Formation of Crown Gall Tumor in Panax ginseng C.A. Meyer (인삼의 Crown Gall Tumor형성에 관한 연구)

  • These studies were carried out to obtain the basic information about transformation of ginseng plant by potential vector system, utilization of opine compound by Agrobacterium sap. , and initiation of crown gall tumor callus. Crown gall tumors were induced from stem of Panax ginseng C.A. Meyer by infection of Agrobacterium tumefaciens. Therefore, it was clarified that transformation of ginseng by Ti plasmid was possible. The crown gall tumors induced by Agrobacterium tumefaciens isolated. from the soil were different in a shape, size, and growth rate. Especially, infection of ginseng by Agrobacterium tumefaciens Y104 led to the amorphic tumor, Tumor tissue derived from stem crown gall could not be continuously cultured on the medium which did not contain phytohormone, and did not form the callus even on the medium supplemented with 2,4-D. On the other hand, the root crown gall tumors formed the calli but the formation rate of callus was quite low. As for the utilization of octopine and nopaline, it was found that 3 strains of Agrobacterium app., Y104, Y110 and C58, utilized nopaline only, Y109 utilized octopine, and Y101 failed to utilize either compound.

Antioxidant Compounds of Oryza sativa L (벼의 항산화성분)

  • In search for the plant-derived antioxidant compound, it was found that the EtOAc, BuOH extracts obtained from the leaves parts of Oryza sativa L. which exhibited a significant antioxidant activity from 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical cleavage. Activity-guided fractionation on the basis of the inhibitory activity upon the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical cleavage. and repeated column chromatography afforded several antioxidant compounds from Oryza sativa L. The structures and stereochemistry of these compounds were established on the basis of analysis of spectra and some chemical transformations as follows: 5,7-dihydroxy- 8-methoxyflavone, $acacetin-7-O-{\beta}-rutinoside$, $pectolinarigenin-7-O-{\beta}-rutinoside$. At antioxidant activity test for isolated three compounds, antioxidant activity was showed too.

Inhibitory Effect of Genistein on Agonist-Induced Modulation of Vascular Contractility

  • The present study was undertaken to determine whether treatment with genistein, the plant-derived estrogen-like compound influences agonist-induced vascular smooth muscle contraction and, if so, to investigate related mechanisms. The measurement of isometric contractions using a computerized data acquisition system was combined with molecular experiments. Genistein completely inhibited KCl-, phorbol ester-, phenylephrine-, fluoride- and thromboxane $A_2$-induced contractions. An inactive analogue, daidzein, completely inhibited only fluoride-induced contraction regardless of endothelial function, suggesting some difference between the mechanisms of RhoA/Rho-kinase activators such as fluoride and thromboxane $A_2$. Furthermore, genistein and daidzein each significantly decreased phosphorylation of MYPT1 at Thr855 had been induced by a thromboxane $A_2$ mimetic. Interestingly, iberiotoxin, a blocker of large-conductance calcium-activated potassium channels, did not inhibit the relaxation response to genistein or daidzein in denuded aortic rings precontracted with fluoride. In conclusion, genistein or daidzein elicit similar relaxing responses in fluoride-induced contractions, regardless of tyrosine kinase inhibition or endothelial function, and the relaxation caused by genistein or daidzein was not antagonized by large conductance $K_{Ca}$-channel inhibitors in the denuded muscle. This suggests that the RhoA/Rho-kinase pathway rather than $K^+$- channels are involved in the genistein-induced vasodilation. In addition, based on molecular and physiological results, only one vasoconstrictor fluoride seems to be a full RhoA/Rho-kinase activator; the others are partial activators.

  • A graphical user interface (GUI) program for steam tracing system selection was developed by using a theoretical model. We derived the model on the basis of the one-dimensional heat transfer theory of conduction and convection through a composite wall. Computational fluid dynamics (CFD) and experiments were performed for validation at steam temperatures of 120.4[℃] and 158.9[℃]. The temperature of a pipe's outer surface obtained through CFD matched well with that predicted by the proposed model for both conditions. By contrast, the experiment results showed a small error at 120.4[℃] and a large error at 158.9[℃] because of the melting of the heat transfer compound and water phase transition. Thus, the steam temperature range of the proposed model is below 120.4[℃].

An Isocoumarin with Hepatoprotective Activity in Hep G2 and Primary Hepatocytes from Agrimonia pilosa

  • In connection with our studies on the isolation of hepatoprotective constituents from natural products. we have recently reported hepatoprotective compounds including phenolic bakuchiol. diarylheptanoids. furocoumarins. In the course of continuing efforts. the aqueous extract of the roots of Agrimonia pilosa Ledeb. (Rosaceae) was found to exhibit promising hepatoprotective activity. A. pilosa is a perennial herb distributed throughout South Korea. and its roots have been used as the hemostatic. antimalarial. and antidysenteric agent in oriental medicine. Chemical investigation of the aqueous extract of the roots of this plant. as guided by hepatoprotective active catechin (2). Compound 1 showed hepatoprotective effects on both tacrine-induced cytotoxicity in human level derived Hep G2 cells and tert-hydroperoxide-induced cytotoxicity in rat primary hepatocyles with $EC_{50}$ values of 66.2 $\pm$ 2.8 and 22.9 $\pm$ 2.6 $\mu\textrm{M}$ respectively.

D-Pinitol Promotes Apoptosis in MCF-7 Cells via Induction of p53 and Bax and Inhibition of Bcl-2 and NF-κB

  • Development of drugs from natural products has been undergoing a gradual evoluation. Many plant derived compounds have excellent therapeutic potential against various human ailments. They are important sources especially for anticancer agents. A number of promising new agents are in clinical development based on their selective molecular targets in the field of oncology. D-pinitol is a naturally occurring compound derived from soy which has significant pharmacological activitites. Therefore we selected D-pinitol in order to evaluate apoptotic potential in the MCF-7 cell line. Human breast cancer cells were treated with different concentrations of D-pinitol and cytotoxicity was measured by MTT and LDH assays. The mechanism of apoptosis was studied with reference to expression of p53, Bcl-2, Bax and NF-kB proteins. The results revealed that D-pinitol significantly inhibited the proliferation of MCF-7 cells in a concentration-dependent manner, while upregulating the expression of p53, Bax and down regulating Bcl-2 and NF-kB. Thus the results obtained in this study clearly vindicated that D-pinitol induces apotosis in MCF-7 cells through regulation of proteins of pro- and anti-apoptotic cascades.

Growth Promotion and Induction of Systemic Resistance Against Phytophthora capsici on Red-pepper Plant by Treatment of Trichoderma harzianum MPA167 (근권 Trichoderma harzianum MPA167 처리에 의한 생육촉진과 고추 역병균에 대한 고추의 유도저항성)

  • Trichoderma harzianum is one of rhizosphere fungus usually lives near the plant root regions in the soil. T. harzianum plays an important role in plant growth promotion and increases disease resistance against various plant pathogens on crops. In this study, the strain T. harzianum MPA167 was isolated from the barley rhizosphere soil in Suwon, Korea. Among 183 isolates, the strain T. harzianum MPA167 was selected as promising strain in which based on hyperparasitical activity against Phytophthora capsici and estimated disease control activity against P. capsici in the greenhouse conditions. The strain T. harzianum MPA167 was identified using 23s rDNA internal transcribed spacer(ITS) region sequences. MPA167 treatment ($1{\times}10^6$ spores/ml) showed greater disease suppression against Phytophthora blight of red-pepper caused by P. capsici in greenhouse compared with the water-treated control. Volatiles derived from T. harzianum MPA167 elicit growth promotion of tobacco and Arabidopsis seedlings in I-plate assay. In addition, T. harzianum MPA167 strain was also found to be effective for the growth promotion and induction of systemic resistance on red-papper plant. These results suggest that MPA167 might be used as one of the potential biocontrol agents.

Isolation and HPLC-DAD validation of xanthoangelol in Lespedeza bicolor extract (싸리나무 추출물의 Xanthoangelol 분리 및 HPLC-DAD 밸리데이션)

  • This study was undertaken to determine the characteristics of xanthoangelol, the major chalcone constituent derived from the extracts of different parts of Lespedeza bicolor. Xanthoangelol was isolated from the root extract using column chromatography and used as a standard for quantitative analysis. The structure of the isolated compound was established based on spectroscopic evidence. The HPLC-DAD method was validated for specificity, linearity, precision, accuracy, limit of detection, and limit of quantitation. The calibration curve of xanthoangelol had significant linearity (R2>0.9999). Limit of detection and limit of quantitation 0.018 and 0.059 ㎍/mL, respectively. The relative standard deviation values of precision test, and intra- and inter-day tests were less than 0.22 and 0.40%, respectively. In the recovery test, the accuracy ranged from 98.98-102.78% with RSD values less than 0.13%. The method validation parameters indicate the applicability of the HPLC method for quality control of food or drug formulations containing L. bicolor.

Contrasting Sources of Plant Wax n-alkanes and n-alkanoic Acids in Gulf of Mexico Sediments (ODP 625B) (멕시코만 코어 퇴적물(ODP 625B)의 식물왁스 탄화수소(n-alkanes)와 지방산(n-alkanoic acids)의 생성기원 비교 연구)

  • Long chain plant waxes (n-alkanes, n-alkanoic acids, and n-alcohols) and their carbon isotopic compositions (${\delta}^{13}C$) in geologic archives are valuable tools for paleovegetation reconstruction. However, the sensitivity of different plant wax constituents to vegetation shift is not well understood. This study explores controls on the variation in ${\delta}^{13}C$ values of long-chain n-alkanes ($C_{27}$ to $C_{33}$) and n-alkanoic acids ($C_{26}-C_{30}$) in the Gulf of Mexico core sediments (ODP 625B) near the Mississippi River delta. n-Alkanoic acids' ${\delta}^{13}C$ values were higher than those of n-alkanes by 1-2‰ on average and such a pattern is the opposite from their isotope fractionation observed in living plants: 1-2‰ smaller in n-alkanes than n-alkanoic acids. We attribute this offset to contributions from aquatic plants or microbes that produce high concentrations of $^{13}C-enriched$ long-chain n-alkanoic acids. The sensitivity of n-alkanes and n-alkanoic acids to vegetation and climate varied among chain lengths. The $n-C_{33}$ alkanes were most sensitive to $C_4$ grassland expansion among n-alkane homologues, while no specific trend was observed in n-alkanoic acids. This is due to the similarity in n-alkanoic acid concentrations between $C_3$ and $C_4$ plants by homologues and low terrestrial plant-derived n-alkanoic acid contributions to the sediments. The results of this study suggest that long chain n-alkanoic acids' ${\delta}^{13}C$ values in sediments may be influenced by contributions from different sources such as aquatic plants or microbial inputs and therefore interpretations regarding this matter should be cautiously formulated. We suggest that there is a need for further studies on characterizing long-chain n-alkanoic acids ($C_{26}-C_{34}$) in aquatic plants and microbes from various climates and environments in order to investigate their production and integration into sedimentary archives.

Evaluation of the functional phenolic compounds as to various ripening phases of the reddish rice cultivar(Oryza sativa, Superhongmi)

  • Recently, consumer's demands are increasing for new natural functional and healthful food. These demands have encouraged a better understanding of the biochemical, chemical and nutritional composition of plant products. Superhongmi rice variety was derived from a cross between CG2-3-5-2-6-2 (Heugjinju/Suwon 425) having reddish brown color and Daeribbyeo 1 having a large grain size. Superhongmi rice headed on Sep. 5th and has 94.7 cm culm length. Taxifolin, a phenolic compound in superhongmi rice extract had high ${\alpha}-glucosidase$ inhibitory activity. The ${\alpha}-glucosidase$ inhibitory activity of the superhongmi rice extracts correlated to the taxifolin content and antioxidant activity of the extracts. A new method for quantitative determination of taxifolin in superhongmi rice variety by high performance liquid chromatography was established. A reversed-phase system with Tosoh TSK-gel $C_{18}$ column using 60% methanol in water(pH 2.4) as a mobile phase was developed. Taxifolin was detected at 280 nm and the analysis was successfully carried out within 40 min. In ripening phase, the amount of taxifolin showed a mild decreasing slope. The highest contents was 59.27mg in 100g seed on the 30 days after heading(DAH). The optimum harvesting time, considering taxifolin content, maturity rate and thousand seed weight(TSW) was DAH 50. These results suggest that superhongmi rice which has high taxifolin content has the potential to contribute as a dietary supplement for controlling hyperglycemia and oxidative stress-linked diabetes complications.
