Effect of Capsicum Components on Liver Microsomal Cytochrome $P_{450}$ in Rat (고초(苦椒)가 백서(白鼠)의 간(肝) 마이크로좀 Cytochrome $P_{450}$에 미치는 영향(影響))

  • The investigation is involved with the effect of Capsicum component on the drug metabolism. To investigate the effects of Capsicum component on, in vivo, drug metabolism in rat, Capsicum acetone extract was given intraperitoneally to mice or rats. The duration of loss of righting reflex was determined as hexobarbital sleeping time in mice. Plasma hexobarbital concentration was also measured by Brodie's method. The rats were pretreated with Capsicum extract acutely or chronically. As the results, hexobarbital sleeping time and plasma hexobarbital concentration were increased by 31.2% and 12.3% in acute study, whereas were decreased by 27.5% and 23.0% in chronic study. An attempt was made to determine if there were any influences on enzyme activities in rats pretreated with Capsicum extract chronically. Microsomal fraction was isolated from rat liver and quantity of cytochrome $P_{450}$ and $b_5$ in the microsomal fraction were determined by Omura's method. It was found that the quantity of cytochome $P_{450}$ was increased by 22.4%. The results suggest that microsomal drug metabolizing enzyme may be induced by chronic administration of Capsicum component, whereas it may be inhibited by acute administration of Capsicum component.

Effects of Uncooked Korean Food on Blood Glucose and Antioxidant Fnzyme Activities of Streptozotocin-induced Diabetic Rats (당뇨환자용 식사대용식이 Streptozotocin으로 유발된 당뇨쥐의 혈당 및 항산화 효소 활성에 미치는 영향)

  • Objectives : This study has been carried out to understand the effect of Uncooked Korean Food(F-DM) on blood glucose and antioxidant enzyme activities in streptozotocin-induced Diabetic Rats. Methods : SD rats were separated into four groups(each with 20 rats). Except normal two group, the other two groups were injected into intra-peritoneal with streptozotocin 60 mg/kg. Experimental group was eated Feed with 25% F-DM for 4 weeks. The change of plasma glucose level, body weight were observed. After 4 weeks, liver and kidney weight, antioxidant enzyme activities, survival rate were observed with histological changes on liver, kidney and pancreas. Results : In experimental group, body weight and survival rate increased, plasma glucose level were decreased significantly. Liver and kidney weight, XOD activity were decreased in experimental group compared to control group. GSH-px and CAT activities, insulin- immunoreactive granules in p-cells were increased significantly in experimental group compared to control group. Conclusions : This study shows that the F-DM might be effective for treatment of diabetes and its complications, as well as reduction of the oxidative stress.

  • Fabry disease (FD) is an X-linked inborn error of glycoshpingolipid metabolism resulting from mutation in the enzyme ${\alpha}$-galactosidase A gene. The disease is an X-linked lipid storage disorder and the lack of ${\alpha}$-Gal A causes an intracellular accumulation of glycosphingolipids, mainly globotriaosylceramide (Gb-3). Measurement of Gb-3 in plasma has clinical importance for monitoring after enzyme replacement therapy for confirmed FD patients. Using electrospray ionization MS/MS we had developed, a simple, rapid, and highly sensitive analytical method for Gb-3 in plasma was used for the purpose of screening FD among high risk groups in Korean population. To date, no comprehensive results for FD screening have been performed and reported in Korea. We screened 1,100 outpatients from 13 hospitals (including clinics) to assess the incidence of FD among patients in high risk groups. For patients with borderline level amount of Gb-3, we repeated Gb-3 or performing complementary or confirmative assay with ${\alpha}$-Gal A activity and DNA mutaion analysis for confirmation diagnosis. Of 1,100 we diagnosed 3 FD with 2 classical type and 1 carrier (0.27%).

Enhancement of Lipid Metabolism and Antioxidant Defense Status in Mice Fed with High Fat Diet Supplemented with Antheraea pernyi Silk Fibroin Powder

  • The effect of diet supplementation of Antheraea pernyi (A. pernyi) silk fibroin on the lipid metabolism and antioxidant defense status in high fat-fed mice was investigated. The animals were given normal control diet (NC group), high fat diet (HF group), or high fat diet supplemented with A. pernyi silk fibroin powder (HFS group) for 7 weeks. After the experimental period, the HF group showed significant increase in body weight, plasma and hepatic total cholesterol levels, and hepatic triglyceride concentration, and decreased activities of hepatic antioxidant enzymes relative to NC group. However, the HFS group exhibited marked reduction in body weight, plasma cholesterol and hepatic triglyceride levels, hepatic lipogenic enzyme activities, and lipid peroxidation rate, and higher high-density lipoprotein (HDL)-cholesterol level, fecal triglyceride content, and antioxidant enzymes activities compared with that of HF group. These findings demonstrate that dietary feeding of A. pernyi silk fibroin could improve the lipid metabolism and antioxidant defense system via regulation of hepatic antioxidant and lipogenic enzymes activities. Hence, this silk fibroin may be beneficial as a functional biomaterial for the development of therapeutic agent against high fat diet-induced hyperlipidemia and its related diseases.

Production of Polyclonal Antibodies Specific to Porcine Adipocyte Plasma Membrane Proteins in Sheep (면양을 이용한 돼지 지방제포 원형질막 단백질 특이 항체의 생산)

  • The objectives of this study were to produce polyclonal antibody to adipocyte plasma membrane (APM) proteins isolated from pig, and to investigate its tissue specificity. Plasma membrane proteins from adipocyte, brain, heart, kidney, liver and spleen were isolated using a self-forming Percoll gradient. Sheep (40kg) was immunized three times at three week interval with the purified APM proteins. Blood was taken from non-immunized sheep (NS) and from immunized sheep at 10 (AS-1), 12 (AS-2), and 14 (AS-3) days after the third immunization. Antisera titers and cross-reactivity against other tissues were determined by enzyme-linked immunosorbent assay (ELISA). Antisera reacted strongly to APM proteins showing detectable amounts of antibody at 1:81,000 dilution. And antisera showed much stronger reactivity to APM proteins than any other tissue plasma membrane proteins. Furthermore, tissue specificity of antisera against APM was reconfirmed by immunoblotting using anti-sheep immunoglobulin G-horseradish peroxidase conjugate as a secondary antibody Antisera to APM proteins showed adipocyte specificity compared with other tissues. In conclusion, polyclonal antibody against APM proteins isolated from pig was developed successfully in our laboratory, and these antisera showed tissue specificity with APM.

Induction of B Lymphocyte Differentiation by a Colostral Immunomodulatory Protein MIEF (초유에 함유되어 있는 면역조절물질인 MIEF가 B 세포의 분화에 미치는 영향)

  • The levels of maternal immunity enhancing factor(MIEF), which is an immunomodulatory protein identified from bovine colostrum, were determined by indirect competitive enzyme-linked immunosorbent assay(ELISA) for the colostrum and normal milk collected during the first two weeks of lactation. The mean concentration of MIEF in the colostrum of the first day of lactation was $109\;{\mu}g/ml$, and fell from the third day of lactation to $3{\sim}4\;{\mu}g/ml$. The molecular weight of the purified MIEF determined by reducing SDS-PAGE and TSK G2000SW column chromatography was 22,000 and 24,000 daltons, respectively, showing that MIEF is a monomeric peptide in its native form. To examine the capacity of MIEF to induce differentiation of B Lymphocytes, human tonsillar Iymphocytes were cultured in the presence of different concentrations of MIEF, and then antibody secreting cells were enumerated by enzyme-linked immunospot(ELISPOT) assay. When added to cultures of human tonsillar Lymphocytes, MIEF induced differentiation of resting B Iymphocyte to antibody secreting plasma cells as efficiently as LPS.

EFFECTS OF DIETARY PROTEINS ON THE ACTIVITIES OF LIPOGENIC ENZYMES IN THE LIVER OF GROWING CHICKS

  • In Experiment 1, when fasted chicks were fed diets containing various sources of protein for 3 days, the activities of lipogenic enzymes (acetyl-CoA carboxylase, fatty acid synthetase, citrate cleavage enzyme and malic enzyme) in the liver of growing chicks were significantly lower in the soybean protein or gluten diet than in the casein or fish protein diet. Triglycride contents of the liver and plasma of chicks fed the casein or fish protein diet were significantly lower than that of those fed soybean protein or gluten diet. In Experiment 2, the effects of dietary amino acid mixture simulating casein or protein on the activities of hepatic lipogenic enzymes were examined. The activities of acetyl-CoA carboxylase and fatty acid synthetase in the liver of chicks fed the casein diet were significantly higher than that of those fed the soybean protein diet or two diets of amino acid mixtures. Furthermore, there were no significant differences between the two diets of amino acid mixture based on casein or soybean protein. However, the activities of malic enzyme and citrate cleavage enzyme tended to be lower in the soybean-type amino acid diet than in the casein-type amino acid diet. Thus, some effects can be ascribed to the protein itself and some to the amino acid composition of the protein sources.

Alteration in Plasma BDNF Level after Repetitive Transcranial Magnetic Stimulation(rTMS) in Treatment-Resistant Schizophrenia : A Pilot Study (치료저항성 정신분열병 환자에서 반복적 경두개자기자극술 병행치료시 혈장 BDNF 농도 변화 : 예비 연구)

  • Objectives : To assess clinical improvement and change in plasma brain-derived neurotrophic factor(BDNF) level after repetitive transcranial magnetic stimulation(rTMS) in patients with treatment-resistant schizophrenia. Methods : Seven patients with DSM-IV schizophrenia, who were proven to be treatment-resistant, were treated with 15 sessions of rTMS for three weeks as an adjuvant therapy to antipsychotic treatment. Clinical improvement and change in plasma BDNF level were measured after the treatment period. The symptom severity was assessed with the Positive and Negative Syndrome Scale(PANSS) and the Korean Version of Calgary Depression Scale for Schizophrenia(K-CDSS) at baseline and 7 days after the treatment. Plasma BDNF level was measured by enzyme-linked immunosorbent assay(ELISA) at baseline and 7 days after the treatment. Results : After the rTMS treatment, there was no significant improvement in PANSS total score(Z=-1.693, p=0.090) and no significant change in plasma BDNF was found(Z=-1.183, p=0.237). Negative correlations were found between percentage change in PANSS positive subscale score and duration of illness(rho=-0.991, N=7, p<0.0005, two-tailed), and PANSS negative subscale score at baseline and percentage change in plasma BDNF level(rho=-0.821, N=7, p=0.023, two-tailed). Conclusion : This preliminary study suggests that rTMS didn't make a significant change in clinical symptoms nor in plasma BDNF level in treatment-resistant schizophrenia. Percentage change in plasma BDNF, however, might be correlated with treatment resistance in schizophrenic patients. This is a pilot study with a small sample size, therefore, a further study with a larger sample size is needed.

Evaluation of Coffee Ground as a Feedstuff in Practical Diets for Olive Flounder Paralichthys olivaceus

  • A 10-week feeding trial was carried out to investigate the feasibility of using spent coffee ground (CG) as a potential feed ingredient for olive flounder. Growth, feed utilization, body composition and antioxidant enzyme activity were examined. A control diet was formulated and three other diets were prepared to contain 5, 10 or 15% CG (designated as Con, CG5, CG10 and CG15, respectively) by replacing for wheat flour. Two hundred forty fish ($104{\pm}0.7g/fish$) were allotted to 12 circular tanks of 400 L capacity at a density of 20 fish per tank and fed the experimental diets twice daily. At the end of the feeding trial, fish fed the CG5 diet exhibited significantly (P < 0.05) higher growth performance than those fed the control diet. Also, fish fed the CG10 diet had a comparable growth to that of the control group, but further increase of dietary CG inclusion level to 15% resulted in significant decrease of growth performance. Fish fed the CG15 diet showed significantly lower feed efficiency and protein efficiency ratio than other treatments. Significantly lower muscle protein content was observed in fish fed CG15 diet compared to the control. Significant reduction in plasma cholesterol concentration was found in fish fed CG15 diet compared to control. No significant changes were found in alkyle and superoxide radicals scavenging activities of plasma, muscle and liver among dietary treatments. Also, liver total protein, total antioxidant capacity, catalase and glutathione peroxidase activities were not significantly influenced by dietary inclusion of CG. According to these results, we concluded that CG can be included up to 10% in the diet for olive flounder without any adverse effects on growth, feed utilization and antioxidant enzyme activity.

Effects of $\beta$-Carotene Supplementation on Lipid Peroxide Levels and Antioxidative Enzyme Activities in Diabetic Rats ($\beta$-Carotene 첨가식이가 당뇨쥐의 지질과산화물 수준과 항산화효소 활성에 미치는 영향)

  • This study investigated the effect of dietary $\beta$-carotene supplementation on lipid peroxidation and anti oxidative enzyme activity as indices of oxidative stress in diabetic rats. Fifty Sprague-Dawley male rats aging 7 weeks were used as experimental animals, which were divided into the non-diabetic control group and the diabetic group. The diabetic group received an intraperitoneal injection with streptozotocin to induce diabetes. Then the diabetic rats were divided into four dietary groups which contained different amounts of $\beta$-carotene; 0%, 0.002%, 0.02%, or 0.2% of the diet. The diabetic rats were fed the experimental diets and the non-diabetic rats were fed the basal diet without $\beta$-carotene supplementation for 2 weeks and then sacrificed. The diabetic group had a significantly higher blood glucose level than the non-diabetic group. However, blood glucose level were not significantly changed by the level of dietary $\beta$-carotene supplementation. Compared to the non-diabetic control group, the diabetic control group indicated a significant increase of plasma thiobarbituric acid reactive substance (TBARS). Liver TBARS level also tended to be higher in diabetic control group, although it was not significant. The $\beta$-carotene supplementation did not reduce plasma TBARS level. However, Liver TBARS level was significantly decreased when 0.02% or more $\beta$-carotene was supplemented in the diet. The liver lipofuscin level in the diabetic control group was higher than in the non-diabetic control group, but the effect of $\beta$-carotene supplementation did not show any differences. Superoxide dismutase activity was significantly lower in the diabetic group, but it was increased in groups receiving 0.02% or more $\beta$-carotene. Compared to the non-diabetic control group, lower activities of catalase and glutathione peroxidase were observed in the diabetic control group, although it was not significant. Catalase and glutathione peroxidase activities tended to increase as the levels of $\beta$-carotene supplementation increased, although it was not statistically significant. Therefore, it seems that dietary $\beta$-carotene supplementation might reduce diabetic complications by partly decreasing the lipid peroxidation and increasing the activity of antioxidative enzyme in diabetes.
