The serine threonine kinase RIP3: lost and found

  • Receptor-interacting protein kinase-3 (RIP3, or RIPK3) is an essential protein in the "programmed", or "regulated" necrosis cell death pathway that is activated in response to death receptor ligands and other types of cellular stress. Programmed necrotic cell death is distinguished from its apoptotic counterpart in that it is not characterized by the activation of caspases; unlike apoptosis, programmed necrosis results in plasma membrane rupture, thus spilling the contents of the cell and triggering the activation of the immune system and inflammation. Here we discuss findings, including our own recent data, which show that RIP3 protein expression is absent in many cancer cell lines. The recent data suggests that the lack of RIP3 expression in a majority of these deficient cell lines is due to methylation-dependent silencing, which limits the responses of these cells to pro-necrotic stimuli. Importantly, RIP3 expression may be restored in many cancer cells through the use of hypomethylating agents, such as decitabine. The potential implications of loss of RIP3 expression in cancer are explored, along with possible consequences for chemotherapeutic response. [BMB Reports 2015; 48(6): 303-312]

Effect of different exercise intensities on biomarkers of oxidant-antioxidant balance, inflammation, and muscle damage

  • We investigated the effect of different exercise intensities on biomarkers of oxidant-antioxidant balance, inflammation, and muscle damage. Eighteen healthy and untrained male subjects were enrolled.Subjects were randomly and equally assigned to a moderate-intensityexercise(MIE, $65%VO_2max$) group(n=9) or a high-intensity exercise(HIE, $85%VO_2max$) group(n=9).Blood samples were collectedimmediately pre-exercise, post-exercise, and 60min post-exercisetoexamine oxidant-antioxidant balance(d-ROMs; BAP), inflammation(CRP; fibrinogen), muscle damage(CK; LDH), and lactate. Serum d-ROMs and BAP levels were significantly increased post-exercise compared with pre-exercise levels in HIE group(p<0.05). Lactate levels were significantly increased post-exercise compared pre-exercise levels in both the MIE and HIE groups(p<0.05). In addition, post-exercise serum d-ROMs and plasma lactate levels were significantly higher in the HIE group than in the MIE group(p<0.05). These results suggest that although relatively high-intensity exercises may increase oxidative stress levels in the body, they do not produce inflammatory response and/or muscle damage.

Central Effects of Ginsenosides on the Feeding Behavior and Response to Stress in Rats

  • To clarify central mechanisms of ginsenosides, changes in ingestive and ambulatory behaviors were investigated in rats after single or continuous infusion into the third cerebroventricle or various hypothalamic loci. Following single infusion into the third cerebroventricle, ginsenoside Rbl at doses of 0.05, 0.10 and 0.20 $\mu$mol dose-dependently decreased food intake. None of the doses tested affected ambulation. Drinking suppression was only observed at the maximum dose of 0.20 $\mu$mol. Equimolar injections into the peritoneum had no effects on ingestive behavior or ambulation. These findings indicated that ginsenoside Rbl specifically and centrally inhibited food intake. According to analyses of daily feeding patterns, this feeding suppression was the result of a decrease in meal size, not from changes in the postprandial intermeal interval or eating speed. The suppressed food intake was accompanied by hyperglycemia, leaving plasma insulin unaffected. Unilateral micro injection of 0.01 u mot ginsenoside Rb, into the ventromedial hypothalamus specifically decreased food intake, although equimolar injection into the lateral hypothalamic area did not affect food intake. Following continuous infusion of Rg, into the third cerebroventricle, the feeding inhibition due to surgical operation was attenuated. Rbs administered by the same procedure abolished the toxic effect of toxohormone-L on food intake. Taken together, these findings suggest that ginsenoside as a whole may have pharmacological potency to maintain feeding at a certain physiological level.

  • We investigated growth, survival and stress response depending on salinity changes in hybrid grouper (Epinephelus fuscoguttatus ♀×E. polyphekadion ♂, BCG) juvenile. Experiment I (Exp I), the hybrids were stocked at 32 psu and reared for 30 days under decreasing salinity at a rate of 5-7 psu per day (25, 20, 15, 10, 5 psu). Experiment II (Exp II), the salinity was decreased from 32 psu to 5 psu during a period of 24, 48 and 72 h. Experiment III (Exp III), acclimatized BCGs were exposed from 5 psu to decreasing salinity at 0, 1, 2, 3, 4 psu. In the Exp I, 90% survival only in the 5 psu. Growth were 5 and 32 psu had significantly low growth than other treatment. In the Exp II 100% survived by the end of the experiment. The plasma cortisol level was highest in the treatment the salinity rapidly dropped, but all the treatment recovered quickly after 24 h of salinity reduction. In the Exp III, the lower the salinity, the faster the mortality occurred in salinity below 5 psu. It is therefore concluded that the BCGs are highly tolerant of low salinity and the salinity tolerance threshold is 5 psu.

홍어 콜라겐 펩타이드의 산화적 스트레스 완화를 통한 항염증효과 (Anti-inflammatory Effect of Skate Collagen Peptide through Attenuation of Oxidative Stress)

  • 홍어 껍질 유래 콜라겐 펩타이드를 한외여과막을 이용하여 분자량의 크기를 1,000 Da 이하와 1,000 Da 이상으로 분리하고 그 효과를 비교하고자 하였다. 각각의 시료를 비만 유발 실험동물인 db/db mice에 체중당 200 mg의 콜라겐 펩타이드를 8주간 투여하였다. 본 연구의 결과는 콜라겐 펩타이드를 투여한 군에서 비만대조군에 비해 체중 증가량의 감소, 혈액 및 간조직의 활성산소 농도의 감소, 간조직의 산화적 스트레스가 완화된 것으로 나타났다. 또한 콜라겐 투여군에서 염증반응과 관련된 간조직의 핵전사인자($NF-{\kappa}B$) 및 효소(COX2, iNOS), 염증성 사이토카인(IL-6)발현이 비만대조군에 비해 유의적으로 감소하였다. 분자량의 크기에 따라 약간의 차이가 나타났지만, 전반적으로는 유의적이지 않았다. 따라서 홍어 콜라겐 펩타이드의 비만에 의해 유발된 염증반응이 억제되었는데, 이는 콜라겐의 산화적 스트레스 완화작용에 기인한 것으로 사료된다.

고수온에 따른 잉어(Cyprinus carpio)의 혈장 코티졸 농도 변화 (Changes of Plasma Cortisol Concentration to High Water Temperature on Common Carp (Cyprinus carpio))

  • 잉어, Cyprinus carpio를 대상으로 고수온 스트레스 발생 시 코티졸 호르몬 변화 및 누적 사망률을 조사하였다. 수온 변화에 따른 20℃, 24℃ 및 28℃ 실험군의 코티졸 변화는 수온 변화 후 6시간에 코티졸 함량이 증가하다가 24시간 이후부터 감소하여 20℃ 및 24℃ 실험군은 실험 전의 코티졸 함량에 가까운 수치로 감소하였다. 48시간의 28℃ 실험군 코티졸 함량은 실험 전의 코티졸 함량보다 높았으나 24시간의 코티졸 함량보다는 유의한 차이를 보이며 감소하였다. 반면, 32℃ 실험군은 수온 변화 후 6시간부터 코티졸 함량이 증가하여 24시간에 코티졸 함량이 최대치로 증가하였으며 48시간까지 코티졸 함량이 감소하지 않고 유지되는 경향이 나타났다. 20℃, 24℃ 및 28℃ 실험군은 수온 변화 후 12시간까지 누적 사망률이 증가하다가 그 이후부터 사망하는 개체가 발생하지 않았으나, 32℃ 실험군은 실험 종료 시(48시간)까지 지속적으로 누적 사망률이 증가하는 경향이 나타났다. 본 연구로 도출된 결과를 이용하여 호르몬 분석을 통해 특정지역의 폐사 위험 정도를 확인할 수 있는 방안으로 활용 가능할 것이다.

전기침이 결박 스트레스 반응에 미치는 영향 : 억제성 회피 과제, 강제 수영 시험, 스트레스 호르몬 반응 연구 (Effects of Electroacupuncture on Immobilization Stress Responses : A Study on Inhibitory Avoidance Task, Forced Swimming Test, and Stress Hormones)

  • 목적 : 본 연구는 결박 스트레스를 시행한 백서의 기억력, 우울, 스트레스 호르몬의 혈중 농도에 대한 전기침의 효과를 관찰하였다. 재료와 방법 : 결박 그룹은 10일 동안 2시간의 결박 스트레스를 받았다. 결박 스트레스 및 고주파수 전침 그룹은 2시간의 결박 스트레스를 받는 동시에 100Hz의 전침을 우측 족삼리 (ST-36)에 시행하였다. 기억력과 우울에 미치는 결박 스트레스와 침의 효과를 비교하기 위하여 억제성 회피 과제와 강제 수영 검사를 실시하였다. 결박 스트레스 및 전기침으로 유도된 코르티코스테론과 멜라토닌, 부신피질자극호르몬, 노르아드레날린의 농도를 측정하기 위하여 결박 스트레스 및 전기침 자극이 시작된 30분, 60분, 90분, 120분 후에 백서의 꼬리에서 혈액 샘플을 채취하였다. 결과 : 억제성 회피 과제에서 전기침을 시행한 그룹은 시행하지 않은 그룹에 대해서 장기 기억의 증진에 유의한 변화가 있었다. 강제 수영 검사 결과에서는 우울에 유의한 변화가 없었으며, 스트레스 호르몬 반응 결과에서도 유의한 변화가 없었다. 결론 : 1. 고주파수 전기침 (100Hz)은 억제성 회피 과제 결과 결박 스트레스에 기억력을 상승시켰다. 2. 고주파수 전기침 (100Hz)은 강제 수영 시험 결과 결박 스트레스로 유도된 우울을 감소시키지 않았다. 3. 고주파수 전기침 (100Hz)은 혈액 채취 결과 스트레스 호르몬을 감소시키지 않았다.

Protective Effect of Administrated Glutathione-enriched Saccharomyces cerevisiae FF-8 Against Carbon Tetrachloride ($CCl_4$)-induced Hepatotoxicity and Oxidative Stress in Rats

  • The present work is aimed to evaluate the protective effect of glutathione-enriched Saccharomyces cerevisiae FF-8 strain on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity and oxidative stress in rats. The activities of liver markers (alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, lactate dehydrogenase), lipid peroxidative index (thiobarbituric acid-reactive substances), and the antioxidant status (reduced glutathione) were used to monitor those protective roles of FF-8 strain. The liver marker enzymes in plasma and the lipid peroxidation in the liver were increased when $CCl_4$ was treated but these were significantly decreased by FF-8 strain treatment. The hepatic concentration of glutathione in the current glutathione-enriched FF-8 strain fed animal was approximately twice as high as the normal, but this was slightly increased in response to $CCl_4$ plus glutathione-enriched FF-8 strain. The increased liver triglyceride concentration due to the $CCl_4$ treatment was significantly decreased by FF-8 strain and the reduced level reached to that of normal group. Administration of FF-8 strain in normal rat did not show any signs of harmful effects. Therefore, the current findings suggest that FF-8 strain could be an effective antioxidant with no or negligible side-effects and it might be useful for the purpose of protection treatment of hepatotoxicity and oxidative stress in $CCl_4$-treatment in rat.

Epac2a-knockout mice are resistant to dexamethasone-induced skeletal muscle atrophy and short-term cold stress

  • Exchange protein directly activated by cAMP (Epac) 2a-knockout (KO) mice exhibit accelerated diet-induced obesity and are resistant to leptin-mediated adipostatic signaling from the hypothalamus to adipose tissue, with sustained food intake. However, the impact of Epac2a deficiency on hypothalamic regulation of sympathetic nervous activity (SNA) has not been elucidated. This study was performed to elucidate the response of Epac2a-KO mice to dexamethasone-induced muscle atrophy and acute cold stress. Compared to age-matched wild-type mice, Epac2a-KO mice showed higher energy expenditures and expression of myogenin and uncoupling protein-1 in skeletal muscle (SM) and brown adipose tissue (BAT), respectively. Epac2a-KO mice exhibited greater endurance to dexamethasone and cold stress. In wild-type mice, exogenous leptin mimicked the responses observed in Epac2a-KO mice. This suggests that leptin-mediated hypothalamic signaling toward SNA appears to be intact in these mice. Hence, the potentiated responses of SM and BAT may be due to their high plasma leptin levels.

Expression of Hepatic Vascular Stress Genes Following Ischemiai/Reperfusion and Subsequent Endotoxemia

  • Hepatic ischemia and reperfusion (l/R) predisposes the liver to secondary stresses such as endotoxemia, possibly via dysregulation of the hepatic microcirculation secondary to an imbalanced regulation of the vascular stress genes. In this study, the effect of hepatic I/R on the hepatic vasoregulatory gene expression in response to endotoxin was determined. Rats were subjected to 90 min of hepatic ischemia and 6 h of reperfusion. Lipopolysaccharide (LPS, 1 mg/kg) was injected intraperitoneally after reperfusion. Plasma and liver samples were obtained 6 h after reperfusion for serum aminotransferase assays and RT-PCR analysis of the mRNA for the genes of interest: endothelin-1 (ET-1), its receptors $ET_A$ and $ET_B$, endothelial nitric oxide synthase (eNOS), inducible nitric oxide synthase (iNOS), heme oxygenase-1 (HO-1), cyciooxygenase-2 (COX-2), and tumor necrosis factor-a (TNF-${\alpha}$). The activities of serum aminotransferases were significantly increased in the I/R group. This increase was markedly potentiated by LPS treatment. The ET-1 mRNA was increased by LPS alone, and this increase was significantly greater in both the I/R alone and I/R + LPS groups compared to the sham. There were no significant differences in ETA receptor mRNA levels among any of the experimental groups. $ET_B$ mRNA was increased by both LPS alone and I/R alone, with no significant difference between the I/R alone and I/R + LPS groups. The eN OS and HO-1 transcripts were increased by I/R alone and further increased by I/R + LPS. The iNOS mRNA levels were increased by I/R alone, but increased significantly more by both LPS alone and I/R + LPS compared to I/R alone. The TNF-${\alpha}$ mRNA levels showed no change with I/R alone, but were increased by both LPS alone and I/R + LPS. The COX-2 expression was increased significantly by I/R alone and significantly more by I/R + LPS. Taken collectively, significantly greater induction of the vasodilator genes over the constriction forces was observed with I/R + LPS. These results may partly explain the increased susceptibility of ischemic livers to injury as a result of endotoxemia.
