Root Yield and Saikosaponin Content in Local Strains of Bupleurum falcatum L. (시호 수집종의 근 수량 및 Saikosaponin 함량)

  • Six local strains collected in Korea and one cultivar (Samdo) introduced from Japan were applied for comparative cultivation to investigate growth characteristics, root yield potentials and saikosaponin contents of root in Bupleurum falcatum L. The one and two year old plants of the native local strains flowered in August 2~3 and July 16~18, respectively, whereas Samdo was late flowering type, being delayed by 18~32days and sustained the growth of above ground parts to the end of November. All the native local strains have similiar characteristics but Samdo was completely different in plant and root type. Local strain collected from Chunchon showed better in growth and root characteristics, consequently resulting high yield of dry roots and saikosaponin contents in both one and two year old plants. The average yield of two year cultivation system was 2.6 times high as 105Kg /10a, compared with that of one year.

Review on the origin of herbal name, Sik-bang-pung, for the root of Peucedanum japonicum Thunberg (갯기름나물 약재명(식방풍(植防風))의 연원에 대한 고찰)

  • Objectives : Sik-bang-pung (植防風, Peucedani Japonici Radix), the root of Peucedanum japonicum Thunberg (Apiaceae), has often been used as a substitute for Bang-pung (防風, Saposhnikoviae Radix) in the past, but recently it is treated as a counterfeit of Bang-pung or a kind of 'Jeonho (Peucedani Radix)'. Hence, it is crucial to investigate the origin of herbal name to provide appropriate usage of Sik-bang-pung. Methods : The classic and contemporary literatures regarding to the herbal medicines and botanical nomenclature were searched to find the herbal and botanical origins of Bang-pung and Sik-bang-pung, respectively. Result : The botanical descriptions of Bang-pung (防風) were consistent throughout the classic literatures and its botanical species was determined as Saposhnikovia divaricata Schischkin (Apiaceae) in the compendium. In the literatures published in the Joseon Dynasty, 'Byeong-pung-na-mul' was recorded as the medicinal plant of Bang-pung, but its botanical species could not be confirmed. In Japanese literatures, Bang-gyu (防葵) was confirmed as Mok-dan-bang-pung (牡丹防風); botan-bōfū in Japanese), which was determined as the herbal name of the root of 'Gaet-girum-namul (P. japonicum)' in Korea during the period of Japanese rule. The roots of Gaet-girum-namul was medicinally named as Mok-dan-bang-pung (牡丹防風), Sik-bang-pung (植防風), or San-bang-pung (山防風) in the past decades in Korea. Among them, Sik-bang-pung might be chosen as the herbal name, but its nominal origin could not be found. Conclusions : The herbal name, Sik-bang-pung, was presumably affected by Japanese botanical nomenclature. Although its medicinal application is still controversial, Sik-bang-pung should be considered an independent herbal medicine.

Observation of Mitotic Chromosome behavior according to Different Treatment Methods of DNA Methylation Inhibitor

  • Chromosome breakage occurred by DNA methylation inhibitor. Zebularine is known as DNA methylation inhibitor and suitable for water solubility among different DNA methylation inhibitors as 5-Azacytidine and 5-aza-2'-deoxycytidine. We used zebularine as mutagen according to different methods by roots absorption and seed imbibition. After zebularine treatment, DNA methylation inhibitor, we observed mitotic chromosome behavior what is different according to two different treatment methods. First, seed imbibition treatment in 1,000 μM of zebularine solution for 72 hours in dark conditions. The second treatment to seedlings of Keumkang was also treated in 1,000 μM of zebularine solution for 72 hours after germination. Root and shoot showed different elongations in each treatment. Root absorption treatment(3.01±0.48, 2.00±0.26) showed the shortest elongation in root and shoot than control(8.16±0.61, 4.03±0.48) and seed imbibition treatment(4.33±0.80, 2.48±0.36). It can be explained root tip meristematic cell activity was damaged by DNA methylation inhibitor. Primary root tips were collected in DW for 24 hours at low temperature(0℃) and fixed in fixation solution for 3 days to chromosome observation in mitosis. Mitotic index, chromosome structure and chromosome aberration were observed by phase-contrast microscope. Mitotic index of the control(0.29) showed twice mitotic cells as the treated groups(imbibition 0.15, absorption 0.14). Observation of chromosomes showed some short chromosomes and loosen chromosomes affected by zebularine. It is considered because of zebularine damage DNA in mitosis. We observed "gap by chromosome breakage" in chromosomes that have loose parts between centromere and telomere. It seems demethylation of zebularine occurs chromosome breakage.

Profiling Metabolites Expressed Corn Root Under Waterlogging

  • Waterlogging tolerance of corn is one of the important factor for cultivate in paddy soil condition to increase cultivation area and self-sufficiency of corn in Korea. In order to develop elite waterlogging tolerance corn, the new corn lines bred by crossing wild corn, Teosinte, and cultivated corn inbred lines. Five accessions among the 2 species, Zea mays sub spp. mexicana and Zea mays spp. parviglumis, of 81 Teosinte were selected through the waterlogging treatment. The waterlogging treatments were implemented for 7 days at the seedling(V3) stage. The inbred lines were developed by crossing 5 teosinte accessions and cultivated corn lines and they were estimated waterlogging tolerance. It was screened and analyzed the metabolites extracted from roots of 19KT-32(KS141 × teosinte) that was treated waterlogging. We selected 8 of 180 metabolites like as γ-aminobutyric acid(GABA), putrescine, citrulline, Gly, and Ala that expression was remarkably changed over 2.5-times, 7 metabolites increased and 1 metabolite decreased in waterlogging, respectively. Glutamate decarboxylase(GAD) catalyzing GABA accumulation gene have 10 haplotypes, and exon1 was highly conserved, but identified to 135 SNPs after the first intron. Among the 135 SNPs, the number of transversion mutations (52) surpassed the number of transition mutations (38). Most of metabolites were related to abiotic stress in plant that it regulated to pH, osmotic pressure K+/Ca++ and ATPase activity. We are analyzing the association using these results for increase breeding efficiency.

Mapping Quantitative Trait Loci Associated with Arsenic Toxicity Stress in a Double Haploid Population of Rice (Oryza sativa L.)

  • Arsenic (As) is a toxic heavy metal that affects the major rice-growing regions of the world and can cause cancer in humans. Rice paddy fields in South Asia are mostly dependent on arsenic-contaminated water sources due to which rice takes up the arsenic from the soil through roots and accumulates it in plant different parts. Here, we present a quantitative trait locus (QTL) mapping study to find out candidate genes conferring As toxicity tolerance in rice (Oryza sativa L.) at the seedling stage. Three weeks old, 120 double haploid CNDH lines derived from a cross between the Indica variety Cheongcheong and the Japonica variety Nagdong and their parental lines were used by treating with 25 μM As. After 2 weeks ofAs stress, 5 traits such as; shoot length (SL), root length (RL), shoot fresh weight (SFW), root fresh weight (RFW), and chlorophyll contents (CHC) were measured. A linkage map of 12 rice chromosomes was constructed from genotypic data DH lines using 778 SSR markers. The linkage map covered a total genetic distance of 2121.7 cM of the rice genome with an average interval of 10.6 cM between markers. A total of seventeen QTLs (LOD>2) were mapped on chromosomes 1, 2, 3, 6, 7, 8, 9, 11, and 12 using composite interval mapping with trait-increasing alleles coming from both parents. Five QTLs for SL, Two QTLs for RL, Five QTLs for SHL, Three QTLs for RFW, and Two QTLs for CHC were detected. The QTLs related to CHC were selected for forther study.

A comparative analysis of characteristics and antioxidant capacity of Korean mulberries for efficient seedling cultivation

  • Mulberry exhibits unique characteristics and functionalities across various components, including the roots, branches, leaves, and fruits. However, despite numerous studies on mulberry, research on this plant at the seedling stage is insufficient. Therefore, this study aimed to compare the suitability for seedling cultivation and antioxidant effects of four Korean mulberry cultivars, namely, Daesim, Suhong, Simgang, and Cheongsu. In terms of seed weight, germination rate, and growth rate, Daesim was the most suitable cultivar for seedling production. Polyphenol and flavonoid content analysis showed that all cultivars, except for Cheongsu, showed the highest phenolic content at the 2-week seedling stage. Similarly, antioxidant assays using 2,2'-azino-di-3-ethylbenzthiazoline sulfonic acid (ABTS) and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radicals revealed that the antioxidant effect of all cultivars, except for Cheongsu, increased with cultivation at 2- 4- and 6-week. However, mulberry seedlings had a slower reaction rate against DPPH radical removal than mulberry leaves. In addition, ABTS radical scavenging activity showed a different correlation with polyphenol content. This phenomenon may be due to the different polyphenol compositions between mulberry leaves and seedlings. The results of this study suggest that mulberry seedlings exhibit different bioactivities from mulberry leaves, and component analysis is required in further research.

Anti-Oral Microbial Effect of Ethanol Extract of Angelica gigas Nakai

  • Background: The Korean name for Angelica gigas Nakai (AGN) is Cham-dang-gui, which grows naturally or is cultivated, and its dried roots are used in traditional herbal medicines. The AGN root exert various pharmacological effects. Despite the various pharmacological effects of the AGN root, there are no reports on its anti-oral microbial effects. The purpose of this study was to reveal the anti-oral microbial effect and the microbial and biochemical changes in oral microorganisms according to the concentration of the ethanol extract of AGN (EAGN) root, and to confirm the possibility of using EAGN as a plant-derived functional substance for controlling oral infectious microorganisms. Methods: Disk diffusion test, growth measurement, biofilm formation assay, and measurements of acid production and buffering capacity were performed to confirm the antibacterial effect of EAGN. Results: EAGN showed anti-oral bacterial effects against Streptococcus mutans and Aggregatibacter actinomycetemcomitans at all concentrations, with S. mutans showing a more susceptible effect at concentrations above 5.0 mg/ml and A. actinomycetemcomitans at 3.75 mg/ml. EAGN treatment significantly reduced A. actinomycetemcomitans growth at all concentrations tested. Biofilm formation was significantly reduced at concentrations above 3.75 mg/ml for S. mutans and 2.5 mg/ml for A. actinomycetemcomitans. Acid production in S. mutans and A. actinomycetemcomitans was significantly increased by treatment with EAGN, and the buffering capacities of S. mutans and A. actinomycetemcomitans increased from an EAGN concentration of 3.75 mg/ml and above. Conclusion: EAGN showed anti-oral bacterial effects against both S. mutans and A. actinomycetemcomitans at concentrations above 3.75 mg/ml, which were thought to be related to the inhibition of their growth and biofilm formation. Therefore, EAGN can be used as a safe functional substance derived from medicinal plants owing to its antibacterial effects against S. mutans and A. actinomycetemcomitans.

An option to provide water and fertilization for rice production in alkaline soil: fertigation with slow release fertilizers (SRFs)

  • An increasing global population requires a greater food supply, and accordingly there is demand for enhanced production of rice, as a major crop plant that covers half of the world's population. Rice production in arid area is extremely difficult due to poor soil fertility, salinity, deficit of irrigation water, and weather conditions. The aim of the present study was to determine whether various fertilization recipes could provide a countermeasure to allow rice production while also providing soil amendment such as soil pH adjustment. The study was conducted at an experimental field of the United Arab-Emirates (UAE) from January to April, 2022. Rice seedlings (cv. Asemi, alkaline-resistant) were transplanted in plastic containers, and different types of water and nutrient managements were employed as follows: water management (flooding and aerobic for NPKs treatment group) and nutrient management (NPKs, slow release fertilizers [SRFs] and SRFs + NPK-1 treatment groups with flooding). Water and nutrient management did not show any effect on soil pH adjustment. Rice growth was significantly enhanced in the flooding compared to the aerobic condition, whereas the effect of nutrient management clearly differed among the treatment groups, with SRFs + NPK-1 showing the best results followed by SRFs and NPKs. Most of the fertilization groups markedly accumulated soluble sugars in the shoots and grains of rice plants, but concomitantly a decrease in the roots. Overall, the level of starch showed a tendency of relatively slight perturbation by fertilization. Taken together, the results indicate that soil physical structure should be preferentially amended to find the key for suitable rice production.

Arctigenin from Burdock Root Exhibits Potent Antibacterial and Anti-Virulence Properties against Pseudomonas aeruginosa

  • Arctium lappa (Burdock) root is used in various culinary applications especially in Asian Cuisine. Arctigenin (ARC) is a polyphenolic compound abundant in the roots of the burdock plant from which it derives its name. The emergence of bacterial resistance is a growing global worry, specifically due to the declining availability of new antibiotics. Screening for the antibacterial candidates among the safe natural products is a promising approach. The present study was aimed to assess the antibacterial activity of ARC against Pseudomonas aeruginosa exploring its effect on the bacterial cell membrane. Furthermore, the anti-virulence activities and anti-quorum sensing (QS) activities of ARC were in vitro, in vivo and in silico assessed against P. aeruginosa. The current results showed the ARC antibacterial activity was owed to its disruption effect of the cell membrane. ARC at sub-MIC significantly decreased the formation of biofilm, motility, production of extracellular enzymes and in vivo protected mice against P. aeruginosa. These anti-virulence activities of ARC are owed to its interference with bacterial QS and its expression. Furthermore, ARC showed mild effect on mammalian erythrocytes, low probability to induce resistance and synergistically combined with antibiotics. In summary, the promising anti-virulence properties of ARC indicate its potential as an effective supplement to conventional antibiotics for treating severe P. aeruginosa infections.

Growth and Rooting Rate of 'Maehyang' Strawberry as Affected by Irrigation Method on Cutting Propagation in Summer Season (하절기 '매향' 딸기의 삽목 번식 시 관수방법에 따른 생육 및 발근율)

  • This study was conducted to investigate the optimum irrigation method for propagation of cutting strawberry ($Fragaria{\times}ananassa$ Duch. cv. Maehyang) plantlet in summer season. The cutting strawberry plantlets were planted in 24-cell tray ($60{\times}34{\times}10cm$) filled with commercial mixed medium (Tosilee) and placed in semi closed-type small plastic tunnel. Each semi closed-type small plastic tunnel was treated as follows; control (non-treatment), over head irrigation (twice a day), capillary mats irrigation (twice a day), or fog irrigation (30 minutes turn on and 10 minutes off from 8:00 to 18:00). The strawberry plantlets were rooted during 8 days in the semi closed-type small plastic tunnel, and then plastic film was removed. Growth parameters, such as plant height, root length, number of primary roots, petiole length, leaf length, leaf width, crown diameter, SPAD, leaf area, fresh and dry weights of the shoot and root, were measured at 61 days after cutting. Relative humidity in tunnel was high in the order by fog irrigation, over head irrigation, capillary mats, and the control as 72.5, 56.3, 45.8, and 29% on average, respectively. However, the air temperature was similar in all treatments. On the 4 and 8 days, the rooting rate of strawberry plantlet was significantly higher in the over head irrigation and fog irrigation treatments. Plant height, petiole length, crown diameter, and leaf area were highest in the over head irrigation and fog irrigation treatments. In addition, fresh and dry weights of shoot were greater in over head irrigation and fog irrigation treatments than the others. Dry weight of root was differed significantly heaviest in the fog irrigation treatment. However, root length, no. of primary roots, SPAD value, and fresh weight of root were not significantly different in all treatments. These results indicated that growth and rooting for propagation of cutting strawberry plantlet 'Maehyang' were best achievement in the over head irrigation and fog irrigation treatments.
