This experiment were carried out to investigate the effect of the rice bran added into spawn various amounts on the cultivation. Our results show that 10 to 20 percent addition of the rice bran as a supplement results in a good mycelial growth and density. However we didn't find a significant variance among the different species of oyster mushroom using poplar sawdust as a medium. When it inoculated spawn with various amounts of rice bran on the medium of rice straw, the mycelial density was increased according to the increase of the added supplement, while there was no significant in the mycelial growth among the treatments. Through the field test it was showed that 15 to 20 percent addition of the supplement results in the highest yield, the shortest days from spawing to pinhead, and the lowest infection rate.
The diversity of microflora according to growth stage of Pleurotus ostreatus and the correlation between microbe and medium fermentation were investigated. In farmhouse I, the aerobic bacteria and fungi as longer of growing period were increased. And, thermophilic bacteria and fluorescent Pseudomonas sp. showed high density at the early stage of spawn inoculation. The thermophilic actinomycetes were distributed evenly during all the growing period, but mesophilic actinomycetes were not observed. In farmhouse II, thermophilic actinomycetes were not observed in fermented medium and density of fungi were suddenly increased at 60 days after spawn inoculation. And also, mushrooms can hardly be harvested due to Penicillium spp. After medium fermentation, density of aerobic bacteria, thermophilic bacteria, and fluorescent Pseudomonas sp. was higher at farmhouse I than those of farmhouse II. In farmhouse I, Bacillus sp. and Pseudomonas sp. dominated at early stage of mushroom growth but as time goes by, density of Bacillus sp. was higher than the others. And also, the kind of microbe showed a few at early stage of mushroom growth but increased as time goes by. In farmhouse II, Bacillus sp. was dominated at early stage of mushroom growth. And the growth of Bacillus sp. and Pseudomonas sp. showed intersect aspect each other in the farmhouse I but Bacillus sp. dominated during all growth periods in the farmhouse II.
This studies investigated the effect of concentrations of sodium chloride (NaCl) on occurrence and growth of fruitbody in oyster mushrooms, Pleurotus spp. Our experiments divided into two parts. When the water contents in substrate were added with sodium chloride solution in cotton waste box cultivation as a first experiment, the growth of mushroom was damaged as the concentration was increased, even though there was a little difference according to the strains. The yield in 1.0% NaCl solution was decreased to 72% compared to non-treated plot while that in 3.0% solution was only 2% of the non-treated plot. Morphological characteristics of mushrooms cultivated in substrate with the different concentration of the solution showed different results. For example, the size and thickness of pilei were not influenced by NaCl concentration, but the length of stipes and individual weight were much influenced. In plastic box cultivation filled with cotton waste, watering treatment with the different concentrations of sodium chloride solution, the second experiment, did not show any difference according to the concentration until 1.0% solution but there was a little difference according to the strains. The productivity of fruitbody started to decrease at 2.0% of the solution and the yield and quality of mushroom in 3.0% solution treatment were generally low. After the second flush, days for mushroom sprouting were generally prolonged in proportion to the solution concentration. Taken altogether, the second experiment did not show a clear effect as the case of the first experiment.

To elucidate how cultivation temperature affected various traits including pileus color, yield and morphology of Pleurotusspp. Main results were as follows. Pileus lightness of all cultivars of Pleurotustested became higher as cultivation temperature increased, while those of Santari, Hwang-geumsantari and Sunjung at 21℃ were lower than at 18℃. Redness and yellowness of pileus decreased as cultivation temperature increased; those of chromatic pileus cultivars showed noticeable difference. Yellowness of cultivar with chromatic pileus was higher than that of cultivar with achromatic pileus. Yield was increased as cultivation temperature increased, Wonhyeung 1ho; low temperature favored cultivar showed high yield when it was cultivated at low temperature andno fruiting body at 21℃. Valid number of stipes were generally higher at 18℃, and its correlation coefficient with yield was low. Length and stipe thickness changed consistently (larger and thicker) upon cultivation temperature; the coefficient of determination(R2) 0.514 for lengthof Heuktari and 0.963for stipe thickness of Santari were high. Correlation coefficient of one trait was highly related with multiple traits. In the future, we will conduct research on the changes of expressed genes involved in the pigments for pileus color by RNA expression analysis.
This study was carried out to investigate effect of adding tobacco waste to promote oyster mushroom, Pleurotus ostreatus, mycelial growth and control of Trichoderma disease. Tobacco waste was treated on the surface of rice straw inoculated with oyster mushroom strains. The mycelial growth treated with 0.1g tobacco waste was somewhat faster than non-treatment. In the case of mixed media with 1.0g tobacco waste, mycelial growth was increased compare to control, but adding 2.0 g tobacco waste decreased the mycelial growth. We also treated the tobacco waste on Trichoderma disease. All of the tobacco waste treatment force down the growth of Trichoderma disease. We treated tobacco waste on the surface of rice straw substrates. ASI 2072 mycelial growth was decreased regardless of amount of tobacco waster and method of treatment. Mycelial growth of ASI 2180 was somewhat decreased with treatment of 0.1g and 0.2g In the case of media mixed, mycelial growth of ASI 2072 was decreased in the overall treatment. Mycelial growth of other strains were decreased as only 2.0g treatment. Field test of mycelial growth was conducted using rice straw and waste cotton with substrates. Rice straw culture with treated 3% tobacco waste and cotton waste culture with treated 6% tobacco waste showed the highest yield. There was no effect to Trichoderma disease.
Twelve species of fungi were isolated from rice straw media for oyster mushroom cultivation. Trichoderma, Aspergillus and Rhizopus were the predominant fungi. Seven species of Trichoderma were isolated and identified from the rice straw media and the order of their frequency in the media was pseudokonigii, aureoviride, viride, harzianum and koningii. Occurrence of harmful fungi in mushroom houses become more severe as the number of cultivation times increased, and that was more severe in spring culture than in autumn culture. Mycelial growth and sporulation of Trichoderma, Aspergillus and Rhizopus were fovorable on the media appended with extracts of rice straws and oyster mushrooms. This results indicate that the rice straw media and mushrooms give favorable conditions for the occurrence of the fungi in the mushroom houses. Mycelial growth of Trichoderma spp. was favorable on sawdust extraction media and rice bran extraction media, and the spawns inoculated at the mushroom beds present media of the fungi.
The characteristics and spore production of Gonji7ho, Bunhong, and Sunjung fruiting bodies were assessed at different growth stages. The shape of the Pleurotus species fruiting body starts out short and small, then takes on a typical mushroom shape as it grows. Gonji7ho has a long stalk, Bunhong has a short stalk and a wide cap, and Sunjung's cap and stalk dimensions are intermediate. Each variety displayed deep color at the beginning of growth but became steadily lighter with continued growth. The shape of the linkage between the mushroom stalk and cap changed from an initial central position to a lateral position after the growing stage. Gonji7ho cap diameter increased 7-fold from 15.5 mm (5 days of growth) to 37.9 mm (9 days of growth). Growth rates for each growth day measured using the growth percentage of the previous day were 285.5% (5 → 6th day), 182.2% (6 → 7th day), 129.4% (7 → 8th day), and 103.8% (8 → 9th day). This trend was also observed in Bunhong and Sunjung, but Bunhong's growth rate was more rapid (4.9 fold on day 6, 2.7 fold on day 7) and continued to increase through day 9. Harvest yield, which is of greatest interest to farmers, displayed a similar trend spanning the growth period, as did cap diameter. Gonji7ho harvest yield increased rapidly until day 7 of growth (more than 177%), then growth slowed down beginning around day 8, and further decreased on day 9 (98%). Similar trends were observed in Bunhong and Sunjung. Bunhong showed characteristic rapid growth in harvest yield (4.9 fold compared to the previous day on day 6 and 2.7 fold on day 7), and the increase continued through day 9. A decrease in mushroom harvest yield commonly seen in the late growth stage is thought to be due to the death of some mushrooms and decomposition of cap tissue. Basidiospore content increased with number of growth days but decreased after day 8. Gonji7ho yielded the highest production on day 7 of growth, coinciding with harvest time, with 209,000,000 spores. This trend was also observed in Bunhong and Sunjung. These results will provide researchers with basal data and guide farmers in selecting the optimal harvest day.
This study was carried out to investigate effect of NaCl treatment on growth oyster mushroom and Trichoderma spp. on PDA and in rice straws. We also investigated the possibility of oyster mushroom cultivation using rice straw harvested from the reclaimed land having been drained sea water. Mycelial growth of oyster mushroom was increased by treatment of 0.2% NaCl but decreased by treatment of the higher concentration of NaCl. In the case of the mycelial growth on PDA of Trichoderma spp., no change was found in 0.5%~1.0% NaCl, but decreased in the range of 1.0%~3.0% NaCl and drastically decreased at 5.0% NaCl. In the rice straws treated with different concentration of NaCl solution, mycelial growth of oyster mushroom showed almost same result compare to PDA. The spore formation of Trichoderma mould was almost same in both 0% and 0.3% NaCl, decreased in 0.5% and was not found spores in the higher concentration of NaCl. As increasing salt concentration in the rice straws, the NaO contents were increased. The $K_2O$ contents were decreased before and after sterilization. The moisture content of rice straws showed no difference by treatment of 3.0% NaCl but decreased the moisture by treatment of 5.0% NaCl. No pH change was found in the rice straws treated with NaCl. Mushroom yield in the rice straw of reclaimed land was a little higher than that of normal paddy land straw. The duration of primordium formation was not affected by NaCl concentration in rice straws. The yield of fruiting body in 0.3% NaCl treatment was 2,700kg, which was almost same to non-treated plot, but decreased in 0.5% NaCl. EC value of soaking water after submerging rice straw was higher than before, but NaO content was not changed at both condition.
The purpose of this study was to investigate the change of water content in mushroom substrate, paddy straw, and the mycelial growth of oyster mushroom by various soaking time. The water content of paddy straw was gradually increased as the soaking time was getting longer. These experiments also showed that the water content in the upper part of paddy straw was less than that of the low part. The mycelial growth was the good between 4 and 12 hours soaking treatment, showing faster growth and higher density in the lower part than the upper part. However we could not found any significant variation among 5 strains of oyster mushroom in the effects of the various soaking times and soaking part. The pH of the lower part substrates had a low pH compared with that of the upper part. When pathogens and mushroom mycelia were co-inoculated on paddy straw under various soaking conditions, the mushroom mycelial growth showed fast and high density according to the increase of the soaking time. The experiment related to water transport reveals that it is not easy to move into upper part, but only soaked part by adding water is available to absorb enough water. Old straw was much better than the straw new this year one for water absorption, mycelial growth and mycelial density.
These studies were investigated for effect of concentration of sodium chloride on occurrence and growth of fruitbody in oyster mushroom, Pleurotus ostreatus. When the mycelia were inoculated and cultured on the PDA plate added with the different concentrations of sodium chloride, the growth of them were not affected at the concentration of 0.5%, started to decrease at 1.0% as compared with the PDA plate without sodium chloride, and they did not grow at 5.0%. When tested at the column filled with sawdust, the tendency was similar, but the decreasing rate of mycelial growth was gentle. In sawdust bottle cultivation sprinkled with the different concentration of sodium chloride solution at the different stage, the productivity and quality of fruitbody started to decrease at 3.0% of the solution and the yield at the treatment of 10.0% solution was only 47% of that in non-treated bottles. Treated at the different stages of mushroom development, treatment just after mushroom sprout did not show any different from that just after scraping spawn from the complete grown bottles. The sprinkle treatment at this stage looked better than that of the soaking with the solution before mushroom sprout. The yield at this treatment with 10% solution showed little damage with that of 90% of non-treated bottle. Sprinkle treatment during mushroom growth stage did not affect and showed almost same to non-treated bottle except the treatment with 10.0% solution. The treatment of this stage showed better results of growing characters than the treatment after and before mushroom sprouting even 10% solution. Morphological characteristics of oyster mushroom do not distinguish to treatment of different stage and concentration of sodium chloride solution. The analyzing data of quality on underwater usually used for oyster mushroom in Inchon, Yeongjong and Kanghwa area showed over-concentration to the standard for edible water and agricultural water. But it was not confirmed whether this water quality could affect to mushroom cultivation.
