Changes in expression of monocarboxylate transporters, heat shock proteins and meat quality of Large White Yorkshire and Ghungroo pigs during hot summer period

  • Objective: Present study explores the effect of hot summer period on the glycolytic rate of early post-mortem meat quality of Ghungroo and Large White Yorkshire (LWY) pig and comparative adaptability to high temperature between above breeds by shifting the expression of stress related genes like mono-carboxylate transporters (MCTs) and heat shock proteins (HSPs). Methods: Healthy pigs of two different breeds, viz., LYW and Ghungroo (20 from each) were maintained during hot summer period (May to June) with a mean temperature of about $38^{\circ}C$. The pigs were slaughtered and meat samples from the longissimus dorsi (LD) muscles were analyzed for pH, glycogen and lactate content and mRNA expression. Following 24 h of chilling, LD muscle was also taken from the carcasses to evaluate protein solubility and different meat quality measurements. Results: LWY exhibited significantly (p<0.01) higher plasma cortisol and lactate dehydrogenase concentration than Ghungroo indicating their higher sensitivity to high temperature. LD muscle from LWY pigs revealed lower initial and ultimate pH values and higher drip loss compared to Ghungroo, indicating a faster rate of pH fall. LD muscle of Ghungroo had significantly lower lactate content at 45 min postmortem indicating normal postmortem glycolysis and much slower glycolytic rate at early postmortem. LD muscle of LWY showed rapid postmortem glycolysis, higher drip loss and higher degrees of protein denaturation. Ghungroo exhibited slightly better water holding capacity, lower cooking loss and higher protein solubility. All HSPs (HSP27, HSP70, and HSP90) and MCTs (MCT1, MCT2, and MCT4) in the LD muscle of pigs inclined to increase more in Ghungroo than LWY when exposed to high temperature. Conclusion: Effect of high temperature on the variation of HSPs and MCTs may play a crucial role in thermal tolerance and adaptation to different climatic conditions, pH regulation, muscle acidification, drip loss, protein denaturation and also in postmortem meat quality development.

  • Sloan-Kettering virus gene product of a cellular protooncogene c-Ski is an unique nuclear pro-oncoprotein and belongs to the Ski/Sno proto-oncogene family. Ski plays multiple roles in a variety of cell types, it can induce both oncogenic transformation and terminal muscle differentiation when expressed at high levels. Ski protein is implicated in proliferation/differentiation in a variety of cells. The alternative fate of granulosa cells other than apoptosis is to differentiate to luteal cells, however, it is unknown whether Ski is expressed and has a role in granulosa cells undergoing luteinization. Thus, the aim of this study was, by means of immunohistochemical methods, to locate Ski protein in the rat ovaries during ovulation and corpora lutea (CL) formation to predict the possible involvement of Ski in luteinization. In addition, we performed to examine whether the initiation of luteinization with luteinizing hormone (LH) directly regulates expression of Ski in the luteinized granulosa and luteal cells after ovulation by in vivo models. In order to examine the expression pattern of Ski protein along with the progress of luteinization, follicular growth was induced by administration of equine chorionic gonadtropin to immature female rat, and luteinization was induced by human chorionic gonadtropin treatment to mimic luteinizing hormone (LH) surge. While no Ski-positive granulosa cells were present in preovulatory follicle, Ski protein expression was induced in response to LH surge, and was maintained after the formation of corpus luteum (CL). These results indicate that Ski is profoundly expressed in the luteinized granulosa cells and luteal cells of CL during luteinization, and suggest that Ski may play a role in luteinization of granulosa cells.

Bacterial Communities of Biofilms Sampled from Seepage Groundwater Contaminated with Petroleum Oil

  • The diesel-degrading activities of biofilms sampled from petroleum-contaminated groundwaters in urban subway drainage systems were examined in liquid cultures, and the microbial populations of the biofilms were characterized by denaturing gel gradient electrophoresis (DGGE) and 16S rDNA sequence analysis. Biofilm samples derived from two sites (19 K and 20 K) at subway Station N and Station I could degrade around $80\%$ of applied diesel within 20 and 40 days, respectively, at $15^{\circ}C$, and these results were strongly correlated with the growth patterns of the biofilms. The closest phylogenetic neighbor of a dominant component in the 19 K biofilm was Thiothrix fructosivorans strain Q ($100\%$ similarity). Four dominant strains in the 20 K biofilm were closely related to Thiothrix fructosivorans strain Q ($100\%$ similarity), Thiothrix sp. CC-5 ($100\%$ similarity), Sphaerotilus sp. IF14 ($99\%$ similarity), and Cytophaga-Flexibacter-Bacterioides (CFB) group bacterium RW262 ($98\%$ similarity). Three dominant members in the Station I biofilms were very similar to uncultured Cytophagales clone CRE-PA82 ($91\%$ similarity), Pseudomonas sp. WDL5 ($97\%$ similarity), and uncultured CFB group bacterium LCK-64 ($94\%$ similarity). The microbial components of the biofilms differed depending on the sampling site. This is the first report on the isolation of clones highly similar to Thiothrix fructosivorans and Thiothrix sp. from biofilms in petroleum-polluted groundwaters, and the first evidence that these organisms may play major roles in petroleum degradation and/or biofilm-development.

  • Global navigation satellite system (GNSS) providing position, navigation and timing (PNT) services consist of satellite, ground, and user systems. Monitoring stations, a key element of the ground segment, play a crucial role in continuously collecting satellite navigation signals for service provision and fault detection. These stations detect anomalies such as threats to the signal-in-space (SIS) of satellites, receiver issues, and local threats. They deliver received data and detection results to the master station. This paper introduces the main monitoring algorithms and measurement pre-processing processes for quality assessment and fault detection of received satellite signals in current satellite navigation system monitoring stations. Furthermore, it proposes a strategy for the development of components, architecture, and algorithms for the new regional navigation satellite system (RNSS) monitoring stations.

Development of Evaluation Indicators of Greenhouse for Tomato Cultivation Using Delphi Survey Method (델파이 설문조사를 통한 토마토 재배시설 평가지표 개발)

  • This study aimed to develop the comprehensive indicators which can be used for evaluating greenhouse for tomato cultivation. To achieve this aim, the study developed the evaluation indicators composed of evaluation items, grades and criteria by extracting preliminary evaluation items through analyzing the related papers and preceding studies, and conducting Delphi survey on an expert group. During the three surveys, the questions of closed-ended type were given to a panel of 100 experts - professors related to tomato cultivation and facilities, researchers and farmers (practical users). As a result, the finally established evaluation indicators consist of 4 categories and 39 specific evaluation items. The 4 categories are the structural factor of greenhouse, equipment factor of greenhouse, cultivation factor, and infrastructure factor. These factors consist of specific evaluation items of 9, 15, 7 and 8, respectively. In addition, on 39 specific evaluation items, weighted values were calculated and grades and criteria were established by collecting opinions of the experts. The newly developed evaluation indicators through this study will play an important role in developing new greenhouse models, considering things that should be complemented preferentially regarding in-use facilities, and improving the efficiency of projects supported by the government.

  • Lichens play an important role in breaking down rocks physically and chemically. Physical weathering occurs mechanically through hyphae and rhizines growing into the rocks. Lichen substances such as oxalic acid, acidic polysaccharides and depsides may assist altering rocks chemically. These actions partly contribute to soil formation but prove harmful for the substratum. Some people do not like to keep lichens on stained glassof churches or on gravestones mainly because of its unglamorous appearance. A biocide, 'Koretrel', is very effective in removing lichens and blue-green algae from the surface of concrete. After spraying 'Koretrel' in the open air, lichens (Caloplaca sp., Endocarpon petrolepideum, Physciella melanchra, Phaeophyscia hispidula, etc.) growing on buildings died within three months and peeled off fromthe substratum. One year after treatment, concrete surfaces were almost lichen free for three or four years. The biocide makesuse of a kind of lichen substance and harmless both for substances and environment. Our recent experiments show that it is also useful for water grasses growing on the surface of an aqueduct of a hydroelectric power station.

Circuit card inspection method through digital circuit design based AITS

  • Previous test equipment was bulky, took a long time to check, and was somewhat less economical. Since most of the checks were about analog signals, we preferred to check them using reference equipments. In this paper, a digital circuit design based on AITS is used to implement signals that can not utilize commercial measurement resources, and also designed and manufactured equipment that can inspect SRU. These test equipments were tested and evaluated by development, operation, and field evaluation, and they were installed to the Korean Field Force. This contributed to the improvement of operability by shortening the inspection time from 83.2 minutes to 7.8 minutes on average In addition, it did not utilize the reference equipment, so it could play a big role in lowering the mass production cost.

  • Nurses play a great role in the nursing unit and their behaviour should be evaluated in order to find user needs in terms of ward design of general hospital. This study aims to investigate the ways in which nurses activities and traffic occur in real behaviour setting of five general hospitals. For that purpose, intensive field survey was undertook to measure the nurses' attitudes and their activities were observed in the day time for 8 hours.

  • Less researches on AGV(Automated Guided Vehicle) path configurations have been conducted so far while more studies have been placed in determining AGV guide path directions and pick-up/drop-off station locations, and routing/dispatching/scheduling strategies. In practice plenty of concerns fall in preventing deadlock and simplifying AGV system control through an appropriate AGV path configuration. In order to meet those requirements, a new AGV path configuration, inter-loop stocker type is introduced here. The stocker serves as relaying material between loops as well as stocking material. Automated material handling systems using AGVs play an important role in semiconductor industry including TFT LCD and memory/non-memory chip. A practical example of implementing the inter-loop stocker concept successfully in the TFT LCD fab is presented in this paper.

  • As a method for effective utilization of the limited power and frequency resources in satellite communications, this paper develops software which can assigns the optimal power of input carrier groups inthe multi-level SCPC systems and performs evaluation to a various satellite link environment. Using the developed software, the paper presents performance according to the frequency plan of carrier groups and suggest a frequency play method for performance improvement of satellite link. A various interference and noise caused by satellite link are analyzed for reliable performance evaluation. The performance criteria for evaluation is classified into 2 categories according to the CNR of destination receiver earth station.
