, and medical skill. Conclusion: To increase the satisfaction of people with disabilities at community oral care centers, it is necessary to establish a facility environment and service according to the patients' need and increase the reduction or exemption benefits between different treatment cost categories. Oral health management policies for the disabled should be developed based on these factors, so that the oral care of vulnerable groups in blind spots can be maintained.

Electrochemical Method for Detecting Hippuric Acid Using Osmium-antigen Conjugate on the Gold Nanoparticles Modified Screen-printed Carbon Electrodes

  • This paper describes an electrochemical immunoassay for simple, fast and quantitative detection of a urinary hippuric acid which is one of major biological indicator in toluene-exposed humans. The electrochemical system of immunoassay was based on the directly osmium complex conjugated with hippuric acid. With the competition between free hippuric acid (HA) and the osmium-hippuric acid conjugate (Os-HA) to bind with antibody hippuric acid (Anti-HA) coated onto gold nanoparticles, the electrical signals were proportional to urinary hippuric acid (HA) in the range of 0.01-5 mg/mL which is enough range to be used for in-field or point-of-care (POC) diagnosis. The proposed electrochemical method can be extended to the applications to detect a wide range of different small molecules in the field of health care.

임상간호사의 건강정보이해능력 역량이 환자중심 간호에 미치는 영향 (The influence of health literacy competencies on patient-centered care among clinical nurses)

  • Purpose: The aim of this study was to identify the relationships between health literacy competencies and patient-centered care among clinical nurses. Methods: The participants of this study were 254 nurses working in two hospitals in the D region. The data were collected from July to August 2020. The health literacy competencies for registered nurses scale and individualized care scale were utilized. Descriptive statistics, independent t-test, ANOVA, Pearson's correlation coefficient and multiple regression analysis were used for data analysis. Results: The mean of health literacy competencies was 3.16±0.31 points on a four-point scale, and the average of patient-centered care was 3.69±0.50 points on a five-point scale. Regarding the nurses' general characteristics, patient-centered care showed significant differences according to age (F=4.68, p=.010), marital status (t=-2.38, p=.018), religion (F=3.03, p=.030), total clinical experience (F=2.94, p=.021) and prior health literacy knowledge (t=3.20, p=.002). As a result of a hierarchical multiple regression analysis, health literacy competencies (β=.63) were found to significantly influence patient-centered care. The explanatory power of the model was 41.0% (F=25.58, p<.001). Conclusion: The study suggests that nurse's health literacy competencies should be developed in order to improve patient-centered care. Nursing education should include an emphasis on integrating health literacy into the nursing school curriculum.

형광 영상 기반 CD4 신속 검사법 개발을 위한 항원-항체 반응 조건 정량화 (Quantitation of Antigen-Antibody Reaction Condition for Development of Fluorescence Image-based CD4 Rapid Test)

  • CD4+ T-cell count determines the effectiveness for antiretroviral therapy (ART) in patients with human immunodeficiency virus (HIV). Although ART slows the progression of HIV to AIDS, rapid counting of CD4+ T lymphocytes with a drop of patient's blood sample is urgently needed to ensure timely ART treatment in rural areas. Recently point-of-care CD4 testing devices have been developed by using non-flow based imaging cytometer incorporated with a sample cartridge where CD4+ T cells are reacted with fluorescently tagged specific antibodies. Here we conducted an experimental study using a conventional fluorescence microscope-based imaging system to quantitate the interaction of CD4 antibodies with CD4+ T cells at different reaction conditions. We demonstrated that a fast and affordable point-of-care CD4 test is feasible with a far less amount of antibodies and a shorter incubation time compared with a conventional sample preparation protocol for flow cytometry. We also proposed a general method to evaluate and compare the detection limit across different CD4 counting platforms by using fluorescently labelled microbeads for intensity calibration.

Adaptation of Evidence-based Surgical Wound Care Algorithm

  • Purpose: This study was designed to adapt a surgical wound care algorithm that is used to provide evidence-based surgical wound care in a critical care unit. Methods: This study used, the 'ADAPTE process', an international clinical practice guideline development method. The -'Bonnie Sue wound care algorithm' - was used as a draft for the new algorithm. A content validity index (CVI) targeting 135 critical care nurses was conducted. A 5-point Likert scale was applied to the CVI test using a statistical criterion of .75. Results: A surgical wound care algorithm comprised 9 components: wound assessment, infection control, necrotic tissue management, wound classification by exudates and depths, dressing selection, consideration of systemic factors, wound expected outcome, reevaluate non-healing wounds, and special treatment for non-healing wounds. All of the CVI tests were ${\leq}$.75. Compared to existing wound care guidelines, the new wound care algorithm provides precise wound assessment, reliabilities of wound care, expands applicability of wound care to critically ill patients, and provides evidence and strength of recommendations. Conclusion: The new surgical wound care algorithm will contribute to the advancement of evidence-based nursing care, and its use is expected as a nursing intervention in critical care.

현장진단용 단백질 칩 사출금형기술 (Injection Mold Technology of Protein Chip for Point-of-Care)

  • A multi-cavity injection mold system of protein chip for point-of-care with cavity temperature and pressure sensors was proposed in this work. In advance of manufacturing for the multi-cavity injection mold system, a single cavity injection mold system to mold protein chip was considered. Injection molding analysis for the presented system was performed to optimize the process of the molding and suggest guides to design. On the basis of the results for the single cavity system, a multi-cavity injection mold system for protein chip was analyzed, designed and manufactured with cavity temperature and pressure sensors. Results of balanced filling for protein chip models were obtained from the presented mold system.

  • This paper presents a simple and reliable one-touch type multi-immunosensing lab-on-a-chip (LOC) detecting antibodies as multi-disease markers using electrochemical method suitable for a portable point-of-care system (POCS). The multi-stacked LOC consists of a PDMS space layer for liquids loading, a PDMS valve layer with 50 im in height for the membrane, a PDMS channel layer for the fluid paths, and a glass layer for multi electrodes. For the disposable immunoassay which needs sequential flow control of sample and buffer liquids according to the designed strategies, reliable and easy-controlled on-chip operation mechanisms without any electric power are necessary. The driving forces of sequential liquids transfer are the capillary attraction force and the pneumatic pressure generated by air bladder push. These passive fluid transport mechanisms are suitable for single-use LOC module. Prior to the application of detection of the antibody as a disease marker, the model experiments were performed with anti-DNP antibody and anti-biotin antibody as target analytes. The flow test results demonstrate that we can control the fluid flow easily by using the capillary stop valve and the PDMS check valves. By the model tests, we confirmed that the proposed LOC is easily applicable to the bioanalytic immunosensors using bioelectrocatalysis.

Development of reverse-transcription loop-mediated isothermal amplification assays for point-of-care testing of human influenza virus subtypes H1N1 and H3N2

  • Influenza A virus (IAV) is the most widespread pathogen causing human respiratory infections. Although polymerase chain reaction (PCR)-based methods are currently the most commonly used tools for IAV detection, PCR is not ideal for point-of-care testing. In this study, we aimed to develop a more rapid and sensitive method than PCR-based tools to detect IAV using loop-mediated isothermal amplification (LAMP) technology. We designed reverse-transcriptional (RT)-LAMP primers targeting the hemagglutinin gene. RNAs from reference H1N1 and H3N2 showed specific RT-LAMP signals with the designed primers. We optimized the reaction conditions and developed universal reaction conditions for both LAMP assays. Under these conditions, the detection limit was 50 copies for both RT-LAMP assays. There was no non-specific signal to 19 non-IAV respiratory viruses, such as influenza B virus, coronaviruses, and respiratory syncytial viruses. Regarding the reaction time, a positive signal was detected within 25 min after starting the reaction. In conclusion, our RT-LAMP assay has high sensitivity and specificity for the detection of the H1 and H3 subtypes, making it suitable for point-of-care IAV testing.

  • 본 논문은 이동 단말(mobile Node)의 핸드오프 발생시 끊김없는 핸드오프(seamless handoff)를 지원하기 위한 기법을 제안하고자 한다. 신호세기의 감소로 인한 핸드오프가 발생 가능한 상태일 때 이동단말은 기존의 CoA(Care-of Address) 외에 감지되는 신호를 통한 새로운 nCoA들을 만들어 자신의 영역을 담당하는 MAP(Mobility Anchor Point)에게 이들 nCoA에 대한 일시적인 바인딩 업데이트 tempBU(temporary Binding Update) 메시지를 보낸다. MAP은 전달받은 새로운 nCoA 주소를 포함한 바인딩 업데이트 메시지를 받아서 기존의 바인딩 정보 외에 새로운 바인딩 정보를 일시적으로 바인딩 캐쉬에 저장한다. 결국 이동단말이 새로운 액세스 라우터(nAR) 영역에 진입하여 확정된 바인딩 업데이트 메시지를 보낼 때까지 멀티캐스팅을 함으로써 끊김없는 핸드오프를 보장하는 기법을 제시한다.
