  • Pine(Pinus densiflora Sieb et Zucc.) is one of rhe popular plant drugs which has been used as a medicine in Asia. To investigate the effect of pine needle extract (PNE) on oxygen radicals and their scavenger enzymes in brain membranes of Sprague- Dawley (SD), make SD rats were fed basic diets(control group), and experimental diets (PNE group) with 0.5 and 1.0% of PNE 6 weeks. Mitochondrial hydroxyl radical levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 30% and 25%, respectively, and microsomal hydrogen peroxide levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 15% compared with control group. Cytosolic superoxide rdical levels in 1.0%-PNE group were significantly inhibited to 20% compared with control group. Lipid peroxide(LPO) levels in brain mitochondria of 0.5%-PNE and 1.0%-PNE groups were significantly lower(25% and 35%) than that in control group. Mn-superoxide disumtase (SOD) activities in brain of 0.5%-PNE and 1.0%-PNE groups were significantly higher(18% and 12%) than those in control groups, but Cu,Zn-SOD activities in brain of 0.5%-PNE were significantly activated to 15% compared with control group. Glutathione peroxidase(GSHPx) activities in brain of 1.5%-PNE and 1.0% PNE groups were significantly higher(14% and 12%) than those in control group. These results suggest that more beneficial effects such as inhibition of oxygen radicals and lipid peroxide(LPO). and oncreases of scavenger enzymes in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE)

  • To investigate the effect of pine needle extract (PNE) on membrane fluidirt and neurotransmiter-related enzymes in brain of Spragu-Dawley(SD), male SD rats were fed basic diets (control group), and experimantal diets (PNE group)with 0.5% and 0.1% fo PNE for 6 weeks. pine (pinus tabulaeformis C$_{ARR}$ is one of the popular plant drugs which has used as a medicine in Asia. Cholesterol levels in brain mitochondria of 0.5%-PNE and 0.1%-PNDE groups were significantly decreased in 15% and 25%, respectively, compared with control group, but cholesterol levels in brain microsomes of these PNE groups howed almost no change compared with control group. Lipofuscin accumulations in brain membranes of 0.5%-PNE and 0.1%-PNE groups were sgnificantly inhibited in 18% and 21%, respectively, compared with control group. Brain memberance fluidity was also activated in 50% and 100% by the administration of 0.5%-PNE and 0.1%-PNE. higher acetylcholinesterase(15% and25%) and lower monoamine oxidase B (25% and 15%0 activities were effectively modulated by the administration of 0.5%-PNE and 0.1%-PNDE. These results suggest that more beneficial effects such as inhibition of cholesterol and lipofuscin, increase of membrane fluidity, higher acetylcholinesterase and lower monoamone oxidase activities in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE).

  • This study was edsigne to investgate the effect of pine needle extract (PNE) on lipid adn oxygen radical metabolisms in serum of Sprague-Dawly (SD). Pine(Pinus densiflora Sieb et Zucc.) is one of the popular plant drugs which gas been used as a medicine in Asia. Male SD rats were fed basic (control group) and experimental diets(RGE group) with 0.5 and 1.0% of PNE for 6 weeks. Body weights gain in 0.5-PNE and 1.0-PNE groups were slightly lower than that in control group, but ther is no significant differences brtween these groups. Total energy intake, feed and gross efficiencies showed almost no change in these groups. Total cholesterol and triglyceride (TG) levels in serumof SD rats in .50-PNE and 0.1-PNE froups sho9wed almost no change colpared with control group. Serum LDL-cholesterol levels significantly decreased (12%) in 0.5-PNE group, while serum HDL-cholesterol levels significantly increased(14%) in 1.0-PNE group compared with control group. Malondialdehyde (MDA) and lipid peroxide (LPO) levels in 50-PNE and 0.1-PNE groups were significantly lower (17 and 13%, 24% and 12%, respectively) than those in control group. Superoxide disumutase (SOD) activity in 50-PNE and 0.1-PNE groups significantly increased about 30 to 15% compared with control group. There results suggest that lower LDL-cholesterol and LPO levels, higher HDL-cholsterol level, and higher SOD activity in serum of rats may be effectively modulated by administration of pine needle extract(PNE).

Application of hybrid material, modified sericite and pine needle extract, for blue-green algae removal in the lake

  • The present study assessed the efficient removal of nutrients and Chlorophyll-a (Chl-a) by using methyl esterified sericite (MES) and pine needle extracts (PNE), a low cost and abundant green hybrid material from nature. For this purpose, the optimal conditions were investigated, such as the pH, temperature, MES and PNE ratio, and MES-PNE dose. In addition, a Microcystis aeruginosa control using MES-PNE was also analyzed with various inhibition models. The removal of the nutrient and Chl-a onto MES-PNE was optimized for over 95% removal as follows: 2-2.5 for the MES-PNE ratio, 7-8 pH and a $22-25^{\circ}C$ temperature. In this respect, approximately 1.52-2.20 g/L of MES-PNE was required to remove each 1 g of dry weight/L of Chl-a. Total phosphorus (TP) has a greater influence on the increase in Chl-a than total nitrogen (TN) according to the correlation between TN, TP and Chl-a. Moreover, the Luong model was the best model for fitting the biodegradation kinetics data from Chl-a on MES-PNE from lake water. The novel hybrid material MES-PNE was very effective at removing TN, TP and Chl-a from the lake and can be applied in the field.

Removal of Microcystis aeruginosa using Pine Needle Extracts (솔잎추출액을 이용한 Microcystis aeruginosa 제거 연구)

  • Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of pine needle extract (PNE) to control nuisance algae by allelopathic inhibition will be environmentally friendly and promising. PNE removed successfully upto 98% of M. aeruginosa at the following optimal conditions: pH 7, $25^{\circ}C$ of temperature, 100 rpm of mixing rate, 5 min of mixing time. These results was indicated that the amount of 1 g/L PNE was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 0.5 g/L of initial concentration of PNE. Different substrate inhibition models were fitted to the kinetic data and found the Luong model was best. The model predicted kinetic parameters were in agreement with the experimental findings. The natural extract, PNE, can be a promising inhibition due to its high efficiency and low dose requirements.

  • Pinus densiflora belongs to the Pinaceae family which has been widely used for health promoting purposes as folk medicine or as a food. Various curative effects of different parts of the pine have been reported including as a remedy for carcinoma. We examined the effects of pine needle water extracts (PNE) on macrophage function using peritoneal macrophage, pre-osteoclast bone macrophage (Raw 264.7 cell) and brain macrophage (C6 microglia). When peritoneal macrophages were treated with various concentrations of PNE ($1{\sim}100{\mu}g/mL$) for 24 hours, phagocytic activity was significantly increased, whereas it had no effect on tumoricidal activity and NO production. However, the treatment of Raw 264.7 with PNE resulted in the enhancement of NO production at high concentration ($100{\mu}g/mL$). Furthermore, the treatment of C6 with PNE increased the production of NO in a concentration-dependent manner, whereas PNE suppressed NO production in $LPS/IFN-{\gamma}-stimulated$ microglia. These results suggest that PNE has differential immunomodulatory effects on macrophages.

  • These studies were designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc.) needle extract (PNE) on oxygen radicals and their scavenger enzymes in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. There were no significant differences in hydroxyl radical (·OH) formations of liver mitochondria and microsomes in 0.5%-PNE group, while ·OH formations were significantly decreased (10% and 18%, respectively) in liver mitochondria and microsomes of 1.0%-PNE group compared with control group. Microsomal hydrogen peroxides and cytosolic superoxide radicals were remarkably decreased (20% and 20∼25%, respectively) in 0.5% and 1.0%-PNE groups compared with control group. Mn-SOD activities in mitochondria were significantly increased about 10% in 1.0%-PNE group, while Mn-SOD activities in mocrosomes were remarkably increased (16∼20%) in 0.5% and 1.0%-PNE groups compared with control group. There were no significant differences in Cu, Zn-SOD activities of liver cytosol in 0.5% and 1.0%-PNE groups, while glutathione peroxidase (GSHPx) and catalase (CAT) activities were significantly decreased (28∼30% and 15∼30%, respectively) in liver cytosols of 0.5% and 1.0%-PNE groups compared with control group. These results suggest that these PNE may play a effective role in a attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

  • This study was designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc) needle extract (PNE) on membrane fluidity and oxidative stress in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. Administrations of 0.5% and 1.0%-PNE resulted in a marked decreases (15∼25% and 23∼26%, respectively) in cholesterol accumulations of liver mitochondria and microsomes compared with control group. Membrane fluidities were significantly increased (15∼25%) in liver microsomes of 0.5% and 1.0%-PNE groups compared with control group. Formations of basal and induced oxygen radicals (BOR and IOR) in liver mitochondria were significantly inhibited (11∼12% and 10∼15%, respectively) by administrations of 0.5% and 1.0%-PNE compared with control group. Lipid peroxide (LPO) levels were remarkbly decreased about 20% in liver mitochondria and microsomes of 0.5% and 1.0%-PNE groups compared with control group. Oxidized protein levels calculated with carbonyl group were significantly decreased about 15% in liver mitochondria of 1.0%-PNE group compared with control group. These results suggest that PNE may play a effective role in a attenuating a oxidative stress and increasing a membrane fluidity.

Effect of Pine Needle and Green Tea Extracts on the Survival of Pathogenic Bacteria (솔잎과 녹차 추출물이 식중독세균의 생존에 미치는 영향)

  • The sensitivity of various pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O157:H7 and Salmonella typhimurium) to the pine needle and green tea extracts was tested. Water extract of pine needle(PNW), 70% ethanol extract of pine needle(PNE), water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activty. Tryptic soy broth(TSB) containing 0∼2%(w/v) of pine needle and green tea extracts were inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacterium and incubated at 35$\^{C}$ for 24 hours. The standard plate count method was used to measure the inhibitory effect of the extracts. Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were derived from the survival curves of pathogenic bacteria. Antibacterial activities of the pine needle and green tea extracts were compared with that of sodium benzoate, a preservative, by clear zone test. L. monocytogenes, S. aureus and A. hydrophila were completely inhibited at 0.4∼1.6% level while E. coli and S. typhimurium were very resistant to the pine needle extracts. Green tea extracts completely inhibited all strains tested at 0.2∼1.0% level and bactercidal to all strains except L. monocytogenes at 0.5∼2.0% level. Antibacterial activities of pine needle and green tea extracts were stronger than that of sodium benzoate. The order of antibacterial activities of pine needle and green tea extracts to the pathogenic bacteria was GTE > GTW > PNE > PNW. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.

Antioxidative and Nitrite Scavenging Activities of Mugwort and Pine Needle Extracts (쑥과 솔잎의 항산화작용 및 아질산염 소거작용)

  • Mugwort and pine needle were extracted with water and 70% ethanol. Electron donating ability(EDA) of extracts were ranged from 50% to 57% in mugwort water extract(MGW) and ranged from 51% to 64% in mugwort ethanol extract(MGE) at 300-1,000ppm. EDA of extracts were ranged from 52% to 60% in pine needle water extract(PNW) and ranged from 68% 71% in pine needle ethanol extract(PNE) at 100-500ppm. EDA of PNW was 70% and that of PNE was 77% at 1,00(ppm. Nitrite scavenging ability(NSA) of extracts measured at various pH(1.2, 3.0, 4.2, 6.0) was the highest in all extracts at pH 1.2 and decreased with increasing pH, suggesting it is pH dependent. NSA of mugwort extracts at 1,000ppm, water extract was 37% and ethanol extract was 27% at pH 1.2. NSA of pine needle extracts at 1,000ppm, water extract was 65%and ethanol extract was 53% at pH 1.2. EDA and NSA of pine needle extracts were higher than mugwort in both of water and ethanol extract. EDA of ethanol extracts were higher than water extracts while NSA of water extracts were higher than ethanol extracts in both of mugwort and pine needle.
